背景技术
洁牙剂等口腔用组合物中,一般配合有羧甲基纤维素钠、卡拉胶或黄原胶等粘合剂,这些成分通过溶于水中来表达粘性,使研磨剂等粉体成分和液体成分结合,赋予保型性,赋予适当的粘性。
因此,在不含水的洁牙剂等口腔用组合物中,不表达粘度,故难于保持保型性,口腔用组合物将发生经时的粉体成分和液体成分的分离。
另一方面,在洁牙剂等口腔用组合物中,以赋予功能·功效为目的,有时在制剂中配合各种有效成分。作为其有效成分,可列举出再钙化促进成分、氟化物、杀菌剂、抗炎剂、止血剂、各种酶等。
作为再钙化促进成分,可列举出羟基磷灰石等磷酸钙化合物,其中,已知α-磷酸三钙(简称为α-TCP)在口腔内再钙化促进效果极高,对龋齿的预防·修复等有效。但是,α-TCP在水存在下具有转换为磷灰石化合物的性质,该反应在氟化物或其它水溶性的磷酸钙的存在下被加速,引起自我固化反应,因此无法稳定地配合在通常含有水的洁牙剂等口腔用组合物中。
另外,作为氟化物,可列举出氟化钠、氟化钾、单氟磷酸钠、氟化锡等,氟化物的功效在于氟离子氟化牙齿的羟基磷灰石来强化牙质。若在通常的含有水的洁牙剂等口腔用组合物中配合氟化物,在口腔用组合物中,有时氟离子将溶出至水中,吸附研磨剂等其它成分,无法发挥氟离子本来的效果。例如,已知在配合有氟化钠或氟化锡等和磷酸钙或碳酸钙等研磨剂、并含有水的口腔用组合物中,氟离子吸附在研磨剂上,氟离子失活。
酶可以列举出,具有溶菌作用和蛋白分解作用的溶霉菌、变位酶(mutanase)、蛋白水解酶、淀粉酶、糊精酶等。这些酶的多数配合于含有水的组成中,会有引起水解、酶活性降低的问题。因此,在含有水的组成中配合酶时,有必要选择难以引起水解的酶、或探索稳定配合不同酶的方法。
另外,有时也将天然杀菌剂桧醇、止血剂ε-氨己酸配合于洁牙剂等口腔用组合物,但是将这些有效成分配合于含有水的组成中,会因为水解有定量值降低的倾向。此外,已知抗坏血酸及其盐类对牙龈炎和牙周病等引起的出血有效,但在水存在下容易进行分解反应,发生含量降低及着色。
有关这些有效成分的稳定性的问题,均是由于洁牙剂等口腔用组合物中含有水。因此,对于可稳定地配合α-TCP、氟化物、各种酶、桧醇、ε-氨己酸及抗血酸等的、几乎不含有水的洁牙剂等口腔用组合物,期望具有保型性、不随时间发生粉体成分与液体成分的分离的组成的口腔用组合物。
迄今为止,作为适用于几乎不含水或非水系的口腔用组合物的口腔用组合物用基质,例如特开2002-114656号中提出了含有琼脂或明胶的基质、特开2002-114657号公报中提出了含有聚乙烯粉末或聚乙烯蜡的基质、特开2002-255772号公报中提出了含有聚乙烯吡咯烷酮的基质。
发明内容
本发明的目的在于提供适用于口腔用组合物的制造、特别是适用于不含水或几乎不含水的口腔用组合物的制造的口腔用组合物用基质。本发明的目的还在于,提供保型性优良不发生成分的分离等经时稳定的、特别是适用于不含水或几乎不含水的口腔用组合物的制造的口腔用组合物用基质。本发明的目的进一步在于,提供通常在水的存在下可稳定地配合不稳定的有效成分的口腔用组合物用基质。本发明的进一步的目的在于,提供通常在水的存在下稳定地配合有不稳定的有效成分的口腔用组合物、特别是不含水或几乎不含水的口腔用组合物。
本发明人为了达成上述目的,进行了锐意研究,结果发现通过使用特定组成的口腔用组合物用基质,可提供稳定性高的口腔用组合物,从而完成了本发明。
因此,本发明涉及一种非水系口腔用组合物用基质,其特征在于,含有羟丙基纤维素,和选自浓甘油、二甘油、丙二醇、1,3-丁二醇及聚乙二醇中的至少一种。作为本发明的优选实施方式,有进一步含有选自无水硅酸及结晶纤维素中的至少一种的非水系口腔用组合物用基质。
本发明进一步涉及含有上述口腔用组合物用基质的口腔用组合物。作为本发明的实施方式,上述口腔用组合物优选为非水系口腔用组合物。
本说明书中的非水系口腔用组合物是指,相对于口腔用组合物全体,水的含量为0~3质量%、优选为0~1质量%、更优选完全不含水的口腔用组合物。进而,在本说明书中将这种适用于非水系口腔用组合物的制造的基质称作非水系口腔用组合物。
本发明还涉及上述口腔用组合物用基质、及含有在水存在下含有不稳定的有效成分的口腔用组合物。作为该在水存在下不稳定的有效成分,可列举出,α-磷酸三钙(α-TCP)、氟化物、酶类、桧醇、ε-氨己酸、抗血酸及抗血酸盐类等。
因此,本发明的口腔用组合物具体地含有上述口腔用组合物用基质及上述在水存在下不稳定的有效成分的至少一种。
本发明更具体地为含有上述口腔用组合物用基质、以及作为在水存在下不稳定的有效成分的选自α-磷酸三钙(α-TCP)、氟化物、酶类、桧醇及ε-氨己酸中的至少一种的口腔用组合物。
本发明更具体地为含有上述口腔用组合物用基质、以及选自α-磷酸三钙(α-TCP)、氟化物、酶类、桧醇及ε-氨己酸中的至少一种的口腔用组合物。

作为本发明的实施方式,有含有上述口腔用组合物用基质及α-磷酸三钙(α-TCP)的口腔用组合物。作为本发明的实施方式,还有含有上述口腔用组合物用基质及至少一种氟化物的口腔用组合物。作为本发明的实施方式,还有含有上述口腔用组合物用基质及至少一种酶类的口腔用组合物。作为本发明的实施方式,有含有上述口腔用组合物用基质及桧醇的口腔用组合物。作为本发明的实施方式,还有含有上述口腔用组合物用基质及ε-氨己酸的口腔用组合物。作为本发明的实施方式,还有含有上述口腔用组合物用基质、及选自抗坏血酸及其盐类的至少一种的口腔用组合物。
作为本发明的口腔用组合物的优选实施方式,有在上述口腔用组合物用基质中组合配合有α-TCP和氟化物的口腔用组合物。
具体实施方式
本说明书中的口腔用组合物包括牙膏、液体洁牙剂、湿洁牙剂等洁牙剂类,乳膏剂,软膏剂,贴剂,口腔清凉剂,漱口剂,口香糖或漱口药等。希望本发明的口腔用组合物为非水系口腔用组合物。
本发明的口腔用组合物用基质中所用的羟丙基纤维素为使环氧丙烷与纤维素反应而得到的非离子系纤维素衍生物。根据分子聚合度的不同,存在表现不同粘度的级别。低粘度级别的为1~4mPa/S左右,高粘度级别的为1000~5000mPa/S左右。该粘度为使用旋转粘度计测定各羟丙基纤维素2%水溶液的值(mPa/S)。
本发明可使用低粘度级别至高粘度级别的羟丙基纤维素,即,可以选自具有1~5000mPa/S的粘度的羟丙基纤维素。另外可以使用粘度不同的2种以上的羟丙基纤维素。本发明优选使用粘度在150~4000mPa/S的羟丙基纤维素。
这种的羟丙基纤维素,以药品和化妆品为代表,一般作为大楼、电机部件的陶瓷、荧光灯管内的烧结剂广泛来使用,本发明中可使用用于这些的市售品。
口腔用组合物中1种或粘度不同的2种以上的的羟丙基纤维素的含量,相对于口腔用组合物总质量为0.1~15质量%,优选为0.1~10质量%。考虑到本发明所要达到的效果和口腔用组合物的使用性,上述范围内的羟丙基纤维素的量是适当的。
本发明的口腔用组合物用基质进一步含有浓甘油、二甘油、丙二醇、1,3-丁二醇及聚乙二醇中的至少一种。其中特别优选使用丙二醇、1,3-丁二醇。
浓甘油、二甘油、丙二醇、1,3-丁二醇及聚乙二醇,以药品和化妆品为代表,一般广泛应用于食品、杂货品、石油化工、涂料工业等。本发明可使用市售品。
口腔组合物中的浓甘油、二甘油、丙二醇、1,3-丁二醇及聚乙二醇的至少一种的含量,相对于口腔用组合物的总质量为10~85质量%是适当的,优选为20~60质量%。考虑到发挥口腔用组合物良好的使用性,它们的含量位于上述范围内是适当的。
本发明的口腔用组合物用基质中还可含有无水硅酸和/或结晶纤维素。
无水硅酸为二氧化硅,性状为白色粉末。用酸人工处理硅酸钠使二氧化硅析出,进行凝胶化老化后,干燥、粉碎而得到。另外,干燥减重为13%以下、强热减重为18%以下是适当的。在此,干燥减重是指105℃下加热1g无水硅酸2小时后的蒸发成分的质量%,强热减重是指850℃下加热1g无水硅酸30分钟后的蒸发成分的质量%。
这种无水硅酸以食品和化妆品为代表,一般广泛地应用于药品、农艺用品、饲料类、油墨工业、橡胶工业、塑料工业等。
口腔用组合物中无水硅酸的含量,相对于口腔用组合物的总质量为0.5~30质量%是适当的,优选为3~15质量%。考虑到本发明所要达到的效果及口腔用组合物的使用性,上述范围中的量是适当的。
结晶纤维素为酸水解或碱水解由纤维性植物作为纸浆得到的α-纤维素而得到的、基本上具有一定聚合度的纤维素微晶集合体。
这种结晶纤维素以食品和化妆品为代表,一般广泛地应用于药品、农艺用品、饲料类等。
口腔用组合物中结晶纤维素的含量,相对于口腔用组合物的总质量为0.5~30质量%是适当的,优选为3~10质量%。考虑到本发明所要达到的效果及口腔用组合物的使用性,上述范围中的量是适当的。
本发明中,在上述的口腔用组合物用基质中,可以组合以往口腔用组合物中使用的添加剂、有效成分等各种成分来制成口腔用组合物。
本发明的口腔用组合物用基质特别是组合通常在水的存在下不稳定的有效成分是有利的。这种成份可列举出α-TCP、氟化物、酶类、桧醇、ε-氨己酸、抗血酸及抗血酸盐类等。
本发明的口腔用组合物中使用的α-TCP为α-磷酸三钙(3Ca3(PO4)2·Ca(OH)2),以药品和化妆品为代表,一般广泛地用于食品、杂货品、石油化工等。本发明中可使用市售品。
本发明的口腔用组合物中α-TCP的含量,相对于口腔用组合物的总质量为0.1~50质量%是适当的,优选为1~30质量%。为了发挥α-TCP的再钙化效果,α-TCP的含量为0.1质量%以上是适当的,而超过50质量%则有时会损害口腔用组合物的使用性。
配合α-TCP时,若进一步并用氟化物,氟离子的供给会增加再钙化作用,因此可发挥极其优良的效果。因此,优选使用α-TCP与后述氟化物的组合。
本发明中所使用的氟化物具体可列举出氟化钠、氟化钾、单氟磷酸钠、氟化锡等,可使用其中至少一种。这些氟化物一般广泛应用于药品和化妆品等。本发明中可使用市售品。
本发明的口腔用组合物中氟化物的含量,相对于口腔用组合物的总质量为0.01~3质量%是适当的,优选洁牙剂等口腔用组合物中氟含量为100~1000ppm。
氟化物优选氟化钠及单氟磷酸钠。
作为本发明的口腔用组合物所使用的酶类可具体列举出溶霉菌、变位酶、蛋白水解酶、淀粉酶、糊精酶等,可使用其中的1种或2种以上。这些酶一般广泛应用于药品和化妆品、食品等。本发明中可使用市售品。
本发明的口腔用组合物中酶含量相对于口腔用组合物的总质量为0.01~5质量%是适当的,优选为0.1~2质量%。
酶优选溶霉菌及淀粉酶。
本发明的口腔用组合物中所使用的桧醇为天然树木青森扁柏所含有的特有成分,为结晶性酸性化合物。含有桧醇的其它树种可列举出台湾扁柏、北美的西部红松等。桧醇具有强的抗菌活性和广的抗菌谱,为少数的天然杀菌剂之一。
这种桧醇以药品和化妆品为代表,一般广泛地用于食品、农艺用品、建筑材料、饲料类等。本发明中可使用用于其中的市售品。
本发明的口腔用组合物中桧醇含量相对于口腔用组合物的总质量为0.005~0.5质量%是适当的,优选为0.01~0.2质量%。
本发明的口腔用组合物中所使用的ε-氨己酸是具有抗纤维蛋白溶酶效果、止血效果、抗炎效果的成分,以化妆品和药品为代表,一般广泛地用于食品、农艺用品等。本发明中可使用市售品。
本发明的口腔用组合物中ε-氨己酸含量,相对于口腔用组合物的总质量为0.001~1质量%是适当的,优选为0.006~0.2质量%。
本发明的口腔用组合物中所使用的抗坏血酸就是所谓的维生素C,具体是指L-抗坏血酸(C6H8O6=176)及其盐。作为使用的抗坏血酸盐的具体例子有抗坏血酸钠、抗坏血酸钾、抗坏血酸钙、抗坏血酸镁等。在本发明的口腔用组合物中,可含有选自抗坏血酸及其盐的1种或2种以上。抗坏血酸盐中优选使用抗坏血酸钠。
这些抗坏血酸及其盐为具有抗炎作用、胶原蛋白纤维合成促进作用的成分,以化妆品和药品为代表,一般广泛用于食品、农艺用品等。本发明中可使用市售品。
本发明的口腔用组合物中选自抗坏血酸及其盐的至少1种的含量,相对于口腔用组合物的总质量为0.001~20质量%是适当的,优选为0.01~10质量%。
如上所述,作为组合使用于本发明的口腔用组合物用基质中的有效成分,有(1)α-TCP;(2)氟化物;(3)酶类;(4)桧醇;(5)ε-氨己酸;及(6)抗坏血酸及其盐。可含有选自它们中的(1)~(6)组中化合物的1种或2种以上,而且不限于1组,可以由2组以上选择化合物来并用。其中,优选并用α-TCP和氟化物。
本发明的口腔用组合物中,根据其种类,除了上述成分,应需要可在通常的使用量的范围内配合以下的成分。
<研磨剂>
可列举出硅胶、沉降性氧化硅、火成岩性氧化硅、含水硅酸、沸石、硅酸铝、硅酸锆等氧化硅类研磨剂,磷酸氢钙二水合物、磷酸氢钙无水物、焦磷酸钙、磷酸三镁、磷酸三钙、氢氧化铝、氧化铝、轻质碳酸钙、重质碳酸钙、碳酸镁、硅酸锆、合成树脂类研磨剂等,可并用其中1种或2种以上来使用。上述研磨剂的配合量相对于组合物全体,优选为3~60质量%,更优选为10~45质量%。
<润湿剂>
可使用山梨糖醇、麦芽糖醇等多元醇等的1种或2种以上。
<发泡剂>
可列举出月桂基硫酸钠、十二烷基肌氨酸钠、烷基磺化琥珀酸钠、椰子油脂肪酸单甘油磺酸钠、α-烯烃磺酸钠、N-麸胺酸等N-酰基氨基酸盐、2-烷基-N-羧甲基-N-羟乙基咪唑啉甜菜碱、麦芽糖醇脂肪酸酯、蔗糖脂肪酸酯、聚甘油脂肪酸酯、脂肪酸二乙醇酰胺、聚氧乙烯山梨醇酐单月桂酯、聚氧乙烯硬化蓖麻油、聚氧乙烯脂肪酸酯等,可并用其中的1种或2种以上来使用。
<甜味剂>
可列举出糖精钠、阿斯巴坦、海藻糖、甜菊糖苷、甜叶菊提取物、对甲氧基肉桂醛、新橙皮甙二氢查尔酮、紫苏葶等,可并用其中的1种或2种以上来使用。
<防腐剂>
可列举出尼泊金甲酯、尼泊金乙酯、尼泊金丙酯、尼泊金丁酯等尼泊金类,安息香酸钠、苯氧基乙醇、烷基二氨基乙基甘氨酸盐酸盐等,可并用其中的1种或2种以上来使用。
<香料成分>
可并用下列物质中的1种或2种以上来使用:1-薄荷醇、茴香脑、薄荷酮、桉油精、柠檬烯、香芹酮、水杨酸甲酯、丁酸乙酯、丁香酚、麝香草酚、肉桂醛和反-2-己烯醛。这些成分可以以单品形式来配合,也可以配合含有它们的精油等。
除了上述香料成分,还可以在不妨碍本发明的效果的范围内配合脂肪醇或其酯、萜类烃、酚醚、醛、酮、内酯等香料成分,精油。上述香料的配合量相对于组合物全体优选为0.02~2质量%。
<有效成分>
可列举出沸石、氯己定盐类、西吡氯铵、苯扎氯铵、苄索氯铵、甜没药、三氯生、异丙基甲酚、生育酚醋酸酯、氨甲环酸、二氢胆固醇、聚乙烯吡咯烷酮、甘草次酸、甘草酸盐类、叶绿素铜盐、氯化钠、愈创 磺酸盐等,可并用其中的1种或2种以上来使用。
<其它>
可使用青色1号等色素、氧化钛等颜料、二丁基羟基甲苯等抗氧剂、茶干镏液、谷氨酸钠等矫味剂等。
本发明的口腔用组合物中,根据其种类,在3质量%以下范围内配合水时,除了上述成分,根据需要也可以配合以下的粘结剂。
<粘结剂>
可列举卡拉胶(1.、λ、κ)、褐藻酸、褐藻酸钠、褐藻酸丙二醇酯、含钙的褐藻酸钠、褐藻酸钾、褐藻酸钙、褐藻酸铵等褐藻酸盐及其衍生物,黄原胶、瓜尔胶、明胶、琼脂、羧甲基纤维素钠、羟乙基纤维素钠、聚丙烯酸钠等,其中可并用1种或1种以上进行使用。
需要说明的是,组合上述成分而成的口腔用组合物可依常法进行制造,无特殊限定。
得到的洁牙剂等组合物,可以填充到铝管、层压管、玻璃蒸镀管、塑料管、塑料瓶、气雾剂容器等来使用。
(实施例)
下面,通过实施例及比较例对本发明进行详细说明,本发明并不限于下述实施例。依表1~4所示的组成(单位:质量%)利用常法调制各种牙膏,供以下试验使用。结果汇总于表1~4。
1.牙膏保存稳定性试验
调制表1中的牙膏后,在下述条件下进行恶劣环境下的保存。恶劣环境下的保存结束后,依下述评价基准对各牙膏的状态进行感官评价。

2.氟离子测定试验
调制表2中的牙膏后,在下述条件下进行恶劣环境下的保存。恶劣环境下的保存结束后,量取各牙膏一定量,用水分散后立即离心分离,分离为液层和固层。对于这些液层部分,使用氟离子计,测定了氟离子浓度。在此,对于检测出的氟离子,被认为是在牙膏中不吸附其它成分、具有活性的氟离子。需要说明的是,具有活性的氟离子的残存率通过下述公式(1)求得。

4.抗坏血酸钠定量试验
调制表4中的牙膏后,在下述条件下进行恶劣环境下的保存。恶劣环境下的保存结束后,对于各牙膏中抗坏血酸钠,在如下条件下利用液相色谱法进行定量试验。
[恶劣环境下的保存条件]
1.60℃的恒温槽中,保存1个月
2.50℃的恒温槽中,保存2个月
[试验条件]
检测器:紫外分光光度计(测定波长:245nm)
柱:在内径4.6mm、长250mm的不锈钢管中填充5μm的高效液相色谱用十八烷基硅烷键合硅胶。
柱温:40℃
流动相:偏磷酸溶液
流量:每分钟1.0ml
抗坏血酸钠的残存率由下述公式(3)求得。

膏调制时的抗坏血酸钠的量)]×100
需要说明的是,对于恶劣条件下的牙膏保存品,抗坏血酸钠残存率为90%以上时,判断为抗坏血酸钠稳定。
另外,对于表4所示的牙膏,也与上述1一样进行了保存稳定性试验。

由以上实验结果、首先由表1结果可知,通过使用本发明的口腔用组合物用基质,牙膏的状态为不分离,保存稳定优良。然后由表2~4结果可知,作为本发明的口腔用组合物的牙膏,状态稳定性良好,氟化物、ε-氨己酸和抗坏血酸及其盐的保存稳定性优良。
对于依常法调制的下述实施例9~22的牙膏也进行了上述的实验,得到了与上述相同的结果。

工业实用性
利用本发明的口腔用组合物用基质,可得到保型性好、且稳定性高的非水系口腔用组合物。通过将本发明的口腔用组合物用基质与通常在水存在下不稳定的有效成分α-TCP、氟化物、各种酶、桧醇、ε-氨己酸、抗血酸、抗坏血酸盐等组合,可得到具有保型性、且可稳定维持有效成分的、不发生固体成分与液体成分的分离的经时稳定的非水系口腔用组合物。
Claims (12)
1.一种非水系口腔用组合物用基质,其特征在于,含有羟丙基纤维素,和选自浓甘油、二甘油、丙二醇、1,3-丁二醇及聚乙二醇中的至少一种。
2.如权利要求1所述的非水系口腔用组合物用基质,其中,进一步含有选自无水硅酸及结晶纤维素中的至少一种。
3.一种口腔用组合物,其中,含有权利要求1或2所述的口腔用组合物用基质。
4.一种口腔用组合物,其中,含有权利要求1或2所述的口腔用组合物用基质、及在水存在下不稳定的成分。
5.如权利要求4所述的口腔用组合物,其中,该在水存在下不稳定的成分为选自α-磷酸三钙(α-TCP)、氟化物、酶类、桧醇及ε-氨己酸中的至少一种。
6.一种口腔用组合物,其中,含有权利要求1或2所述的口腔用组合物用基质,和选自α-磷酸三钙(α-TCP)、氟化物、酶类、桧醇及ε-氨己酸中的至少一种。
7.如权利要求5或6所述的口腔用组合物,其中,含有α-磷酸三钙(α-TCP)。
8.如权利要求5或6所述的口腔用组合物,其中,含有至少一种氟化物。
9.如权利要求5或6所述的口腔用组合物,其中,含有至少一种酶类。
10.如权利要求5或6所述的口腔用组合物,其中,含有桧醇。
11.如权利要求5或6所述的口腔用组合物,其中,含有ε-氨己酸。
12.如权利要求3~11任一项所述的口腔用组合物,其为非水系口腔用组合物。
