In particular, the present invention relates an enzymatic process in which in a coupled enzymatic reaction system NAD (P) H for the production of organic Connection consumed by an enzyme and the NAD (P) H at the same time is regenerated by a second enzyme system.

Die Gewinnung optisch aktiver organischer
Verbindungen, z.B. Alkohole und Aminosäuren, auf biokatalytischem
Wege gewinnt zunehmend an Bedeutung.
gezeigt (
Schema 1: Scheme 1:
The efficiently used in the aqueous medium In addition to their catalytic properties, biocatalysts have and Efficiency also has the advantage of being unlike a multitude of synthetic metal-containing catalysts on the use of metal-containing, especially heavy metal and therefore toxic feedstocks can be dispensed with. Can also rely on the use of expensive and also dangerous Reducing agents such as borane in the asymmetric Reduction can be dispensed with.
The previously used in these systems Successful FDH, e.g. from Candida boidinii, has the disadvantage that the specific activity this enzyme class with 4-8 U / mg is very low. This requires the use of a large amount on expensive enzyme with apparatus recycling difficult, if such a process on a technical scale under economic Aspects should be carried out advantageously.
The "malic enzyme" called malate dehydrogenase (MDH) catalyzes the oxidative decarboxylation of malate to pyruvate. There are numerous malate dehydrogenases from different organisms known, e.g. from higher Animals, plants and microorganisms. There are four types distinguished from malate dehydrogenases, which are found in the E.C. 1.1.1.37 to EC 1.1.1.40 are classified (http://www.genome.ad.jp). Dependent on of the type of malate dehydrogenase, NAD and / or NADP is used as a cofactor needed.
Because of the irreversibility of the oxidative Decarboxylation reaction of L-malic acid to pyruvate offers the use of malate dehydrogenase also with regard to a favorable Cofactor regeneration in the systems described above.
Suye, M. Kawagoe, S. Inuta, Can. J. Chem. Eng. 1992 , 70, 306–312) . Suye, Recent Res. Devel. Ferment. Bioeng. 1998, 1, 55–64).The use of malate dehydrogenase to regenerate NAD can be found, for example, in Suye et al. in a work from 1992 (S.-I. Suye, M. Kawagoe, S. Inuta, Can. J. Chem. Eng. 1992, 70, 306-312). The regeneration of NADH using malate dehydrogenase is used for the reductive amination of pyruvate using an alanine dehydrogenase using NADH. The pyruvate is formed by the oxidative decarboxylation from L-malic acid and is immediately used up again in the following step by the alanine dehydrogenase. An accumulation of pyruvate in the reaction solution is thus avoided, which also eliminates the problem of possible inhibitions of the enzymes involved by the presence of Py is eliminated in the stoichiometric range. However, the system presented here is only restricted to the production of alanine (see also S.-I. Suye, Recent Res. Devel. Ferment. Bioeng. 1998, 1, 55-64).
Interestingly, despite the technically high potential - besides this Working from the Suye working group no further work using the "malate dehydrogenase" for regeneration of the oxidized cofactor NAD + in the case of reductions of chains or reductive Aminations of keto acids described.
Object of the present invention was therefore the specification of a further process for the production of chiral organic compounds such as amino acids or alcohols, which by a coupled enzymatic reaction system as described above can be obtained using malate dehydrogenase and which is not limited to the representation of a substance. In particular, this process should be economical on an industrial scale like ecological Points of view can be used particularly advantageously.
The task is accomplished through a process solved with the characterizing features of the subject claim 1. Claims 2 to 7 relate to preferred embodiments. Claims 8 and 9 protect an inventive reaction system and a correspondingly working whole cell catalyst. claim 10 protects preferred plasmids.
The fact that in a process for the production of enantiomerically enriched organic compounds in a coupled enzymatic reaction system comprising a first enzymatic transformation of an organic substrate using NAD (P) H and the regeneration of NAD (P) H in a second enzymatic transformation by a Malate dehydrogenase with the oxidation of L-malic acid to pyruvate and CO 2 , which does not use the resulting pyruvate of the second enzymatic transformation as a substrate in the first enzymatic transformation, is achieved in an extremely surprising but no less advantageous manner in solving the problem. It can be regarded as a surprise that the simultaneous use of, for example, an amino acid dehydrogenase with the decarboxylating malate dehydrogenase is possible without the occurrence of cross-reactions. In addition, no inhibitory effects of substrates and products of the primary reaction on malate dehydrogenase and vice versa can be demonstrated. In particular, it is positive that the pyruvate formed as a by-product does not have an inhibitory effect on the malate dehydrogenase itself or on the alcohol or amino acid dehydrogenases used in parallel.
Advantageously, with the method according to the invention enantiomerically enriched alcohols or amino acids. In this Well-known and cheap alcohol dehydrogenase is the case or amino acid dehydrogenase as enzymes for the first enzymatic transformation in question. The specialist is in principle free to choose, depending on the type of substrate spectrum, stability and conversion rate of the enzyme under consideration. Known enzymes this provenance are in K. Drauz, H. Waldmann (ed.), Enzymes Catalysis in Organic Synthesis, Volume III, Wiley-VCH, Weinheim, 2002, Chapter 15.
Besonders vorteilhaft ist der Einsatz der Alkoholdehydrogenase aus den Organismen Rhodococcus erythropolis (S-ADH) oder Lactobacillus kefir (R-ADH) (ADH aus R. erythropolis: J. Peters, T. Zelinski, M.-R. Kula, Purification and characterization of a novel carbonyl reductase silated from Rhodococcus erythropolis, J. Biotechnol. 1994, 33, 283–292)(ADH aus Lactabacillus kefir: C. w. Bradshaw, W. Hummel, C.-H. Wong, Lactobacillus kefir Alcohol Dehydrogenase: A Useful Catalyst for Synthesis, J. Org. Chem. 1992, 57, 1532–1536.). Phenylalanindehydrogenasen (A. Bommarius in: Enzyme Catalysis in Organic Synthesis (Hrsg.: K. Drauz, H. Waldmann), Volume III, Wiley-VCH, Weinheim, 2002, Kapitel 15.3) verwiesen. The use is particularly advantageous the alcohol dehydrogenase from the organisms Rhodococcus erythropolis (S-ADH) or Lactobacillus kefir (R-ADH) (ADH from R. erythropolis: J. Bommarius in: Enzyme Catalysis in Organic Synthesis (Ed .: K. Drauz, H. Waldmann), Volume III, Wiley-VCH, Weinheim, 2002, chapter 15.3).
Genisolierung und Klonierung sind beschrieben in S. Naamniehs Dissertation, Universität Düsseldorf, in Vorbereitung S. 70ff.Malate dehydrogenases are known to the person skilled in the art also common (Lit. see above or dissertation S. Naamnieh, University of Düsseldorf, in preparation). Here, too, the specialist will look for his Select the most efficient dehydrogenase for the purpose. in the In principle, those malate dehydrogenases are preferred which contain the NAD (P) H to such an extent Regenerate that there is no bottleneck for the reaction process of the other enzyme used occurs. The known malate dehydrogenase is preferred from E. coli K12 in this context. Gene isolation and cloning are described in S. Naamnieh's dissertation, University of Düsseldorf, in preparation p. 70ff.

In principle, the specialist is at the choice of during reaction temperature free. He prefers to orient himself in getting one if possible high yield of product in as much as possible high purity in as much as possible short time. In addition, the enzymes used should be among those used Temperatures should be sufficiently stable and the reaction should take place with one if possible high enantioselectivity run. With regard to the use of enzymes from thermophiles Organisms can temperatures of 100 ° C represent the upper limit of the temperature range in the reaction. As the lower limit in aqueous systems are safe at –15 ° C meaningful. A temperature interval between 10 and is advantageous 60, particularly preferably between 20 and 40 ° C.
The pH during the reaction is from Expert determined based on the enzyme stabilities and conversion rates and accordingly for the inventive method set. Preferably one pH range from 5.5 to 10.0, in particular 6.0 to 9.0.

The invention also relates to a coupled enzymatic reaction system for the production of enantiomerically enriched organic compounds comprising a first enzymatic transformation of an organic substrate using NAD (P) H and the regeneration of NAD (P) H in a second enzymatic transformation by a malate dehydrogenase under oxidation from L-malic acid to pyruvate and CO 2 , the resulting pyruvate of the second enzymatic transformation not being used as a substrate in the first enzymatic transformation. In principle, the same advantages and preferred embodiments apply to this reaction system as have already been mentioned in relation to the process according to the invention. The reaction system is advantageously used, for example, in a stirred tank, a stirred tank cascade or in membrane reactors which can be operated both in batch mode and continuously.

The advantage of such an organism is the simultaneous expression of both polypeptide systems, with which just a rec organism for the reaction of the invention must be tightened.
Dies ist dem Fachmann jedoch hinlänglich bekannt (Gellissen, G.; Piontek, M.; Dahlems, U.; Jenzelewski, V.; Gavagan, J. W.; DiCosimo, R.; Anton, D. L.; Janowicz, Z. A. (1996), Recombinant Hansenula polymorpha as a biocatalyst. Coexpression of the spinach glycolate oxidase (GO) and the S. cerevisiae catalase T (CTT1) gene, Appl.-Microbiol. Biotechnol. 46, 46–54; Farwick, M.; London, M.; Dohmen; J.; Dahlems, U.; Gellissen, G.; Strasser, A. W.; DE19920712).In order to coordinate the expression of the polypeptides with regard to their conversion rates, the corresponding coding nucleic acid sequences can be accommodated on different plasmids with different copy numbers and / or promoters of different strengths can be used for differently strong expression of the nucleic acid sequences. With such coordinated enzyme systems advantageously, an accumulation of an interconnect does not occur and the reaction under consideration can proceed at an optimal overall speed. However, this is well known to the person skilled in the art (Gellissen, G .; Piontek, M .; Dahlems, U .; Jenzelewski, V .; Gavagan, JW; DiCosimo, R .; Anton, DL; Janowicz, ZA (1996), Recombinant Hansenula polymorpha as a biocatalyst. Coexpression of the spinach glycolate oxidase (GO) and the S. cerevisiae catalase T (CTT1) gene, Appl.-Microbiol. Biotechnol. 46, 46-54; Farwick, M .; London, M .; Dohmen ; J .; Dahlems, U .; Gellissen, G .; Strasser, AW; DE19920712).
It is particularly advantageous that the whole-cell catalyst according to the invention if necessary, in the reaction according to the invention the resulting pyruvate is further metabolized, it is more or less a food source used. Whole-cell catalysts designed in this way lead to that the pyruvate is not a by-product of the reaction and therefore not in further process steps from the actual desired chiral product must be separated.
For the application can the polypeptides under consideration of the method according to the invention in free form as homogeneously purified compounds or as recombinantly produced Enzyme can be used. Furthermore, these polypeptides can also be used as part of an intact host organism or in connection with the open-minded and arbitrarily highly purified Cell mass of the host organism.
Möglich ist ebenfalls die Verwendung der Enzyme in immobilisierter Form (Sharma B. P.; Bailey L. F. und Messing R. A. (1982), Immobilisierte Biomaterialiern – Techniken und Anwendungen, Angew. Chem. 94, 836–852). Vorteilhafterweise erfolgt die Immobilisierung durch Lyophilisation (Paradkar, V. M.; Dordick, J. S. (1994), Aqueous-Like Activity of α-Chymotrypsin Dissolved in Nearly Anhydrous Organic Solvents, J. Am. Chem. Soc. 116, 5009–5010; Mori, T.; Okahata, Y. (1997), A variety of lipicoated glycoside hydrolases as effective glycosyl transfer catalysts in homogeneous organic solvents, Tetrahedron Lett. 38, 1971–1974; Otamiri, M.; Adlercreutz, P.; Matthiasson, B. (1992), Complex formation between chymotrypsin and ethyl cellulose as a means to solbilize the enzyme in active form in toluene, Biocatalysis 6, 291–305). Tech. 11, 375–378).Possible is also the use of the enzymes in immobilized form (Sharma B.P .; Bailey L.F. and Messing R.A. (1982), Immobilisiert Biomaterialization - techniques and applications, Angew. Chem. 94, 836-852). This is advantageously done immobilization by lyophilization (Paradkar, V. M .; Dordick, J. S. (1994), Aqueous-Like Activity of α-Chymotrypsin Dissolved in Nearly Anhydrous Organic Solvents, J. Am. Chem. Soc. 116, 5009-5010; Mori, T .; Okahata, Y. (1997), A variety of lipicoated glycoside hydrolases as effective glycosyl transfer catalysts in homogeneous organic solvents, tetrahedron Lett. 38, 1971-1974; Otamiri, M .; Adlercreutz, P .; Matthiasson, B. (1992), Complex formation between chymotrypsin and ethyl cellulose as a means to solbilize the enzyme in active form in toluene, Biocatalysis 6, 291-305). Is very particularly preferred the lyophilization in the presence of surface-active substances, such as Aerosol OT or polyvinylpyrrolidone or polyethylene glycol (PEG) or Brij 52 (diethylene glycol mono-cetyl ether) (Kamiya, N .; Okazaki, S.-Y .; Goto, M. (1997) Surfactant-horseradish peroxidase complex catalytically active in anhydrous benzene, biotechnol. Tech. 11 375-378).
Äußerst bevorzugt ist die Immobilisierung an Eupergit® , insbesondere Eupergit C® und Eupergit 250L® (Röhm) (Eupergit.RTM. C, a carrier for immobilization of enzymes of industrial potential. Katchalski-Katzir, E.; Kraemer, D. M. Journal of Molecular Catalysis B: Enzymatic (2000), 10(1–3), 157–176).Immobilization on Eupergit ® , in particular Eupergit C ® and Eupergit 250L ® (Röhm) (Eupergit.RTM. C, a carrier for immobilization of enzymes of industrial potential. Katchalski-Katzir, E .; Kraemer, DM Journal of Molecular Catalysis B: Enzymatic (2000), 10 (1-3), 157-176).
Gleichfalls bevorzugt ist die Immobilisierung an Ni-NTA in Kombination mit dem His-Tag (Hexa-Histidin) ergänzten Polypeptid (Purification of proteins using polyhistidine affinity tags. Bornhorst, Joshua A.; Falke, Joseph J. Methods in Enzymology (2000), 326, 245–254).Immobilization is also preferred on Ni-NTA in combination with the His tag (hexa-histidine) supplemented polypeptide (Purification of proteins using polyhistidine affinity tags. Bornhorst, Joshua A .; Falke, Joseph J. Methods in Enzymology (2000), 326, 245-254).
Die Verwendung als CLECs ist ebenfalls denkbar (St. Clair, N.; Wang, Y.-F.; Margolin, A. L. (2000), Cofactor-bound cross-linked enzyme crystals (CLEC) of alcohol dehydrogenase, Angew. Chem. Int. Ed. 39, 380–383).The use as CLECs is also conceivable (St. Clair, N .; Wang, Y.-F .; Margolin, A.L. (2000), Cofactor-bound cross-linked enzyme crystals (CLEC) of alcohol dehydrogenase, Angew. Chem. Int. Ed. 39, 380-383).
With these measures it can succeed Polypeptides that become unstable due to organic solvents, to generate those in mixtures of aqueous and organic solvents or can work entirely in organic matter.
Das erfindungsgemäße Verfahren kann so ausgeführt werden,

Reinigung und biochemische Eigenschaften der MDH aus E. coli:Cleaning and biochemical Properties of MDH from E. coli:
a) Reinigunga) Cleaning
Die Reinigung der recMDH aus E. coli-Rohextrakten (Expressionsstamm: E. coli-Derivat JM105) erfolgte in Anlehnung an das Reinigungsprotokoll (Stols L., and Donnelly M. I. (1997). Production of succinic acid through overexpression of NAD(+)-dependent malic enzyme in an Escherichia coli mutant. Appl Environ Microbiol 63: 2695-701.). Purification of recMDH from E. coli crude extracts (Expression strain: E. coli derivative JM105) was based on to the cleaning protocol (Stols L., and Donnelly M. First the rec bacterial cells were disintegrated with glass beads digested (digestion buffer Tris / HCl 100mM pH 7.5). In connection followed by a cleaning step using a Q-Sepharose. After Purification using Q-Sepharose could have a specific activity of MDH of about 7.3 U / mg can be determined. By cleaning this up Enzymes using further chromatographic steps, hydroxyapatite and phenylsepharose was able to achieve homogeneity with a specific activity of 133 U / mg can be cleaned.

For A km value of 0.29 mM was measured for L-malate and for the coenzyme NAD + a Km of 0.14 mM.
Both km values are low Range from <1 mM, they show that both substrates have good affinity for the enzyme be recognized. Both values suggest that the MDH for the regeneration of NADH can be used.

The standard transformation was following the Hanahan protocol (Hanahan D. (1983). Studies on transformation of Escherichia coli with plasmids. J Mol Biol 166: 557-80.). For this became 100-200 μl more competent E. coli cells thawed on ice and 40 ng DNA from the ligation batch added. The plasmid cell suspension was cooled on ice for 30 min and subsequently for 90 sec to 42 ° C heated and immediately chilled on ice again. After adding 300 μl LB - medium the cells were incubated at 37 ° C. for about 45 minutes for regeneration. Subsequently were 200 ul spread this culture on an antibiotic-containing LB plate and over Night at 37 ° C incubated.
With the cloning of malate dehydrogenase 3 'to PheDH at the Pst1 and HindIII interfaces with their own ribosomal binding site could express both enzymes done simultaneously.

Some of the positive clones were selected for expression. A single colony of the respective clones was inoculated into 5 ml LB amp medium and, after reaching OD 580 = 0.6, induced with 1 mM IPTG. Induction took place overnight and the harvested cells were disrupted using ultrasound.
The recombinant strain HB101 shows a malate dehydrogenase activity of 100 U / ml and also a PheDH activity of 130 U / ml. The activity of both Enzymes in the recombinant strain JM 105 is clearly higher and is ~ 600 U / ml for Malate dehydrogenase and 1200 U / ml for PheDH.
The two recombinant strains were cultivated in a 10 L fermenter and the activity of both enzymes determined.
Table 5: Activity determination of the expressed enzymes in a 10 L fermenter with LB medium as batch fermentation.
From the data of the expression is it can be seen that the E. coli strain JM 105 has significantly better activity for both Enzymes shows, therefore all further attempts with this strain carried out.
Beispiel 6: Optimierung der AktivitätExample 6: Optimization of activity
To get the maximum activity of the received Use raw extracts to be able several parameters were examined and varied.
Both heterologously expressed enzymes show the best stability properties under different conditions. Of interest was the optimal properties for both Identify enzymes in the same system. The subsequent attempts were carried out with this in mind.
a) Optimierung des Aufschlusspuffera) Optimization of the digestion buffer
1 g Zellen JM105 wurde in 0,1 M Tris- bzw. 0,1 M Kpi-Puffer mit/ohne BSA (1,5 g/l) 30 %ig aufgeschlossen. Der Aufschluss erfolgte mit Ultraschall bei 70 cont. Cycles. Zusätzlich zu den Puffern wurden 1,5 g/l BSA zur Stabilisierung der Enzyme zugegeben.1 g of cells JM 105 was digested in 0.1 M Tris or 0.1 M Kpi buffer with / without BSA (1.5 g / l) 30%. The digestion was carried out with ultrasound at 70 cont. Cycles. In addition to the buffers, 1.5 g / l BSA was added to stabilize the enzymes.
Tabelle 6: Aktivitätsvergleich in Abhängigkeit des Aufschlusspuffers Table 6: Activity comparison depending on the digestion buffer
The addition to BSA resulted in both cases to an increase in activity. The digestion buffer also influenced the activities. in this connection it was observed that the appropriate digestion buffer was different for the individual enzymes was. The Kpi buffer was for the PheDH is more suitable than for the malate dehydrogenase, but since the activity decrease of the malate dehydrogenase in the Kpi buffer was relatively low, the recombinant cells after the heterologous expression continued to be digested in this buffer.
b) Aufschlussdauer zur Untersuchung der Stabilitätb) Duration of information for the investigation of stability
Ebenso wurde die Aufschlussdauer überprüft und der
kritische Punkt zur Stabilität
der Enzyme während
dieses Vorganges bestimmt. Aus den Daten ist die optimale Aufschlussdauer

Für
diesen Versuch wurde folgende Suspension verwendet:
1 g rekombinante
Zellen (JM105)
3 ml Kpi-Puffer 0,1 MThe following suspension was used for this experiment:
1 g recombinant cells (JM105)
3 ml Kpi buffer 0.1 M
With prolonged treatment of the cells with ultrasound, the activity of PheDH decreases dramatically, whereas the activity the malate dehydrogenase is retained. The ideal digestion conditions for one 25% digestion of 1 g of recombinant E. coli JM105 is therefore 4 × 30 s ultrasound treatment with meanwhile 3 × 30s cooling down in the ice bath.
For longer treatments with ultrasound the sample is heated, which can lead to denaturation of the enzymes. The amount of protein was determined in this experiment and is from the following refer to.

The cells were temporarily cooled for 30 seconds after a 60-, 30-second treatment.
When cleaning the malate dehydrogenase to homogeneity could have a specific activity of 466 U / mg can be achieved. The cleaning steps are in table 8 summarized.

c) Km-Wertbestimmungc) Km value determination
The K M values were determined for the substrate and the coenzyme of malate dehydrogenase. The K M values were determined on homogeneous or partially purified rec-malate dehydrogenase samples.

Important factors for a coupled Reaction are the pH optimum as well as the temperature stability of the two enzymes. additionally to stability the enzymes play other factors such as the influence of different Substrates on the enzymes matter.

The malate dehydrogenase is for a long time at 45 ° C stable. However, since the PheDH becomes unstable at this temperature, the syntheses were at 30 ° C carried out, thus stability both enzymes and the coenzyme guaranteed for a long time could be.
Both enzymes were made with two different buffers tested in one reaction batch each (Table 9).

Table 9: Comparison of PheDH and malate dehydrogenase activity in different reaction buffers. The activities can be seen in percent of the optimal.
Because the activity of PheDH in HEPES buffer did not decrease significantly, the coupled enzyme reaction was in this Buffer performed.
By determining the buffer, pH and temperature values could create suitable conditions and media for the synthesis of phenylalanine through a coupled enzyme reaction with regeneration of the cofactor (NADH) is selected become.
