The invention relates to an electrode arrangement for electrochemically supported biosynthesis, a bioreactor with such an electrode arrangement and a method for electrochemically supported biosynthesis.
The object of the present invention is to provide an electrode arrangement for BES systems that is as simple in construction and robust as possible and can be used in particular for electrolytic water splitting during ongoing biosynthetic processes.

- a) a first electrode designed as a rod electrode with a first end and a second end,
- b) a second electrode designed as a hollow electrode and
- c) a housing holding the first electrode and the second electrode made of non-electrically conductive material with a first housing end and a second housing end, and wherein
- - the first electrode is surrounded by the second electrode in a first electrode region, so that an inner electrode chamber is formed in the first electrode region between the outside of the first electrode and the inside of the second electrode,
- - the first electrode in a second electrode region is not surrounded by the second electrode and is surrounded by the housing in such a way that a gas discharge channel is formed between the outside of the first electrode and an inside of the housing,

The electrode arrangement according to the invention is particularly well suited for electrolytic water splitting during ongoing biosynthetic processes. Its design makes it possible to separate the products of water splitting (O 2 and H 2 ) and make them usable individually. Depending on the polarity, ie the direction of the current, either oxygen or hydrogen can be produced at the (outer) second electrode, thus creating a reductive or oxidative environment. By repolarizing the electrode surfaces, a rapid change from cathode to anode, for example in a bioreactor, is possible, which means that conditions can be created that can be used advantageously by microorganisms or enzymes in the biosynthesis process to produce valuable products. Abiotic processes in aqueous media can also be supplied with oxygen or hydrogen in situ. The electrode arrangement according to the invention can advantageously be used to initiate or support various redox processes, especially those in which gases play a role, for example also in the reduction of CO 2 to formate.
The term “bioelectrochemical system (BES)” refers to systems in which microbial metabolic processes are influenced by extracellular electron transfer processes at electrodes. In bioelectrochemical systems, for example, electrodes are used to supply or remove electrons from microbial metabolic processes directly or indirectly, for example using mediators gen. The term also includes systems in which the redox balance of microbial metabolic processes is influenced electrochemically.
The term “electrochemically assisted biosynthesis” refers to a biosynthesis process, i.e. the synthesis of a chemical compound through a microbial metabolic process, with the help of a bioelectrochemical system, e.g. biosynthesis in a bioreactor, in the medium of which hydrogen gas is produced by electrolysis.
The expression that "the gas discharge channel is in fluid communication with a gas outlet opening and with the inner electrode chamber" means that the gas discharge channel is connected to both a gas outlet opening and to the inner electrode chamber so that gas formed in the inner electrode chamber passes through the gas discharge channel to the gas outlet opening can reach.
The expression according to which the “outside of the second electrode is exposed” means that the outside of the second electrode is essentially freely accessible and is therefore, for example, exposed to a medium surrounding the electrode arrangement and can essentially come into free contact with this medium. The term includes in particular that gas formed on the outside of the electrode can essentially escape freely into the environment or be absorbed by a medium surrounding or flowing against the electrode. The term includes that the outer surface of the electrode is optionally at least partially coated or covered, and the electrode is exposed with its coated or covered outside. For example, the electrode can be coated with a preferably gas-permeable plastic film to protect against biofouling, or it can be covered by a protective cover to protect against mechanical damage caused by particulate media components, which, however, preferably remains permeable to other media components. This can be, for example, a suitably perforated cover. The coating of the electrode can also consist of or include immobilized catalysts, e.g. enzymes. For example, the electrode can only be coated on the outside.
The term “working volume” in relation to a bioreactor refers to the usable volume of a bioreactor, i.e. the volume of the medium with which the bioreactor can be used. The term not only denotes the numerical amount of the volume, but is also used if necessary to identify the corresponding spatial area, i.e. the spatial area in the bioreactor that the medium occupies.
In relation to a bioreactor, a “connection”, possibly also referred to as a “port”, is understood to mean access through the reactor wall or a top plate closing the reactor to the interior of the reactor and thus to the working volume of the bioreactor. These are usually openings into which devices designed for this purpose can be installed, for example measuring probes, e.g. pH probes, oxygen probes or level probes, pipes and the like. A “standard connection” here is understood to mean a connection that corresponds to a standard and, for example, provides a specific diameter for the opening (e.g. 19 mm) and a specific type of fastening for the equipment to be installed, e.g. a thread.
The electrode arrangement according to the invention is designed in such a way that a gas formed with sufficient polarization voltage on the outside of the first electrode facing the inner electrode chamber, in the case of electrolytic water splitting depending on the polarity of the electrodes, oxygen or hydrogen, from the inner electrode chamber via a gas discharge channel to a gas outlet opening can be derived from the outside of the housing. The gas outlet opening is spaced from the location or area at or in which the gas is formed, to the extent that the gas outlet opening is preferably outside the vessel when the electrode arrangement is installed in a vessel, for example a bioreactor. The gas can also be collected or forwarded via the gas outlet opening.
For example, the gas can be returned to a bioreactor or transferred to another bioreactor. For this purpose, a corresponding connector, preferably detachable, for example by means of a thread, can be attached to the gas outlet opening, to which, for example, a hose can be attached. Of course, if necessary, several separate gas discharge channels and/or several gas outlet openings can also be provided. As a result, gas formed can be used for various purposes.
The housing of the electrode arrangement according to the invention serves both to hold the electrodes and the separator and to form at least part of the gas discharge channel. The housing consists of an electrically non-conductive material, for example ceramic or plastic, preferably a plastic material. Biocompatible plastic materials that are resistant to higher temperatures, such as those required for sterilization under superheated steam, and are resistant to chemicals such as acids and alkalis. Examples of suitable plastics are polyetheretherketone (PEEK), polytetrafluoroethylene (PTFE), perfluoroalkoxylalkane (PFA) or polyphenylene sulfide (PPS). The housing can be formed in one piece or in several pieces. For example, the housing can be divided into a housing cap, a shaft and a base plate.
In a preferred embodiment, a first electrode region of the electrode arrangement with the inner electrode chamber is at least partially not surrounded by the housing, so that the outside of the second (outer) electrode is preferably at least partially exposed. When installed in a bioreactor, the second electrode can be exposed to the reactor medium, for example. For this purpose, the housing can be completely missing in this area or openings can be provided in the housing.
The electrode arrangement according to the invention is preferably generally longitudinally cylindrical. “Longitudinal cylindrical” here means that the electrode arrangement is generally cylindrical, extending predominantly in the longitudinal direction of the first electrode. Such a configuration is particularly advantageous for installation in a bioreactor, for example.
In a preferred embodiment of the electrode arrangement according to the invention, the second electrode region is arranged closer to the first housing end than the first electrode region. The second electrode area includes the gas discharge channel, which opens into a gas outlet opening on the outside of the housing. The gas outlet opening lies towards the first housing end, which in the installation situation, for example when the electrode arrangement is installed in a bioreactor, is preferably arranged outside the reactor.
In the electrode arrangement according to the invention, the first electrode region with the inner electrode chamber is preferably arranged towards the second housing end. However, in other embodiments, it is also possible for the first electrode region to be arranged closer to the first than to the second housing end, for example in the event that a further electrode region adjoins the first electrode region towards the second housing end.
In a longitudinally cylindrical preferred embodiment of an electrode arrangement according to the invention, the first and second electrode regions lie one behind the other in the longitudinal direction, for example, with the first electrode region being further spaced from the first housing end than the second electrode region. It is preferred, but not necessary, that the first and second electrode areas immediately follow one another. This results in a relatively compact arrangement.
In a preferred embodiment of the electrode arrangement according to the invention, the first electrode is fixed with its first end on or in the first housing end and with its second end on or in the second housing end. The electrode can, for example, be screwed, clamped or glued into the housing ends. The housing ends can, for example, also be designed as separate caps or plates, so that the electrode can, for example, be screwed, clamped or glued into a housing cap with its first end and into a base plate with its second end.
The electrodes can be made of any suitable materials. The electrodes are preferably made of platinized titanium. However, they can also be made of stainless steel, tin or carbon. For example, electrodes made of tin or tin-coated materials are particularly suitable for the reduction of CO 2 to formate. The first electrode is preferably designed as a solid, preferably cylindrical, rod electrode, but can also be designed as a hollow electrode. The second electrode is preferably designed as a cylindrical grid electrode. The electrodes can be electrically contacted through appropriate holes or recesses in the housing.
The electrodes, in particular the second electrode, can, for example, also have immobilized catalysts (e.g. enzymes) on their surface or can be coated or covered in another way suitable for certain purposes. For example, the outside of the second electrode can be coated with a preferably gas-permeable plastic film in order to avoid biofouling, for example. The second electrode can also be provided with a coating that facilitates the formation of a biofilm.
The separator is an electrically non-conductive but ion-permeable element that separates the first and second electrodes from one another. It can be, for example, a porous ceramic cylinder or an ion exchange membrane. The pore size of the ceramic cylinder is preferably so small that microorganisms, such as bacteria, cannot penetrate the pores. For example, the pore diameter can be ≤ 0.4 µm, ≤ 0.3 µm or ≤ 0.2 µm. For example, membranes made of a sulfur are used as ion exchange membranes fonated tetrafluoroethylene polymer (PTFE) in question. Such membranes can be steam sterilized and are available, for example, under the brand “Nafion” (DuPont). The type and required properties of suitable separators are known to the person skilled in the art or can be easily determined. It is preferably a porous ceramic cylinder.
It is preferred if the separator is spaced from the first electrode so that the inner surface of the separator and the outer surface of the first electrode do not touch each other. It is therefore preferred if the space between the (inner) first electrode and the (outer) second electrode forming the inner electrode chamber is not completely filled by the separator.
In a particularly preferred embodiment of the invention, the electrode arrangement is dimensioned and designed so that it can be installed in a bioreactor in such a way that the first electrode region in the installation situation is arranged within the working volume of the bioreactor, i.e. immersed in the bioreactor medium, and the gas outlet opening opens outside the reactor vessel of the bioreactor. In a further preferred embodiment, the electrode arrangement is dimensioned and designed so that it can be inserted or screwed into a 19 mm standard connection of a bioreactor. For example, a thread can be provided on the housing, with the help of which the electrode arrangement can be screwed into such a standard connection.
In a further particularly preferred embodiment, the electrode arrangement according to the invention is designed as an agitator. For example, the electrode arrangement can be designed so that it can be rotated about its longitudinal axis. The electrodes can be contacted using sliding contacts. The electrode arrangement can, for example, also be designed in such a way that only the first electrode is rotatable and, for example, is equipped with stirring blades at one end. Alternatively, the first electrode can also be designed as a hollow electrode and a stirrer shaft can be arranged coaxially in its interior.
The electrode arrangement is particularly preferably steam sterilizable. In such a configuration, the electrode arrangement consists of materials that are resistant to the temperatures of, for example, 121 ° C that occur during steam sterilization. For example, the housing can be made of polyetheretherketone (PEEK), the electrodes can be made of platinized titanium and the separator can be made of porous ceramic material.
In a particularly preferred embodiment of the electrode arrangement according to the invention, the gas outlet opening can be closed. For example, the gas outlet opening can be closed by means of a screwable seal. This is particularly advantageous in the case of a steam-sterilizable electrode arrangement. Particularly preferably, the gas outlet opening can be connected to a connector, preferably in a detachable manner, so that escaping gas can be forwarded or collected, for example by means of a hose. For this purpose, for example, an internal thread can be provided in the gas outlet opening, into which a corresponding connector can be screwed.
In a further aspect, the invention also relates to a bioreactor, comprising a reactor vessel with a working volume and at least one electrode arrangement according to the invention arranged in a connection of the bioreactor.
The working volume of the bioreactor according to the invention is preferably significantly larger than the volume of the inner electrode chamber. For example, the ratio between the working volume of the bioreactor to the volume of the internal electrode chamber is preferably ≥ 5, more preferably ≥ 10, ≥ 15, ≥ 20, ≥ 50 or ≥ 100, and particularly preferably ≥ 200, ≥ 500, ≥ 750 or ≥ 1000. When we talk about the working volume of the bioreactor in this context, what is of course meant is its numerical amount - expressed in liters, for example. Due to the small volume of the inner electrode chamber compared to the working volume of the reactor, undesirable effects that would otherwise occur, such as uncontrolled pH value changes and the like, are often negligible.
In a preferred embodiment of the bioreactor according to the invention, the first electrode region of the electrode arrangement according to the invention is arranged within the working volume of the bioreactor. The working volume here means the spatial area of the bioreactor that contains the reactor medium. A medium here is understood to mean water or an aqueous medium. It can, for example, be a cultivation medium for a microorganism. The medium can contain inorganic compounds, for example salts, and also organic compounds, for example organic compounds, substrates, etc. contained in yeast extract.
In the bioreactor according to the invention, the gas discharge channel preferably opens into a gas outlet opening located outside the reactor vessel. As a result, gas formed in the inner electrode chamber can be removed from the reactor vessel. Gas aust is further preferred rite opening, for example by means of a thread, can be connected to a nozzle through which the gas can be drained, forwarded or collected. If necessary, the gas can also be returned to the bioreactor at another point or introduced into another bioreactor or reused in another way.
In one embodiment of the bioreactor according to the invention, the electrode arrangement is arranged in a connection in a top or bottom plate closing the reactor vessel. However, the electrode arrangement can also be arranged in a connection integrated into the wall of the reactor vessel.
In a further aspect, the invention also relates to a method for electrochemically supported biosynthesis, wherein at least one living cell is cultivated in an aqueous medium in a bioreactor according to the invention. Preferably, water is split electrolytically by means of the electrode arrangement during the cultivation of the at least one living cell. For this purpose, an electrical voltage is applied to the electrodes of the electrode arrangement, which corresponds at least to the decomposition voltage for the electrolytic splitting of water. However, if necessary, a voltage below the decomposition voltage for electrolytic water splitting can also be applied to the electrodes, for example in the event that the formation of a biofilm on the second electrode is desired.
In a preferred embodiment of the method according to the invention, the electrode arrangement is operated, i.e. subjected to an electrical voltage, in such a way that oxygen is formed in the inner electrode chamber and hydrogen is formed in the working volume of the bioreactor. In an alternative embodiment of the method according to the invention, the electrode arrangement is operated, i.e. subjected to an electrical voltage, in such a way that hydrogen is formed in the inner electrode chamber and oxygen is formed in the working volume of the bioreactor.

An

In a
The

In the
In the example described, the
The power (P) provided by the electrode increased uniformly as the current flow increased and was between 0.3 and 2.4 W in the applied current ranges. The hydrogen production rate was close to the theoretically possible rate at lower currents. The efficiency was between 30 and 35%. The pH value was only influenced at higher currents. Since the electrode arrangement is essentially intended to be used at low currents, the influences that only occur at high currents are negligible.
Although the efficiency of the tested embodiment of an electrode arrangement according to the invention of 35% is below the efficiency of a modern electrolyzer, it produces finely distributed hydrogen in situ in the bioreactor and thus directly influences the environment of cultivated bacteria.
A fermentation of C. pasteurianum 525 was carried out using the
For example, compared to the control, 1,3-PDO production was increased using the electrode. The yield of 1,3-PDO was significantly increased from 0.14 to 0.27 mol/mol glycerol.
The use of an electrode arrangement according to the invention in the bioreactor therefore had, for example, a positive effect on the 1,3-PDO production of the microorganisms. The preference of 1,3-PDO over butanol as a product in this process is advantageous due to the value of 1,3-PDO and inhibition processes by butanol. By using an electrode arrangement according to the invention, more preferred 1,3-PDO could be produced and the bioprocess could be made more effective. For example, by using larger or more electrodes in larger reactors, such electrobiofermentation can be “up-scaled”.
