Abstract
Wasserloesliche Ueberzuege auf Arzneiformen werden aus einer waessrigen, gegebenenfalls pigmentierten Loesung eines niedrig substituierten Staerkeesters mit anorganischen Saeuren wie Staerkemonophosphat hergestellt. Darueber hinaus enthaelt die Loesung gleichzeitig sowohl Weichmacher als auch Polyethylenglykol. Als Weichmacher eignen sich Polyole und Polyalkohole. Als nachteilig erweist es sich weiterhin, daß zur Erzielung eines ansprechenden Überzuges ein Massezuwachs von 50 bis 100% erforderlich ist. Aus diesen Gründen wird in zunehmendem Maße das Filmdragierverfahron eingesetzt. Durch Einsatz leistungsfähiger Dragierkessel- und Wirbelschichtmethoden weiden bei der Filmdragiorung hohe Durchsatzleistungen und eine weitgehende Automatisierung möglich. Bereits ein Massezuwachs von 1 bis 2O0O verleiht der Arzneiform die gewünschten Eigenschaften.Aqueous coating mixtures for solid dosage forms have long been known and commonly used. Beim Einsatz wäßriger Polymerlösungen als Überzugsmaterialien treten eine Reihe von Nachteilen auf. Auf Grund der mit steigender Polymerkonzentration schnell ansteigenden Viskosität muß bei einer Vielzahl von Polymeren mit niudrig konzentrierten Lösungen gearbeitet werden. Dadurch wird der gesamte Prozeß ökonomisch Mngür:stiger. Der einfache Ersatz der organischen Lösungsmittel durch Wasser bringt während des Verarbeitungsprozesses eine Reihe von Problemen mit sich. Als nachteilig erweist es sich bei der Verwendung dieser Cellulosederivate, daß der Gesamtfeststoffgehalt der Überzugsrezepturen in der Regel weit unter 10% liegt und dadurch der Prozeß durch das Verdunsten des hohen Wasseranteiles ökonomisch ungünstiger wird. Die Herstellung von Amylosefilmen ist aus den Pattntschriften DE 2103620 und DE 2137767 bekannt. Können einerseits solche Filme nur aus mit Amylose angereicherter Stärke nergostellt werden, was zu einer merklichen Verteuerung des Endproduktes führt, erweist sich andererseits die Herstellung der Amyloselösung durch Dispergierung des weichgemachten Amylosegemisches durch Erhitzen bei 120 bis 170°C als kompliziert. Außerdem ist ein kontinuierlicher Sprühauftrag dieser Lösungen nicht möglich. Weiterhin ist der Einsatz eines Polysaccharide und eines Netzmittels als temporärer Schutzanstrich auf Gegenständen aus der Patentschrift DE 3223443 bekannt. Durch Zusatz von Weichmachungs- und Hydrophobisierungsmitteln wird die Auflösung des Schutzüberzuges so stark zurückgedrängt, daß er für unseren Anwendungszweck nicht einsetzbar ist. Freie Filme werden aus wäßrigen Lösungen von Monostärkemonophosphat mit oder ohne Zusatz von Weichmacher bereits bei Zimmertemperatur erhalten (DE 2428133). Diese Rezepturen eignen sich jedoch auf Grund hoher Klebrigkeit während der Trockenphase nicht zum Aufbringen auf Arzneiformen.There has been no lack of attempts to use aqueous solutions of cellulose derivatives for film draging. Maltodextrine sind niedrig verzuckerte Stärkon mit einem Dextroseäquivalent bis zu 20.
