Abstract
Die Erfindung betrifft eine Verbindung der allgemeinen Formel (I) umfassend eine Thioether-Struktur, ein Verfahren zu deren Herstellung und Verbindungen erhalten oder erhältlich nach diesem Verfahren. Weiterhin betrifft die Erfindung eine Verbindung der allgemeinen Formel (I), oder eine Verbindung der allgemeinen Formel (I) erhalten oder erhältlich nach dem Herstellungsverfahren oder ein pharmazeutisch akzeptables Salz einer Verbindung der allgemeinen Formel (I) zur Verwendung als Arzneimittel, insbesondere zur Verwendung als Schmerzmittel und/oder zur Verwendung bei der Tinnitusbehandlung oder -prävention und/oder zur Verwendung bei der Behandlung oder Prävention von Krampfzuständen und/oder zur Verwendung bei der Behandlung oder Prävention einer Erkrankung ausgewählt aus der Gruppe bestehend aus Inkontinenz, Angstzustand, Abhängigkeit, Manie, bipolare Störung, kognitive Störung und Dyskinesie. Eine neuronale Übererregbarkeit spielt eine entscheidende Rolle bei chronischen Schmerzen, Epilepsie oder Migräne. In diesem Kontext stellt die Hyperpolarisation des Ruhemembranpotentials bestimmter Neurone durch Öffnung von Kaliumkanälen und die daraus folgende verringerte Generierung von Aktionspotentialen einen therapeutischen Ansatz dar, der bereits durch die Arzneistoffe Flupirtin und Retigabin klinisch genutzt wird. In den meisten Neuronen überwiegen Kv7.2- und Kv7.3-Untereinheiten.[2] Daneben kommen Kv7-Kaliumkanäle auch in den primären Sinneszellen des Innenohrs, in Skelettmuskelzellen und in Zellen der glatten Muskulatur vor.[3-5] Ähnlich wie bei anderen spannungsgesteuerten Kaliumkanälen bilden jeweils vier Untereinheiten einen Kanal. Dabei können sowohl Homo- als auch Heterotetramere gebildet werden.[6,7] Der Effekt der Arzneistoffe Flupirtin und Retigabin auf Kv7-Kanäle ist abhängig davon aus welchen Untereinheiten sie aufgebaut sind.[8] Flupirtin und Retigabin wirken auf molekularer Ebene insbesondere über eine Stabilisierung der offenen Konformation der heterotetrameren Kv7.2/Kv7.3-Kanäle, die genaue Bindungsstelle konnte bisher nur durch Berechnungen abgeschätzt aber nicht durch Kristallstrukturanalyse bestimmt werden.[7-10]A voltage-dependent, slowly activating and nonactivating potassium ion current that occurs in the central and peripheral nervous systems is involved in the control of the firing behavior and thus in the regulation of the excitability of a large number of neuronal cells. Flupirtin wurde vom Chemiewerk Hornburg (später ASTA Medica) entwickelt. Es gehört chemisch betrachtet zur Klasse der Triaminopyridine und wird in Europa seit 1984 als Analgetikum mit muskelrelaxierenden Eigenschaften eingesetzt.[7] Es ist durch die EMA zur Schmerzbehandlung zugelassen und als Maleat-Salz in Form von Kapseln, Tabletten oder Injektionslösungen erhältlich. Der Wirkstoff erreicht seinen schmerzlindernden Effekt auf spinalen und supraspinalen Ebenen.[11] Opiodrezeptoren sind dabei genauso wenig an der Wirkung beteiligt wie eine Beeinflussung der Prostaglandinsynthese, alpha1- oder alpha2-Adreno- oder 5-HT-Rezeptoren.[12-14] Flupirtin ist ein positiver Modulator von Kv7-Kaliumkanälen und öffnet Kv7.2/7.3-Kanäle mit einer EC50 von ungefähr 5 µM.[15] Bei niedrigen mikromolaren Plasmakonzentrationen (2-6 µM), welche für einen analgetischen Effekt nötig sind, gilt dies als der hauptsächliche Wirkmechanismus von Flupirtin.[16]Flupirtin was developed by Chemiewerk Hornburg (later ASTA Medica). [15] At low micromolar plasma concentrations (2-6 μM), which are necessary for an analgesic effect, this is considered to be the main mode of action of flupirtine. [16]
Des Weiteren gibt es Indizien dafür, dass Flupirtin den Effekt von GABA am GABAA-Rezeptor verstärkt.[17] Flupirtin ist zugelassen für die Therapie von akuten und chronischen Schmerzen, beispielsweise bei schmerzhaften Verspannungen, degenerativen Gelenkerkrankungen, Spannungskopfschmerzen, Tumorschmerzen, Regelschmerzen oder postoperativen Schmerzen. Basierend auf dem Wirkungsmechanismus von Flupirtin sind theoretisch weitere Indikationen möglich. Dazu zählen multiple Sklerose, Tinnitusprophylaxe, überaktive Blase, neuropathische Schmerzen, periphere Neuropathien und Fibromyalgien.[13] Weiterhin verfügt Flupirtin über neuroprotektive Eigenschaften, die potentiell therapeutisch nutzbar sind.[18] Studien an Patienten mit postoperativen oder Tumorschmerzen haben gezeigt, dass Flupirtin in etwa die gleiche analgetische Wirkstärke wie schwache Opioide besitzt.[19-20] Flupirtin galt lange Zeit allgemeinhin als gut verträglich. Schlaflosigkeit, Schläfrigkeit, Schwindel, trockener Mund, Erbrechen und Unwohlsein im Magen-Darm-Bereich sind die häufigsten Nebenwirkungen. Im Rahmen der Pharmakavigilanz wurde jedoch ein ernst zu nehmendes Problem ersichtlich. Bis 2013 wurden 800 unerwünschte Arzneimittelwirkungen gemeldet. Mehr als ein Drittel dieser Meldungen beschreiben Leber- oder Gallenerkrankungen. Aus der Gesamtzahl aller Fallberichte endeten 24 tödlich und für 17 von diesen wurden wiederum lebertoxische Reaktionen berichtet. Nach einer erneuten Nutzen-Risiko-Bewertung besteht für Flupirtin-haltige Arzneimittel eine Einschränkung der therapeutischen Zielgruppe und eine Begrenzung der Behandlungsdauer auf zwei Wochen. [12] Mittlerweile hat der Pharmakavigilanz-Ausschuss (PRAC) der Europäischen Arzneimittelagentur EMA empfohlen, die Zulassungen für alle Flupirtin-haltigen Arzneimittel (Katadolon® und andere) zu widerrufen.[22]Furthermore, there is evidence that flupirtine enhances the effect of GABA on the GABA A receptor. Insomnia, drowsiness, dizziness, dry mouth, vomiting and discomfort in the gastrointestinal area are the most common side effects. In the context of pharmacovigilance, however, a serious problem became apparent. By 2013, 800 adverse drug reactions had been reported. More than a third of these reports describe liver or biliary disorders. Of the total number of case reports, 24 were fatal and 17 of these were again reported to have liver toxic reactions. Following a reassessment of benefit-risk, flupirtine-containing drugs are limited in their therapeutic target group and have a two-week treatment limit. [12] Meanwhile, the Pharmacavigilance Committee (PRAC) has recommended that the European Medicines Agency (EMA) withdraw authorizations for all medicinal products containing flupirtine (Katadolon® and others). [22]
In höheren Dosen verfügt Flupirtin auch über antikonvulsive Eigenschaften. Eine strukturelle Optimierung führte zur Entwicklung des deutlich potenteren, hochanalogen Wirkstoffs Retigabin, der in der Folge als Antiepileptikum zugelassen wurde.[23] Flupirtin und Retigabin sind nachfolgend dargestellt:
Der postulierte Wirkmechanismus von Retigabin ist vergleichbar mit dem von Flupirtin. Damit besitzt Retigabin einen gänzlich anderen Angriffspunkt als alle anderen aktuell verfügbaren Antiepileptika. Retigabin ist in nahezu jedem Epilepsie-Tiermodell aktiv mit einem Gesamtprofil, welches sich von jedem anderen Antiepileptikum unterscheidet.[23] Das macht es zu einem unverzichtbaren Arzneistoff bei Arzneimittel-resistenten Epilepsien.[24, 25]The postulated mechanism of action of retigabine is comparable to that of flupirtine. Die häufigsten unerwünschten Arzneimittelwirkungen während einer Behandlung mit Retigabin (Trobalt ®) sind Benommenheit, Müdigkeit, Kopfschmerz, Verwirrtheit und Schwäche. Darüber hinaus wurden bei 12% der Behandelten renale und urologische Störungen beobachtet. Retigabin verursacht eine Harnretention, die über die Aktivierung von Kv7-Kanälen in der glatten Muskulatur der Blase zu erklären ist.[26] Im Gegensatz zu einigen anderen Antiepileptika ist Retigabin jedoch nicht mit kardialer, hämatologischer oder hepatischer Toxizität assoziiert und ist weder teratogen noch reproduktionstoxisch. Des weiteren besitzt Retigabin nur ein geringes Potential für Arzneimittelinteraktionen, da es Leberenzyme weder induziert noch inhibiert.[6] Diese Eigenschaften machen Retigabin im Allgemeinen zu einem gut verträglichen Antiepileptikum. Berichte aus klinischen Langzeitstudien über Pigmentveränderungen (Verfärbung) von Augengeweben einschließlich der Retina und blaugraue Verfärbungen der Haut, Lippen und Nägel führten jedoch zu einer Anwendungsbeschränkung. Retigabin ist inzwischen nur noch als Zusatztherapie für pharmakaresistente fokale Krampfanfälle mit oder ohne sekundärer Generalisierung bei erwachsenen Patienten mit Epilepsie zugelassen, wenn andere geeignete Arzneimittel unzureichend wirkten oder nicht vertragen wurden.[27] Im Juni 2017 erfolgte letztendlich die weltweite Marktrücknahme von Trobalt® durch GlaxoSmithKline. Eine mögliche Erklärung für die beobachtetet Toxizität ist die Bildung toxischer Metabolite.[28] In Greifswald wurde deshalb eine klinische Studie durchgeführt, in der Hinweise auf entstandene Oxidationsprodukte (Chinondiimine) gefunden wurden, die in Folgereaktionen Dimere bzw. Trimere bildeten und sich mit Glutathion zu entsprechenden Konjugaten umsetzen ließen.[29] Diese Glutathionkonjugate werden in Folgereaktionen wahrscheinlich zu Cystein- und Mercaptursäurekonjugaten metabolisiert.[30] Es wurde aus diesen Befunden abgeleitet, dass die Chinondiiminintermediate des Flupirtins für die Hepatotoxizität verantwortlich sind.[31] Dies ist beispielsweise für Paracetamol und Diclofenac bekannt, die ebenfalls Chinoniminintermediate bilden.[32,33]The most common adverse drug reactions during retigabine (Trobalt ®) treatment are drowsiness, fatigue, headache, confusion, and weakness. Retigabine is now approved as adjunctive therapy for pharmacoresistant seizures with or without secondary generalization in adult patients with epilepsy if other appropriate drugs are inadequate or intolerable. [27] In June 2017, the worldwide market withdrawal of Trobalt® by GlaxoSmithKline finally took place. One possible explanation for the observed toxicity is the formation of toxic metabolites. [28] In Greifswald, a clinical study was therefore conducted in which evidence for resulting oxidation products (quinonediimines) were found, which formed dimers or trimers in subsequent reactions and could be converted with glutathione into corresponding conjugates. [29] These glutathione conjugates are likely to be metabolized to cysteine and mercapturic acid conjugates in subsequent reactions. [30] It was deduced from these findings that the chinondiimine intermediates of flupirtine are responsible for hepatotoxicity. [31] This is known, for example, for paracetamol and diclofenac, which also form quinoneimine intermediates. [32,33]
In einer weiteren in vitro Reaktion konnte die Carbamatgruppe des Flupirtins durch Schweineleberesterasen gespalten werden. Die so entstandene primäre, aromatische Aminogruppe wurde wiederum von menschlichen N-Acetyltransferasen zum Metaboliten D13223 acetyliert. Dieser zeigte sich im Vergleich zu Flupirtin als weniger reaktiv gegenüber Peroxidasen. Die entsprechenden Chinondiimine konnten in vitro detektiert werden, d.h. sie sind relativ stabil.[29] Dies erklärt eventuell auch, dass der Metabolit D13223 des Flupirtins weniger hepatotoxisch ist als Flupirtin selbst.[31] Der N-acetylierte Metabolit von Flupirtin verfügt darüber hinaus noch über etwa ein Viertel der analgetischen Wirkung der Muttersubstanz.[34] Der Metabolismus von Retigabin unterscheidet sich etwas von dem des Flupirtins. Gemeinsam ist, dass auch Retigabin durch Acetylierung und Glucuronidierung verstoffwechselt wird. In einem 3-Hals-Kolben wurden 3,65 mmol 6-[(4-Fluorbenzyl)thio]-3-nitropyridin-2-amin (1,02 g) mit 18,6 mmol SnCl2•2H2O (5,1 eq, 4,2 g) vorgelegt und in 40 ml Ethanol suspendiert. Der Reaktionsansatz wurde unter Argon-Atmosphäre für 25 h auf 70 °C erhitzt. Anschließend wurde auf 40 °C abgekühlt und 37,2 mmol Et3N (10,2 eq, 5,16 ml) sowie 4,6 mmol Chlorameisensäureethylester (1,26 eq, 0,44 ml) hinzugefügt. Nach 30 min wurde die Temperatur auf 55 °C erhöht und für eine Stunde gehalten. Nach abkühlen auf Raumtemperatur wurde der Ansatz über Kieselgur filtriert und der Rückstand mit Ethanol gewaschen. Das Filtrat wurde am Rotationsverdampfer eingeengt, in DCM aufgenommen und mit Wasser gewaschen. Evaporation der organischen Phase und waschen des Rückstandes mit n-Hexan ergab das saubere Produkt. Ethyl-[2-amino-6-(benzylthio)pyridin-3-yl]carbamatEthyl [2-amino-6- (benzylthio) pyridin-3-yl] carbamate
2,31 mmol 6-(Benzylthio)-3-nitropyridin-2-amin (0,60 g) wurden in einer Mischung aus 15 ml Wasser und 25 ml Isopropanol suspendiert. 6,93 mmol Eisenpulver (3 eq, 0,39 g) und 10,4 mmol NH4Cl (4,5 eq, 0,56 g) wurden hinzugefügt und die Suspension für 6 h zum Rückfluss erhitzt. Die resultierende dunkelbraune Mischung wurde filtriert und der Rückstand mit 50 ml Isopropanol gewaschen. Anschließend wurden 13,9 mmol Et3N (6,0 eq, 1,91 ml) und 5,54 mmol Chlorameisensäureethylester (2,4 eq, 0,53 ml) hinzugefügt. Zur Vervollständigung der Reaktion wurden nach einer sowie zwei Stunden jeweils 5,54 mmol Chlorameisensäureethylester hinzugefügt. Nach zusätzlichen 5 Stunden wurde die Lösung am Rotationsverdampfer eingeengt und der Rückstand in 100 ml Ethylacetat aufgenommen. Nach weiteren 5 h wurde die Lösung am Rotationsverdampfer eingeengt, der Rückstand in 100 ml Ethylacetat aufgenommen und mit 2×100 ml Wasser sowie 100 ml gesättigter Kochsalzlösung gewaschen. Das Rohprodukt wurde mittels Flashchromatographie (Mobile Phase: n-Hexan/Ethylacetat 7:3) aufgereinigt. Der Reaktionsansatz wurde unter Argon-Atmosphäre für 23 h auf 70 °C erhitzt. Anschließend wurde der Ansatz auf 40 °C abgekühlt und 15,4 mmol Et3N (10,2 eq, 2,14 ml) sowie 1,9 mmol Chlorameisensäureethylester (1,27 eq, 0,18 ml) hinzugefügt. Nach 30 min wurde die Temperatur auf 55 °C erhöht und für eine weitere Stunde gerührt. Nach anschließendem Abkühlen auf Raumtemperatur wurde die Suspension über Kieselgur filtriert und der Rückstand mit Ethanol gewaschen. Das Filtrat wurde am Rotationsverdampfer eingeengt und der Rückstand in DCM aufgenommen. Nach waschen der organischen Phase mit Wasser und anschließender Trocknung über Na2SO4 wurde das Lösemittel im Teilvakuum entfernt und der Rückstand mit n-Hexan gewaschen. Die Zellen wurden mit den vorbereiteten Substanzkonzentrationen, welche im Verhältnis 1:100 (V/V) hinzugefügt wurden, weitere 30 min im Dunkeln bei Raumtemperatur inkubiert. Die Negativkontrolle beinhaltete nur Lösungsmittel (1 % DMSO). Für die Fluoreszenzintensitätsmessungen (485 nm Exzitation, 535 nm Emission, Messmodus Boden) wurde das Mikrotiterplattenlesegerät Infinite® F200 Pro (Tecan) verwendet. Die zu vermessende Platte wurde ins Gerät eingebracht. In einer Zeitspanne von 20 s, wobei alle 2,5-3 s Messwerte vom Gerät aufgenommen wurden, wurde die Fluoreszenzintensität jedes Wells bestimmt (Hintergrundmessung). Danach wurde zügig im Verhältnis 1:5 (V/V) Stimulus Buffer, welcher 7,5 mM Tl2SO4, 12,5 mM K2SO4 und 1x Chloride-free Buffer in destilliertem Wasser enthielt, zu den einzelnen Wells gegeben. Die Fluoreszenzintensität der jeweiligen Wells wurde ungefähr für weitere 2 min periodisch alle 2,5-3 s bestimmt. Die Bestimmung der Kaliumkanalöffnungsaktivität einer Verbindung anhand der aufgenommenen Daten erfolgte in mehreren Schritten. Zunächst wurde für jede Verdünnungsstufe einer Substanz deren Fluoreszenzintensitätsänderung zu einem definierten Zeitpunkt gegenüber der gemittelten Fluoreszenzintensitäten der Hintergrundmessung berechnet (ΔF/F). Des Weiteren wurde dieses ermittelte Ergebnis um den analog berechneten Wert der Negativkontrolle bereinigt (korr. ΔF/F). Beim erwähnten definierten Zeitpunkt (t) war die Summe der jeweiligen um die Negativkontrolle korrigierten Fluoreszenzintensitätsänderungen der Verdünnungsreihe einer Substanz maximal. Durch visuelle Inspektion eines zugehörigen Fluoreszenzintensität-Zeit-Diagramms (Fluoreszenzintensität korrigiert um Negativkontrolle, relativ auf jeweiligen Hintergrund bezogen) wurde die Auswahl des Zeitpunkts kontrolliert. Die Nebenwirkung von Flupirtin, die auch für seinen Entzug vom Markt verantwortlich war, ist die Hepatotoxizität. Aus diesem Grund wurden zwei Zellkulturmodelle für Hepatotoxizität entwickelt, um auf toxische Wirkung der neuen Substanzen zu testen und toxische Kandidaten frühzeitig zu identifizieren. Als Zellviabilitätsassay wurde der etablierte MTT-Viabilitätsassay durchgeführt.[41] Das Prinzip des Assays beruht darauf, dass viable Zellen in der Lage sind, das gelbe, wasserlösliche MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromid) zum unlöslichen violetten Formazanprodukt durch mitochondriale Dehydrogenasen zu reduzieren (unter Verbrauch von NADH bzw. NADPH), dessen Absorption dann photometrisch bestimmt werden kann. Der erste zellbasierte Assay wurde mit der TAMH-Zelllinie der Maus (transforming growth factor-a transgenic mouse hepatocytes) durchgeführt, der bereits in früheren Arbeiten mit Flupirtin [41] als auch mit anderen hepatotoxischen Wirkstoffen Anwendung fand und als Prädiktor von Hepatotoxizität nützlich ist.[42] Zum anderen wurde eine humanen Hepatokarzinomzelllinie, die HEP-G2 (human hepatocellular carcinoma cells) als zweites Hepatotoxizitätsmodell benutzt [43], um mit einem humanen Zelllinien die Ergebnisse aus dem TAMH-Zellinie zu vergleichen. Beide Zelllinien wachsen als Monolayer in Kultur, die TAHM-Zelllinien in serumfreiem Medium, die HEP-G2-Linie in Kulturmedium mit 10 % fötalem Kälberserum.The side effect of flupirtine, which was also responsible for its withdrawal from the market, is hepatotoxicity. TAMH-Zellen, welche eine Konfluenz von mindestens 80 % aufwiesen, wurden mit HBSS (Hanks balanced salt solution, P04-34500, PAN-Biotech) gewaschen, mithilfe von Trypsin-EDTA-Lösung (T3924, SigmaAldrich), die ca. 10 s auf den Zellen verweilte, vom Zellkulturflaschenboden abgelöst (noch 2 min bei 37 °C), dann mit Soybean Trypsin Inhibitor (T6522, Sigma Aldrich) suspendiert, anschließend bei 400 g 5 min zentrifugiert und nach Überstandabnahme mit DMEM/F12 (1:1 Mix) resuspendiert. HEP-G2-Zellen wiederum, welche eine Konfluenz von ungefähr 70 % aufwiesen, wurden mit phosphatgepufferter Salzlösung (z. B. L1825, Biochrom) gewaschen, mithilfe von Trypsin-EDTA-Lösung vom Zellkulturflaschenboden abgelöst (ca. 5 min bei 37 °C), dann mit RPMI 1640 suspendiert, anschließend bei 125 g 5 min zentrifugiert und nach Überstandabnahme erneut mit RPMI 1640 resuspendiert. Die Zellzahl beider Zellsuspension wurde mit dem automatischen Zellzählmessgerät EVE™ (NanoEnTEK) bestimmt. Von den murinen Zellen wurden anschließend 20 000 Zellen/Well/200 µL in 96 Well TC-Platten, Cell+, F (#83.3924.300, Sarstedt) ausgesät und 2-3 Tage bis zur vollständigen Konfluenz bei 37 °C, 5 % CO2 und 95 % Luftfeuchtigkeit inkubiert. Von den humanen Zellen wurden 15 000 Zellen/Well/100 µL in 96 Well TC-Platten, Standard, F (#83.3924, Sarstedt) ausgesät und 24 h bei 37 °C, 5 % CO2 und 95 % Luftfeuchtigkeit inkubiert. Von den zu testenden Verbindungen wurde jeweils eine serielle Verdünnungsreihe (mindestens fünf Konzentrationen) in DMSO (#A994.2, Carl Roth) angefertigt, wobei die Konzentrationen dem 100fachen der gewünschten späteren in-Well Konzentrationen betrugen. Nach erfolgter Inkubation beider Zelllinien wurde das jeweilige in den Platten befindliche Medium verworfen und die Wells nach einem bestimmten Muster mit jeweils 200 µL/Well mit den im nachfolgenden beschriebenen verschiedenen Lösungen beladen. Die angefertigte Verdünnungsreihe einer Substanz wurde dem entsprechendem Medium im Verhältnis 1:100 (V/V) beigemischt und zu den Zellen gegeben. Kontroll-Wells enthielten die entsprechenden Zellen, welche nur mit dem jeweiligen Medium und 1 % DMSO behandelt wurden. Wells, welche keine Zellen, sondern nur das entsprechende Medium mit 1 % DMSO enthielten, wurden zur Bestimmung des Hintergrunds herangezogen (Blank). Jede Bedingung wurde mindestens als Triplikat bestimmt. Die Platten wurden 24 h bei 37 °C, 5 % CO2 und 95 % Luftfeuchtigkeit inkubiert. Anschließend wurde jegliche Flüssigkeit aus den Platten entfernt und eine Mischung aus dem jeweiligen Medium und einer 2,5 mg/ml MTT-Lösung (in Dulbecco's Puffer, L11939.06, Alfa Aesar) im Verhältnis 1:5 (V/V) zu den Zellen gegeben. Es folgte eine Inkubation von 1,5-4 h bei 37 °C, 5 % CO2 und 95 % Luftfeuchtigkeit. Nun wurden die Platten komplett von der zugegebenen Mischung befreit und entstandene Formazankristalle mithilfe von DMSO (#7029.2, Carl Roth) in Lösung gebracht. Die Absorption der einzelnen Wells wurde bei λ = 570 nm mit dem SpectraMax 190 Mikroplattenlesegerät inkl. SoftMax Pro Software (Molecular Devices) ermittelt. Die Viabilität der mit der zu testenden Substanz behandelten Zellen wurde im Verhältnis zur Kontrolle angegeben. Dazu wurde der Absorptionswert eines einzelnen Wells, welches Zellen beinhaltete, die mit einer Verdünnungsstufe der Substanz behandelt wurden (T), durch den Mittelwert der Absorptionswerte der Kontroll-Wells (<C>) dividiert und mit 100 multipliziert. Beide Terme wurden zuvor um den Mittelwert der gemessenen Absorptionen der Wells für die Blank-Bestimmung (<Blank>) korrigiert. Somit ergibt sich folgende Formel:
Zur Beurteilung des hepatotoxischen Potentials einer Substanz wurde der IC50-Wert verwendet. Als IC50 wurde die Konzentration einer Verbindung definiert, bei welcher die Viabilität der Zellen auf 50 % sank (
Die Ergebnisse der Hepatoxizitäts-Untersuchung sind in Tabelle 3 zusammengefasst. In den meisten Fällen konnte kein IC50-Wert berechnet werden, weil die obere Löslichkeitsgrenze der Substanzen in Zellkulturmedium unterhalb die 50% toxische Konzentration lag; d.h. die Substanzen fiel aus, bevor eine IC50-Konzentration erreicht werden konnte.The results of the hepatotoxicity study are summarized in Table 3. Tabelle 3 fast die gesamten biologischen Testergebnisse der synthetisierte Substanzen zusammen und vergleicht sie mit der Referenzsubstanz Flupritinmaleat. Aus dem Zusammenspiel der Ergebnisse von Kv7.2/3-Öffnungsaktivität und den Hepatotoxizitäts-Ergebnissen in vitro ist ersichtlich, dass insgesamt Verbindungen der allgemeinen Strukturformen (I) die Wirkung von der Referenzsubstanz Flupirtinmaleat deutlich übertreffen. Insbesondere Verbindungen mit einem niedrigen EC50-Wert, bevorzugt einem EC50-Wert von weniger als 0,9 mikromolar (Mikromol pro Liter), weiter bevorzugt von weniger als 0,1, weiter bevorzugt von weniger als 0,01 mikromolar, sind besonders geeignet. Bei der potentesten Substanz 11 werden so niedrige Substanzkonzentrationen (0,0014 µmol pro Liter) zur Erreichung des halbmaximalen Effekts auf Kaliumkanäle benötigt, dass toxische Effekte bei der resultierenden Dosierung sehr unwahrscheinlich sind. Die ermittelten Toxizitätsdaten zeigen, dass bei den durch Löslichkeit im Testmedium limitierten Konzentrationen von 63 µmol pro Liter für Verbindung 11 keine toxischen Effekte beobachtet wurden und der IC50 daher als größer als 63 µmol pro Liter angegeben werden kann. Entsprechend ist das Verhältnis von IC50 zu EC50 mindestens 85-mal größer bei Substanz 11 als bei Flupirtinmaleat. Leberschädigungen während der Therapie sollten damit weniger wahrscheinlich sein. Auch der biologische Effekt der Testsubstanz sollte größer sein (beispielsweise stärkere Analgesie), da Emax bei über 100 % lag und somit ein stärkerer Kaliumefflux zu erwarten ist als mit Flupirtinmaleat.
