Ein wichtiges Ziel der medizinischen Diagnostik ist der Nachweis von Antigenen in verschiedenen Proben, z.B. aus Gewebe oder Blut. Antigene können durch spezifische Antikörpern gebunden und erkannt werden. Eine häufige Anwendung ist der „Enzyme Linked Immunosorbent Assay“ (ELISA), der für zahlreiche medizinisch-diagnostische Verfahren eingesetzt wird, z.B. zum Nachweis von Antigenen aus Blutproben im Nachweis verschiedener Erreger, Autoimmunkrankheiten oder der Quantifizierung von Blutbestandteilen. Hierbei wird eine Antigen-Antikörper-Bindung im Assay in ein colorimetrisches oder fluoreszentes Signal umgewandelt, um ein nachweisbares Messsignal zu erhalten. Ein ausreichend starkes colorimetrisches oder fluoreszentes Signal erhält man beim ELISA Assay dadurch, dass an den Antikörper ein Enzym, beispielsweise eine Peroxidase, gebunden ist, die die chemische Umwandlung von nicht farbigen oder nicht fluoreszenten Substratmolekülen in farbige oder fluoreszente Substratmoleküle katalysiert. Die so erzeugten Substratmoleküle sind allerdings nicht an den Antikörper gebunden, sondern diffundieren frei im Medium, beispielsweise in einer Lösung. Dadurch ist zwar die quantitative Bestimmung eines Antigens möglich, aber nicht dessen direkte Beobachtung und Lokalisation, etwa auf einer Zelloberfläche.An important goal of medical diagnostics is the detection of antigens in various samples, e.g. from tissue or blood. Es müssen also viele Antikörper und fluoreszenzmarkierte Zweitantikörper an die Zellen gebunden werden, um ein nachweisbares Fluoreszenzsignal zu erzeugen. Dies setzt aber voraus, dass auch viele Antigene auf der Zelloberfläche vorhanden sind, was oft nicht der Fall ist. So ist aus diesem Grund zum Beispiel der Nachweis des Her2-Antigens auf Zellen nicht direkt nachweisbar. Das Her2-Protein wird in ca. 20% der Patientinnen mit invasivem Mammakarzinom überexprimiert, so dass ein immunhistochemischer Nachweis für die Diagnose, Prognose und Behandlung wünschenswert ist. Um dieses Problem zu lösen, wird zur Verstärkung des Fluoreszenzsignals der Antigen-Antikörper-Bindung, aber auch der Interaktion eines Biomoleküls mit einem anderen der O-Link-Assay im Handel angeboten. Hierbei ist an einem Biomolekül eine kurze Nukleinsäure gebunden, an die zunächst eine weitere Nukleinsäure hybridisiert wird. Durch eine Polymerase wird in einer „Rolling Circle Amplifikation“ dieser Strang verlängert. An den verlängerten Strang können dann viele Oligonukleotide gebunden werden, die jeweils mit einem Fluorophor markiert sind. So kann ein verstärktes Fluoreszenzsignal erzeugt und gemessen werden. Hierfür ist allerdings sowohl die Fixierung der Zellen als auch verschiedene Wasch-, Polymerisations- und Hybridierungsschritte notwendig. Diese Aufgabe wird dadurch gelöst, dass der Marker ein Dendrimer umfasst, an dem mehrere Chromophore und eine reaktive Kopplungsgruppe gebunden sind. Ein Marker im Sinne der Erfindung ist eine optische Sonde zum Nachweis von Biomolekülen. Dendrimere sind Kaskadenpolymere, die ausgehend von einem Verzweigungskern („Core“) repetitive Molekülketten aufweisen. Es kann eine Verzweigung zu zwei oder auch mehr Verknüpfungsstellen geben. Durch die baumartige Struktur können sehr viele Chromophore in günstiger räumlicher Anordnung, bevorzugt endständig, an die Molekülketten des Dendrimers gebunden werden. Der erfindungsgemäße Marker weist außerdem eine reaktive Kopplungsgruppe auf, die an Biomoleküle bindet, wodurch diese durch den Marker markiert werden. So können vor einer Messung beispielsweise Antikörper zu dem Marker zugegeben werden. Ein Antikörper bindet dann kovalent jeweils an die eine reaktive Kopplungsgruppe des Markers. Unmarkierte Antikörper oder ungebundene Marker sollten abgetrennt werden. Das Biomolekül, der im genannten Beispiel als Antikörper ausgebildet ist, kann dann wiederum hochspezifisch an eine Bindungsstelle binden. Eine Bindungsstelle ist ein Epitop eines Antigens, an das der hierfür spezifische Antikörper bindet. Die für den Nachweis der Bindung bzw. des Antigens notwendige Verstärkung des optischen Signals erfolgt dadurch, dass nicht ein oder wenige, sondern sehr viele Chromophore an das Dendrimer gebunden sind, wobei bis etwa 1000 Chromophore an das Dendrimer gekoppelt sein können. Hierdurch wird ein sehr intensives optisches Signal geliefert, wodurch es möglich ist, lokalisiert einzelne Biomoleküle, insbesondere Antikörper, mikroskopisch sichtbar zu machen und nachzuweisen, die an eine Bindungsstelle bzw. ein Antigen gebunden haben. Durch das starke optische Signal ist das einzelne Biomolekül, bzw. der einzelne Antikörper, also in einem Fluoreszenzmikroskop direkt sichtbar. Somit kann auch eine Bindung dieses Antikörpers, beispielsweise an ein Protein auf einer Zelloberfläche oder in einer Zelle, direkt sichtbar gemacht werden. Wenn die Chromophore als, insbesondere hoch-wasserlösliche, Fluorophore ausgebildet sind, kann als optischer Nachweis das Fluoreszenzsignal des Farbstoffes verwendet werden. Außerdem kann das Antigen beispielsweise auf einer Zelloberfläche lokalisiert nachgewiesen und im Fluoreszenzmikroskop sichtbar gemacht werden, d.h. es kann beispielsweise nachgewiesen werden, an welchem Ort auf oder in der Zelle das Antigen lokalisiert ist.
