Description
The The invention relates to a method for the quantitative determination of Colocalization of at least two molecular markers in tissue sections, especially in skin and Mucosal tissue sections.
The objective of applying such methods is to derive from the thus gained knowledge about the molecular arrangement and distribution diagnostic To obtain data on specific diseases or findings on the Organization of cells or cell systems at the molecular level to win.
The known in the art and in biology, biochemistry and medicine used by default Procedures are based largely on manual methods. In these the solution to be examined is treated with a solution, which is a specific reagent, eg. As a dye or an antibody contains. This marks a group of molecules to be examined in that it is attaches to this or with her example, to form a reacts specific complex. After that, the so marked molecular groups contained tissue section by various, mostly optical methods examined, with a to be determined arrangement and distribution the molecular group of interest reproducing signal distribution pattern is recorded.
A proven The method is fluorescence microscopy, in which the Fluorescence from with a fluorophore, for example fluorescein iso-thiocyanate or phycoerythrin, labeled molecules is measured. This becomes a fluorescence signal distribution pattern which shows the distribution and arrangement of the labeled molecule in the reproduces the microscopic object. To determine the colocalization several labeled molecule species is the method described above under the use respectively Repeat different fluorophores often. The for the different molecular species respectively obtained fluorescence signal distribution pattern are then to Determination of the correlative distribution of the investigated molecular species together evaluate.

From that The invention is therefore based on the object, a method indicate which is also reliable quantitative statements on the colocalization of molecular markers in tissue sections, especially in skin and mucosal tissue sections enabled.
- Generation of a digitized image of the tissue section for each of the at least two molecular markers to represent the respective molecular marker,
- - superimposition of the digitized images into a summation image,
- Pixel-wise quantization of the molecular marker signal for each of the at least two molecular markers in the summation image and
- Generation of a matrix, wherein each pixel is assigned a vector whose components are the quantized molecular marker signals of the at least two molecular markers.
In the method according to the invention, an image which represents the signal distribution pattern of the respective molecular marker is first recorded and digitized for each of the at least two molecular markers. The images can be generated by different analysis methods. The fluorescence proves to be particularly suitable zenzmikroskopie as a proven in biology and medicine for years analysis method. In this case, fluorophores are used as molecular markers whose fluorescence signals are displayed in a fluorescence image. However, other methods of analysis may be used to generate the digitized images of the tissue section, such as autoradiography, in which the digitized images are read by reading a radiation detector which records the radioactive radiation emitted by isotope-labeled molecules, or by digitizing one by the radiation blackened photographic film are generated.
The digitized images are then according to the invention to a Sum picture superimposed, where It is particularly important to or pixel-precise overlay arrives to exact statements about to get the relative positions of each molecule marker to each other. For this it is advantageous, in addition to the individual images for presentation the respective molecular marker in a conventional manner also record a phase contrast image.
According to the invention after image overlay the signals for each of the at least two molecular markers quantized pixel by pixel in the summation image. This can be done through different ones Types of coding take place, for example by means of a trinary coding (Base 3) or generally by coding to the base of a number n.
According to the teaching the invention is exhaustive generates a matrix, with each pixel of the summation image Vector is assigned, whose components are the quantized molecular marker signals which correspond to at least two molecular markers. This now makes it possible those obtained in the methods known from the prior art qualitative data for the colocalization of molecular markers also be able to evaluate quantitatively precisely. this makes possible among other things, a valid comparability of measurement results, which were obtained for example on different samples.
To an advantageous embodiment of the invention is provided that the tissue section before generating the digitized images anatomically standardized. In case of a skin tissue cut For example, the standardized alignment can be done in such a way that the basal membrane zone (BMZ) of the skin tissue in each of the digitized Images arranged essentially horizontally. Alternatively, too the digitized images are anatomically standardized, so that alignment of the tissue section itself is not required is.
To a further advantageous teaching of the invention will become immediate before the overlay of the Pictures to a sum picture of each of the pictures with regard to non-informative Contents, in particular with regard to any artifacts, checked with non-informative content marked image areas and this Image areas removed from all images. This ensures that that superimposed to a sum picture digitized images no the quantitative analysis of molecular marker colonization falsifying Image content more included. The removal of at least one Image with non-informative content occupied image areas of all allows digitized images, that the summation image subsequently generated by superposition Usable information on all molecular markers in every pixel contains. As test criteria come for example the integrity of the tissue section or the contamination the tissue section by particles in question. Also obvious unnatural high signal values justify an exclusion of the relevant Image area.
To A further advantageous teaching of the invention, the each Pixel associated vectors on a defined horizontal Normalized width of skin tissue cut. This can be done by vectorial Addition of the signal vectors of the defined horizontal Width of Hautgewebeschnitts juxtaposed pixels respectively. This carries the biological situation takes into account that it is the skin around a nonhomogeneous, stratified, perpendicular to the BMZ line permanently differentiating epithelial tissue, so that a standardization therefore only along the BMZ line, d. H. in horizontal Direction, and not perpendicular to it makes sense. The defined horizontal width of Hautgewebeschnitts may be for example 100 microns.
The Analytics of for each pixel can be represented as a vector quantized molecular marker signals refer to the entire tissue section, but it is also possible that Colocalization of certain molecular markers, z. B. certain T-cell subpopulations, toposelective in certain microcompartments, like the epidermis, in the blood or lymphatic system or in the dermis, to be determined quantitatively. Similarly, the quantitative determination the colocalization of the molecular markers take place in at least one focal field of the tissue section. As a focus field can be any, for example, a rectangular, circular or Any polygonal area in the tissue section to be examined be determined.
Aufgrund des hohen Automatisierungspotentials des erfindungsgemäßen Verfahrens ist es sinnvoll, dieses unter Ausführung eines Computerprogramms auf einer Datenverarbeitungsanlage durchzuführen.by virtue of the high automation potential of the method according to the invention it makes sense to do this by running a computer program on a data processing system.
Of the Invention is also the object of a device for quantitative determination of the colocalization of at least two molecular markers in tissue sections, in particular in skin and mucosal tissue sections, to create which is made up of commercially available standard components composed and thus is relatively inexpensive to implement.
The The object is achieved with a device according to claim 15.
For example, the unit for generating digitized images is designed as a fluorescence microscope and includes a CCD camera.

Diese Normierung wird auch als "normalized number of Pixels (NORP) with respect to a defined horizontal skin width" bezeichnet. It proves to be appropriate, the for every pixel received signal vectors to a defined horizontal width normalize the skin tissue cut. The standardization is preferred by vectorial addition of the signal vectors juxtaposed Pixels on the defined horizontal width of the skin tissue section, the present expediently to 100 microns is determined. Standardization takes into account the biological situation that it is permanent in the skin around a perpendicular to the BMZ line differentiating epithelial tissue, so a normalization only along the BMZ line, d.
