Abstract
本发明属于载体递送领域,具体涉及具有靶向性的载体及其靶向宿主细胞的方法,其包含:与靶细胞的内吞受体结合的第一分子和促进靶向载体搭载的物质释放进入细胞质的第二分子;当靶向载体为病毒载体时,所述第一分子不是病毒包膜蛋白的一部分。本方案利用内吞与内体/溶酶体逃逸相结合的方式,对载体进行改造,使其带有能与靶细胞的内吞受体结合的第一分子,得到靶向载体。根据所要靶向的细胞的内吞受体设计第一分子,可以靶向不同种类的细胞,应该范围广泛;对载体进行合理的突变之后,可以避免感染不需要靶向的细胞,精确性大大提高。

技术领域
本发明属于载体递送领域,具体涉及具有靶向性的载体及其靶向宿主细胞的方法。
背景技术
目前,药物递送主要通过两种方式:(1)病毒载体,如腺病毒、腺相关病毒(AAV)、逆转录病毒和慢病毒等,和(2)非病毒载体,如脂质体纳米粒(LNP)、外泌体、类病毒颗粒(VLP)、抗体偶联药物(ADC)等。其中,腺病毒、腺相关病毒、类病毒颗粒、抗体偶联药物为无脂质体包膜的递送载体,逆转录病毒、慢病毒、纳米脂质体、外泌体等为有脂质体包膜的递送载体。有包膜的病毒载体通过包膜蛋白,靶向识别细胞膜上的特定受体蛋白,如水疱性口炎病毒包膜糖蛋白(VSVG)可以靶向识别低密度脂肪酸受体(LDL-R),狒狒内源性逆转录病毒(BaEV)包膜糖蛋白可以靶向识别ASCT1和ASCT2受体,从而介导病毒载体进入到表达上述受体的靶细胞中。
目前基于慢病毒骨架和VSVG包膜蛋白的载体已经广泛应用于临床治疗,如在嵌合抗原受体T细胞疗法(CAR-T)中,通过携带VSVG的慢病毒包裹CAR分子感染T细胞制备CAR-T。
慢病毒载体是一类改造自人免疫缺陷病毒(HIV)的病毒载体,是逆转录病毒的一种,基因组是RNA,其毒性基因已经被剔除并被外源性目的基因所取代,属于假型病毒。可利用逆转录酶将外源基因整合到基因组中实现稳定表达,具有感染***期与非***期细胞的特性。原始HIV病毒携带gp120和gp41复合物,可以识别CD4分子,从而促进的HIV对细胞的感染。经改造的慢病毒载体,不表达gp120和gp41,而表达VSVG包膜蛋白。由于VSVG可以靶向识别LDL-R,且LDL-R表达广泛,从而使得携带VSVG的慢病毒可以广泛感染T细胞、肝细胞、心肌细胞、神经元、内皮细胞、干细胞,以及多种肿瘤细胞等多种类型的细胞。
慢病毒载体优势为:(1)表达时间长:慢病毒可将外源基因整合到宿主细胞基因组上,实现基因长时间稳定的表达,不随细胞***传代而丢失,是细胞实验的首选,常用于构建稳转株;(2)安全性高:未发现致病性,慢病毒载体改造T细胞用于CAR-T细胞治疗;(3)免疫原性低:直接注射活体组织不易造成免疫反应,适用于动物实验。其缺点是,由于LDL-R在许多类型的广谱表达,因此靶向性不强;且难以感染低表达LDL-R的细胞,如静息态的造血干细胞、静息态T细胞、NK细胞和B细胞等。
目前,已有报道开发靶向慢病毒。比如:1、基于CD3/CD28抗体的靶向慢病毒,通过在慢病毒表面表达带有跨膜序列的CD3和CD28抗体或其它可以激活T细胞的抗体,可以促使病毒课题靶向T细胞,同时促使T激活,激活后的T细胞会高表达LDL-R,使得可以与VSVG结合从而促使病毒颗粒进入T细胞中。该方法可以有效促使慢病毒载体在体内感染T细胞,虽然由于CD3抗体的存在可以使得该病毒具有一定的靶向性,然而由于VSVG的存在,使得其仍然可以感染其它类型的细胞,其靶向性仍然有带提高。2、基于融合素(Fusogen)的靶向病毒。与VSVG和BaEV等同时结合受体识别和包膜融合的包膜蛋白不同,麻疹病毒(MV)和尼帕病毒(NiV)包膜糖蛋白为H/G蛋白和F蛋白,其中H/G蛋白负责受体的识别,在H识别受体后,F蛋白(Fusogen)发生构想改变介导病毒包膜和细胞膜的融合。通过将H/G蛋白与抗体相连,如CD4或者CD8抗体相连,别使抗体递送到胞外,便可介导病毒特异地识别CD4+或者CD8+的T细胞。由于在H/G蛋白上增加抗体会增加H/G蛋白的空间位阻,使得变构后的F蛋白可能会够不着靶细胞的细胞膜,从而难以介导病毒与包膜的融合。
除慢病毒外,AAV载体被广泛应用于递送基因治疗药物。腺相关病毒是一种单链DNA病毒,目前的科学界共识是它不会导致任何人类疾病。作为基因疗法载体的重组腺相关病毒(rAAV)携带的蛋白衣壳与野生型AAV几乎完全相同,然而衣壳内的基因组中编码病毒蛋白的部分完全被删除,取而代之的是治疗性转基因。AAV基因组中唯一被保留的部分是ITRs,它起到指导基因组的复制和病毒载体组装的作用。目前,AAV的临床试验大部分集中在4大器官/组织上:眼睛、肝脏、肌肉和中枢神经***。靶向肝脏的静脉基因治疗显示可以解决代谢和血液疾病。帕金森病、血友病A和血友病B是临床试验最多的3个适应症。
AAV载体的优势为:(1)非致病性,载体原自非致病的病毒;(2)颗粒小、已穿透组织;(3)容易制备、其包装滴度极高。同时虽然不同血清型的AAV载体具有不同的感染偏好性,但其靶向性依旧不足。
其它载体,包括LNP和ADC药物递送体系也已经广泛应用于临床。
LNP包裹的信使核糖核酸(mRNA)被用于制备新冠疫苗,并在肿瘤疫苗领域开展了大量的研究。由于脂质双分子层不具备靶向性,因此难以靶向感染特定细胞。通过LNP连接CD5抗体,可以将表达CAR-T分子的mRNA,靶向递送至T细胞,从而制备CAR-T。但是由于LNP天然容易被树突状细胞(DC)和巨噬细胞等吞噬,且容易在肝脏中富集,因此即使带上抗体,其靶向性依旧不强。
ADC药物将细胞特定内吞受体的抗体与等药物通过铰链(linker)连接,linker在血液中稳定,因此在静脉输注时药物不会释放,而当抗体与细胞特定内吞受体结合后,被溶酶体包裹,此时linker被溶酶体中的酶降解,从而使得药物释放出来。药物不会被溶酶体降解,无需依赖溶酶体逃逸即可发挥作用。ADC药物具有很强的靶向性,但是基于现有的ADC递送方式,难以向细胞递送除小分子化合物毒素外的其它药物,特别是生物大分子药物,这是由于蛋白和核酸等生物大分子容易被溶酶体降解。
综上所述,现有递送载体,普遍难以实现对特定细胞的靶向递送,特别是将生物大分子靶向递送到特定细胞中,并发挥功能。
发明内容
本发明的第一方面,提供一种靶向载体,其包含:与靶细胞的内吞受体结合的第一分子和促进靶向载体搭载的物质释放进入细胞质的第二分子;当靶向载体为病毒载体时,所述第一分子不是病毒包膜蛋白的一部分。
靶向载体有多种形式,可以是有包膜的载体或无包膜的载体,有包膜的载体如逆转录病毒载体、慢病毒载体、脂质纳米颗粒;无包膜的载体如腺病毒、腺相关病毒、类病毒颗粒。
内吞受体,是指在细胞表面表达的,在与抗体、配体或特定物质结合后,可以诱发内吞作用(endocytosis,或称为胞饮作用)的膜蛋白,如CD7、CD5、HER2、Mesothelin等,通常内吞受体含有YXXPhi、[D/E]XXXL[L/I]、FXNPXY等结构域;此外一些膜蛋白,如CD8分子,由于不具备内吞受体结构域,不具备内吞的能力。因而在内吞结构域的筛选中,常用胞内序列尾端嵌合随机序列的CD8分子进行筛选。
不同的细胞表达不同的、特定的内吞受体,第一分子可以根据所要靶向的细胞的内吞受体进行设计,通过与内吞受体特异性结合的抗体或者配体(第一分子的一部分),使得载体被靶细胞内吞,从而使载体针对特定的细胞具有感染能力,而对其它细胞没有感染能力。
第一分子与内吞受体结合后,诱发受体介导的内吞作用。受体介导的内吞作用是细胞依靠细胞表面的受体特异性地摄取细胞外蛋白或其他化合物的过程。细胞表面的受体具有高度特异性,与相应配体结合形成复合物,继而此部分质膜凹陷形成有被小窝,小窝与质膜脱离形成有被小泡,将细胞外物质摄入细胞内。有被小泡进入细胞后,脱去外衣,与胞内体的小囊泡结合形成大的内体或内体。
第二分子的作用是促使靶向载体被靶细胞内吞之后发生内体逃逸(endosomalescape)或者溶酶体逃逸(lysosomal escape)以进入细胞质。如果靶向载体被内吞后,不能发生内体逃逸或者溶酶体逃逸,则会最终会被溶酶体降解。因此有效的提送载体负载的物质进入细胞质,需要载体具备内体逃逸或者溶酶体逃逸的能力。
常见的内体/溶酶体逃逸的方式主要包括:(1)破坏内体/溶酶体,如聚乙烯亚胺,其通过“质子海绵”效应使内体/溶酶体膨胀而破裂,进入是所搭载物质逃逸并进入细胞质;(2)利用载体表面正电荷与带负电的溶酶体膜相互作用降低溶酶体膜的稳定性;(3)通过载体与内体/溶酶体发生膜融合,实现逃逸,如在载体表达进行融肽GALA修饰,或者载体带有具有膜融合能力的病毒包膜蛋白,如VSVG或其变体。
在一个具体的方案中,第二分子为病毒包膜蛋白和/或非病毒包膜蛋白。
在一个具体的方案中,所述病毒包膜蛋白选自水疱性口炎病毒包膜糖蛋白VSVG及其变体、狒狒内源性逆转录病毒包膜糖蛋白BaEV及其变体、猫内源性逆转录病毒包膜糖蛋白RD114及其变体、长臂猿白血病病毒包膜糖蛋白GALV及其变体中的至少一种;所述非病毒包膜蛋白选自腺相关病毒AAV的VP1及其变体、腺相关病毒AAV的VP2及其变体、GALA多肽及其变体、聚乙烯亚胺中的至少一种。
病毒包膜蛋白,如VSVG,可以在内体/溶酶体中促使病毒包膜与内吞体/溶酶体膜融合,从而促使载体搭载的物质释放。非病毒包膜蛋白,如AAV的VP1和VP2可以在酸性条件发生构象改变,并在内体/溶酶体上形成穿孔,从而使载体搭载的物质释放;聚乙烯亚胺可以在内体/溶酶体中发挥质子海绵效应,即当溶酶体内的pH下降时,PEI能够大量捕获质子,并引起氯离子的内流,导致内体/溶酶体内渗透压升高,最后内体/溶酶体破裂从而促使载体搭载的物质释放。
变体是指与非突变体(野生型)的氨基酸序列具有至少75%同一性的突变体,“至少75%同一性”指与非突变体(野生型)的氨基酸序列的70%、71%、72%、73%、74%、75%、76%、77%、78%、79%、80%、81%、82%、83%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%或100%相同。
由于VSVG的受体LDL-R广泛表达于激活后的T细胞、肝细胞、心肌细胞、内皮细胞、干细胞等多种细胞,以及多种肿瘤细胞,因此携带VSVG及其变体的假型慢病毒可以感染多种类型的细胞。虽然基于VSVG的假型病毒具备广谱感染能力,但是许多细胞不表达或者低表达VSVG的受体LDL-R,如NK细胞、静息态的T细胞等,此时基于VSVG的假型病毒难以进行感染;此外,由于LDL-R广泛表达在多种细胞中,因此基于VSVG的假型病毒的靶向性不足,比如有多种细胞都表达LDL-R,慢病毒难以精准只转染其中的一种。
本方案利用慢病毒的内体/溶酶体逃逸能力,对其进行改造,在慢病毒上连接第一分子,得到的靶向载体,可以灵活应用于针对不同细胞的不同场景,根据所要靶向的细胞设计第一分子,适用范围大大拓宽,而且靶向的精确性也得以提高。
在一个具体的方案中,所述第二分子选自VSVG及其变体、GALA多肽及其变体。
在一个具体的方案中,第一分子包含跨膜肽段、与靶细胞的内吞受体结合的抗体或配体,在一些方案中,第一分子还包含胞外铰链区。膜表达的蛋白一般需要铰链区,有利于膜蛋白的伸展,常用的是CD8铰链区。
在不影响其功能的情况下,并不限定第一分子的氨基酸序列,比如第一分子包括CD33抗体时,可以是与CD33抗体的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白。
不同靶细胞的内吞受体有所不同,常见的内吞受体如:HER2、CD20、CD19、CD79A、CD79B、CD56、CD22、CD138、CD37、CD98、CD309、CD33、CD163、CD163B、CD5、CD7、CD169、CD204、CD205、CD209、CD280、CD302、TROP-2、CD19、NECTIN4、5T4、CD30 TROP2、FRα、STEAP1、ENPP3、GCC、SLC44A4、NaPi2b、CA9、SC-16、CD142、P-Cadherin、PSMA、ED-B、endothelin receptorsETB、TN-C、Collagen IV、Periostin、CEACAM、c-MET、TDGF1、IGF1R、Mesothelin、TIM1、NCAM1、ZIP6、CD166、GPNMB、SDC1、glycosphingolipid、TfR、Ganglioside、CD74、CLDN18、DPEP3、SLITRK6、PRL-R、LY75、CD48、MUC1、CDKs、B7-H4、STING、KAAG1、CD70、CDH3、LRRC15、EGFR、ASGPR。
在一个具体的方案中,所述靶向载体为慢病毒载体;所述慢病毒载体表达的第一分子为跨膜蛋白,所述慢病毒载体表达的第二分子为病毒包膜蛋白,病毒包膜蛋白具备促进内体逃逸或溶酶体逃逸的能力。在靶细胞不表达病毒包膜蛋白受体的时候,可以通过第一分子介导的内吞,实现病毒对靶细胞的感染。
在一个具体的方案中,所述跨膜蛋白选自CD7抗体、CD79B抗体、CD33抗体、ASGRP抗体或配体、Mesothelin抗体、HER2抗体;所述跨膜蛋白还可以是与CD7抗体的CD8信号肽、TH-69重链(VH)、GS连接肽(linker)、TH-69轻链(VL)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白;所述跨膜蛋白还可以是与CD33抗体的CD8信号肽、Gemtuzumab轻链(VL)、GS连接肽(linker)、Gemtuzumab重链(VH)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白;所述跨膜蛋白还可以是与CD79B抗体的CD8信号肽、SN-8重链(VH)、GS连接肽(linker)、SN-8轻链(VL)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白;所述跨膜蛋白还可以是与ASGRP抗体的CD8信号肽、ASGPR轻链B11(VL)、GS连接肽(linker)、ASGPR重链(VH)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白;所述跨膜蛋白还可以是与HER2抗体的CD8信号肽、Pertuzumab轻链(VL)、GS连接肽(linker)、Pertuzumab重链(VH)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白;所述跨膜蛋白还可以是与MESOTHELIN抗体的CD8信号肽、PE38重链(VH)、GS连接肽(linker)、PE38轻链(VL)、CD8铰链区、CD8跨膜区中至少之一的氨基酸序列具有至少约75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%相同的蛋白。
在一个具体的方案中,病毒包膜蛋白发生突变,使其受体识别能力减弱,比如对LDL-R结合能力减弱或消失,使其仅保留内体逃逸或溶酶体逃逸的能力,从而完全依赖第一分子发挥靶向作用。通过携带该突变的包膜蛋白,进一步提升载体的靶向能力,使之只能感染特定的靶细胞。
在一个具体的方案中,第一分子包含抗体或配体,抗体与靶细胞上对应的抗原(内吞受体)结合后,会发生内吞;第二分子采用突变的VSVG,破坏VSVG与LDL-R的结合能力,但同时不影响VSVG的溶酶体逃逸能力,突变为***、缺失或替换,比如,水疱性口炎病毒Indian毒株包膜糖蛋白VSVG包含以下位点突变的一种或多种组合:H8突变、N9突变、Q10突变、K47突变、K50突变、A51突变、S183突变、S179突变、N180突变、I182突变、M184突变、Y209突变、I347突变、T350突变、T352突变、E353突变、R354突变、第1-18位氨基酸缺失、第19-36位氨基酸缺失、第37-51位氨基酸缺失、第314-384位氨基酸缺失、第321-374位氨基酸缺失、第331-364位氨基酸缺失、第344-354位氨基酸缺失、第345-353位氨基酸缺失。
进一步地,VSVG包含以下位点突变的一种或多种组合:H8的替换、N9的替换、Q10的替换、K47的替换、K47的缺失、K50的替换、A51的替换、S183的替换、S179的替换、N180的替换、I182的替换、M184的替换、Y209的替换、I347的替换、T350的替换、T352的替换、E353的替换、R354的替换、R354的缺失、第1-18位氨基酸缺失、第19-36位氨基酸缺失、第37-51位氨基酸缺失、第314-384位氨基酸缺失、第321-374位氨基酸缺失、第331-364位氨基酸缺失、第344-354位氨基酸缺失、第345-353位氨基酸缺失。
进一步地,VSVG的氨基酸序列的K47和/或R354发生突变,比如,VSVG的第47位氨基酸由赖氨酸K替换为谷氨酰胺Q,和/或第R354氨基酸由精氨酸R替换为谷氨酰胺Q,比如,VSVG的第47位氨基酸缺失,和/或第R354氨基酸缺失。
以慢病毒感染T细胞为例,在慢病毒包膜表达突变的VSVG和CD7抗体,当CD7抗体与T细胞表面的CD7抗原结合后,可以介导慢病毒的内吞,然后突变的VSVG介导溶酶体逃逸,从而完成慢病毒对T细胞的靶向感染。而且慢病毒只感染表达CD7的T细胞,对于不表达CD7的细胞,没有感染能力。
靶向载体搭载的物质不受限制,可以根据实际需要选取,比如小分子化合物、蛋白质、多肽、RNA、DNA,包括嵌合抗原受体CAR和TCR等。CAR类型不受限制,所述CAR包含抗原结合结构域。在一些实施方案中,所述抗原结合结构域是含有重链和轻链可变区的单链可变片段(scFv),所述重链和轻链可变区与所需抗原特异性结合。scFv选自单克隆抗体、嵌合单克隆抗体、人源化单克隆抗体、人抗体、纳米抗体以及合成抗体。在一些实施方案中,所述CAR还包含跨膜结构域(例如CD8跨膜结构域)和包含一个或多个基于免疫受体酪氨酸的激活基序(ITAM)的信号传导结构域(例如CD3ζ)。在一些实施方案中,所述CAR包含一个或多个共刺激结构域。CAR不受共刺激结构域类型的限制。事实上,可以使用本领域已知的任何共刺激结构域,包括但不限于CD28、4-1BB、DAP10、DAP12。
小分子化合物指分子量小于900道尔顿的有机化合物分子,尤其是可以作为药物使用的小分子。
在一个具体的方案中,所述靶细胞是淋巴细胞、髓系细胞、造血干/组细胞或非血液细胞,尤其是髓系细胞、造血干/组细胞或非血液细胞,包括正常细胞和肿瘤细胞。

本发明的另一方面,靶向载体的制备方法,包括以下步骤:
根据靶细胞的内吞受体,设计第一分子,
选取第二分子,
将第一分子、第二分子与载体搭载的物质进行组装,制备靶向载体。
第一种情况,通过设计第一分子,即在病毒表面修饰可以靶向识别特定受体的抗体/配体,选择野生型的病毒包膜蛋白为第二分子,若靶细胞表达病毒所携带包膜蛋白受体,那么第一分子的存在,可以促进靶向载体对靶细胞的感染。
第二种情况,通过设计第一分子,即在病毒表面修饰可以靶向识别特定受体的抗体/配体,选择野生型的病毒包膜为第二分子,若靶细胞低表达或者不表达病毒所携带包膜蛋白受体,那么依赖第一分子,病毒仍然可以完成对靶细胞的感染。
第三种情况,通过设计第一分子,即在病毒表面修饰可以靶向识别特定受体的抗体/配体,选择与受体结合受限、但具备内体/溶酶体逃逸能力的突变型的病毒包膜蛋白作为第二分子,不管靶细胞是否表达病毒所携带包膜蛋白受体,那么依赖第一分子,病毒均可以对靶细胞进行靶向感染,同时病毒不会感染其它细胞,大大提高了靶向的精确性。
有包膜载体和无包膜载体的组装方式:
有包膜的载体的组装,以VSVG为例:将表达与内体受体结合的抗体/配体的质粒,与慢病毒包装质粒,如psPAX2和pMD.2G(VSVG或VSVG突变体),以及慢病毒表达载体混合,即可以构建靶向表达载体。
无包膜的载体的组装,以AAV为例:将与内吞受体结合的抗体/配体通过跨膜肽段连接到AAV的包膜蛋白上,即可实现AAV靶向载体的构建。
有包膜载体和无包膜载体进入细胞的方式:
有包膜的载体,如基于VSVG的假型慢病毒载体,通过VSVG与细胞膜表面的LDL-R结合,通过网格蛋白(clathrin)介导的内吞作用进入细胞。在内吞形成的内体酸化之后,VSVG构象发生变化,导致病毒包膜与内体膜发生融合,从而使病毒从内体/溶酶体中脱离,并且通过核孔进入细胞核。
无包膜的载体,无重组AAV病毒颗粒通过与宿主细胞表面表达的糖化受体相结合,通过网格蛋白(clathrin)介导的内吞作用进入细胞,在内吞形成的内体酸化之后,病毒衣壳的VP1/VP2部分构象发生变化,导致病毒从内体中脱离,并且通过核孔进入细胞核。
本发明的另一方面,靶向载体可以用于药物或疫苗递送,尤其是用于递送小分子化合物、蛋白质、多肽、RNA或DNA。
本发明的另一方面,一种将物质引入细胞的方法,所述方法包括:使所述细胞与靶向载体相接触。
在一个具体的方案中,所述细胞是哺乳动物的细胞。
在一个具体的方案中,所述细胞是正常细胞或癌细胞。
在一个具体的方案中,所述细胞是T细胞、NK细胞、B细胞、巨噬细胞、粒细胞、树突状细胞、造血干细胞、肝细胞、胰岛细胞、神经细胞、肌肉细胞。
在一个具体的方案中,所述接触在体内或体外进行,例如,可以在静脉内、腹膜内、肿瘤内、骨内或结节内给药,使靶向载体进入体内,与靶细胞接触;或将靶病毒在体外直接感染靶细胞。
术语“哺乳动物”指任何哺乳动物物种诸如人、小鼠、大鼠、狗、猫、仓鼠、豚鼠、兔、家畜等。
本发明的另一方面,一种组合物,包含靶向载体;该组合物可以用作药物;组合物可以用于制备基因疗法、免疫疗法、细胞疗法、治疗基因缺陷疾病、治疗癌症的药物。
本方案利用内吞与内体/溶酶体逃逸相结合的方式,对载体进行改造,使其带有能与靶细胞的内吞受体结合的第一分子,得到靶向载体,具有以下有益效果:
根据不同细胞类型特异的高效内吞受体,可以针对性地快速开发针对不同细胞类型的靶向载体,而无需盲目地筛选或者试错验证;
根据所要靶向的细胞的内吞受体设计第一分子,可以靶向不同种类的细胞,特别是不表达或低表达已广泛应用的病毒包膜糖蛋白受体(如LDL-R)的细胞,应该范围广泛;
对载体进行合理的突变之后,可以避免感染不需要靶向的细胞,精确性大大提高。

由于NK低表达VSVG的受体LDL-R,因此普通的VSVG慢病毒很难感染NK细胞,因此发明人在慢病毒构建一个膜表达的CD7抗体,使之与NK细胞上的CD7发生结合并诱导慢病毒内吞,接着VSVG介导病毒包膜和内体/溶酶体膜的融合,从而发生溶酶体逃逸,进而使得载体负载的GFP进入细胞表达。慢病毒靶向载体的结构如图1所示,1为可以靶向内吞受体的第一分子CD7抗体,2为介导内体/溶酶体逃逸的第二分子VSVG。
1、设计膜表达的CD7抗体

3、包被慢病毒
用携带膜表达CD7抗体的质粒、psPAX2和pMD2.G(VSVG野生型)包装质粒,包被表达pGClenti-GFP(GFP荧光蛋白)的慢病毒(骨架如图2所示)。具体的做法是,将上述4种质粒混合,并通过PEI转染进293T细胞,48小时收集培养基上清,离心即可获得可以靶向NK细胞的慢病毒。将慢病毒添加到NK细胞培养体系中,即可感染NK细胞。结果如图3所示,左图为VSVG慢病毒感染的NK细胞表达GFP的情况;右图为携带模型CD7抗体的VSVG慢病毒感染的NK表达GFP的情况;普通VSVG慢病毒难以感染NK细胞,而携带模型CD7抗体的VSVG慢病毒可以有效地靶向感染NK靶细胞,这是因为NK细胞不表达LDL-R,而高表达CD7。
实施例2
筛选不具备受体结合能力,但保留内体/溶酶体逃逸能力的VSVG突变体
根据VSVG与LDL-R的结合位置,发明人设计了一系列VSVG的突变体,如图4所示,Mut代表突变体,Δ代表碱基缺失,以测试其与受体的结合能力以及内体/溶酶体的逃逸能力。
实施例3
使用突变型VSVG构建靶向Jurkat细胞的慢病毒
使用突变型VSVG,保留其溶酶体逃逸的能力,丧失与LDL-R结合的能力,使其特异性靶向表达CD7的细胞,对于不表达CD7、表达LDL-R的细胞,慢病毒不能感染。
1、设计CD7抗体
与实施例相同。
2、构建突变的VSVG
VSVG突变体(第354位氨基酸突变为Q)的氨基酸序列为SEQ ID NO:8:

3、包被慢病毒

结果如图5所示,用VSVG慢病毒、靶向性慢病毒分别感染不表达CD7的Raji细胞和表达CD7的Jurkat细胞,左列为非靶向性慢病毒载体感染的结果,右列为靶向性慢病毒载体感染的结果;对照组为Raji细胞,该细胞不表达CD7,实验组为CD7+Jurkat细胞。非靶向性慢病毒载体对Raji细胞和Jurkat细胞都具有感染能力,靶向性慢病毒载体只对Jurkat细胞具备感染能力,对Raji细胞没有感染能力。本实施例靶向CD7的慢病毒不能有效地感染Raji细胞,只能有效地感染Jurkat细胞,这是因为靶向性慢病毒所使用的突变型VSVG对细胞没有感染能力,其靶向性依赖于CD7抗体,只能靶向表达CD7的细胞,非靶向性慢病毒所携带的野生型VSVG可以与表达LDL-R细胞结合,不管该种细胞是否表达CD7。
实施例4
构建靶向CD33+细胞的慢病毒
1、设计模型表达的CD33抗体
模型表达的CD33抗体序列,从5’到3’端依次包括CD8信号肽、Gemtuzumab轻链(VL)、GS连接肽(linker)、Gemtuzumab重链(VH)、CD8铰链区、CD8跨膜区。

构建靶向去唾液酸糖蛋白受体阳性(ASGPR+)肝细胞的慢病毒。
靶向去唾液酸糖蛋白受体(ASGPR)可以为N-乙醯半乳糖胺(GalNAc)或者ASGPR抗体。GalNAc可以通过修饰添加到慢病毒包膜表面,或者,可以通过膜表达ASGPR抗体,制备靶向肝细胞的慢病毒。
1、设计膜表达的ASGPR抗体
模型表达的CD33抗体序列,从5’到3’端依次包括CD8信号肽、ASGPR轻链B11(VL)、GS连接肽(linker)、ASGPR重链(VH)、CD8铰链区、CD8跨膜区。

基于GALA的靶向慢病毒的构建
GALA是一种具有谷氨酸-丙氨酸-亮氨酸-丙氨酸(EALA)重复序列的30个氨基酸合成肽。GALA多肽蛋白可以介导内体/溶酶体膜融合,通过在GALA的3端添加对病毒出芽具有重要作用的包膜蛋白尾端序列,如BaEV、RD114包膜蛋白或者VSVG包膜蛋白的尾端(439-529号氨基酸)序列,可以构建基于GALA多肽第二分子的靶向慢病毒。第二分子的五端,有信号肽。

结果发现,CD8+细胞和慢病毒接触之后,虽然CD8受体与抗体结合,但不能有效内吞,基于膜表达CD8抗体的慢病毒载体不能有效介导对CD8+细胞的感染。因此,不具备内吞能力的受体,不能作为第一分子,用于构建本发明的靶向载体。

Claims (10)
1.一种靶向载体,其包含:与靶细胞的内吞受体结合的第一分子和促进靶向载体搭载的物质释放进入细胞质的第二分子;当靶向载体为病毒载体时,所述第一分子不是病毒包膜蛋白的一部分。
2.根据权利要求1所述的靶向载体,其特征在于,所述第二分子促使靶向载体发生内体逃逸或者溶酶体逃逸。
3.根据权利要求1所述的靶向载体,其特征在于,所述靶向载体为有包膜的载体或无包膜的载体;所述有包膜的载体选自逆转录病毒载体、慢病毒载体、脂质纳米颗粒;所述无包膜的载体选自腺病毒、腺相关病毒、类病毒颗粒。
4.根据权利要求1~3任意一项所述的靶向载体,其特征在于,所述第二分子为病毒包膜蛋白和/或非病毒包膜蛋白;所述病毒包膜蛋白选自水疱性口炎病毒包膜糖蛋白VSVG及其变体、狒狒内源性逆转录病毒包膜糖蛋白BaEV及其变体、猫内源性逆转录病毒包膜糖蛋白RD114及其变体、长臂猿白血病病毒包膜糖蛋白GALV及其变体中的至少一种;所述非病毒包膜蛋白选自腺相关病毒AAV的VP1及其变体、腺相关病毒AAV的VP2及其变体、GALA多肽及其变体、聚乙烯亚胺中的至少一种。
5.根据权利要求4所述的靶向载体,其特征在于,所述内吞受体选自:HER2、CD20、CD19、CD79A、CD79B、CD56、CD22、CD138、CD37、CD98、CD309、CD33、CD163、CD163B、CD5、CD7、CD169、CD204、CD205、CD209、CD280、CD302、TROP-2、CD19、NECTIN4、5T4、CD30 TROP2、FRα、STEAP1、ENPP3、GCC、SLC44A4、NaPi2b、CA9、SC-16、CD142、P-Cadherin、PSMA、ED-B、endothelinreceptors ETB、TN-C、Collagen IV、Periostin、CEACAM、c-MET、TDGF1、IGF1R、Mesothelin、TIM1、NCAM1、ZIP6、CD166、GPNMB、SDC1、glycosphingolipid、TfR、Ganglioside、CD74、CLDN18、DPEP3、SLITRK6、PRL-R、LY75、CD48、MUC1、CDKs、B7-H4、STING、KAAG1、CD70、CDH3、LRRC15、EGFR、ASGPR。
6.根据权利要求1所述的靶向载体,其特征在于,所述靶向载体为慢病毒载体;所述慢病毒载体表达的第一分子为跨膜蛋白,所述慢病毒载体表达的第二分子为病毒包膜蛋白。
7.根据权利要求6所述的靶向载体,其特征在于,所述跨膜蛋白选自CD7抗体、CD79B抗体、CD33抗体、ASGRP抗体或配体、Mesothelin抗体、HER2抗体。
8.根据权利要求6所述的靶向载体,其特征在于,所述病毒包膜蛋白发生突变,使其受体识别能力减弱;所述病毒包膜蛋白为水疱性口炎病毒Indian毒株包膜糖蛋白VSVG。
9.根据权利要求1所述的靶向载体,其特征在于,所述物质为小分子化合物、蛋白质、多肽、RNA、DNA中的至少一种。
10.一种将物质引入细胞的方法,所述方法包括:使所述细胞与权利要求1~9任意一项所述的靶向载体相接触。
