Abstract
Die Erfindung bezieht sich auf ein Invertierungsverfahren von reinen oder feststoffbehafteten saccharosehaltigen Loesungen unterschiedlicher Konzentration. Zum Einsatz kommen invertaseproduzierende Hefen der Arten Saccharomyces cerevisiae, Candida utilis und Hansenula anomala, die als Suspension oder Filtrate der Hefesuspensionen direkt der saccharosehaltigen Loesung zugesetzt werden. Die Invertierung kann oekonomisch guenstiger als bei vergleichbaren anderen Invertierungsverfahren in einem vertretbaren Zeitaufwand durchgefuehrt werden.The invention relates to an inversion process of pure or solid-laden sucrose-containing solutions of different concentrations. Description
Die Erfindung betrifft ein Verfahren zur Invertierung von saccharosehaltigen Lösungen unterschiedlicher Konzentration. Die Erfindung kann in solchen Industriezweigen genutzt werden, die Substrate für die Kultivierung von invertaseinaktiven Mikroorganismen benötigen bzw. in denen Invertzuckerlösungen gebraucht und so ökonomisch günstiger gewonnen werden können.The invention relates to a method for inverting sucrose-containing solutions of different concentrations. Invnrtzuckerlösungen werden in vielen Industriezweigen, wie z.B. der Süßwarer-, Backwaren- oder Getränkeindustrie angewandt. Die Gewinnung dieses Invertzucker basiert meist auf der Behandlung von Saccharoselösungen mit anorganischen oder organischen Säuren. So kommen organische Säuren, wie Citronensäure (SU-PS 730809) und anorganische Säuren, wie Schwefel- oder Salzsäure, zur Anwendung. Diese Verfahren bringen aber den Nachteil mit sich, daß die erhaltenen Inventzuckerlösungen einer intensiven Reinigung unterzogen werden müssen, um die Anionen, die durch die Säuren eingetragen werden, zu entfernen. Dadurch gestaltet sich die Verfahrensführung ökonomisch uneffektiv. Außerdem kann die Verschiebung des pH-Wertes eine Neutralisation erforderlich machen, wodurch zusätzliche Ionen in die Lösung gelangen. Eine weitere Möglichkeit besteht in der enzymatischen Hydrolyse der Saccharosemoleküle unter Verwendung industriell hergestellter Invertasepräparate. Die Gewinnung dieser Invertase (ß-D-Fructo-furanosido-Fructohydrolase, EC3.2.1.26.) erfolgt vorrangig aus Back- und Bierhefe durch Plasmolyse und Autolyse. Das gereinigte Produkt wird in flüssiger Form, z. B. mit 70% Sorlirt stabilisiert in den Handel sebracht. Ebenfalls gibt es die Möglichkeit, die Zellen mechanisch oder enzymatisch aufzusch aßen, um die Invertase freizusetzen. (SUlTON und LAMPEN, Biochim. Biophys. acta 56 [19621, S.303-312). Großtechnisch korniit ein Verfahren zur Anwendung, in dem Kontinuierlich aus Backhefe Invertase gewonnen wird (GB-PS 2198731). Dazu wird die Hefezellsuspension in einer Spindelmühle kontinuierlich durch Mikroglaskugeln aufgeschlossen. Über verschiedene fteinigungsschritte wird die invertasehaltige Lösung von den Hefezellen getrennt und konzentriert. Stabilisiert mit Glycerol gelangt sie dann in den Handel. Die Herstellung einer solchen Invertaselösung birgt auf Grund der umfangreichen Reinigungsund Stabilisierungsschritte ökonomische Nachteila in sich. Dementsprechend ist die Verwendung so hergestellter Präparate aus ökonomischer Sicht oft nicht vorte'lhaft.Invalid sugar solutions are used in many industries, such as the confectionery, bakery or beverage industry. Bei dem Verfahren wird weiterhin die zu hydrolysierende Saccharosolösung vor der Invertierung kurzzeitig erhitzt und durchläuft noch einen adsorptiven Reinigungsschritt. Durch die Nutzung solcher immobilisierter Enzyme entstehen eine Reihe von Nachteilen für die Verfahrensführung. So wird die Aktivierungsenergie heraufgesetzt und die Substratumsatzgeschwindigkeit nimmt ab. Rentabel gestaltet sich eine solche Verfahrensführung nur für große Mengenströme, wobei ausreichend urd ökonomisch günstig Trägermaterialien und Enzyme vorhanden sein müssen. Erfindungsgemäß wird die Aufgabe dadurch gelöst, daß man eine dickbreitige Hefesuspension, die keine physikalisch oder enzymatisch aufgeschlossenen Zellen enthält, der Hefen Saccharomyces cerevisiae, Candida utilis oder Hansenula anomala direkt zugibt oder mit Bierwürze oder Wasser auswäscht und filtriert, so daß die Hefezellzahl maximal 106HZ/ml beträgt, und diese Lösung in einer Konzentration von 1 % bis 20%, vorzugsweise 5%-15% einer saccharosehaltigen Lösung, die unterschiedliche Konzentrationen aufweisen kann, zusetzt. Die Temperatur dieser Lösung wird dabei auf 20°C-70°C, vorzugjweise 50°C-60°C, und der pH-Wert auf 3-6, vorzugsweise 4-5,5 eingestellt. Dieser pH-Wert braucht nicht durch Zusatz anorganischer oder organischer Säuren einreguliert zu werden, da eine Saccharoselösung selbst einen solchen pH-Wert aufweist. Es besteht ebenfalls die Möglichkeit, ungereinigte saccharosehaltige Lösungen (Zwischerprodukte bei der Zuckelherstellung, Dünnsaft) zu invertieren, solange die enthaltenen Ionen, z. B. Ca* *, nicht zu einer Inhibierung der Invertase führen. Sonst müßten sie vorher durch geeignete Maßnahmen abgetrennt werden.
