背景技术
T细胞的体外产生和扩增可用于广泛的临床应用,包含癌症治疗。例如,临床数据显示,具有肿瘤抗原特异性受体的经工程化的T细胞在一些患者中对于引起转移性癌症的消退是有用的。不幸的是,并不是所有的患者都受益于这种治疗。一种解释是在体外扩增期间,一些T细胞变得耗尽或衰老,这限制了治疗功效和体内持久性。
发明内容
本公开提供了用于产生具有增强的抗肿瘤表型的T细胞的***和方法。具体地,本公开提供了从用包含T细胞受体(TCR)、嵌合抗原受体(CAR)和至少一种另外的转基因在内的多种转基因工程化的干细胞(例如,HSC)制备T细胞的方法。通过从HSC开始,本发明的***和方法利用干细胞的自我再生和细胞分化能力来制备具有改善的抗肿瘤表型的T细胞。具体地,本公开提供了将核酸引入到编码TCR、CAR和至少一种另外的转基因的CD34+干细胞中。TCR和CAR的组合表达可用于提供具有特异性癌细胞靶向性质的T细胞。此外,通过本发明的方法产生的被赋予至少一种另外的转基因的T细胞装备有货物(例如,细胞因子),当与靶癌细胞接触时,该货物对于治疗癌症是有用的。
干细胞(例如,HSC)的优点包含这些干细胞的再生和扩增的能力。同种异体细胞疗法通常需要数十亿个细胞用于单剂量的治疗。由于干细胞潜在的无限扩增能力,本发明的方法非常适于大规模产生高质量细胞产品并且使那些产品可快速地用于治疗。此外,干细胞的标志是其分化为不同细胞类型的能力。在本公开的上下文中,分化能力提供了可以产生各种T细胞亚型(包含例如自然杀伤T细胞、αβT细胞、γδT细胞等)的细胞制造平台。
在一方面,本发明提供了产生经工程化的恒定型自然杀伤T细胞的方法。该方法涉及将编码T细胞受体(TCR)、嵌合抗原受体(CAR)和至少一种另外的转基因的一种或多种核酸引入到造血干细胞(HSC)中;以及将该HSC转化为表达该TCR和该CAR并且包括该转基因的恒定型自然杀伤T(iNKT)细胞。在优选的实施例中,该一种或多种核酸是由单一载体如慢病毒载体提供的。然而,在一些情况下,例如,当整合大的转基因时,使用至少两种不同的核酸分子,如多个载体(例如,慢病毒载体),整合该一种或多种核酸可能是有用的。该至少两种不同的核酸分子可以依次或同时被引入到这些HSC中。
在优选的实施例中,本发明的方法涉及通过慢病毒转导将核酸引入到HSC中。该一种或多种核酸的引入提供了表达至少一种TCR、CAR和另外的转基因的HSC。该另外的转基因的并入可用于提供具有改善的功能性质(如改善的细胞扩增、持久性、安全性和/或抗肿瘤活性)的治疗性T细胞。该一种或多种另外的转基因可以包含以下中的任何一种或多种:细胞因子、检查点抑制剂、转化生长因子β信号传导的抑制剂、细胞因子释放综合征的抑制剂或神经毒性的抑制剂。例如,可以提供编码以下中的一种或多种的转基因:IL-2、IL-7、IL-15、IL-12、IL-18或IL-21。
因此,在一些情况下,本发明的方法提供了通过例如引入编码IL-2、IL-7、IL-1-15中的一种或多种的核酸来制备具有改善的扩增和持久性能力的CAR T细胞。在一些情况下,方法可以通过例如引入编码IL-12、IL-18中的一种或多种的转基因来为CAR T细胞提供增加的IFN-g产生并且因此改善的T细胞效力。在其它情况下,本发明的方法可用于通过引入包含IL-21在内的转基因来增强原初T细胞产生。在一些情况下,本文所述的方法通过例如引入IL-6、GM-CSF的抑制剂或细胞因子释放综合征和神经毒性的其它介质来提供具有改善的安全性的CAR T细胞的产生。方法可以通过提供可用于对抗肿瘤微环境的有效载荷(例如,通过TGF-B的抑制剂、检查点)来提供具有改善的功效的CAR T细胞。

具体实施方式
嵌合抗原受体(CAR)T细胞的临床研究在治疗某些病理(如B细胞恶性肿瘤)中示出了显著效果。然而,目前,涉及CAR T细胞疗法的商业方法涉及自体CAR T细胞,这些细胞的广泛使用受到物流和与特定世代相关的高成本的限制。同种异体CAR T细胞疗法通过提供立即可用于患者治疗的预制细胞库解决了自体细胞的限制。然而,尽管具有潜力,但是尚未建立用于持续产生治疗有效的同种异体CAR T细胞的方法。例如,大多数方案需要长时间的离体培养,具有至少两个激活步骤,这潜在地导致过度分化、T细胞耗尽和/或细胞衰老,从而破坏体内功效。参见,Jafarzadeh,2020,在过继性癌症免疫疗法中嵌合抗原受体(CAR)T细胞的延长的持久性:挑战和前进方式(Prolonged Persistence of Chimeric AntigenReceptor(CAR)T Cell in Adoptive Cancer Immunotherapy:Challenges and WaysForward),《免疫学前沿(Frontiers in Immunology)》,11(702):1-17,该文献通过引用并入。
本公开提供了用于制备具有改善的表型和细胞功能的T细胞的可靠方法。根据一些方面,本公开提供了使用缩短的离体培养时间进行T细胞产生的方法。本发明的某些方法通过省略离体激活来减少离体培养,这将离体制备过程减少了至多2至3周。本发明的其它方法提供了单个激活步骤。本发明的方法认识到在施用于受试者之后,在体内可能发生漫长的激活和/或扩增过程。通过缩短离体细胞培养,本发明的方法使在T细胞产生期间发生转录和/或表型变化的机会最小化。此外,缩短离体培养时间减少进行T细胞维持所需的昂贵的细胞培养消耗品的量。
在不同方面,本公开提供了用于使用单个激活步骤制备T细胞产品的多步骤工作流程。该方法涉及进行包括将HSC体外分化和成熟为T细胞的过程,该过程具有不超过一个体外T细胞激活步骤。该T细胞产品可以用于治疗或研究。常规的T细胞制备方法需要至少两个单独的体外T细胞激活步骤。多个激活步骤通常涉及多种培养基类型,即,含有不同激活因子的培养基。有利地,通过单个激活步骤产生T细胞减少细胞培养的时间,并且产生表达与T细胞耗尽相关的更少标志物的更有效的T细胞产品。
本发明的方法可用于使用较少体外细胞培养产生T细胞产品。通过减少体外细胞培养,本发明的方法产生更有效的T细胞产品并且含有更少的耗尽/功能失调的T细胞。相较于通过2个或更多个T细胞激活步骤产生的T细胞,该T细胞产品可以表达更低水平的涉及T细胞耗尽的蛋白质,例如PD-1、CTLA-4、LAG-3、TIM-3、2B4/CD244/SLAMF4、CD160、TIGIT。这些T细胞产品还可以表达更高水平的IL-2。
某些实施例涉及用于产生T细胞的多步骤过程,其中这些步骤中仅一个步骤是T细胞激活。单个激活步骤可以通过在激活培养基(例如,包括T细胞激活试剂如抗体的培养基)中培养T细胞来进行。
在一些情况下,激活步骤涉及基于外周血单核细胞(PMBC)的激活。基于PMBC的激活可以涉及将T细胞引入到负载α半乳糖基神经酰胺(aGC)的PBMC、可溶性抗CD3/28阳性PBMC和可溶性抗CD2/3/28阳性PBMC。在一些情况下,该激活步骤涉及基于抗原呈递细胞(aAPC)的T细胞激活步骤。因此,激活步骤可以涉及将T细胞引入到aAPC。优选地,aAPC被辐射。该aAPC可以是表达CD80-CD83-CD137L-CAR-抗原的经工程化的K562细胞。该aAPC可以是aAPC+CD1d和/或aAPC+CD1d+/-aGC。在其它情况下,该激活步骤包括基于无饲养层的T细胞激活步骤。基于无饲养层的T细胞激活步骤可以涉及将包含抗CD3、抗CD28、抗CD2/3/28、CD3/28在内的可溶性抗体引入到T细胞。在一些实施例中,该方法涉及培养基,该培养基包括以下中的一种或多种:IL-7/15、IL-2、IL-2+21、IL-12或IL-18。在一些实施例中,培养基含有IL-15。
此外,根据其它方面,本发明的方法可用于产生具有增强的抗肿瘤活性的T细胞。本公开提供了用于从与包含TCR、CAR和至少一种另外的转基因在内的多种转基因结合的干细胞(例如,HSC)产生T细胞的***和方法。通过启动从干细胞的产生过程,本发明的***和方法利用自我更新和细胞分化能力来制备具有“更年轻的”表型和增强的抗肿瘤活性的T细胞。具体地,本公开提供了将核酸引入到CD34阳性干细胞中,这些阳性干细胞编码至少一种TCR、CAR和至少一种另外的转基因。TCR和CAR以及另外的转基因的组合表达可用于提供具有可用于治疗癌症的特异性癌细胞靶向性质的T细胞。
图1图示了用于产生T细胞的方法101。具体地,所展示的是提供根据本发明的各方面的用于产生T细胞的方法的一般概述的简单流程图。方法101包含:获得105干细胞(例如,CD34+造血干细胞/祖细胞);将一种或多种核酸(例如,编码TCR、CAR和另外的转基因)引入109到干细胞中;进行111干细胞的体外分化和成熟以产生T细胞;以及在不进行T细胞激活步骤的情况下提供113(例如,用于同种异体疗法或研究)细胞。
方法101涉及获得105干细胞。优选地,细胞是CD34+细胞。在一个非限制性实例中,CD34+干细胞是造血干细胞/祖细胞。造血干细胞或祖细胞是在被称为造血作用的过程中产生其它血细胞的干细胞。
造血干细胞/祖细胞可以从健康供体获得。造血干细胞/祖细胞可以从例如骨髓、外周血、羊水或脐带血获得。造血干细胞/祖细胞可以通过夹持脐带的末端并且用针从夹持的末端之间抽吸血液来从脐带血获得。可以使用阳性免疫磁性分离技术从脐带血分离造血干细胞/祖细胞,并且可以将柠檬酸盐-磷酸盐-右旋糖(CPD)作为抗凝血剂添加到脐带血中。可以将来自脐带血的细胞冷冻保存并且储存在例如-80摄氏度的温度下直到使用。
在实践本公开的方法中,获得105干细胞优选地涉及接收一小瓶经冷冻保存的CD34+脐带血细胞,包含造血干细胞/祖细胞。可以从例如在干冰上的隔热容器中的细胞库接收一小瓶经冷冻保存的脐带血细胞。
可以根据本领域中已知的方法将一小瓶经冷冻保存的脐带血细胞解冻。例如,可以通过将小瓶置于37度的水浴中持续大约1至2分钟来解冻一小瓶细胞。在一些优选的实施例中,一旦细胞解冻,就将细胞铺板到预先包被有促进病毒与靶细胞共定位以增强转导效率的试剂的组织培养皿上。
可以将CD34+细胞以例如每孔10,000个细胞、每孔15,000个细胞或每孔20,000个细胞或每孔25,000个细胞或更多铺板在标准6孔培养皿中。优选地,将细胞以每孔15,000个细胞铺板。
方法101进一步包含将编码TCR、CAR和/或另外的转基因中的一者或多者的一种或多种核酸引入109到CD34+干细胞中。优选地,该一种或多种核酸编码TCR、CAR或另外的转基因中的至少两者。例如,在一些实施例中,引入109到干细胞中的该一种或多种核酸至少编码TCR和CAR,以产生能够靶向在癌细胞的表面上表达的特异性蛋白的T细胞。在其它实施例中,引入109到干细胞中的该一种或多种核酸编码TCR、CAR和另外的转基因中的每一者。
在优选的实施例中,通过核酸引入的TCR是iNKT TCR。iNKT TCR可以包含iNKT细胞受体的α链、iNKT细胞受体的β链中的一者或两者。优选地,iNKT细胞受体由干细胞表达,使得干细胞识别α-半乳糖基神经酰胺。另外,优选的实施例包含引入编码至少一种CAR的核酸。如下文所讨论的,CAR可以是第一代、第二代或第三代CAR。CAR可用于为细胞提供对与癌症相关的抗原具有特异性的受体。例如,在一些实施例中,抗原包括间皮素、磷脂酰肌醇蛋白聚糖3、CD19或BCMA中的一种。通过本发明的方法产生的T细胞可以被遗传修饰以表达至少一种另外的转基因。转基因可以是例如以下中的一种:细胞因子、检查点抑制剂、转化生长因子β信号传导的抑制剂、细胞因子释放综合征的抑制剂或神经毒性的抑制剂。因此,本发明的方法可以用于产生具有增强的效应子功能的T细胞。
例如,在一些情况下,本发明的方法可用于制备具有改善的扩增和持久性能力的CAR T细胞,这是通过引入编码IL-2、IL-7、IL-1-15中的一种或多种的转基因来提供的。在一些情况下,方法可以通过例如引入编码IL-12、IL-18中的一种或多种的转基因来为CAR T细胞提供增加的IFN-g产生并且因此改善的T细胞效力。在一些情况下,本发明的方法可用于通过引入包含IL-21在内的转基因来增强原初T细胞产生。在一些情况下,本文所述的方法通过例如引入IL-6、GM-CSF的抑制剂或细胞因子释放综合征和神经毒性的其它介质来提供具有改善的安全性的CAR T细胞的产生。方法可以通过提供可用于对抗肿瘤微环境的有效载荷(例如,通过TGF-B的抑制剂、检查点)来提供具有改善的功效的CAR T细胞。
将该一种或多种核酸引入109到干细胞中可以通过病毒转导方法或非病毒转染来实现。在一些情况下,例如,用于引入该一种或多种核酸的方法涉及非病毒方法,例如使用睡美人转座子(Sleeping Beauty transposon)/转座酶***。睡美人转座子***涉及被设计成将精确定义的DNA序列引入到细胞的染色体中的合成DNA转座子。该***使用Tc1/mariner型***,其中转座酶从多个无活性鱼类序列复活。有利地,非小瓶方法可以提供节约成本的益处并且降低与使用某些病毒相关的风险。然而,非病毒方法可能与降低的效率相关。因此,优选的实施例通过病毒转导(例如,通过逆转录病毒)将核酸引入到干细胞中。
病毒转导方法因其多功能性而得到公认,并且涉及可用于用显著量的核酸转导***细胞和非***细胞两者的慢病毒载体的使用。慢病毒载体的使用被认为是安全的并且通常提供长期转基因表达。因此,方法101优选地通过慢病毒载体将该一种或多种核酸引入109到34+干细胞中。关于干细胞的慢病毒转导的讨论,参见Jang,2020,优化用于基因疗法的造血干细胞的慢病毒载体转导(Optimizing lentiviral vector transduction ofhematopoietic stem cells for gene therapy),《基因疗法(Gene Therapy)》(27):545-556,该文献通过引用并入。
本发明的方法不受用于将核酸引入到干细胞中的任何一种过程或实验室程序的限制。在一些情况下,使用单个慢病毒载体。单个慢病毒载体可以编码TCR、CAR和另外的转基因中的每一者。在其它情况下,使用至少两种不同的慢病毒载体,其中该至少两种慢病毒载体中的每一种慢病毒载体编码TCR、CAR和另外的转基因中的至少一者,使得在转导时,TCR、CAR和另外的转基因中的每一者被转导到干细胞中。此外,在使用多于一种慢病毒载体的情况下,方法101不受引入两种(或更多种)慢病毒载体干细胞的时间序列的限制。这些载体可以在同一转导中同时引入或依次引入。
方法101进一步涉及进行111a过程,该过程包括将干细胞(例如,HSC)体外分化和成熟为T细胞。
细胞制备方法101的一个优点在于从单一起始材料(即,干细胞)产生各种T细胞亚型的能力。通过本发明的方法产生的T细胞可以包含例如辅助T细胞、细胞毒性T细胞、记忆T细胞、调节T细胞、自然杀伤T细胞、恒定型自然杀伤T细胞、αβT细胞、γδT细胞。在优选的实施例中,方法101涉及产生恒定型自然杀伤T细胞。恒定型自然杀伤T(iNKT)细胞的产生对于其同种异体细胞疗法应用是优选的。具体地,其激活和扩增抗原特异性T细胞应答以治疗癌症而不诱导移植物抗宿主病的能力。
因此,方法101包含进行111干细胞(例如,HSC)向T细胞(例如,iNKT)的体外分化和成熟过程。如下文详细地讨论的,优选地进行111干细胞的体外分化和成熟,条件是该过程不涉及T细胞的激活和扩增的后续体外步骤。

在一些实施例中,CD4阳性CD8阳性T细胞在冷冻保存和/或施用于同种异体细胞疗法接受者之前在体外扩增。CD4阳性CD8阳性T细胞的扩增可以涉及在存在IL-7、IL-15、CD3、CD28、CD2、α-半乳糖基神经酰胺中的一者或多者的情况下培养细胞。
本发明的方法利用体内激活机制来减少体外培养步骤。一旦在没有经历两个激活步骤的情况下产生T细胞并且将其引入到受试者体内,当T细胞在次级淋巴器官遇到适当激活的抗原呈递细胞(APC)(如树突状细胞)时,T细胞被完全激活。如果APC通过主要组织相容性复合物(MHC)II类分子显示适当的肽配体,则APC被TCR识别。这对于激活T细胞非常重要。还可能需要由APC递送的两种其它刺激信号。这些信号可以由APC表面的两种不同配体(如CD80和CD86)提供给T细胞上的表面分子,例如,CD28。对于激活重要的其它因子包含参与引导T细胞分化为效应子T细胞的不同亚群的那些因子,例如,细胞因子,如IL-6、IL-12和TGF-β。经激活的T细胞的CD28依赖性共刺激可以导致经激活的T细胞自身产生IL-2。IL-2表达后,除了如ICOS和CD40L等其它调节分子之外,还可以上调IL-2受体的第三组分(称为α链),也称为CD25。IL-2与其高亲和力受体的结合促进细胞生长,而APC(主要是树突状细胞)产生各种细胞因子或表达诱导CD4+T淋巴细胞分化为产生细胞因子的效应子细胞的表面蛋白,这取决于环境条件。
图2展示了通过三种不同过程进行离体T细胞制备的阶段。具体地,展示了与本发明的简化离体过程205、207相比用于制备T细胞的常规离体过程203。在常规过程203中,使成熟的T细胞经受至少两轮体外激活步骤,其中两轮被展示。此过程203可能需要至少35天的离体细胞培养完成,例如,至少42天,并且通常更长时间。相反地,过程205省略T细胞激活,因此可以在少至21天内产生T细胞。通过省略激活步骤,T细胞的离体培养得到显著减少,例如,2至3周。第三过程207涉及单个激活步骤。单个激活步骤可能有助于产生有效量的T细胞。通过使用仅一个激活步骤而不是两个激活步骤,本发明的方法可以在比现有技术的T细胞制备过程少至少14天内产生T细胞。
图3提供了用于体外产生T细胞的两种培养过程的比较。第一过程303包含两个激活步骤。通过用例如包括CD3/CD28/CD2和IL-15的不同的激活培养基处理双阳性T细胞来实现的两步骤激活过程使制备过程增加了至少一周并且通常更久。第二过程305省略体外激活。
本发明的方法可以涉及单个激活步骤。单个激活步骤可以涉及用激活试剂培养T细胞,持续不超过7天的时段。在其它实施例中,单个激活步骤涉及在激活培养基中培养T细胞,持续不超过6天或5天或4天或3天或2天或1天。在其它实施例中,单个激活步骤包括不在激活培养基中培养T细胞,持续超过8天或9天或10天或11天或12天或13天或14天。单个激活步骤可以涉及用单一类型的激活培养基培养T细胞。单一类型的激活培养基可以包含激活试剂,如可溶性抗体。单个激活步骤可以涉及将T细胞与抗原递呈细胞(如aAPC)共培养。单个激活步骤可以涉及将T细胞与PBMC共培养。
本发明的方法涉及从造血干细胞/祖细胞产生T细胞。造血干细胞/祖细胞通常与可以在脐带血中发现的CD34+细胞相关。在一些情况下,细胞可以源自祖细胞。在一些情况下,细胞是多能干细胞,如胚胎干细胞。
由HSC产生的同种异体CAR T细胞可以为某些病理提供治愈性治疗方法。然而,一些限制包含GVHD,一种供体T细胞介导的同种异体反应过程,其负责与同种异体细胞疗法相关的许多发病率和死亡率。一些临床研究显示,供体iNKT细胞可以在不增加疾病复发风险的情况下预防GVHD。供体CAR iNKT细胞的过继性转移以及随后的体内激活和/或扩增可以预防或减轻GVHD的症状。这种保护作用可以通过同种异体反应性T细胞的Th2极化和供体调节性T细胞(Treg)的扩增来介导。由于如通过本发明的方法产生的同种异体iNKT细胞不会引起GVHD,因此本文所述的方法为‘现成的’CAR免疫疗法提供理想的平台。
本发明的方法可用于制备通过TCR的抗原识别区的功能性重排获得抗原特异性的治疗活性T细胞。TCR是在T细胞(或T淋巴细胞)的表面上发现的分子,该分子负责识别与主要组织相容性复合物分子结合的抗原。TCR可以由至少两条不同的蛋白质链(例如,异二聚体)构成。在大多数(例如,95%)T细胞中,TCR由α链和β链组成,而在一些(例如,5%)T细胞中,TCR由γ链和δ链组成。此类T细胞可以在细胞表面中具有抗原特异性。TCR分子在体内分化为不同的表型亚群,包含但不限于经典CD3阳性、α-βTCR CD4阳性、CD3阴性α-βTCR CD8阳性、γδT细胞、自然杀伤T细胞等。此外,T细胞可以进一步包含各种激活状态,包含但不限于原初、中枢记忆、效应子记忆、末端效应等。
在优选的实施例中,本发明的方法提供了CAR T细胞的制备。CAR T细胞是被遗传工程化以产生用于免疫疗法中使用的人工T细胞受体的T细胞。CAR(即,嵌合抗原受体)可以用于经由通过例如逆转录病毒载体促进其编码序列的转移将单克隆抗体的特异性移植到具有TCR的干细胞上。CAR是被工程化以向T细胞赋予靶向特异性蛋白质的新能力的受体蛋白。受体是嵌合的,因为这些受体将抗原结合功能和T细胞激活功能两者组合到单个受体中。因此,本发明的方法通过产生经修饰的T细胞提供用于免疫疗法的产品,这些经修饰的T细胞识别癌细胞以便更有效地靶向并破坏这些癌细胞。

CAR通常包含胞外结构域、跨膜结构域和胞内结构域。胞外结构域可以包含抗原结合/识别区/结构域。CAR的抗原结合结构域可用于与特异性抗原(例如,肿瘤抗原)、病原体抗原(例如,病毒抗原)、CD(分化簇)抗原结合。胞外结构域还可以包含指导新生蛋白进入内质网中的信号肽。如果CAR将被糖基化并锚定在细胞膜中,则信号肽可能是必不可少的。跨膜结构域是跨越膜的疏水性α螺旋。不同的跨膜结构域产生不同的受体稳定性。在抗原识别之后,受体聚集,并且信号传递到细胞。最常用的胞内组分是含有3个ITAM的CD3Xi。这在结合抗原之后将激活信号传递到T细胞。CAR还可以包含将抗原结合结构域连接到跨膜结构域的间隔子区。间隔子区应当是足够柔性的,以允许抗原结合结构域在不同方向上定向,从而促进抗原识别。间隔子可以是来自IgG1的铰链区或免疫球蛋白的CH2CH3区以及CD3的各部分。

将一种或多种转基因引入到CAR T细胞中可以改善性质(如T细胞扩增和持久性(例如,使用IL-2、IL-7/15)、IFN-g产生和T细胞效力(例如,用IL-12、IL-18)),增强原初亚群(例如,IL-21),改善安全性(例如,通过IL-6的抑制剂、GM-CSF或CRS和神经毒性的其它介质)或通过对抗肿瘤微环境(TGF-B、检查点等)来改善功效。
例如,IL-12和IL-18在增强CAR T细胞的某些效应子功能中起主要作用。已知IL-12激活某些NK细胞和T淋巴细胞,诱导Th-1型应答,并且增加IFN-γ分泌。IL-12的诱导型表达可以增强CAR T细胞对抗某些病理(如淋巴瘤、肝细胞癌、卵巢肿瘤和B16黑色素瘤)的抗肿瘤能力。IL-18还已用于改善CAR T细胞的治疗潜力。最初鉴定为IFN-γ的有效诱导剂,IL-18可能有助于T和NK细胞的激活以及Th-1细胞极化。例如,可以向中间靶向的CAR T细胞提供编码IL-18的转基因,以增强IFN-γ的分泌并根除癌细胞。关于进一步讨论,参见Tian,2020,下一代CAR T细胞对抗实体癌的基因修饰策略(Gene modification strategies fornext-generation CAR T cells against solid cancers),《血液学与肿瘤学杂志(Journal of Hematology&Oncology),第13(5)卷,该文献通过引用并入。
IL-7、IL-15和IL-21可用于促进干细胞样记忆T细胞表型的产生。此表型可以提供T细胞在体内的增加的扩增和持久性。在一些情况下,可以提供编码IL-2的转基因。用IL-2产生T细胞可以通过例如促进参与营养感应和摄取的蛋白质的诱导和产生来为T细胞提供改善的对肿瘤环境有应答的能力。
在一些实施例中,转基因是细胞因子释放综合征的抑制剂。细胞因子释放综合征涉及通常与CAR T细胞疗法相关的严重的、可能危及生命的副作用。细胞因子释放综合征表现为由过度炎性细胞因子释放(包含例如IFN-γ和IL-6)驱动的快速(超)免疫反应。已知涉及细胞因子释放综合征的许多细胞因子通过JAK-STAT通路起作用。因此,在一些实施例中,本发明的方法涉及产生表达JAK通路的抑制剂的CAR-T细胞,以改善体内CAR T细胞增殖、抗肿瘤活性和细胞因子水平。例如,可以提供抑制IL-6、JAK-STAT或BTK的功能的转基因。此外,抑制剂可以进一步用于抑制神经毒性。CAR T细胞相关的神经毒性是一种通常导致严重神经紊乱(如癫痫和昏迷)的综合征。
在其它情况下,方法涉及将转基因引入到HSC中,其中该转基因是检查点抑制剂,例如,免疫检查点抑制剂。免疫检查点是免疫***的某些方面的调节剂。在正常的生理条件下,检查点使免疫***能够对宿主抗原做出反应,从而保护健康组织。在癌症中,这些分子促进肿瘤细胞逃避。在一些情况下,转基因可以编码抗体或抗体片段,如抗组织蛋白酶抗体、半乳凝素-1阻断和抗OX40激动性抗体。抗体可以在细胞表面上分泌或表达。可以分泌例如靶向PD1或PDL1的抗体。
在实施例中,产生表达转化生长因子β的抑制剂的CAR T细胞。经工程化的细胞面临限制其功效的不利的微环境。调节环境可以转化用于促进CAR T细胞增殖、存活和/或杀死癌细胞的能力。肿瘤环境内的主要抑制机制之一是转化生长因子β。因此,本发明的一些方面涉及引入编码转化生长因子β的抑制剂的转基因。抑制剂可以是例如抗体或其片段。抗体或抗体片段可以从CAR T细胞分泌以干扰转化生长因子β的正常功能。
在一些实施例中,方法包含使CAR T细胞通过肿瘤微环境的标志物靶向实体瘤类型。在其它实施例中,方法制备具有基于单结构域抗体(VHH)的嵌合抗原受体的CAR T细胞,其可以用于识别肿瘤微环境的标志物而不需要肿瘤特异性靶标。根据本发明,基于VHH的CAR T细胞可以通过免疫检查点受体或通过基质和细胞外基质标志物靶向肿瘤微环境,其在同基因免疫活性动物模型中有效对抗实体瘤。因此,本发明的方法可用于制备靶向可能缺乏肿瘤特异性抗原表达的肿瘤的CAR T细胞。仅重链抗体(VHH或纳米抗体)的可变区是具有与传统短链可变片段(scFv)相当的亲和力的小的、稳定的、骆驼源性单结构域抗体片段。VHH的免疫原性通常低于scFv的免疫原性,并且由于其较小的尺寸,可以获得与scFv所见到的表位不同的表位。因此,如本发明提供的,VHH可以用作CAR T细胞中的合适的抗原识别结构域。与scFv不同,VHH不需要V区配对所附带的另外的折叠和组装步骤。VHH允许表面显示,而不需要大量的接头优化或其它类型的重新格式化。能够切换出各种基于VHH的识别结构域产生高度模块化平台,无需将每种新的常规抗体重新格式化为scFv即可获得。
此外,许多微环境涉及如PD-L1等抑制分子的表达。使用VHH作为识别结构域,例如,PD-L1特异性CAR T细胞,通过本发明的方法产生的CAR T细胞可以靶向肿瘤微环境。PD-L1在肿瘤细胞以及浸润性髓样细胞和淋巴细胞上广泛表达。识别PD-L1的CAR应当减轻免疫抑制,并且同时允许肿瘤微环境中的CAR T细胞激活。因此,PD-L1靶向的CAR T细胞可以对肿瘤微环境进行重新编程,从而抑制免疫抑制信号并促进炎症。例如,如在Xie,2019,靶向肿瘤微环境的基于纳米抗体的CAR T细胞抑制免疫活性小鼠中的实体瘤的生长(Nanobody-based CAR T cells that target the tumor microenvironment inhibit the growthof solid tumors in immunocompetent mice),《美国国家科学院院刊(PNAS)》2019年4月16日116(16)7624-7631中讨论的,该文献通过引用并入。

本公开的一些方面涉及在HSC中引入和表达多种转基因。为了促进多种基因的表达,在具有2A序列(即,2A肽的编码结构域)的核酸上分离转基因可能是有用的。2A自切割肽或2A肽是一类18至22个aa长肽,其可以在蛋白质翻译期间诱导核糖体跳跃。在细胞内,当2A肽的编码结构域被***在两种蛋白质(例如,TCR和CAR)的两个编码结构域之间时,该肽将由于核糖体跳跃而被翻译成独立折叠的两种蛋白质。

对于使用本发明的抗原特异性T细胞的过继性免疫疗法,可以输注在10,000,000至10,000,000,000的范围内的细胞剂量。在将T细胞施用到受试者体内时,T细胞可能经历抗原依赖性激活过程。

本发明的方法可用于产生安全且有效的同种异体疗法。在一些情况下,方法可以涉及在制造期间在一个或多个点处表征细胞产品以确保产品质量。在一些实施例中,本发明的方法涉及分析T细胞以鉴定由这些T细胞表达的一种或多种蛋白质。该一种或多种蛋白质可以包含一种或多种CCR7、CD62L或CD45RA。蛋白质可以包含与原初干细胞相关的标志物。分析优选地包含分析细胞表面蛋白的高通量方法,例如,基于大量单独细胞的荧光信号的方法,如FACS。
根据需要,治疗的可用性是同种异体细胞疗法的一个益处。由于方法可以涉及在临床应用之前制备细胞,因此一些优选的方法可以包含冷冻保存T细胞。冷冻保存T细胞可用于安全且有效地储存细胞,直到患者需要这些细胞。冷冻保存还可用于将细胞产品运输至临床设施,在这些临床设施中这些细胞可以施用于患者。在一些优选的实施例中,将双阳性T细胞(例如,原初T细胞)在不进行体外激活步骤的情况下冷冻保存。
在过去十年里,免疫疗法已经成为新一代癌症药物。具体地,基于细胞的细胞疗法已经显示出巨大的前景。突出的实例是经CAR工程化的过继性T细胞疗法,该疗法以令人印象深刻的功效靶向某些血癌。然而,大多数目前的治疗方案由自体过继性细胞转移组成,其中从患者收集的免疫细胞被制备并且用于治疗此单个患者。此类方法是昂贵的、制造劳动力密集的,并且难以广泛地递送给所有有需要的患者。通过本文所述的方法,同种异体免疫细胞产品可以大规模制造,并且可以容易地分发以治疗更高数量的患者,因此有很大的需求。
一些实施例涉及经工程化的iNKT细胞或经工程化的iNKT细胞群体。在至少一些情况下,经工程化的iNKT细胞包括CAR和/或经工程化的T细胞受体。在细胞的上下文中讨论的任何实施例都可以应用于此类细胞群体。在特定实施例中,经工程化的iNKT细胞包括核酸,该核酸包括以下中的1者、2者和/或3者:i)恒定型αT细胞受体编码序列的全部或部分;ii)恒定型βT细胞受体编码序列的全部或部分;或iii)***基因。在另外的实施例中,存在一种经工程化的iNKT细胞,该经工程化的iNKT细胞包括核酸,该核酸具有编码以下的序列:i)恒定型αT细胞受体的全部或部分;ii)恒定型βT细胞受体的全部或部分;和/或iii)***基因产物。
另外的方面涉及具有增加的NK激活受体水平、降低的NK抑制受体水平和/或增加的细胞毒性分子水平的经工程化的iNKT细胞。在一些实施例中,NK激活受体包括NKG2D和/或DNAM-1。在一些实施例中,细胞毒性分子包括穿孔素和/或颗粒酶B。在一些实施例中,抑制剂受体包括KIR。增加或降低可能与从健康个体分离的非工程化的iNKT中的相同标志物的水平有关。另外的方面涉及经工程化的iNKT细胞群体,其中该细胞群体具有增加的NK激活受体水平、降低的NK抑制受体水平和/或增加的细胞毒性分子水平。
在一些实施例中,经工程化的iNKT细胞包括在异源启动子的控制下的核酸,这意味着启动子不是控制核酸转录的同一基因组启动子。设想的是,经工程化的iNKT细胞包括外源性核酸,该外源性核酸包括一个或多个编码序列,在本文所述的许多实施例中,这些编码序列中的一些或所有编码序列在异源启动子的控制下。

在另外的实施例中,细胞缺乏或具有减少的至少一种HLA-I或HLA-II分子的表面表达。在一些实施例中,HLA-I和/或HLA-P分子的表面表达的缺乏是通过以下方式实现的:破坏编码单独HLA-I/II分子的基因;破坏编码B2M(β2微球蛋白)的基因,该B2M是所有HLA-I复合物分子的共同组分;或破坏编码CIITA(II类主要组织相容性复合物反式激活因子)的基因,该CIITA是控制所有HLA-II基因的表达的关键转录因子。在具体实施例中,细胞缺乏一种或多种HLA-I和/或HLA-II分子的表面表达,或者该细胞的此类分子的表达水平降低了(或至少降低了)50%、60%、70%、80%、90%、100%(或其中可得出的任何范围)。在一些实施例中,HLA-I或HLA-II在iNKT细胞中不表达,因为该细胞是通过基因编辑操纵的。

术语“细胞”在本文中以其在本领域中最广泛的意义使用,并且是指活体,该活体是多细胞生物体的组织的结构单元,被将其与外界隔离的膜结构包围,具有自我复制的能力,并且具有遗传信息和用于表达该遗传信息的机制。本文所使用的细胞可以是天然存在的细胞或人工修饰的细胞(例如,融合细胞、经遗传修饰的细胞等)。
iNKT细胞是从小鼠到人高度保守的αβT淋巴细胞的小群体。iNKT细胞被认为在调节许多疾病中起重要作用,包含癌症、感染、过敏和自身免疫。当受到刺激时,作为细胞群体和在单细胞水平下两者,iNKT细胞快速释放大量的效应子细胞因子,例如,像IFN-γ和IL-4。然后,这些细胞因子通过经激活的DC激活各种免疫效应子细胞,如先天性免疫***的自然杀伤细胞和树突状细胞(DC),以及适应性免疫***的CD4辅助细胞和CD8细胞毒性常规αβT细胞。由于其独特的激活机制,iNKT细胞可以攻击多种疾病而不受抗原和MHC限制,从而使其成为有吸引力的通用治疗剂。
先前,已经进行了一系列主要靶向癌症的基于iNKT细胞的临床试验。最近的试验报告显示,患有头颈部鳞状细胞癌的患者的抗肿瘤免疫得到增强,证明了基于iNKT细胞的免疫疗法的潜力。然而,迄今为止,大多数临床试验产生了不令人满意的结果,这是由于这些临床试验是基于内源性iNKT细胞的直接激活或离体扩增,由此仅对少数患者产生短期、有限的临床益处。在人类中iNKT细胞的低频率和高变异性(血液中约0.01%至1%)以及这些细胞在激活后的快速耗尽被认为是限制这些试验成功的主要障碍。
iNKT细胞已经从诱导多能干(iPS)细胞工程化。参见美国专利第8,945,922号,该美国专利通过引用并入。iPS细胞是通过用外源性核重编程因子Oct4、Sox2、Klf4和c-Myc等转导体细胞产生的。不幸的是,由于外源性核重编程因子的转录水平随着细胞向多能状态的转变而降低,因此稳定的iPS细胞系产生的效率可能有所下降。另外,由于Oct4、Sox2、Klf4和c-Myc是导致瘤形成的癌基因,因此外源性核重编程因子的转录可以在iPS细胞中恢复并且引起源自iPS细胞的细胞的瘤发育。
本发明的方法可以用于产生iNKT细胞,例如,如在美国公开号US20170283481A1中和在世界申请第2019241400号中讨论的,这些文献中的每个文献通过引用并入。
举例来说,在一些实施例中,本发明的方法产生iNKT细胞,其中iNKT TCR核酸序列从iNKT细胞的亚群(如CD4/DN/CD8亚群或产生Th1、Th2或Th17细胞因子的亚群)获得,并且包含双阴性iNKT细胞。在一些实施例中,iNKT-TCR核酸序列从来自曾患有或患有癌症(如黑色素瘤、肾癌、肺癌、***癌、乳腺癌、淋巴瘤、白血病、恶性血液肿瘤等)的供体的iNKT细胞获得。在一些实施例中,iNKT TCR核酸分子具有来自一个iNKT细胞的TCRα序列和来自不同iNKT细胞的TCRβ序列。在一些实施例中,从其获得TCRα序列的iNKT细胞和从其获得TCRβ序列的iNKT细胞来自同一供体。在一些实施例中,从其获得TCRα序列的iNKT细胞的供体不同于从其获得TCRβ序列的iNKT细胞的供体。在一些实施例中,TCRα序列和/或TCRβ序列被密码子优化用于表达。在一些实施例中,TCRα序列和/或TCRβ序列被修饰以编码与由未经修饰的序列编码的多肽相比具有一个或多个氨基酸取代、缺失和/或截短的多肽。在一些实施例中,iNKT TCR核酸分子编码识别在CD1d上呈递的α-半乳糖基神经酰胺(α-GalCer)的T细胞受体。在一些实施例中,iNKT TCR核酸分子包含在表达载体中。在一些实施例中,表达载体是慢病毒表达载体。在一些实施例中,表达载体是iNKT TCR核酸分子替代MIG载体的IRES-EGFP区段的MIG载体。在一些实施例中,表达载体是phiNKT-EGFP。

在一些方法中,细胞在人工胸腺类器官(ATO)***中培养。ATO***涉及三维(3D)细胞聚集体,其是细胞的聚集体。在某些实施例中,3D细胞聚集体包括表达Notch配体的所选基质细胞群体。在一些实施例中,通过在物理基质或支架上将CD34+转导的细胞与所选基质细胞群体混合来产生3D细胞聚集体。在另外的实施例中,方法包括对CD34+转导的细胞和基质细胞进行离心,以形成置于物理基质或支架上的细胞沉淀。在某些实施例中,基质细胞表达Notch配体,该配体是完整的、部分的或经修饰的DLL1、DLL4、JAG1、JAG2或其组合。在另外的实施例中,Notch配体是人Notch配体。在其它实施例中,Notch配体是人DLL1。

在某些方面,本公开提供了用于产生具有增强的抗肿瘤表型的T细胞的***和方法。具体地,本公开提供了从用包含T细胞受体(TCR)、嵌合抗原受体(CAR)和至少一种另外的转基因在内的多种转基因工程化的干细胞(例如,HSC)制备T细胞的方法。通过从HSC开始,本发明的***和方法利用干细胞的自我再生和细胞分化能力来制备具有改善的抗肿瘤表型的T细胞。具体地,本公开提供了将核酸引入到编码TCR、CAR和至少一种另外的转基因的CD34+干细胞中。TCR和CAR的组合表达可用于提供具有特异性癌细胞靶向性质的T细胞。此外,通过本发明的方法产生的被赋予至少一种另外的转基因的T细胞装备有货物(例如,细胞因子),当与靶癌细胞接触时,该货物对于治疗癌症是有用的。
例如,在优选的实施例中,本发明的方法涉及通过慢病毒转导将核酸引入到HSC中。该一种或多种核酸的引入提供了表达至少一种TCR、CAR和另外的转基因的HSC。该另外的转基因的并入可用于提供具有改善的功能性质(如改善的细胞扩增、持久性、安全性和/或抗肿瘤活性)的治疗性T细胞。
该一种或多种另外的转基因可以包含以下中的任何一种或多种:细胞因子、检查点抑制剂、转化生长因子β信号传导的抑制剂、细胞因子释放综合征的抑制剂或神经毒性的抑制剂。例如,可以提供编码以下中的一种或多种的转基因:IL-2、IL-7、IL-15、IL-12、IL-18或IL-21。
因此,在一些情况下,本发明的方法提供了通过例如引入编码IL-2、IL-7、IL-1-15中的一种或多种的核酸来制备具有改善的扩增和持久性能力的CAR T细胞。在一些情况下,方法可以通过例如引入编码IL-12、IL-18中的一种或多种的转基因来为CAR T细胞提供增加的IFN-g产生并且因此改善的T细胞效力。在其它情况下,本发明的方法可用于通过引入包含IL-21在内的转基因来增强原初T细胞产生。在一些情况下,本文所述的方法通过例如引入IL-6、GM-CSF的抑制剂或细胞因子释放综合征和神经毒性的其它介质来提供具有改善的安全性的CAR T细胞的产生。方法可以通过提供可用于对抗肿瘤微环境的有效载荷(例如,通过TGF-B的抑制剂、检查点)来提供具有改善的功效的CAR T细胞。
本发明的某些方法涉及单个体外激活步骤。在一些情况下,用试剂短暂激活T细胞,例如,持续1至3天,并且在此之后,通常从培养基中去除激活试剂,以便不连续地刺激细胞并且因此耗尽细胞。激活后,经激活的T细胞群体可以快速扩增,例如,每24小时数量翻倍。可以添加一些试剂来促进扩增。在一些实施例中,该方法涉及培养基可以补充有以下中的一种或多种:IL-7/15、IL-2、IL-2+21、IL-12或IL-18。
在一些情况下,单个激活步骤涉及基于PMBC的T细胞激活。因此,该激活步骤可以涉及将负载aGC的PBMC、可溶性抗CD3/28阳性PBMC和可溶性抗CD2/3/28阳性PBMC引入到T细胞。在一些情况下,该激活步骤涉及基于抗原呈递细胞(aAPC)的T细胞激活步骤。因此,激活步骤可以涉及将T细胞引入到aAPC。该aAPC可以是表达CD80-CD83-CD137L-CAR-抗原的经工程化的K562细胞。该aAPC可以是aAPC+CD1d和/或aAPC+CD1d+/-aGC。在其它情况下,该激活步骤包括基于无饲养层的T细胞激活步骤。基于无饲养层的T细胞激活步骤可以涉及将包含抗CD3、抗CD28、抗CD2/3/28、CD3/28在内的可溶性抗体引入到T细胞。在一些实施例中,T细胞激活步骤涉及在存在添加到激活扩增培养基IL-7/15、IL-2、IL-2+21、IL-12、IL-18的不同细胞因子的情况下培养T细胞。
以下实例提供了如由本发明的方法提供的从CD34+细胞制备T细胞(例如,iNKT)的有用的示例性方案。关于进一步的实例和讨论,参见WO2019241400A1,该文献通过引用并入。
实例1:CD34+HSPC细胞培养和慢病毒转导
第-2天:预刺激
1.用在PBS中稀释的0.5毫升/孔的20ug/ml纤维连接蛋白(RN)包被适当数量的24孔非组织培养处理的板的孔(建议接种约15x 103个细胞/孔或6个孔,对于0.1x 106等分试样)。可以将1mg/mL的RN储备等分试样以60微升等分试样储存在-20℃下。
2.在室温(RT)下温育2小时(h)。
3.抽吸RN并用0.5毫升/孔的在PBS中稀释的2% BSA替代。30% BSA等分试样储存在-20℃下。
4.在RT下温育30分钟。
5.抽吸并用0.5毫升/孔的PBS替代。
6.按照标准程序将0.1x106等分试样的CD34+细胞解冻到干细胞培养基中,并且以300g旋转10分钟。
7.抽吸上清液,重悬于干细胞培养基中,并且使用血细胞计数器进行计数,记录细胞计数。在一些情况下,申请人发现0.1x 106等分试样的细胞计数太低而不可靠。
8.用干细胞培养基将CD34+细胞稀释至0.05x 106个细胞/毫升。
9.从RN包被的孔中抽吸PBS,并接种300ul细胞/孔(约15x 103个细胞/孔)。
10.在37℃、5% CO2下温育12至18h。
第-1天:转导
1.将浓缩的慢病毒载体(LVV)解冻,并轻轻移液以混合(不涡旋并且不再冷冻)。
2.制备转导管:
a.通过计算足以达到MOI为100至200的体积,将适当体积的LVV转移到1.5ml管。
b.将适当体积的PGE2添加到同一管,以使总培养体积的最终浓度达到10nM。

第0天收获
1.通过轻轻移液以将细胞从板中去除并转移到锥形管来收集细胞。
2.将孔用等体积的冷X-VIVO-15洗涤,以去除仍粘附到板的任何细胞并转移到锥形管。
3.在显微镜下检查并根据需要用冷X-VIVO-15进行另外的洗涤,以从板中收集所有细胞。
4.以300g旋转10分钟并抽吸上清液。
5.继续进行至阶段2(即,实例2)。
实例2:iNKT-CAR细胞的产生;分化
第0至14天:分化(持续时间:2周)
1.以1毫升/孔用淋巴分化涂层材料(LDCM)(如在PBS中以1:200稀释的由STEMCELL以商品名StemSpan提供的淋巴分化材料)包被适当数量的12孔非组织培养处理的板的孔(建议接种1,000至2,000个细胞/孔或12孔/15,000个细胞)。在4℃下温育12至18h。
2.抽吸LDCM并添加2毫升/孔的PBS。
3.将在阶段1(即,实例1)中收集的经转导的CD34+细胞重悬于淋巴祖细胞扩增培养基(LPEM)(如由STEMCELL以商标名StemSpan出售的淋巴祖细胞扩增培养基)中。
4.用LPEM将细胞密度调整至1-2x 103个细胞/毫升。
5.从LDCM包被的板中抽吸PBS。
6.将0.75ml细胞/孔接种到LCDM包被的板中。
7.将细胞在37℃、5% CO2下温育。
8.在第3天,添加0.25毫升/孔的新鲜LPEM并继续培养。
9.在第7天和第11天,在不干扰细胞的情况下小心地去除<0.5毫升/孔并补充0.5毫升/孔的新鲜LPEM。
10.继续培养至第14天。

实例3:iNKT-CAR细胞的产生;成熟
第14-21+天:成熟(持续时间:1至2周)
1.用在PBS中以1:200稀释的2毫升/孔的LDCM包被适当数量的6孔非组织培养物处理的板的孔(建议接种50-100x 103/孔)。如果在接种前一天制备,则在4℃下温育12至18h,或者如果在接种当天制备,则在37℃下温育2h。
2.抽吸LDCM并添加4毫升/孔的PBS。
3.在第14天,使用细胞活力计数器(如由贝克曼库尔特公司(Beckman Coulter)以商标名Vi-Cell XR提供的细胞活力计数器)收获细胞并对细胞进行计数。
a.通过轻轻移液以将细胞从板中去除并转移到锥形管来收集细胞。
b.将孔用1毫升/4孔的冷SFEMII洗涤,以去除仍粘附到板的任何细胞并转移到锥形管。

7.将细胞以2.5-5x 104个细胞/毫升重悬于T细胞祖细胞成熟培养基(TPMM)(如由STEMCELL以商标名StemSpan提供的T细胞祖成熟培养基)中。
8.从LDCM包被的板中抽吸PBS。
9.将2ml细胞/孔接种到LDCM包被的板中。
10.将细胞在37℃、5% CO2下温育。
11.将剩余细胞以300g沉淀10分钟。
12.抽吸剩余细胞并将剩余细胞重悬于适当体积的冷冻保存溶液(如由STEMCELL以商标名CryoStor CS10出售的冷冻保存溶液)中,以达到2-5x 106个细胞/毫升。
13.将1毫升/冷冻小瓶等分试样并且适当地冷冻(例如,在负80℃的冰箱中);在冷冻24小时内移动至液氮储存。
14.在第17天,添加2毫升/孔的新鲜TPMM并继续培养。
15.在第21天,通过从孔中心中的细胞移液100ul,取出样品进行流式细胞术,并且接种到96孔V形底板中进行流式染色。
16.从孔中心中的细胞取另外100ul并用细胞活力计数器进行计数。
17.如果TCR表达>50%,CD4/CD8双阳性表达>20%,并且细胞尺寸已经减小(指示从HSC发育为T细胞),则细胞可以冷冻保存并提供用于同种异体疗法,或者任选地如由阶段4提供的那样扩增。如果不满足这些参数,则在必要的饲养和***下继续进行培养。去除<2毫升/孔并且每3至4天补充2毫升/孔的新鲜TPMM,并且如果其接近汇合则***培养物。在第24/25天和第28天再次进行此检查,直到满足上述标准。
阶段4:iNKT-CAR细胞的产生;任选的扩增:
第21至28天(假定在第21天满足标准,相应地调整):刺激(持续时间:1周)
1.在第21天,收获细胞并使用细胞活力计数器对细胞进行计数(在相关excel表中记录计数)。
a.通过轻轻移液以将细胞从板中去除并转移到锥形管来收集细胞。
b.将孔用2毫升/4孔的冷细胞扩增培养基(EM)(如由赛默飞世尔公司(ThermoFisher)以商标名OpTmizer提供的细胞扩增培养基)洗涤,以去除仍粘附到板的任何细胞并转移到锥形管。

e.提供CD3/CD28/CD2 T细胞激活剂,如由干细胞技术有限公司以商标名ImmunoCult Human CD3/CD28/CD2 T细胞激活剂出售的激活剂。
i.将细胞用具有10ng/ml的IL-7和IL-15的EM稀释至每ml 1x 106个细胞。
ii.将CD3/CD28/CD2 T细胞激活剂以25ul/ml添加到细胞悬浮液。
iii.接种细胞并在37℃、5% CO2下温育。
f.负载aGC的PBMC
i.将人外周血单核细胞(PBMC)解冻。
ii.以每ml 10x 106个细胞重悬于具有2ug/ml aGC的EM中,并在37℃、5% CO2下温育1h。
iii.收集负载aGC的PBMC并在6,000rads下辐射。
iv.用EM对细胞进行至少两次洗涤以去除未结合的aGC。
v.将PBMC重悬于具有10ng/ml的IL-7和IL-15的EM中。
vi.将iNKT-CAR细胞与PBMC以1:2-3(iNKT-CAR:PBMC)的比率组合,使得iNKT-CAR细胞最终浓度为每ml 1x 106个细胞。
vii.接种细胞并在37℃、5% CO2下温育。
g.K562-CD80-CD83-CD137L-A2ESO人工抗原呈递细胞(aAPC)。
i.从培养物中收集aAPC并以10,000rads辐射。
ii.将iNKT-CAR细胞与aAPC以4:1(iNKT-CAR:aAPC)的比率组合,使得iNKT-CAR细胞最终浓度为每ml 1x 106个细胞。
iii.接种细胞并在37℃、5% CO2下温育。
h.K562-CD80-CD83-CD137L-A2ESO-CD1d人工抗原呈递细胞(aAPC-CD1d)。
i.从培养物中收集aAPC并以10,000rads辐射。
ii.将iNKT-CAR细胞与aAPC以4:1(iNKT-CAR:aAPC)的比率组合,使得iNKT-CAR细胞最终浓度为每ml 1x 106个细胞。
iii.接种细胞并在37℃、5% CO2下温育。
i.负载aGC的K562-CD80-CD83-CD137L-A2ESO-CD1d人工抗原呈递细胞(aAPC-CD1d)。
i.从培养物中收集aAPC。
ii.以每ml 10x 106个细胞重悬于具有2ug/ml aGC(参见α-GalCer Prep SOP)的EM中,并在37℃、5% CO2下温育1h。
iii.收集负载aGC的aAPC并在10,000rads下辐射。
iv.将iNKT-CAR细胞与aAPC以4:1(iNKT-CAR:aAPC)的比率组合,使得iNKT-CAR细胞最终浓度为每ml 1x 106个细胞。
v.接种细胞并在37℃、5% CO2下温育。
3.在第24天,使用细胞计数器对细胞进行计数(在相关excel表中记录计数),并且使用具有10ng/ml的IL-7和IL-15的EM将细胞稀释至每ml 1x 106个细胞,根据需要转移培养容器。如果利用PBMC或aAPC,在此时间点进行的细胞计数可能更难以解释,在这种情况下,在不干扰细胞的情况下小心地去除一半培养基并且添加等体积的具有10ng/ml的IL-7和IL-15的新鲜EM。如果没有PBMC或aAPC的条件不需要稀释,也进行此半培养基更换。
4.在第26天,使用细胞活力计数器对细胞进行计数(在相关excel表中记录计数),并且使用具有10ng/ml的IL-7和IL-15的EM将细胞稀释至每ml 1x 106个细胞,根据需要转移培养容器。
5.在第28天,收获细胞并使用Vi-Cell进行计数(在相关excel表中记录计数)。
a.取出0.2x 106个细胞的等分试样并接种到96孔V形底板中进行流式染色。
b.取出0.2x 106个细胞的等分试样并接种到96孔V形底板中进行另外的流式染色表征。
c.取出1x 106个细胞的等分试样,使用具有10ng/ml的IL-7和IL-15的EM稀释至每ml 1x 106个细胞,并接种进行纵向追踪(任选步骤,在细胞冷冻完成之后优先考虑)。
d.将剩余细胞以300g沉淀10分钟并抽吸上清液。
e.重悬于适当体积的冷冻保存培养基中,以达到每mL 25-50x 106个细胞。
f.将1毫升/冷冻小瓶等分试样并且适当地冷冻,在冷冻24小时内移动至液氮储存。

Claims (15)
1.一种产生经工程化的恒定型自然杀伤T细胞的方法,所述方法包括:
将编码T细胞受体(TCR)、嵌合抗原受体(CAR)和至少一种另外的转基因的一种或多种核酸引入到造血干细胞(HSC)中;以及
将所述HSC转化为表达所述TCR和所述CAR并且包括所述转基因的恒定型自然杀伤T(iNKT)细胞。
2.根据权利要求1所述的方法,其中所述一种或多种核酸是由单一载体提供的。
3.根据权利要求2所述的方法,其中所述单一载体包括慢病毒载体。
4.根据权利要求1所述的方法,其中引入所述一种或多种核酸涉及通过多个载体将至少两种不同的核酸分子并入到所述HSC中。
5.根据权利要求4所述的方法,其中所述至少两种不同的核酸分子包括慢病毒载体。
6.根据权利要求4所述的方法,其中将所述至少两种不同的核酸分子依次或同时引入到所述HSC中。
7.根据权利要求1所述的方法,其中所述HSC源自祖细胞。
8.根据权利要求7所述的方法,其中所述祖细胞是多能干细胞。
9.根据权利要求1所述的方法,其中所述一种或多种核酸进一步编码诱导核糖体跳跃的序列。
10.根据权利要求9所述的方法,其中所述序列编码2A序列。
11.根据权利要求1所述的方法,其中所述iNKT细胞是α/βiNKT细胞或γ/δiNKT细胞。
12.根据权利要求1所述的方法,其中所述一种或多种另外的转基因包括以下中的至少一种:细胞因子、检查点抑制剂、转化生长因子β信号传导的抑制剂、细胞因子释放综合征的抑制剂或神经毒性的抑制剂。
13.根据权利要求12所述的方法,其中所述细胞因子包括以下中的一种:IL-2、IL-7、IL-15、IL-12、IL-18或IL-21。
14.根据权利要求1所述的方法,其中所述CAR包括单结构域抗体。
15.根据权利要求1所述的方法,其中所述CAR包括重链抗体的可变区。
