Abstract
The invention relates to a method for producing a medical patch, which is based essentially on the production of a carrier material by coagulation of a colloidal system provided with active ingredients and on the achievement of an ionic strength in the coagulum decreasing in the direction of the releasing surface. The colloidal system is a polyacrylate dispersion or solution. The polyacrylates used are preferably a carboxyl group-containing copolymer of acrylic, methacrylic, itaconic, crotonic, maleic or fumaric acid with analogous esters of C 1 to C 8 alcohols. A colloidal system of the invention is obtained by reacting an aqueous polyacrylate dispersion with a glycerol-containing drug solution is added. As active ingredients are in particular drugs, preferably procaine, oxybuprocaine, Glyceroltrinitrat, steroids, clonidine, scopolamine and b-blocker into consideration, among the steroids z. Description
The invention relates to a method for the production of a medical patch, which contains active ingredients and provides them timed for a local or systematic effect.
In a review (Pfleget, P. and M. Dittgen, Pharmacy 42, 799-809 [1987/121] the possibilities known for patches for the timing of drug release have been explained. Accordingly, special measures are required in order to set the release quantity constant in the case of the paving stones with matrix diffusion-controlled release. These measures include, for example, the so-called case II diffusion, which allows a time-constant release in gel and swellable matrices, or the so-called carrier fixation, in which a staggered release of free and carrier-fixed drug provides overall for the desirable time constant. The gel-like and swellable matrix is perceived as unpleasant during the main contact. Furthermore, as a function of the hydration and transpiration different skin hydration, fluctuating levels of release are found over time.
Although the carrier fixation brings substantial time constant release, but on the other hand means an additional effort in the production.
Another problem with medical patches is their adhesion to the skin.
For patches for the epidermal application of drugs (eg Nitradisc®) that are already on the market, the lack of adhesion, etc., is by combination with a separate, surrounding the preparation adhesive layer. Also, the film described in DE-AS 2006696, which generally consists of copolymers of acrylates and methacrylates and active ingredients such as Antikontrazeptiva in homogeneous mixture contains, is applied in conjunction with a patch to deliver the drugs transdermally.
All these preparations have the common drawback that the skin contact is not continuously secured due to the lack of adhesion of the applied films and that there are additional difficulties in the application. This deficiency can not be avoided by pasting over with plaster or a different kind of combination with plaster, since due to the necessary surface area of the preparations for therapeutic requirements always with dislocations and bloating under the plaster must be expected. This statement is confirmed, inter alia, by the studies of Prakash R. Keshary and Yie W. Chien (Drug Development and Industrial Pharmacy 10 (6), 883-913 [1984]). The authors found standard deviation of ± 5,14 mg / cm 2 · h 1/2 for the drug-inflammation rate in a Keshary-Chien cell in the already mentioned Nitradisc 9 .
AS 2920500 and 3204551 disclose patches in the form of a polyacrylate film. In the case of DE-AS 2920500 it is provided for producing a drug gradient that one or more times drug-containing acrylic resin solution is applied to one or both sides of the film. The contact of the polyacrylate film with organic solvents causes difficulties, since the film softens and dissolves when it is dissolved with the solvent. As a result, it turns out that film pieces, for. As stamped, have from such a film produced larger standard deviations in the release rate of the drug. Also in the process described in DE-AS 3204551, the polyacrylate is taken up in an organic solvent and poured together with the drugs to form a film. The use of organic solvents in connection with the preparation of the patch always means a residual solvent residue that may later cause skin irritation or drug-release disorder.
The concept of membrane-directed transdermal therapeutic systems (eg Transderm Nitro 8 , Alza Corp., USA) provides a layered structure: drug reservoir, control membrane, adhesive layer. The control membrane used in this case influences the drug transfer to the skin, which is why it must be applied with extreme precision, which makes the preparations more expensive.
Des weiteren lassen sich die Schichten nur schwer reproduzierbar auftragen, und sie haften zwar relativ gut auf derIn another case, an attempt was made to achieve adhesion and, at the same time, as little as possible an influence on the drug substance penetration by means of an outer layer of defined surface area and thickness from a physiologically inert, cross-linked hydrogel, e.g. B. disclosed in DD-WP 66910 or commercially under the name landfill *. These solutions have the disadvantage that hydrogel layers can retard the artefact transfer and cause this to varying degrees by aging. Furthermore, the layers are difficult to reproducibly apply, and they adhere relatively well on the
Skin, but less well on a Unter'dge or a carrier, so that there is a risk of separation of the dosage form in this area.
Tech. In other cases, certain requirements are placed on the mechanical properties of the supports or documents. Thus, according to DE-OS 3200369, it is necessary to select underlays which stretch by at least 10% or which have been subjected to a stretch-contraction treatment in order to prevent peeling or avoid an unpleasant feeling when applied to the skin. However, it has been shown (M. Dittgen in Drug Development and Industrial Pharmacy, Pharm. Tech., Conference Issue 11: 1 & 2 [1985], that a variation in the mechanical properties of drug-containing films causes severe fluctuations in drug delivery. On the other hand, the mechanical properties of drug-containing films, especially those based on polyacrylate, can not be kept sufficiently constant at reasonable expense during storage, since they depend in particular on the humidity during storage (M. Dittgen, W. Stahlkopf and Cornelia Schu! Pharmacy 39 »7) 499-500 [i984)). Thus, it is difficult and economically complicated to ensure reproducible conditions and thus a constant bioavailability of the incorporated active ingredient for such films, which in some way already have an improved application behavior on the skin.
Ziel der Erfindung ist es, die Nachteile der bisher vorliegenden Herstellungsverfahren für medizinische Pflaster und insbesondere die Auswirkungen bezüglich Wirkstofffreigabe aus dem Pflaster sowie Restgehalt an Fremdstoffen zu vermeiden und damit ein Pflaster bereitzustellen, das sich dadurch auszeichnet, daß ein inkorporierter Wirkstoff mit vorausbestimmter, konstanter Geschwindigkeit möglichst vollständig freigesetzt wirdThe aim of the invention is to avoid the disadvantages of the present production of medical plaster and in particular the effects of drug release from the patch and residual content of foreign substances and thus to provide a plaster, which is characterized in that an incorporated drug with predetermined, constant speed released as completely as possible
The manufacturing process should be cost-effective, least require the use of foreign substances and be based on the use of carriers or filin formers, which cause no irritation of Htut.
The invention has for its object to develop a method by which any active ingredients, in particular drugs, can be embedded in a carrier material, so that in simple technology ai ss this material transdermal patches can be produced which contain little foreign substances and zeigesteuert the drug release. According to the invention, this object is achieved by a manufacturing technology which produces a carrier material obtained by coagulation from a colloidal drug-containing system wherein the active-ingredient-containing coagulum has an ionic strength decreasing towards the releasing surface and the carrier material having a cover - And adhesive layer is provided. Suitable colloidal systems are polyacrylate dispersions or solutions. The polyacrylates preferably represent a carboxyl-containing copolymer of acrylic, methacrylic, itaconic, crotonic, maleic or fumaric acid with analogous esters of C r to C 8 -alcohols.
The active substances used are, in particular, medicaments, preferably procaine, oxybuprocaine, glycerol trinitrate, clonidine, scopolamine and steroids and β-blockers. Suitable steroids are, for example, estradiol esters. An embodiment of the invention relates, for example, to the use of an aqueous polyacrylate dispersion with glycerol-containing solutions of procaine (1) or oxybuprocaine (2).
In a preferred embodiment of the invention, the support material by coagulation of a commercial polyacrylate dispersion, the SCOPACRYL * (trademark of VEB Chemische Werke BUNA for carboxyl-containing copolymer of aryl and / or methacrylic acid with esters of C r to C 8 alcohols) existing It is in accordance with the essence of the invention that the film obtained by coagulation of the colloidal system comprises both aqueous polyacrylate dispersions (emulsion polymers) and polyacrylate preparations in organic solvents (US Pat. Solution polymers, block polymers) can be produced. Preferably, an aqueous polyacrylate dispersion (40% solids, formulation A) is mixed with a glycerol-containing drug solution, e.g. For example, a solution of {!) Having an ionic strength I 3 of, for example, 1 a = 6.25 10 ~ 2 mol · 1 "'(batch B a ) is added and the mixture is stirred until uniform After a drying process, for example, 3 min at 80 0 C or 72 h at 2O 0 C to 25 0 C) results in a surface (eg polyethylene film) defined surface (eg 87 m 2 ) Coagulum characterized by a relatively high diffusion resistance for the incorporated drug, a diffusion coefficient for (1) of D "= 7.7 × 10 -5 cm 2 -s-1 was determined.
It also corresponds to the essence of the invention to apply several coagulates on top of each other on this coagulum. For this purpose, a mixture of the mentioned approach A is coagulated on the respective preceding order with an approach B modified in its composition.
The obtained by modifying the approach B a batch B b represents, for example, a glycerol-aqueous solution of (1) represents the ionic strength Ib but is lower in comparison to the preceding batch B, for example, l b = 3.16 10 -2 mol Γ 1 , Then follow corresponding combinations in which the ionic strength in each approach B c "l c " is further lowered in each case.
It is also within the spirit of the invention to depart from the cumbersome layered order and to make the structure of the erf indungsgomäßen, provided with drugs carrier continuously. In this case, work is carried out on a suitable coating system, which makes it possible to apply combinations of such additives as explained above with the approaches A and B 4 ... X in one operation from a plurality of application stations.
B. Diffusionskoeffizient für (1) in Ansatz B„:D„ = 4,4· 10~'°cm2 s"'.Since, in particular, it has been found that the lower the ionic strength I of the batch B, the lower the ionic strength I of the batch B, the diffusion resistance of a coagulum for the incorporated drug is correspondingly staggered according to this principle for all further coagulates, which manifests itself in an increasing diffusion coefficient , z. B. Diffusion coefficient for (1) in batch B ": D" = 4.4 × 10 -6 cm 2 s -1.
It is dependent on the preparation of the carrier coagulum set forth. it is possible to produce a transdermal patch which ensures a time-dependent dependency of the time taken for the incorporated drug, or a specific requirement (for example, chronopharmacology).
The essence of the invention further includes that the described coagulation of the polyacrylate also by a variation of other essential for colloid stability parameters such. B. the temperature and or or the pH is carried out. The carrier material according to the invention may be self-adhesive or, as already known, it is provided with an adhesive layer which ensures epicutaneous adherence of the patch and at the same time mediates drug delivery to the skin. This adhesive layer may also contain drug and be prepared in the same manner as described for the carrier material by coagulation of a polyacrylate.
