Abstract
The invention provides a water-soluble fertilizer, a preparation method and application thereof, and belongs to the technical field of agriculture. The preparation method comprises the following steps: mixing plant immunity protein composition, humic acid, nutrient and adjuvant, and stirring. The water-soluble fertilizer can be prepared into powder, water and granules, and can be applied in the modes of drip irrigation, spray irrigation, seed soaking, leaf surface spraying, root irrigation and flight protection during application. Meanwhile, the water-soluble fertilizer has the effects of effectively improving crop yield and promoting crop growth, and can also be used for preventing and treating plant virus diseases and improving sugar content in fruits.
Description
Technical Field
The invention belongs to the technical field of agriculture, and particularly relates to a water-soluble fertilizer and a preparation method and application thereof.
Background
The plant immunity protein is called as a plant immunity resistance inducer or a plant vaccine, has the effects of obviously preventing and treating diseases, resisting stress, increasing yield and improving quality, and is harmless to human and livestock and does not pollute the environment. The plant immunity protein does not directly act on a target, but excites the plant to generate an immune response, and induces the expression of plant related genes through a signal transduction system, so that corresponding defensive substances are generated, the immunity of the plant is improved, the rooting and the leaf enlargement of the plant are promoted, the drought resistance and the salt tolerance of the plant are enhanced, and meanwhile, the occurrence of plant diseases is lightened. The research and development of the plant immunity protein are the important development direction of the current international novel fertilizer and biological pesticide, and have great development prospect.
Fungal sources of plant immunity proteins are mainly Botrytis, alternaria, magnaporthe grisea (Magnaporthe), verticillium (Verticillium), aspergillus flavus (Aspergillus), rhizoctonia solani (Rhizoctonia), trichoderma, fusarium (Fusarium). Plant immunity proteins induce plants to develop systemic resistance, thereby activating the autoreactive system, and induce plant resistance for about 22 days.
How to better utilize the performance of plant immunity proteins, and provide fertilizer with good effect and wide functions, and further research is still needed.
Disclosure of Invention
In view of the above, the invention aims to provide a plant immunity protein water-soluble fertilizer, a preparation method and application thereof, and the water-soluble fertilizer can promote the growth of crops, improve the yield of the crops, can be used for preventing and treating plant virus diseases, and has the function of improving the sugar content in fruits.
In order to achieve the above purpose, the present invention provides the following technical solutions:
the invention provides a plant immunity protein water-soluble fertilizer, which comprises a plant immunity protein composition, humic acid and nutrient substances.
Preferably, the plant immunity protein composition: humic acid: the weight percentage of the nutrient substances is 1-100: 1 to 100: 1-100, preferably a plant immunity protein composition: humic acid: the weight percentage of the nutrient substances is 1-77: 3 to 79:20 to 96 percent.
Preferably, the plant immunity protein composition comprises one or more of three active proteins PebC1, pebC2 and BcGS1 with molecular weights of 36kDa, 14kDa and 80kDa respectively.
Preferably, the humic acid comprises one or more of potassium fulvate, sodium humate, potassium humate, nitro humic acid, brown fulvic acid and boron humate.
Preferably, the nutrient substances comprise one or more of nitrogenous fertilizer, phosphate fertilizer, potash fertilizer, medium element and trace element.
Preferably, the nutrient is selected from one or more of urea, urea ammonium nitrate solution, ammonium sulfate, ammonium chloride, ammonium polyphosphate, monoammonium phosphate, monopotassium phosphate, potassium chloride, potassium nitrate, potassium sulfate, potassium glycinate, potassium formate, calcium ammonium nitrate, magnesium chloride, zinc sulfate, zinc nitrate, zinc sulfate monohydrate, EDTA-Zn, sodium octaborate tetrahydrate, borax, magnesium sulfate, calcium nitrate, EDTA-Ca, copper sulfate, EDTA-Cu, manganese sulfate, EDTA-Mn, EDTA-Fe and ammonium molybdate.
Preferably, the preparation method of the plant immunity protein composition comprises the following steps:
activating Botrytis cinerea strain, culturing with seed solution, fermenting to obtain fermentation liquid, crushing fermentation liquid with colloid mill cell, heating, filtering, and purifying to obtain plant immunity protein composition.
Preferably, the preparation method of the water-soluble fertilizer comprises the following steps:
mixing plant immunity protein composition, humic acid and nutrient substances to prepare various dosage forms; preferably, 3-10% of humic acid, 20-50% of nutrient substances, 1% of disodium ethylenediamine tetraacetate and required auxiliary agents are added into the plant immunity protein composition, and the mixture is dried by a spray dryer, and then 1% of sodium dodecyl benzene sulfonate is added to prepare the solid water-soluble fertilizer;
preferably, 3-10% of humic acid, 20-50% of nutrient substances, 1% of disodium ethylenediamine tetraacetate and required auxiliary agents are added into the plant immunity protein composition, and then 1% of sodium dodecyl benzene sulfonate is added, so that the liquid water-soluble fertilizer can be prepared.
Preferably, the dosage form of the water-soluble fertilizer comprises powder, water and granules.
The invention provides application of the water-soluble fertilizer in improving crop growth performance and/or crop yield.
Preferably, the application method of the plant immunity protein water-soluble fertilizer comprises drip irrigation, spray irrigation, seed soaking, leaf surface spraying, root irrigation and flight protection.
Preferably, the field crop is selected from wheat, rice, corn, tobacco; the vegetables are selected from tomato, cucumber and green pepper, and the fruits are selected from strawberry, grape, apple and watermelon.
Compared with the prior art, the invention has the following beneficial effects:
(1) The water-soluble fertilizer prepared by the invention can provide nutrient substances required by the growth of crops, promote the rooting of the crops, ensure that the leaves of the crops are dark green after the crops are used, and the crops are strong, disease-resistant and stress-resistant, and improve the quality.
(2) The water-soluble fertilizer prepared by the invention has the function of preventing and treating plant virus diseases, such as tomato yellow leaf curl virus diseases, watermelon fusarium wilt and the like.
(3) The water-soluble fertilizer prepared by the invention can improve the yield of crops, vegetables and fruits (such as corn, wheat, tomatoes and the like) and can improve the sugar content in the fruits (such as watermelons).
Detailed Description
The invention provides a water-soluble fertilizer which comprises the following raw material components in parts by weight: 1-100 parts of plant immunity protein composition, 1-100 parts of humic acid and 1-100 parts of nutrient substances.
In the present invention, the amount of the plant immunity protein composition is 1 to 100 parts, preferably 1 to 77 parts, more preferably 30 to 60 parts, still more preferably 40 to 50 parts; the plant immunity protein composition comprises one or more of three active proteins PebC1, pebC2 and BcGS1 with molecular weights of 36kDa, 14kDa and 80kDa respectively. In the present invention, the method for preparing the plant immunity protein composition preferably comprises the following steps: fermenting the botrytis cinerea to obtain fermentation liquor, and carrying out colloid mill, filtration and purification treatment on the fermentation liquor to obtain the plant immunity protein composition. In the invention, the botrytis cinerea is subjected to activation, primary seed liquid culture and secondary seed liquid culture before liquid fermentation; the activation temperature is preferably 22 to 27 ℃, more preferably 24 to 26 ℃, still more preferably 25 ℃; the activation time is preferably 5 to 9d, more preferably 6 to 8d, still more preferably 7d; the temperature of the enrichment culture is preferably 22-27 ℃, more preferably 24-26 ℃, still more preferably 25 ℃; the time of the enrichment culture is preferably 70 to 75 hours, more preferably 72 to 74 hours, still more preferably 73 hours; the culture mode of the first-stage seed liquid is preferably shake flask culture, and the rotation speed of the shake flask culture is preferably 150-250 r/min, more preferably 170-230 r/min, and even more preferably 200r/min; the temperature of the primary seed liquid culture is preferably 22 to 27 ℃, more preferably 24 to 26 ℃, still more preferably 25 ℃; the culture time of the primary seed liquid is preferably 55-65 h, more preferably 58-63 h, still more preferably 60h; the culture temperature of the secondary seed liquid is the same as that of the primary seed liquid; the time for culturing the secondary seed liquid is preferably 70-75 hours, more preferably 72-74 hours, even more preferably 73 hours; the primary seed liquid culture and the secondary seed liquid culture are accompanied by stirring, and the stirring speed is preferably 90-110 r/min, more preferably 95-105 r/min, and even more preferably 100r/min; the fermentation broth is preferably mixed with potassium dihydrogen phosphate and dipotassium hydrogen phosphate before passing through a colloid mill; the final concentration of the phosphate buffer in the fermentation liquid after adding potassium dihydrogen phosphate and dipotassium hydrogen phosphate is preferably 15 to 25mM, more preferably 17 to 24mM, still more preferably 20mM; the fermentation broth is preferably subjected to heating treatment before filtration, and the heating temperature is preferably 90-110 ℃, more preferably 95-105 ℃, and even more preferably 100 ℃; the heating time is preferably 25 to 35 minutes, more preferably 27 to 32 minutes, and still more preferably 30 minutes. In the present invention, the effect of the heating is to destroy and inactivate other proteins. The purification method of the present invention is not particularly limited, and may be carried out by a method of purifying a protein which is conventional in the art. In the implementation process of the invention, the filtrate collected after filtration can be directly used.
In the present invention, the amount of the humic acid is 1 to 100 parts, preferably 3 to 79 parts, more preferably 30 to 60 parts, still more preferably 40 to 50 parts; the humic acid preferably comprises one or more of potassium fulvate, sodium humate, potassium humate and nitro humic acid, brown fulvic acid and boron humate, more preferably comprises one or more of potassium fulvate, sodium humate and potassium humate, and still more preferably comprises sodium humate; the nutrient preferably comprises one or more of nitrogen fertilizer, phosphate fertilizer, potash fertilizer, medium element and trace element, more preferably comprises one or more of nitrogen fertilizer, phosphate fertilizer and potash fertilizer, and still more preferably comprises nitrogen fertilizer.
In the present invention, the amount of the nutrient is 1 to 100 parts, preferably 20 to 96 parts, more preferably 30 to 60 parts, still more preferably 40 to 50 parts; the nutrient preferably comprises one or more of urea, urea ammonium nitrate solution, ammonium sulfate, ammonium chloride, ammonium polyphosphate, monoammonium phosphate, monopotassium phosphate, potassium chloride, potassium nitrate, potassium sulfate, potassium fulvate, potassium glycinate, potassium formate, calcium ammonium nitrate, magnesium chloride, zinc sulfate, zinc nitrate, zinc sulfate monohydrate, EDTA-Zn, sodium octaborate tetrahydrate, borax, magnesium sulfate, calcium nitrate, EDTA-Ca, copper sulfate, EDTA-Cu, manganese sulfate, EDTA-Mn, EDTA-Fe, ammonium molybdate, further preferably comprises one or more of urea, potassium dihydrogen phosphate, potassium fulvate, magnesium sulfate, EDTA-Ca, ammonium molybdate, still further preferably comprises one or more of ammonium sulfate, potassium chloride, magnesium chloride, zinc sulfate monohydrate.
The invention also provides a preparation method of the water-soluble fertilizer, which comprises the following steps:
mixing and stirring the plant immunity protein combination, humic acid and nutrient substances uniformly to obtain the water-soluble fertilizer.
The specific type of the auxiliary agent is not particularly limited, and the preparation formulation of the water-soluble fertilizer is specifically selected according to the preparation. When preparing the solid water-soluble fertilizer, the preparation method is as follows:
drying the plant immunity protein composition, humic acid, nutrient substances, 1% of disodium ethylenediamine tetraacetate and required auxiliary agents by a spray dryer, and adding 1% of sodium dodecyl benzene sulfonate to prepare a solid water-soluble fertilizer;
when preparing the liquid water-soluble fertilizer, the preparation method is as follows:
humic acid, nutrient substances, 1% of disodium ethylenediamine tetraacetate and required auxiliary agents are added into the plant immunity protein composition, and then 1% of sodium dodecyl benzene sulfonate is added, so that the liquid water-soluble fertilizer can be prepared. In the preparation of the liquid water-soluble fertilizer, the auxiliary agent is preferably ethylene glycol.
In the present invention, the amount of sodium dodecylbenzenesulfonate added is preferably 0.5 to 1.5 parts, more preferably 0.8 to 1.2 parts, and still more preferably 1.0 part.
The invention also provides application of the water-soluble fertilizer in promoting crop growth.
In the present invention, the crops preferably include field crops, fruits and vegetables; the field crops preferably comprise wheat, rice, corn and tobacco; the fruit preferably comprises strawberry, grape, apple, watermelon; the vegetables preferably comprise tomatoes, cucumbers and green peppers.
The invention also provides application of the water-soluble fertilizer in preventing and treating plant virus diseases.
The invention also provides application of the water-soluble fertilizer in improving crop yield and sugar content.
In the invention, the water-soluble fertilizer is applied in the modes of drip irrigation, spray irrigation, seed soaking, leaf surface spraying, root irrigation and flight protection during use. The concentration of the water-soluble fertilizer is preferably 800-1200 times of dilution, more preferably 900-1100 times of dilution, and even more preferably 1000 times of dilution.
The technical solutions provided by the present invention are described in detail below with reference to examples, but they should not be construed as limiting the scope of the present invention.
EXAMPLE 1 preparation of plant immune proteins
(1) Inoculating a botrytis cinerea strain on a PDA culture medium, and performing activation culture for 7d at 25 ℃; meanwhile, the culture medium is sub-packaged in 1000ml triangular flasks for sterilization, and the liquid loading amount of each triangular flask is 500ml;
(2) Inoculating the activated strain into a triangular flask, and shake-flask culturing at 26 ℃ and 200r/min for 72 hours to obtain primary seed liquid;
(3) Inoculating 1L of primary seed liquid cultured in a triangular flask into a 500L fermentation tank, wherein the liquid loading amount in the fermentation tank is 300L, and culturing at 26 ℃ for 60h at 100r/min to obtain secondary seed liquid;
(4) According to the following steps of 1:10, inoculating the secondary seed liquid into a fermentation tank with the liquid loading amount of 2/3, and culturing at 26 ℃ for 72 hours at 100r/min to obtain fermentation liquid;
(5) Adding monopotassium phosphate and dipotassium phosphate into the fermentation liquor to obtain a phosphate buffer solution with the final concentration of 20mM, crushing cells by using three groups of colloid mills, heating at 100 ℃ for 30min, and filtering and purifying to obtain the plant immunity protein composition.
Example 2 preparation of Water-soluble Fertilizer powder
The plant immunity protein composition prepared in the embodiment 1 is added with potassium humate, urea, monoammonium phosphate, potassium nitrate, disodium ethylenediamine tetraacetate and required auxiliary agents according to mass percent, and the mixture is uniformly mixed and stirred, filtered, dried by a spray dryer, and added with sodium dodecyl benzene sulfonate, thus obtaining the plant immunity protein macroelement water-soluble fertilizer powder.
Wherein the plant immunity protein composition comprises 57.8% of plant immunity protein composition, 5% of potassium humate, 16.8% of urea, 8.4% of monoammonium phosphate, 10% of potassium nitrate, 1% of disodium ethylenediamine tetraacetate and 1% of dodecane basic sodium sulfonate.
Example 3 preparation of Water-soluble Fertilizer powder
The preparation method comprises the following steps: same as in example 2
Wherein the plant immunity protein composition comprises 55.7% of plant immunity protein composition, 8.5% of humic acid potassium, 21.8% of urea, 10% of monopotassium phosphate, 2% of potassium sulfate, 1% of disodium ethylenediamine tetraacetate and 1% of dodecane basic sodium sulfonate.
Example 4 preparation of Water-soluble fertilizer aqua
Adding potassium humate, urea, monoammonium phosphate, potassium nitrate, disodium ethylenediamine tetraacetate and required auxiliary agents into the plant immunity protein composition according to mass percent, finally adding sodium dodecyl benzene sulfonate, mixing and stirring uniformly, and filtering to prepare the plant immunity protein macroelement water-soluble fertilizer aqueous agent.
Wherein the plant immunity protein composition comprises 53.7% of plant immunity protein composition, 4.5% of potassium humate, 26% of urea ammonium nitrate, 4.2% of ammonium polyphosphate, 9.6% of dipotassium hydrogen phosphate, 1% of disodium ethylene diamine tetraacetate and 1% of ethylene glycol.
Example 5 preparation of Water-soluble Fertilizer powder
The preparation method comprises the following steps: same as in example 2
Wherein the plant immunity protein composition comprises 59.6% of plant immunity protein composition, 6.8% of potassium humate, 8.7% of zinc sulfate monohydrate, 4% of EDTA-Mn, 5.6% of EDTA-Cu, 8% of EDTA-Fe, 0.4% of ammonium molybdate, 4.9% of sodium octaborate tetrahydrate, 1% of disodium ethylenediamine tetraacetate and 1% of dodecane basic sodium sulfonate.
Test example 1 Effect of Water-soluble fertilizer on wheat growth and yield
2 mu of test wheat fields (pH 6.97, nitrogen content of 150mg/kg, phosphorus content of 150g/kg, potassium content of 287g/kg and organic matter of 30.74 g/kg) with the same soil type and nutrient content are selected from test wheat fields of the agricultural institute of ligusticum city in Hebei province, and wheat is planted. Respectively applying water-soluble fertilizer in the wheat green-returning and jointing period and the booting and heading period; the water-soluble fertilizer obtained in example 2 and example 3 was applied as a control group (CK) which was purchased conventionally in the market. The water-soluble fertilizer is applied in an amount of 15 g/(mu. Times), diluted 1000 times, applied at an interval of 20 days, sprayed 3 times, and applied in a total amount of 45 g per mu. The results are shown in tables 1-1, 1-2 and 2.

As can be seen from tables 1-1, 1-2 and 2, after the water-soluble fertilizer prepared by the invention is applied, compared with a blank control, wheat seedlings grow in order, plants are strong, root systems are developed, tillering numbers are large, the stress resistance, cold resistance and drought resistance are strong, and the green turning is fast; the wheat in the flowering period is large in wheat ears, full in seeds, high in lodging resistance and thousand seed weight, and obvious in yield increasing effect.
Test example 2 Effect of Water-soluble fertilizer on wheat yield
In the test wheat fields in the Shahew towns of the Chen Tai, hebei, 3 mu of test wheat fields with basically the same soil type and nutrient content are selected, and wheat is planted. Applying a water-soluble fertilizer in the booting and heading period of wheat; the water-soluble fertilizer prepared in application example 3 was used as treatment group 1, the water-soluble fertilizer prepared in application example 2 was used as treatment 2, and the water-soluble fertilizer was not used as control group (CK). The application amount of the water-soluble fertilizer is 25 g/mu each time, the water-soluble fertilizer is diluted 1000 times for use, the use time interval is 7 days, the unmanned aerial vehicle sprays for 2 times, and the total fertilization amount per mu is 50 g. The results are shown in Table 3.

As can be seen from Table 3, after the water-soluble fertilizer prepared by the invention is applied, wheat ears are dark green and glossy, the growth vigor is strong, the wheat stalks are strong, and the yield-increasing effect is expected to be obvious.
Test example 3 Effect of Water-soluble fertilizers on maize plants and yield
In the test corn fields in Wucheng county of Shandong province, 2 mu of test corn fields with basically same soil type and nutrient content are selected, and corn of Zhongdan 909 is planted. The conventional water-soluble fertilizer purchased on the market was used as a control CK, and the water-soluble fertilizer prepared in example 2 was used as a test treatment. The corn seedlings were applied 1 time in foliar spray and 1 time in foliar spray after 25 days. Wherein the application amount of the water-soluble fertilizer is 25 g/mu, the dilution is 1000 times, and the total fertilization amount per mu is 50 g. The results are shown in tables 4 and 5.

As can be seen from tables 4 and 5, after the water-soluble fertilizer prepared by the invention is applied, the leaves of the corn at the seedling stage are dark green, the plants are strong, and the leaves are wide; the corn ear length in the harvest period increases the number of grains in rows and the thousand grain weight, the length of the bald tip is reduced, and the yield is obviously increased.
Test example 4 Effect of Water-soluble fertilizer on tomato yellow leaf curl Virus disease control and yield
In the greenhouse of modern agriculture and technology garden in Hebei province, selecting soil with basically same soil type and nutrient content, and planting tomatoes with the variety of golden greenhouse autumn. Applying the water-soluble fertilizer prepared in the example 2 and a commercially available water-soluble fertilizer in a tomato seedling stage; and the water-soluble fertilizer prepared in application example 2 is used as a treatment group 1, and the water-soluble fertilizer of a company conventionally purchased in the market is used as a treatment group control CK. The application amount of the water-soluble fertilizer prepared by the method is 15 g/mu each time, the water-soluble fertilizer is diluted 1000 times and used, the foliar spray is carried out for 3 times, and the application interval of each time is about 20 days. The results are shown in tables 6 and 7.

As can be seen from tables 6 and 7, after the water-soluble fertilizer prepared by the invention is applied, the leaves of tomato seedlings are dark green, the plants are strong, the fruit setting rate is high, and the disease resistance is enhanced; the tomato fruits in the mature period are round, bright in color, good in taste and obviously increased in yield.
Test example 5 Effect of Water-soluble fertilizer on control of watermelon fusarium wilt and sugar content
In the experimental watermelon greenhouse of Rao Yang county in Hebei province, selecting an experimental field with basically same soil type and nutrient content, and planting watermelons. Applying the water-soluble fertilizer prepared in example 2 and a commercially available water-soluble fertilizer in the mature period of the watermelon; the water-soluble fertilizer prepared in application example 2 was used as treatment group 1, and the conventional water-soluble fertilizer purchased in the market was used as control group (CK), and was used as treatment group 2. The application amount of the water-soluble fertilizer is 20 g/mu each time, the water-soluble fertilizer is diluted 1000 times and used, the foliar spray is carried out for 2 times, and the application interval of each time is 25 days. The results are shown in Table 8.

As can be seen from Table 8, after the water-soluble fertilizer prepared by the invention is applied, the watermelon leaves are dark green, the plant is strong, and the fruit setting rate is high; the application in the mature period can advance the time of watermelon marketing, the pulp of the watermelon is ruddy, the sugar content is high, and the taste is good.

1. The water-soluble fertilizer is characterized by comprising the following raw material components in parts by weight: 1-100 parts of plant immunity protein composition, 1-100 parts of humic acid and 1-100 parts of nutrient substances;
the plant immunity protein composition comprises one or more of PebC1, pebC2 and BcGS 1.
2. The water-soluble fertilizer of claim 1, wherein the humic acid comprises one or more of potassium fulvate, sodium humate, potassium humate, nitro humic acid, brown fulvic acid, boron humate.
3. The water-soluble fertilizer of claim 1, wherein the nutrient comprises one or more of a nitrogen fertilizer, a phosphate fertilizer, a potash fertilizer, a medium element, and a trace element.

The water-soluble fertilizer of claim 1, wherein the nutrients comprise one or more of urea, urea ammonium nitrate solution, ammonium sulfate, ammonium chloride, ammonium polyphosphate, monoammonium phosphate, monopotassium phosphate, potassium chloride, potassium nitrate, potassium sulfate, potassium fulvate, potassium glycinate, potassium formate, calcium ammonium nitrate, magnesium chloride, zinc sulfate, zinc nitrate, zinc sulfate monohydrate, EDTA-Zn, sodium octaborate tetrahydrate, borax, magnesium sulfate, calcium nitrate, EDTA-Ca, copper sulfate, EDTA-Cu, manganese sulfate, EDTA-Mn, EDTA-Fe, ammonium molybdate.
5. The water-soluble fertilizer according to claim 1, wherein the method for preparing the plant immunity protein composition comprises the following steps:
activating Botrytis cinerea strain, culturing with seed solution, fermenting to obtain fermentation liquid, crushing fermentation liquid with colloid mill cell, heating, filtering, and purifying to obtain plant immunity protein composition.
6. The method for preparing a water-soluble fertilizer according to any one of claims 1 to 5, comprising the steps of:
humic acid and nutrient substances are added into the plant immunity protein composition, and the water-soluble fertilizer is obtained after uniform mixing and stirring.
7. The method of claim 6, wherein the water-soluble fertilizer comprises a powder, a water or a granule;
auxiliary materials are added in the preparation process of the powder, the water agent or the granule, and the auxiliary materials comprise auxiliary materials or carriers.
8. Use of a water-soluble fertilizer as claimed in any one of claims 1 to 5 for improving crop growth performance and/or crop yield.
9. The use according to claim 8, wherein the water-soluble fertilizer is applied in the form of drip irrigation, spray irrigation, seed soaking, foliar spray, root irrigation and air defense at the time of use.
10. The use according to claim 8, wherein the crops include field crops, fruits and vegetables; the field crops comprise wheat, rice, corn and tobacco; the fruit comprises strawberry, grape, apple and watermelon; the vegetables include tomato, cucumber and green pepper.
