Es wird 1 h bei 0°C gerührt. Die klare Reaktionslösung wird in ca. 500 ml Aceton ausgefällt und mit ethanolischer NaOH neutralisiert. Der Niederschlag wird mehrmals mit Aceton gewaschen, in Wasser (bidest.) gelöst, in Wasser dialysiert und anschließend gefriergetrocknet. Das erhaltene Produkt wird bei 50°C im Vakuum bis zur Gewichtskonstanz getrocknet.

Anhand
der in Tabelle 1 aufgeführten
Substanzen soll beispielhaft die zytotoxische und antivirale Wirkung
von carboxylierten Hyaluronsäuresulfaten
aufgezeigt werden. Tabelle 1: Beispiele synthetisierter Hyaluronsäurederivate
und Kontrollsubstanzen.
3.2. Zytotoxizität3.2. cytotoxicity
Testbeschreibungtest Description
Die Zytotoxizität der Testverbindungen wird auf zwei Tage-alten GMK-Zell-Monolayern bestimmt (Schmidtke et al., J. Virol. Meth. 95, 2001: 133–143). Dazu werden 9 Verdünnungen der Substanz in 100 μl Testmedium zugegeben. Zellen ohne Substanz dienen als Kontrolle. Nach einer Inkubationszeit von 72 h bei 37°C mit 5% CO2, erfolgt die weitere Bearbeitung. Nach Entfernen des Zellkulturüberstandes werden die Zell-Monolayer 3 × mit 300 μl physiologischer Kochsalzlösung gewaschen, um tote Zellen zu entfernen. Die Fixierung und Färbung der Zellen erfolgt mit 50 μl einer 0,3%igen Kristallviolett-Lösung für 10 min. Nach 6-maligem Waschen mit 300 μl Wasser werden die gefärbten Monolayer 20 min mit 100 μl Lyse-Puffer (0,8979 g Natriumcitrat, 1,25 ml 1 N HCl in 98,05 ml 47,5% Ethanol) behandelt und das Kristallviolett herausgelöst. Die optische Dichte der einzelnen Vertiefungen wird bei 550/630 nm bestimmt. Die optische Dichte der 6 Kontrollen ohne Substanz wird 100% gesetzt. Anhand der mittleren Dosis-Wirkungs-Kurve von 3 unabhängigen Testen werden die 50% zytotoxische Konzentration (CC50) und maximal tolerierte Dosis (MTD = CC10) errechnet.The cytotoxicity of the test compounds is determined on two day old GMK cell monolayers (Schmidtke et al., J. zpE-Hemmtest wird auf 2 Tage alten GMK-Zellrasen in Mikrotiterplatten
mit 96 Vertiefungen durchgeführt
(Schmidtke et al., J. Virol. Meth. 95, 2001: 133–143). Nach dem Absaugen des
Zellwachstum-Mediums werden
50 μl der
Testsubstanz, gelöst
im Testmedium, und eine bestimmte Viruskonzentration (HSV-1) in
50 μl des
Testmediums zugegeben. 6 Vertiefungen mit Zellen ohne Virus und
6 Vertiefungen mit Virus, aber ohne Testsubstanz, dienen als Zell-
bzw. Viruskontrolle auf jeder Platte. In jedem Test wird von Phosphonoameisensäure (PFA),
die zuvor im Plaque-Reduktions-Test bestimmte 50%ige und 100%ige
Hemmkonzentration (IC50 = 12,5 μg/ml und
IC100 = 125 μg/ml) als Kontrolle mit geführt. Die
Platten werden bei 37°C
und 5% CO2 inkubiert. Die Entwicklung des
zytopathischen Effektes (zpE) wird mikroskopisch verfolgt. Zirka
48 h nach Infektion, wenn die Viruskontrollen den maximalen zytopathischen
Effekt zeigen und die Referenzverbindung (PFA) eine etwa 50 bzw.
100%-ige Hemmung, werden die Zellen fixiert und mit 0,3% Kristallviolett
in Ethanol/Formalin-Lösung
gefärbt.
Nach dreimaligem Waschen und anschließendem Herauslösen des
Farbstoffes mit einem Lysepuffer wird die optische Dichte bei 550/630
nm bestimmt und die antivirale Aktivität berechnet. Anhand der mittleren
Dosis-Wirkungskurven von 3 unabhängigen

Die antivirale Wirkung der caboxymethylierten Hyaluronsäuresulfate steht in einem engen Zusammenhang mit ihrem Sulfatierungsgrad (Tabelle 2). Je stärker die Sulfatierung, desto stärker war die nachgewiesene Hemmung des HSV-1-induzierten zpE in GMK-Zellen und desto niedriger war die 50%ige Hemmkonzentration. Für die Verbindungen mit einem Schwefelgehalt von > 11% (CHS-3 und CHS-4) wurden IC50-Werte von 0,7 μg/ml ermittelt. Diese Werte entsprachen in etwa der 50%igen Hemmkonzentration des Hyaluronsäuresulfats (HS-1) und liegen um ein Vielfaches niedriger als die der Kontrollsubstanz PFA. Nichtsulfatierte Hyaluronsäure bzw. deren carboxymethyliertes Derivat (CH-1) sind im verwendeten Konzentrationsbereich bis 100 μg/ml nicht antiviral wirksam.The antiviral activity of caboxymethylated hyaluronic acid sulfates is closely related to their degree of sulfation (Table 2). Melisse wurden die unter Punkt 2 bereits beschriebenen zpE-Hemmteste
eingesetzt. Die dabei einzeln sowie in Kombination eingesetzten
Konzentrationen betrugen für
Melisse 12,5, 25, 50, 100 und 200 μg/ml, für MS-1 und CHS-4 0,1, 0,2,
0,4, 0,8 und 1,6 μg/ml.
Die mit HS-1 und Melisse bzw. CHS-4 und Melisse erzielten antiviralen
Wirkungen sind in den Tabellen 3 bzw. 4 zusammenfassend dargestellt. Tabelle 3: Hemmung des HSV-1-induzierten

Aus den Daten wird ersichtlich, dass sowohl die Kombination von Hyaluronsäuresulfat mit Melisse (Tabelle 3) als auch von caboxymethyliertem Hyaluronsäuresulfat (Tabelle 4) mit Melisse zu einer starken Steigerung der antiviralen Wirkung führte.
