Abstract
Description
The invention relates to a transilluminator for the qualitative and quantitative evaluation of labeled with fluorescent or visible dyes material samples, consisting of a support surface for the material sample to be analyzed, below the support surface, a light source and a UV-resistant diffuser and above the support surface an emission filter for the separation of Excitation wavelength and a device for visual or automated detection and evaluation are arranged.
For numerous tasks for the qualitative and quantitative analysis of various objects, illuminated tables are used, which are also called "transilluminators". Typical applications for this are the control or evaluation of banknotes, X-ray images and medical or biological material samples. Regardless of the particular application, transilluminators are used primarily for the excitation of fluorescent dyes (UV / VIS) and for the backlighting of colorimetrically colored samples or any film negatives or film positive (white light).
Thus, the dyeing and marking techniques have evolved clearly and dominantly in the direction of fluorescent dyes in the last 10 to 15 years. In the meantime, more than two-thirds of all DNA, RNA and protein detections are performed with fluorescent dyes.
Die zu markierenden oder färbenden Flächen liegen hierbei in Größenbereichen von 2 cm2 bis 1.000 cm2.The fluorescent dyes are preferably used for labeling cells and internal cell constituents in fluorescence and laser scanning microscopy and for labeling cells for cell sorters. Another main field of application is the labeling and staining of macromolecules and molecular biological probes. For example, up to 100 different fluorescent dyes are used. The areas to be marked or colored here are in size ranges from 2 cm 2 to 1,000 cm 2 .
For new special applications in pharmacy, medicine and biotechnology, a large number of new fluorescence dyes adapted to the requirements of research and production has recently been developed. Thus, market-leading manufacturers offer more than 300 different fluorescent dyes, with the number of dyes frequently used being on the order of more than 200 different dyes.
The variety of fluorescent dyes currently employed present problems for both research laboratories and industrial users due to the limited number of excitation wavelengths available using commercial transilluminators to excite these fluorescent dyes. The common standard variant of a transilluminator used by many laboratories includes a surface illumination source emitting one or at most two discrete wavelengths. In order to carry out the excitation of gels and other support materials, the transilluminators have luminous areas of 10 × 10 cm to about 30 × 47 cm. The samples excited to fluoresce by the transilluminator wavelength emit a dye-specific wavelength. This wavelength can be separated from the excitation wavelength via a matched filter (emission filter) and detected visually or with a CCD detector.
Since different fluorescent dyes are used in the laboratory for different methods, several separate transilluminators are necessary, which inevitably results in a greater purchase and maintenance and space requirements.
Nevertheless, several alternative available transilluminators are suboptimal for most fluorescent dyes because the specific excitation wavelengths are not available. Therefore, many fluorescent dyes either can not be used at all or due to the suboptimal excitation can not develop their full performance and sensitivity. Consequently, due to an insufficient Excitation efficiency diverse information (eg, DNA, RNA, protein fragments in low and lowest concentrations) that are contained in the sample, are not covered for scientific analysis at all.
The object of the invention is to provide a transilluminator, which has a much better functionality over the previously known device technology. This should be achieved in particular by evaluations for different fluorescent dyes with different excitation wavelengths can be realized with only one device.
This object is achieved by an additional unit for setting specific wavelengths is arranged in the beam path between the light source and the diffuser. Advantageous embodiments are the subject of subclaims, whose technical features are described in more detail in the embodiment.
- Possibility of setting specific wavelengths in a bandwidth of X nm (eg 20 nm), preferably in a range of 250 nm to 1000 nm
- - homogeneous illumination of the illuminated area
- - sufficiently high radiation intensity for the sensitive excitation of the fluorescent dyes
- - Control of the wavelength selection and the radiation intensity by means of PC and manual adjustment
- - Laboratory-compatible design with reliable technology and easy operation
The tunable transilluminator according to the invention can also be used without problem as a single device for the visual verification of fluorescent samples.
To do this, the user merely has to select an emission filter suitable for the application and, for example, put on special goggles in order to be able to distinguish the fluorescent regions from the excitation radiation, insofar as this is in the visible range.
B. Klebstoffe. This transilluminator is suitable for numerous qualitative and quantitative evaluations of material samples labeled with fluorescent or visible dyes, for example, for electrophoresis electrophoresis, for gelassing, for stimulating minerals, for stimulating fish parts for controls of any worms present, and for desiccating materials , z. B. adhesives.
With the alternatively possible manual or computer-aided control of the transilluminator can be optimally adapted to the specific application requirements. Thus, a manual control can be used at least for all essential functions, such as in particular intensity, control of the wavelength selection, selection of a wavelength from a broadband light source and driving only this selected wavelength or switching from one to the other wavelength.
These functions that can be controlled manually can also be controlled by computer. In addition, the computer-based control can also be used for other functions, such as a simultaneous control of all X-wavelengths, switching between PC and manual control, an hour meter, an integrated automatic shutdown after a predetermined time limit and the like. Furthermore, with such a functionality by means of the PC control and time-intensity wavelength sequences can be programmed (by the user created by software) and the transilluminator can be controlled automatically via the function. Finally, another advantage of this variant is its suitability for laboratory practice, in that all settings and functions can be permanently documented and thus enable correct verification even at later points in time.

Regardless of the respective specific embodiment in detail, each variant provides a tunable transilluminator with a homogeneous illumination, in the use of which a multiplicity of relevant wavelengths can be set with a single device. In this case, the combination of the broadband light source with a narrow-banded and variable filter ensures that the wavelengths of excitation and emission, which are closely adjacent to many dyes, do not cause any problems in the practical operation of the transilluminator.
