Abstract

The invention belongs to the technical field of traditional Chinese medicine health care, and in particular relates to a traditional Chinese medicine composition with lung-protecting and cough-relieving effects, a preparation method thereof and application thereof in preparing a lung-protecting and cough-relieving medicine product.
Background
At present, western medicines are generally used for treating lung diseases, but have a plurality of side effects, and the symptoms and the root causes are not treated. The Chinese herbal medicine is a treasure in the history development process of China, has the effects of preventing and treating a plurality of diseases, and has no side effect. For possible pulmonary diseases, the occurrence of the pulmonary diseases is relieved or prevented by eating herbs having a lung protecting effect in the early symptomatic or asymptomatic period.
Chinese patent document CN114306529A discloses a traditional Chinese medicine composition, which comprises the following traditional Chinese medicine compositions in parts by weight: 20-15 parts of Chinese yam, 5-7 parts of ginseng, 9-11 parts of bighead atractylodes rhizome, 14-16 parts of reed rhizome, 9-11 parts of radix scrophulariae, 14-16 parts of polygonum cuspidatum, 9-11 parts of peach kernel, 9-11 parts of safflower, 9-11 parts of Chinese angelica, 9-11 parts of platycodon grandiflorum, 9-11 parts of fructus aurantii, 9-11 parts of coltsfoot flower and 2-4 parts of ephedra.
However, the above patent documents add ephedra into the traditional Chinese medicine composition, and the ephedra has strong effects of relieving cough and eliminating phlegm, but has addiction after long-term administration, and the decoction mode of the traditional Chinese medicine has defects in terms of convenience and mouthfeel of medicament administration, so that the development of safer, more effective, more convenient and better-mouthfeel traditional Chinese medicine composition is an urgent need.
Disclosure of Invention
Aiming at the defects existing in the prior art, the invention aims to provide a traditional Chinese medicine composition with the effects of protecting lung and relieving cough, a preparation method thereof and application thereof in preparing a lung-protecting and cough-relieving medicine product.
In order to achieve the above object, the solution adopted by the present invention is as follows:
In a first aspect, the invention provides a traditional Chinese medicine composition with lung protecting and cough relieving effects, which comprises the following raw material medicine powder or water or organic solvent extracts of the raw material medicines in parts by weight as active ingredients:
10-30 parts of chlamydomonas reinhardtii, 10-40 parts of penthorum chinense pursh, 12-35 parts of dendrobium candidum, 13-26 parts of fructus aurantii, 15-41 parts of platycodon grandiflorum and 20-60 parts of chrysanthemum.
Preferably, the pharmaceutical composition comprises the following medicinal powder of the raw materials in parts by weight or water or organic solvent extracts of the raw materials as active ingredients:
18 parts of chlamydomonas reinhardtii, 20 parts of penthorum chinense pursh, 20 parts of dendrobium candidum, 17 parts of fructus aurantii, 20 parts of platycodon grandiflorum and 35 parts of chrysanthemum.
Preferably, the traditional Chinese medicine composition further comprises pharmaceutically acceptable auxiliary materials; the pharmaceutically acceptable auxiliary materials comprise inert diluents, dispersants and/or granulating agents, surfactants and/or emulsifying agents, disintegrating agents, binders, preservatives, buffers, lubricants and/or oils.
Preferably, the formulation is an oral formulation.
Preferably, the oral preparation is granule, powder, pill or solution.
In a second aspect, the invention also provides a preparation method of the traditional Chinese medicine composition with the lung protecting and cough relieving effects, which comprises the following steps:
step (1), weighing bulk drugs according to the proportion of claim 1;
And (2) grinding the raw material medicaments into powder, or obtaining water or organic solvent extract of the raw material medicaments, and then adding pharmaceutically common auxiliary materials to obtain the traditional Chinese medicine composition with the effects of protecting lung and relieving cough.
In a third aspect, the invention also provides an application of the traditional Chinese medicine composition with the lung-protecting and cough-relieving effects in preparing a lung-protecting and cough-relieving medicine product.
In a fourth aspect, the invention also provides a lung-protecting and cough-relieving product, which comprises the traditional Chinese medicine composition with the lung-protecting and cough-relieving effects.
Preferably, the lung-protecting cough-relieving product is an oral medicine or food.
Preferably, the formulation of the lung-protecting cough-relieving product is paste, tablet, capsule, granule, suspension, powder or pill.

Detailed Description
The invention provides a traditional Chinese medicine composition with the effects of protecting lung and relieving cough, and the active ingredients of the traditional Chinese medicine composition comprise medicinal powder of the following raw materials or water or organic solvent extracts of the raw materials: chlamydomonas reinhardtii, penthorum chinense pursh, dendrobium candidum, fructus aurantii, platycodon grandiflorum and chrysanthemum.
The invention also provides a product for protecting lung and relieving cough, which comprises the traditional Chinese medicine composition.
In the present invention, the "composition" and the "product for protecting lung and relieving cough" are usually compositions, which comprise a composition having an active ingredient component (active ingredient for short) and pharmaceutically acceptable auxiliary materials.
The "pharmaceutically acceptable auxiliary materials" in the present invention include inert diluents, dispersants and/or granulating agents, surfactants and/or emulsifying agents, disintegrants, binders, preservatives, buffers, lubricants and/or lubricating oils.
The product for protecting lung and relieving cough comprising the composition is usually an oral medicine or food, the dosage form of which is paste, tablet, capsule, granule, suspension, powder or pill, and can be prepared into oral medicine or food products such as cream, capsule, tablet candy or beverage by adding at least one of excipient, colorant, coating agent, sweetener, flavoring agent and flavoring agent into the composition.
The compositions in the present invention may be prepared by any method known in the art of pharmacy. Generally, these methods of preparation involve associating the active ingredient with a carrier or excipient and/or one or more other auxiliary ingredients, and then shaping and/or packaging the product into the desired single or multi-dose unit.

The invention provides a lung-protecting cough-relieving paste product, which comprises a traditional Chinese medicine composition with the lung-protecting cough-relieving effect, wherein the active ingredients of the traditional Chinese medicine composition comprise medicinal powder of the following raw materials or water or organic solvent extracts of the raw materials: the preferred parts by weight of the chlamydomonas reinhardtii, penthorum chinense pursh, dendrobium candidum, fructus aurantii, platycodon grandiflorum and chrysanthemum are listed in table 1.

The preparation process of the lung-protecting cough-relieving paste product provided by the invention comprises the following steps:
(1) Weighing: weighing chlamydomonas reinhardtii, penthorum chinense pursh, dendrobium candidum, fructus aurantii, platycodon grandiflorum and chrysanthemum according to a formula;
(2) Cleaning: washing with clear water and then placing into a flask;
(3) Extracting: adding 10 times of water, boiling for 2 hours;
(4) And (3) filtering: filtering with 100 mesh gauze, and keeping filtrate for use;
(5) Extracting: adding water 5 times into the filter residue again, boiling for 2 hours;
(6) And (3) filtering: filtering with 100 mesh gauze, and mixing filtrates;
(7) Concentrating: 70+/-5 ℃ and the vacuum degree is 0.07-0.09 MPa until the paste is concentrated to the paste with the relative density of 1.25-1.40;
(8) Cooling to obtain the final product.
The preferred route of administration of the lung-protecting cough-relieving paste product is the oral route.
In the traditional Chinese medicine composition with the effects of protecting lung and relieving cough, each raw material component serving as an active ingredient has the following expected functions:
Chlamydomonas reinhardtii:
Contains abundant chlorophyll, and has strong lung-heat clearing function. Chlorophyll is known to be the best natural antidote, can neutralize pulmonary toxins and discharge the toxins out of the body, can prevent lung infection and prevent diffusion of lung inflammation, and plays a role in clearing lung heat. Chlamydomonas reinhardtii contains abundant chlamydomonas polysaccharide, and the chlamydomonas polysaccharide has the effect of moistening lung and simultaneously has the effect of anti-inflammatory. Chlamydomonas reinhardtii contains more than 30% of chlamydomonas protein, and the chlamydomonas protein has the effect of improving the immunity of organisms.
Grass of Penthorn:
Penthorn grass, the name of penthorum chinense pursh, is originally carried in Ming Dynasty's "Ding Ben Cao", is a seedling medicament used in the generation of Miao nationality in Gulin regions, is named as "Shen Xian Cao", has slightly warm nature, sweet taste and no toxicity, and has the effects of promoting diuresis and removing jaundice, ping Gan invigorating spleen, inducing diuresis and detumescence, clearing heat and detoxicating, promoting blood circulation and dredging channels, removing blood stasis and relieving pain, and protecting liver.
The main effective components of herba Penthori chinensis comprise flavone, gallic acid, quercetin, sitosterol, pinocembrin-7-0-glucoside, and various antioxidant components. The average content of quercetin in penthorum chinense pursh is 0.45% and the average content of gallic acid is 2.35% as measured by RP-HPLC. In the study of comparing the flavone content of different parts of the penthorum chinense pursh, the flavone content in the root is found to be 3.5 percent, the flavone content in the stem is found to be 3.7 percent, and the flavone content in the leaf is found to be 4.1 percent.
Modern researches show that penthorum chinense pursh can inhibit transforming growth factor-beta (TGF-beta) induced pulmonary fibrosis by regulating Wnt4 in Wnt signal channels, and has the effect of protecting lung. And the active substances of the penthorum chinense pursh, namely quercetin and gallic acid, can enhance the capacity of resisting oxidative damage, thereby reducing the level of inflammatory factors in the lung. It is proved by literature that the decoction of Huang Caoshui has good antibacterial effect, and can destroy the formation of biological membrane by inhibiting the secretion of extracellular polysaccharide and DNA. In conclusion, the penthorum chinense pursh plays a role in protecting the lung from several aspects of clearing heat and detoxicating, inhibiting pulmonary fibrosis, reducing pulmonary inflammation and pulmonary bacterial infection.
Dendrobium officinale:
The dendrobium nobile mainly contains 'two alkali-sugar' and various microelements and amino acids, wherein the hydroxy dendrobine and dendrobii subalkali can regulate the pH value in human body, balance yin and yang, remove the toxin of viscera, form black carbon by various surface viruses and garbage remained on the tracheal wall and bronchial wall, and completely remove the outside body. The content of the contained dendrobium polysaccharide is very high, and the dendrobium polysaccharide has the functions of supplementing nutrition, improving the elasticity of lung fiber cells and improving the antiviral capability of the lung. Dendrobium officinale is also called as a lung scavenger by traditional Chinese medicine. The effect of clearing lung is doubled when the Chinese medicinal composition is combined with Chlamydomonas reinhardtii.
Fructus Aurantii: has effects in tranquilizing, activating qi-flowing, eliminating phlegm, moistening lung, and relieving cough, and fructus Aurantii can relieve pain due to cough.
Radix Platycodi: the traditional Chinese medicine is usually used for treating cough, eliminating phlegm, ventilating lung, expelling pus and the like.
And (3) chrysanthemum: the fragrance is good, and the smell of the composition is blended, so that the composition is better to eat.
In the pharmaceutical composition with the effects of protecting lung and relieving cough, the chlamydomonas reinhardtii and the dendrobium candidum have the effects of clearing lung toxins, relieving lung infection and being combined as monarch drugs, and the two are combined to use and have a synergistic effect; the penthorum chinense pursh can repair lung injury, prevent lung lesions, and has the effect of protecting the lung, and is regarded as ministerial drug; fructus Aurantii and radix Platycodi have effects of activating qi-flowing, eliminating phlegm, dispersing lung qi, relieving cough, and relieving lung pressure, and can be used as adjuvant drug. The medicines are combined together to play the roles of protecting lung and relieving cough.
The present invention will be further illustrated by the following examples, which are not to be construed as limiting the scope of the invention, in order to better understand the essential characteristics of the invention. It is also specifically noted herein that the specific experimental methods and apparatus referred to in the examples are conventional methods or carried out under the conditions recommended by the manufacturer's instructions unless otherwise specified, and that the reagents referred to are commercially available without otherwise specified.
Example 1:
The embodiment provides a traditional Chinese medicine composition with the effects of protecting lung and relieving cough, which is prepared from the following raw materials in parts by weight: 18g of chlamydomonas reinhardtii, 20g of penthorum chinense pursh, 20g of dendrobium candidum, 17g of fructus aurantii, 20g of platycodon grandiflorum and 35g of chrysanthemum.
The embodiment further provides a lung-protecting cough-relieving ointment prepared from the traditional Chinese medicine composition, and the preparation process comprises the following steps:
(1) Weighing: weighing chlamydomonas reinhardtii, penthorum chinense pursh, dendrobium candidum, fructus aurantii, platycodon grandiflorum and chrysanthemum according to a formula;

Example 2:
The embodiment provides a traditional Chinese medicine composition with the effects of protecting lung and relieving cough, which is prepared from the following raw materials in parts by weight: 25g of chlamydomonas reinhardtii, 30g of penthorum chinense pursh, 30g of dendrobium candidum, 23g of fructus aurantii, 30g of platycodon grandiflorum and 45g of chrysanthemum.
The preparation process of the lung-protecting cough-relieving ointment prepared by the traditional Chinese medicine composition is the same as that of the embodiment 1.
Comparative example 1:
Unlike example 1, there was no Chlamydomonas reinhardtii.

unlike example 1, no Dendrobium officinale was included.

unlike example 1, there was no Chlamydomonas reinhardtii and dendrobium candidum.

Efficacy experiment:
(1) Strong ammonia water cough-inducing experiment:
The female and male mice of SPF-class Kunming species are respectively half-90, weighed, 18-22 g in mass, randomly divided into 9 groups of 10 mice each, and marked; the experiment group, codeine phosphate (30 mg/kg) is used as a positive control group, and physiological saline is used as a blank control group; each group of mice is respectively subjected to gastric administration (lung-protecting cough-relieving paste of examples 1-2 and comparative examples 1-5), the mice are continuously fed for 10 days according to the dosage of 0.2mL/10g, normal feeding is carried out, after the last 1 gastric administration for 1h, the mice are placed into a bell jar one by one, an ultrasonic atomizer is used for spraying 25% -28% ammonia water mist into the bell jar for 15s with the maximum spraying amount to induce cough, the mice are moved out of the bell jar, a stopwatch is immediately used for timing, and the cough latency period(s) and the cough times within 3min of the mice are recorded. (note: the mice cough was judged to be preferably contracted in the abdominal muscles of the large mouth, a slight cough was heard; and latency was the time from onset of spraying to onset of cough).
The experimental results are shown in Table 1.

The test results show that the cough relieving effect of example 1 is the best. Compared with a blank control group, the examples 1-2, the comparative examples 1-5 and the codeine phosphate group can prolong the incubation period of the cough of the mice, reduce the times of cough and have statistical difference (P is less than or equal to 0.05). Further, from the latency period and the number of times of cough in 3min of comparative example 3, comparative example 3 is significantly deteriorated in cough relieving effect as compared with example 1, comparative example 1 and comparative example 2, it is seen that chlamydomonas reinhardtii and dendrobium candidum in the traditional Chinese medicine composition of the present invention have a synergistic effect and have a role as a monarch drug. From the comparison of the latency period and the number of times of cough within 3min of comparative example 4 with those of examples 1 and comparative examples 1 to 3, and the latency period and the number of times of cough within 3min of comparative example 5 with those of examples 1 and comparative examples 1 to 3, the status of the penthorum chinense pursh, the fructus aurantii and the platycodon grandiflorum in the traditional Chinese medicine composition of the invention as ministerial drugs and adjuvant drugs for assisting in protecting liver and relieving cough can be illustrated.
(2) Experimental study of expectorant effect:
90 mice each weighing 18-22 g were divided into 9 groups according to sex and body weight, and the above samples (lung-protecting cough-relieving pastes of examples 1-2 and comparative examples 1-5) were respectively filled with stomach, and positive control 0.9% ammonium chloride and blank control group (distilled water of the same volume). After each group was administered by gavage for 30min, 1ml of 0.25% phenol red was injected intraperitoneally. After 30 minutes, the mice were sacrificed by cervical vertebrae removal, the trachea was separated, the trachea was repeatedly washed three times with 5% sodium bicarbonate as a washing solution, the three washing solutions were combined, phenol red content was measured by colorimetry, and statistical data were obtained, and the results are shown in table 2.

The test results show that the phlegm eliminating effect of the example 1 is the best. Compared with a blank control group, the phenol red excretion amount can be improved in each of the examples 1-2, the comparative examples 1-5 and the codeine phosphate group, and the statistical difference is that P is less than or equal to 0.05. The effect of example 1 is close to that of the control group, and the ammonium chloride tablet is an expectorant with higher clinical use frequency and better effect, so that the ammonium chloride tablet is used as a positive control. Further, from the fact that the amount of phenol red excreted in comparative example 3 is significantly deteriorated in the phlegm eliminating effect as compared with examples 1, 1 and 2, it is seen that the chlamydomonas reinhardtii and dendrobium candidum in the Chinese medicinal composition of the present invention have a synergistic effect and have a position as a monarch. The status of penthorum chinense pursh, fructus aurantii and platycodon grandiflorum as ministerial drugs and adjuvant drugs to assist in protecting liver and relieving cough in the traditional Chinese medicine composition can be illustrated by the fact that the phenol red excretion amount of comparative example 4 is compared with examples 1 and 1-3 and the phenol red excretion amount of comparative example 5 is compared with examples 1 and 1-3.
The foregoing is merely a preferred embodiment of the present invention and it should be noted that modifications and adaptations to those skilled in the art may be made without departing from the principles of the present invention, which are intended to be comprehended within the scope of the present invention.

1. The traditional Chinese medicine composition with the effects of protecting lung and relieving cough is characterized by comprising the following raw material medicine powder or water or organic solvent extracts of the raw material medicines in parts by weight as active ingredients:
10-30 parts of chlamydomonas reinhardtii, 10-40 parts of penthorum chinense pursh, 12-35 parts of dendrobium candidum, 13-26 parts of fructus aurantii, 15-41 parts of platycodon grandiflorum and 20-60 parts of chrysanthemum.

The traditional Chinese medicine composition with the effects of protecting lung and relieving cough according to claim 1, which is characterized by comprising the following raw material medicine powder or water or organic solvent extract of each raw material medicine as an active ingredient in parts by weight:
18 parts of chlamydomonas reinhardtii, 20 parts of penthorum chinense pursh, 20 parts of dendrobium candidum, 17 parts of fructus aurantii, 20 parts of platycodon grandiflorum and 35 parts of chrysanthemum.

The traditional Chinese medicine composition with the effects of protecting lung and relieving cough according to claim 1 or 2, wherein the traditional Chinese medicine composition further comprises pharmaceutically acceptable auxiliary materials; the pharmaceutically acceptable auxiliary materials comprise inert diluents, dispersants and/or granulating agents, surfactants and/or emulsifying agents, disintegrating agents, binders, preservatives, buffers, lubricants and/or oils.
4. The Chinese medicinal composition with lung protecting and cough relieving effects according to claim 3, wherein the preparation is an oral preparation.

6. A method for preparing a traditional Chinese medicine composition with lung protecting and cough relieving effects according to any one of claims 1 to 5, which comprises the following steps:
step (1), weighing bulk drugs according to the proportion of claim 1;
And (2) grinding the raw material medicaments into powder, or obtaining water or organic solvent extract of the raw material medicaments, and then adding pharmaceutically common auxiliary materials to obtain the traditional Chinese medicine composition with the effects of protecting lung and relieving cough.
7. Use of a Chinese medicinal composition with lung protecting and cough relieving effects according to any one of claims 1-5 in the preparation of a pharmaceutical product for lung protecting and cough relieving.
8. A lung-protecting and cough-relieving product, characterized by comprising the traditional Chinese medicine composition with the lung-protecting and cough-relieving effects according to any one of claims 1-5.
9. The lung-protecting cough product of claim 8, wherein the lung-protecting cough product is an oral pharmaceutical or food product.
10. The lung-protecting and cough-relieving product according to claim 8, wherein the formulation of the lung-protecting and cough-relieving product is a paste, a tablet, a capsule, a granule, a suspension, a powder or a pill.
