Abstract
In confocal three-dimensional scanning absorption, an excitation beam (26) and a measuring beam (32) are focused into an essentially common focus in order to determine the three-dimensional structure of an object. The object (10) and the focus are moved relative to one another by the focus in order to scan the object. The change in the transmission of the measuring beam (32) when passing through the focus is determined as a function of the change in the intensity of the excitation beam (26). By irradiating the sample with light in a well-defined focus, the transmission of the sample in this focus changes locally. This is recorded using the measuring beam. In this way, for example, the inner structure of cells can be recorded three-dimensionally by means of absorption measurements without having to be stained with dyes beforehand.

The invention relates to an arrangement and a method for determining the three-dimensional structure of a Object.

With confocal microscopes, the spatial resolution through a spatial restriction improved focus in front of the lens.
This is done by emitting the sample Light mapped onto a pinhole, which is in a too the focal plane of the lens in the object conjugate plane is. The improved resolution is coming in that two point mapping functions do the mapping determine in the confocal microscope: the imaging function of the focus of the laser beam and the imaging function of the detector, which the imaging of the light emitted by the sample into the pinhole and describes the detector located behind it. The mapping function of the detector describes the probability with which an im Focus emitted photon reaches the detector. Since both the suggestion and also the detection must take place is the point mapping function of a confocal microscope Product from the two probability distributions, i.e. the product the point mapping functions for excitation and detection. This leads to a significantly narrower main maximum the confocal point mapping function compared to one not confocal microscope. This corresponds to a higher resolution of the confocal microscope and causes discrimination against all points, that are not in the immediate vicinity of the focus. The latter is a requirement for the creation of three-dimensional images.

All of these laser scanning microscopes is common that they help clarify the spatial structure of an object use the spontaneous emission of luminescent molecules. Because the one to be examined Sample i. d. R. itself does not fluoresce - native fluorescence is right Rare -, the sample to be examined must i. d. R. marked with suitable dye molecules become. Change this possibly the structure or even the function of the object to be examined and thus the measurement. Furthermore, dye molecules often suffer low photostability, with which the production of phototoxic substances in the sample, z. B. singlet oxygen, is connected. Also. such measurements are always made using fluorescent light of contaminants or frequency-shifted scattered light (so-called Raman light) falsified, especially at low dye concentrations.
The object of the invention is that Determining the three-dimensional structure of an object to enable without having to mark the object with dye molecules beforehand.
This task is shared by the inventions the characteristics of the independent Expectations 1 and 4 solved. Advantageous developments of the inventions are characterized in the subclaims.
According to the invention, an arrangement for determining the three-dimensional structure of an object. As objects are u. a. biological objects of interest, such as cells whose inner and / or outer structure enlightened shall be.
The arrangement has an excitation beam and a measuring beam with an essentially common focus. The rays are i. d. R. electromagnetic radiation, in particular Light rays that are preferably generated by a laser. However, acoustic rays, neutron rays, are also conceivable or other particle rays. The beams should be focusable his.
In order to clarify the three-dimensional structure of the object, the focus is smaller than the object. The focus is usually on the three-dimensional ellipsoid of maximum radiation intensity records. Depending on the size of the object, the focus can be chosen to be relatively large. If the object is small, for example a biological cell, a laser beam is generally used as the beam and the focus is chosen to be as small as possible, in particular diffraction-limited.
The object can be fixed freely in the room his. The object can also be embedded in a medium that essentially transparent for both the excitation and the measurement beam is.
The arrangement has a device for relative movement of object and focus for scanning the object by focusing on so that the structure of the object can be determined can.
There are many options for the transmission properties the sample for to change the measuring beam. By irradiating the part of the object in focus with changed the excitation beam locally the transmission of this part of the object for the measuring beam, dependent on the local nature of the property. The excitation of the sample or some selectively responsive molecules within the excitation beam the sample causes these molecules to go from a ground state to transferred to an excited state. This empties the basic state on the one hand and on the other hand the excited state populates. Both leaves to prove themselves. The proportion of molecules in an excited Condition to be transferred need is very low and is typically on the order of 0.1% to 1%.
Frequently the electronically excited molecules have one against the molecules changed in the basic state Extinction coefficients or a changed absorption spectrum. The change is i. d. R. for every kind of molecule specific. To prove the population A suitable measuring beam of the excited state is selected, for example a laser beam with a wavelength that is an absorption band of the excited state of the molecules, but none Absorption band of the ground state.
The population of the excited state can be can also be demonstrated by stimulated emission. For this, the measuring beam tuned to a wavelength, which corresponds to an emission band of the excited state. comes stimulated emission, the transmission of the Measuring beam apparently. Furthermore, when using the stimulated emission the photostability (the number of on average potential Excitation cycles) of any dyes used, since this is a shorter time in electronically excited states linger from which they i. d. R. enter into chemical reactions can, which their fluorescence ability cancel.
The emptying of the basic state can be demonstrated that the measuring beam, for example the same or similar Wavelength like the excitation beam has and then both generated by a laser become. They both like tuned to an absorption band of the ground state of the molecules his. If the basic state is partially depopulated or faded, then so the absorption of the measuring beam decreases depending on the intensity the excitation beam, d. H. the transmission increases. In this case it is also possible that the measuring beam is matched to another absorption band of the ground state, so a different wavelength has as the excitation beam.
In this way, through the excitation beam a volume within the object that produces specific properties that can be detected selectively. This volume is e.g. B. determined by the range of the maximum concentration of molecules in the excited Status. This area is defined by the focus of the excitation beam generated. Without this targeted creation of a prepared volume in the object could an absorption measurement only provides information about the totality of the molecules in the beam path of the measuring beam.
Depending on the need have the excitation beam and the measuring beam, if it is a laser beam, the same wavelength or different wavelengths. Accordingly, one or more radiation sources are required.
It is not necessary to use suitable probes z. B. dye molecules, to mark. It is sufficient, find an absorption band from parts of the sample and pass them through reversibly and specifically bleach the excitation beam. For biological samples there are often a number of such absorption bands, for example cytochrome or hemoglobin. However, it is possible by marking the object with suitable dyes To bring about a targeted effect or reinforce. You can do this dyes are also used which fluoresce only poorly, such as classic colorants, whose effects on cells are well researched.
The arrangement finally has one Set up to determine the change in transmission of the Measuring beam when passing through the focus, depending on of change the intensity of the excitation beam. As a rule, the change in transmission is proportional to the concentration of the molecules, which have been brought into an excited state by the excitation beam. So that's the relative change the transmission i. d. R. proportional to the intensity of the excitation beam. Typical values for the relative change the transmission of the measuring beam is in the range of 1% or less. Relative transmission changes Up to 3 * 10 ^ -8 can still be demonstrated with suitable equipment. With this technique can individual absorber molecules be detected.
A great advantage of the invention is that the marking with dyes can be dispensed with. This leads, among other things, to a reduction in the production of phototoxic substances zen in the sample, which is strongly catalyzed by fluorescent dyes.
Furthermore, the desired measurement signal i. d. Can be proven free of underground. Is used as a measuring beam one in the wavelength narrow-band laser beam is used, so is the desired signal detected only in this narrow wavelength range. For this, e.g. B. in front of the detector also a narrow band Color filter or monochromator can be used. Ambient light This eliminates background fluorescence or Raman light. If, as in the current state of the art, fluorescent light is detected, so the emission light is over collect a much wider range of wavelengths what to a correspondingly increased Leads underground. Furthermore, a selective state can be prepared by the excitation light which is detected by the measuring beam, a state possibly natural in the sample does not occur and is therefore underground.
The invention can have many applications u. a. for ultra trace analysis be used in a sample and to check the homogeneity of the color of Polymers or to check the internal structure of the dye distribution in samples.
To further increase the spatial resolution, i. d. R. used a microscope to view the excitation beam and the Focus the measuring beam into the sample. Most suitable for carrying out the invention is a modified confocal laser scanning microscope.
The invention allows the spectral Distribution of the change the transmission of the measuring beam as it passes through the focus to be spatially resolved. For this purpose, a lamp is used to generate the measuring beam a broad, if possible emits a continuous spectrum of light. Alternatively, too a narrow-band, tunable light source, such as a tunable one Lasers. The transmission of the measuring beam is then detected using a spectral detector. The so determined Transmission or absorption spectra can also be a function of wavelength of the excitation beam can be determined. This will create a new field Multi-dimensional spectroscopy opens up enormous opportunities the differentiated representation z. B. of tissue or cell structures creates.
