Abstract
Die Erfindung betrifft ein neues Verfahren zur Herstellung einer epidermal anwendbaren pharmazeutischen Zubereitung, fuer die charakteristisch ist, dass eine Haftschicht auf eine arzneistoffhaltige oder inerte Unterlage aufgebracht wird. Die Herstellung der Haftschicht erfolgt in der Weise, dass einer waessrigen Dispersion eines carboxylgruppenhaltigen Copolymerisates von Acryl- und/oder Methacrylestern mit einem maximalen Fremdstoffgehalt von 0,56% nach dem Aufbringen auf die Unterlage das Wasser weitgehend entzogen wird. Bei der Applikation befindet sich die Haftschicht zwischen Haut und Unterlage, wobei sie den Arzneistoffuebertritt nicht oder nur wenig beeintraechtigt, einen festen Sitz der Zubereitung garantiert und die Haut nicht irritiert bzw. kein unangenehmes Gefuehl verursacht.The invention relates to a novel process for the preparation of an epidermally applicable pharmaceutical preparation, for which it is characteristic that an adhesive layer is applied to a drug-containing or inert substrate. Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung einer haftfähigen pharmazeutischen Zubereitung, die für die epidermale Applikation von Arzneistoffen geeignet ist und die eine gesteuerte Arzneistoffabgabe an die Haut ermöglicht.The present invention relates to a process for the preparation of an adhesive pharmaceutical preparation which is suitable for the epidermal application of drugs and which enables a controlled drug delivery to the skin.
Es ist bekannt, Arzneistoffe in Polymere einzubetten oder anderweitig in die Form eines Filmes zu bringen, so daß derselbe in definierter Fläche epidermal appliziert werden kann. So beschreiben beispielsweise die US-PS 3598122,3589123,3742951, 3797494,3996934,3995632,4060084,3731683 und 4031894 Mehrschichten-Zubereitungen. Weder dabei noch bei den Verfahren zur Herstellung von pharmazeutischen Einschichten-Filmen, wie beispielsweise in den DE-OS 2432925,2218200, 2207635 und DD-WP 252654/6 sowie DD-AP 150849 und US-PS 4076798, wird auf eine Haftfähigkeit der Filme orientiert. Bei bereits im Handel befindlichen filmartigen Zubereitungen zur epidermalen Applikation von Arzneistoffen (z. B. Nitradisc") wird dem Mangel fehlender Haftfähigkeit u.a. durch Kombination mit einer separaten, das Präparat umschließenden Haftschicht begegnet. Auch der in der DE-A 2006696 beschriebene Film, der ganz allgemein aus Copolymerisaten und Acrylaten und Methacrylaten besteht und Wirkstoffe wie Antikontrazeptiva in homogener Vermischung enthält, wird in Verbindung mit einem Pflaster appliziert, um die Wirkstoffe transdermal abzugeben.It is known to embed drugs in polymers or otherwise bring in the form of a film so that it can be applied epidermally in a defined area. Dieser Mangel kann auch durch das Überkleben mit Pflaster oder eine andersgeartete Kombination mit Pflastern nicht vermieden werden, da aufgrund der aus therapeutischen Erfordernissen notwendigen Flächengröße der Zubereitungen stets mit Verwerfungen und Aufblähungen unter dem Pflaster gerechnet werden muß. Diese Aussage wird u.a. durch die Untersuchungen von Prakash R. Keshary und Yie W. Chien (Drug Development and Industrial Pharmacy 10[6], 883-913 [1984] bestätigt. Als Folge ergibt sich, daß Filmstücke, z. B. Stanzlinge, aus einem derart hergestellten Film größere Standardabweichungen in der Freigabegeschwindigkeit des Wirkstoffes aufweisen. Auch bei dem in der DE-A 3204551 beschriebenen Verfahren wird das Acrylat in einem organischen Lösungsmittel aufgenommen und zusammen mit den Arzneistoffen zu einem Film ausgegossen. Die Anwendung organischer Lösungsmittel im Zusammenhang mit der Herstellung arzneistoffhaltiger Polyacrylatfilme birgt in jedem Falle den Nachteil des Zurückbleibens von Lösungsmittelresten, die bei der Anwendung möglicherweise eine Hautirritation oder eine Störung der Arzneistoffabgabe verursachen können. Das Konzept der transdermalen therapeutischen Systeme (z. B. Transderm Nitro81, Alza Corp., USA) sieht einen Schichtenaufbau: Arzneistoffreservoir, Kontrollmembran, Haftschicht vor. Die hierbei verwendete Haftschicht ist jedoch nicht hinreichend indifferent. Sie kann einerseits zu Irritationen und Hautreizungen führen. Andererseits beeinflußt sie den Arzneistoffübertritt zur Haut, weshalb sie mit äußerster Präzision aufgebracht werden muß, wodurch sich die Zubereitungen verteuern. Es wurde versucht, durch eine äußere Schicht von definierter Oberfläche und Dicke aus einem physiologisch inerten, vernetzten Hydrogel eine Haftung und zugleich eine möglichst geringe Beeinflussung des Arzneistoffübertritts zu erzielen, z. B. offenbart im DD-WP 66910 bzw. im Handel unter der Bezeichnung Deponit*. Diese Lösungen bergen den Nachteil, daß Hydrogelschichten den Arzneistoffübertritt verzögern können und durch Alterung dies in unterschiedlichem Ausmaß bewirken. Des weiteren lassen sich die Schichten nur schwer reproduzierbar auftragen, und sie haften zwar relativ gut auf der Haut, aber weniger gut auf einer Unterlage bzw. einem Träger, so daß die Gefahr einer Trennung der Arzneiform in diesem Bereich besteht. In anderen Fällen werden bestimmte Anforderungen an die mechanische Beschaffenheit der Träger oder Unterlagen gestellt. So müssen gemäß DE-OS 3200369 Unterlagen ausgewählt werden, die sich um mindestens 10% dehnen oder die einer Dehn-Kontraktion-Behandlung unterworfen worden sind, um bei der Anwendung auf der Haut das Abschälen zu verhindern oder ein unangenehmes Gefühl zu vermeiden. Es hat sich jedoch gezeigt (M. Dittgen in Drug Development and Industrial Pharmacy, Pharm.Tech.Conference Issue 11:1 & 2 [1985]), daß eine Variation der mechanischen Eigenschaften arzneistoffhaltiger Filme gravierende Schwankungen in der Arzneistoffabgabe auslöst. Andererseits lassen sich die mechanischen Eigenschaften arzneistoffhaltiger Filme, insbesondere solcher auf Polyacrylatbasis, mit vertretbarem Aufwand bei der Lagerung nicht ausreichend konstant halten, da sie insbesondere von der Luftfeuchtigkeit bei der Lagerung abhängen (M. Dittgen, W. Stahlkopf und Cornelia Schulz, Pharmazie 39 [7] 499-500 [1984]). Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zu entwickeln, nach dem beliebige arzneistoffhaltige Träger bzw. Erfindungsgemäß wird eine Haftarzneiform hergestellt, die dadurch gekennzeichnet ist, daß ein arzneistoffhaltiger Träger bzw. eine Unterlage, die den Arzneistoff gesteuert freisetzen, und eine Haftschicht verwendet werden. Der Träger bzw. die Unterlage können beispielsweise aus einer Aluminiumfolie, die gegebenenfalls mit einer Acrylat- oder Cellulosederivatschicht beschichtet ist, einem Acrylat- oder anderen Kunststoff-Film bestehen. Bevorzugt wird ein lösungsmittelhaitiger Film gem. DD-WP 253654/6 oder ein Film aus einem bekannten Acrylat (z. B.
