Description
Vynález se týká směsi chemických sloučenin k fotometrickému stanoveni aktivity enzymu alkoholdéhydrogenasy v biologickém materiálu.The invention relates to a mixture of chemical compounds for the photometric determination of the activity of an alcohol dehydrogenase enzyme in a biological material.
Použitelní diagnostické závěry poskytuje většinou teprve poměr změřených aktivit dvou i více enzymů. Je poměrně málo takových enzymů, u nichž by odchylka úrovně aktivity od normálních hodnot specificky indikovala určitý fyziologický stav nebo intenzitu a lokalizaci patologického procesu. Právě stanovení aktivity alkoholdéhydrogenasy v krevním seru by mohlo poměrné selektivně vypovídat o poškození jaterní tkáně, protože jaterni buňky obsahují alkoholdéhydrogenasy mnohonásobně více než některé jiné orgány. Protože i mozková tkáň tento enzxm obsahuje, mohlo by stanovení aktivity alkoholdehydrogenasy v mozkomíšním moku indikovat některé patologické procesy nejvyšší úrovně nervové soustavy.Measurement of enzyme activities in biological material is a valuable diagnostic tool in human and veterinary medicine. Mostly, increased activity in blood serum is monitored when a pathologic condition enters a certain enzyme into the blood from disintegrating cells of diseased tissue. Usable diagnostic conclusions are usually only provided by the ratio of measured activities of two or more enzymes. There are relatively few such enzymes in which a deviation in the level of activity from normal values would specifically indicate a particular physiological state or intensity and location of the pathological process. Determination of the activity of alcohol dehydrogenase in blood serum could proportionally indicate damage to liver tissue, because liver cells contain alcohol dehydrogenases many times more than some other organs. Since brain tissue also contains this enzyme, the determination of alcohol dehydrogenase activity in cerebrospinal fluid could indicate some pathological processes of the highest nervous system level.
CitlivějšíConventional photometric techniques for the determination of alcohol dehydrogenase based on the increase or decrease in absorbance of the reduced coenzyme at 340 nM are not sensitive enough for the low levels of activity found in biological material under normal conditions. These drawbacks are overcome by the composition for the photometric determination of the alcohol dehydrogenase activity of our invention, which consists of an alcoholic substrate consisting of a primary or secondary alcohol such as ethanol, butanol, cyclohexanol or furyl alcohol at a concentration of 10 -4 to 10 - ¼, from the coenzyme a concentration of 10 - 10 to 10 - 3 in an aqueous solution of pH 7 to 11 and of an aryl nitroso compound of the general formula 10 to 10 - 10, where A is phenyl or naphthyl,
R^ a Rg jsou alkyly o jednom až čtyřech uhlících a která obsahuje popřípadě v meta-poloze k N0-skupině - CH^, - OCH^ nebo halogen, přičemž k odlišeni neenzymové reakce obsahuje —/i směs inhibitor alkoholdehydrogenasy do koncentrace 10 až 10 M.R 4 and R 8 are alkyl of one to four carbons and optionally contains meta-position to the NO-group -CH 2, -OCH 2 or halogen, wherein the distillation of the non-enzymatic reaction comprises a mixture of an alcohol dehydrogenase inhibitor up to a concentration of 10 to 10 M.
This mixture, mixed with air, cerebrospinal fluid, or tissue extract, and the like, allows the measurement of low alcohol dehydrogenase activity in these fluids without pretreatment, so that the rate of decrease of the nitro compound's color, measured by a photometer, directly determines this activity.
The components of the proposed mixture allow coupling of the two reactions catalysed by alcohol dehydrogenase such that the reduced form of the coenzyme produced by the oxidation of alcohol by the enzyme is used for the enzymatic reduction of the strongly colored nitro compound, thereby turning it into a colorless product.
Použití směsi podle vynálezu osvětlují dále uvedené příklady:The following examples illustrate the use of the composition of the invention:
To two milliliters of a mixture of 2 mM n-butanol, 0.1 mM nicotinamide adenine dinucleotide and 50 µM p-mitrosodlethylaniline in phosphate buffer pH 8.5 in a spectrophotometer cuvette, 0.5 ml of blood serum is mixed and at a wavelength of 440 mm. at 25 [deg.] C. for 2 to J minutes, there is a decrease in yellow color, which is registered with an automatic recorder. To stop the enzyme reaction, 20 µl of a saturated aqueous inhibitor solution, pyrazole, is then added to the cuvette and the color intensity is registered in the reaction cuvette for 2 minutes: if it decreases further, this is due to non-enzymatic reduction of the nitroso compound.
The enzyme activity in any conventional units or (using a molar absorption coefficient ni4-1-1 troso compound, which is 3.5.10 cm) is calculated from the initial rate of yellow color loss, determined by the tangent to the curve at the beginning of the recording, reduced by any non-enzymatic reaction rate. M) in international units or in katalah.
Přiklad 2Example 2
Připraví se dvě dvoumililitrové porce fosfátového pufru o pH 7,0 s nikotinamidadenindinuklsotidem v koncentraci 50 uM, cyklohexanolem v koncentraci 10 mM a s p-nitrosodimethylanilinem v koncentraci 20 um. Do prvé porce, jako slepého vzorku se přidá 0,1 mlTwo 2 ml portions of phosphate buffer pH 7.0 are prepared with 50 µM nicotinamide adenine dinucleotide, 10 mM cyclohexanol and 20 µm p-nitrosodimethylaniline. 0.5 ml of the blood serum to be examined is then added to both portions of the mixture. After exactly 20 minutes of incubation at 25 ° C, 0.1 ml of a 2M aqueous isobutyramide solution was added to the second portion of the mixture, thereby interrupting the enzyme reaction that had been blocked from the beginning in the first sample. The measured difference in absorbance of the two samples at 450 nm is proportional to the activity of the alcohol dehydrogenase, the numerical value of which is calculated analogously to Example 1.
