Abstract

Zur Entwicklung neuer Wirkstoffe z.B. auf dem Gebiet der Tumorbehandlung werden potentielle pharmakologische Substanzen über einem mehrstufigen Prozess zunächst in der Zellkultur und bei erfolgsversprechenden Ergebnissen in vivo in Tierversuchen untersucht. Es stellt sich immer wieder heraus, dass die Ergebnisse von In-Vitro-Versuchen jedoch nicht ohne weiteres auf In-Vivo-Versuche übertragbar sind, es scheitern viele potentielle Wirkstoffe in diesen Versuchsphasen. Ein effektiveres In-Vitro-Screening insbesondere ein in Bezug auf die späteren In-Vivo-Versuche erfolgreiches In-Vitro-Screenen ist wünschenswert, um den Aufwand zur Entwicklung aktiver Wirkstoffe und gleichzeitig die Versuchstierzahlen zu reduzieren. Mit dem Ziel, eine verbesserte Screening-Plattform zwischen Zellkultur und Tierversuch zu etablieren, werden derzeit unterschiedliche 3D-In-vitro-Tissue-Engineering (TE) Konzepte verfolgt. Ein erster Schritt auf dem Weg zu einem 3D-Tumormodell wurde mit der Untersuchung statisch kultivierter, mit Zellen beladener Mikropartikel gemacht, z.B.
Bioprinting ist ein spezielles Tissue-Engineering-Verfahren, das sich dem Prinzip der generativen Fertigungsverfahren – auch als Rapid-Prototyping bekannt – bedient, um dreidimensionale lebende Gewebe zu generieren. Im Gegensatz zu industriell angewandten generativen Fertigungsverfahren, wie z.B. der Stereolitographie, dem selektiven Laserschmelzen oder dem Fused Deposition Modeling, werden beim Bioprinting mit Zellen beladene Hydrogele als Baumaterial verwendet. Gemäß eines 3D-Modells wird diese Schicht für Schicht aufgetragen. Hydrogele ahmen die extrazelluläre Matrix natürlichen Gewebes nach und bieten so eine zellfreundliche Umgebung für das 3D-Tissue-Engineering, siehe z.B.
In nichtpolymerisierter Form lassen sich Hydrogele gut handhaben und dispensieren. Durch eine chemische, thermische oder durch Licht induzierte Polymerisation verfestigt sich das Gel, so dass nachfolgende Schichten sukzessiv aufgetragen werden können. Über verschiedene Parameter, wie der Gel-Konzentration, der Vernetzer-Konzentration, dem pH-Wert und der Temperatur können Elastizität und Polymerisations-Zeitpunkt sowie Polymerisations-Geschwindigkeit eingestellt werden. Mit Hilfe des Bioprintings lassen sich komplexe 3D-Gewebe Konstrukte aus unterschiedlichen Materialien mit verschiedenen Zelltypen aufbauen, wie vielfältig in der Literatur beschrieben, z.B.
Allerdings gilt es solche dreidimensionalen Tumorangiogenesemodelle zu verbessern, insbesondere die Herstellungsverfahren hiervon zu verbessern, um entsprechende Gewebemodelle für Screening-Verfahren bereitzustellen. Hierbei sollen diese Modelle geeignet sein, das Tumorwachstum, die Tumorangiogenese, sowie Tumortherapieeffekte zu untersuchen. Dabei sollen sowohl der Tumor bzw. die Gewebestruktur als auch die Versorgungsstruktur aus im Wesentlichen natürlich gewonnenen Materialien bestehen, vorzugsweise biologischen Material, wie entsprechende Hydrogele und Zellen. Auf die Trennschicht wird das Tumorgewebe in Form einer Tumorzellen enthaltende Hydrogel Suspension aufgedruckt, z.B. Tumorzellen in Matrigel. Ggf. kann das Tumorgewebe mit weiteren Zelltypen und weiteren Hydrogelen strukturiert werden. Das so hergestellte Konstrukt wird wie in Pkt. 3–5 beschrieben weiter bearbeitet und kultiviert. Die Trennschicht verhindert hierbei das Wuchern der Tumorzellen in Richtung des Versorgungskanals während der ersten Wochen der Kultivierung. Die Trennschicht ist so auszulege, dass sie sich erst dann aufgelöst hat, wenn die Endothelschicht in der Wandung des Kanals voll ausgeprägt ist. Nach Auflösen der Trennschicht wird dem Tumorgewebe der Zugang zum Versorgungskanal gewährt und ein Anschluss mittels Gefäßneubildung ermöglicht.The supply channel is manufactured as described (points 1-2). Tumorzellen in Matrigel. Ggf. kann das Tumorgewebe mit weiteren Zelltypen und weiteren Hydrogelen strukturiert werden.
