The object underlying the invention is to provide a surface-functionalized polymer - with improved compared to the prior art stability of the functionalization - for biological applications and a simple and inexpensive process for its preparation, that is, a method for surface functionalization of polymer substrates with the Aim of a defined immobilization and presentation of the biologically functional molecules at interfaces.
The solution of the object of the present invention consists in a surface-functionalized polymer for biological applications according to claim 1. A polymer is selected which forms a solid hydrophobic phase in aqueous systems in which the hydrophobic regions of amphiphilic molecules at least in the region of the phase boundary to the environment are partially arranged, wherein the hydrophilic regions of the amphiphilic molecules are aligned as free ends with their respective hydrophilic terminus towards the hydrophilic environment. As amphiphilic molecules are selected those from the polymer class of poly (2-oxazoline) s, at the respective hydrophilic terminus of a biologically functional molecule is conjugated.
The term "hydrophobic" is used to describe substances that do not mix with water and usually cause it to "bead off" on the surface. In this context, a hydrophobic phase is to be understood as meaning a phase whose surface has a phase boundary with respect to water Contact angle of more than 90 °. Accordingly, a hydrophilic phase is to be understood as meaning a phase whose surface at its phase boundary with respect to water has a contact angle of less than 90 °, usually close to 0 °. Amphiphilia is a special property of a molecule that has hydrophobic and hydrophilic regions.
Als besonders vorteilhaft erwies sich als Polymersubstrat Polylactid-co-Glycolid (PLGA).Preferably, the surface-functionalized polymer, more particularly the polymer substrate of the surface-functionalized polymer, is a thermoplastic. Advantageously, the polymer substrate is a polyester or has a polyester-based polymer surface. According to a particularly preferred embodiment of the invention, the polyester-based polymer substrate is selected from the group of copolymers, in particular random copolymers, gradient copolymers and block copolymers. Polylactide-co-glycolide (PLGA) proved to be particularly advantageous as a polymer substrate.
As the amphiphilic molecule, there is preferably provided at least one of the compounds of the class of poly (2-oxazoline) s consisting of a combination of hydrophilic and hydrophobic monomer units. The combination of these monomer units makes the poly (2-oxazoline) amphiphilic. The combination of these monomer units is realized as a copolymer and may advantageously be in the form of a random copolymer, gradient copolymer or block copolymer. Here, the poly (2-oxazoline) monomer units are characterized by their substitution in the side chain, which determines their water solubility. In particular, short-chain side groups having n- or iso-alkyl groups or also aryl groups are hydrophilic for a number X of carbon atoms in the side chain lying in the range C1 to C3 (ie X is in the range 1 to 3), and hydrophobic for C4 and larger CX values. The amphiphilia may also be realized by a combination of conjugation of hydrophilic poly (2-oxazoline) s and long-chain hydrophobic moieties. For example, a poly (2-oxazoline) may be bound with an n-alkane, perfluorinated compounds or aromatic molecule segments or lipids or lipopolymers.
The poly (2-oxazoline) s are advantageously conjugated at their hydrophilic terminus to one of the following biologically functional molecules: adhesion peptide sequences, antimicrobial peptides, other biologically active peptides, proteins, for example growth factors, and aptamers.
- a) a surface-functionalized polymer, that is to say the polymer substrate which forms a solid hydrophobic phase in aqueous systems, and a hydrophilic polymer and an amphiphilic polymer consisting of molecules of the polymer class of the poly (2-oxazoline) s by means of a Solvent in a homogeneous solution,
- b) the solvent is separated, so that a phase separation takes place with the formation of a phase boundary between a hydrophobic phase of the polymer to be functionalized and a hydrophilic phase of the hydrophilic polymer and the amphiphilic molecules (AM) align themselves at the phase boundary such that the hydrophobic regions the amphiphilic molecules are at least partially disposed in the hydrophobic phase and the hydrophilic regions of the amphiphilic molecules are at least partially disposed in the hydrophilic phase,
- c) separating the hydrophilic phase so that the hydrophilic regions of the amphiphilic molecules are aligned as free ends with their respective hydrophilic terminus towards the environment, while the hydrophobic regions of the amphiphilic molecules are further arranged in the polymer to be functionalized.
According to the invention, a biologically functional molecule is conjugated to the respective hydrophilic terminus of the amphiphilic polymers, whereby the biologically functional molecules are immobilized and presented.
- 1. Due to the defined synthetic route, the amphiphilic polymers can be adapted in a wide range to the substrate to be modified and the environment.
- 2. The amphiphilic polymers can be functionalized regioselectively and quantitatively with biologically functional molecules.
- 3. The hydrophilic portion of the amphiphilic polymer can be adapted to the specific requirements, for example for the control of biocompatibility and / or the control of cell adhesion.
Possible applications for surface-functionalized polymers in this way are found especially in the field of medicine, here in particular regenerative medicine, for example in bone formation.
The present inventive solution provides a simple but defined possibility for surface functionalization of a widely used in the biomaterials polymer group by a biocompatible, highly defined and variable polymer system, namely the poly (2-oxazoline) s ready.
In this way, all hydrophobic polymer phase solids can be functionalized on their surface. The hydrophilic polymer used is preferably a polyethylene glycol (PEG). Suitable solvents for process step a) are, in particular, dichloromethane and chloroform.
