本发明涉及抗病毒药物技术领域,本发明提供了一种Vidofludimus在制备戊型肝炎药物中的应用。本发明通过实验证明Vidofludimus可以抑制HEV RNA复制和HEVORF2蛋白的表达,并且对临床利巴韦林治疗失败相关的HEV突变毒株同样具有抗病毒效果。因此Vidofludimus可以为治疗慢性戊型肝炎患者提供一种新的药物和治疗思路,也可以为临床利巴韦林耐药的HEV患者治疗提供新方案。
Description
技术领域
本发明涉及抗病毒药物技术领域,尤其涉及一种Vidofludimus在制备戊型肝炎药物中的应用。
背景技术
戊型肝炎是由戊型肝炎病毒(Hepatitis E Virus,HEV)引起的一种病毒性肝炎。HEV是造成全球公共卫生负担的重要病原体。根据世界卫生组织统计,全球每年约有2000万人感染HEV,其中约有330万临床病例,最终导致4.4~7万死亡。HEV对人群普遍易感,临床多表现为隐性感染或急性肝炎。但免疫功能低下或器官移植患者感染HEV会造成HEV持续性感染,逐渐发展为慢性戊型肝炎、肝硬化和肝癌等。越来越多的证据也表明其可导致诸如神经***症状等在内的肝外损伤。据报道,孕妇感染HEV可能发展为暴发性肝炎,死亡率高达20%~30%。对于HEV感染,目前还没有公认的治疗疗法。利巴韦林和干扰素-α是治疗戊肝的首选药物。虽然利巴韦林已成功用于慢性HEV感染,但利巴韦林有致畸性,禁用于妊娠患者的治疗,而且现在临床已经出现了利巴韦林耐药的HEV突变株;而且干扰素-α治疗会导致器官移植受者的移植排斥反应。目前尚无针对HEV的直接抗病毒药物,因此寻找更加安全且有效的新型抗HEV药物对于慢性戊型肝炎的治疗具有重大意义。
Vidofludimus是一种有效的二氢乳清酸脱氢酶(DHODH)抑制剂,可在不影响淋巴细胞增殖的条件下抑制IL-17的分泌。也是法尼醇X受体(FXR)的新型调节剂,具有口服活性。Vidofludimus作为一种免疫调节剂,可用于研究自身免疫性疾病,目前正在进行治疗类风湿关节炎的临床III期研究。Vidofludimus还可通过靶向FXR用于脂肪肝的研究。但目前并没有Vidofludimus在抗戊型肝炎病毒方面的相关报道。
发明内容
本发明的目的在于提供一种Vidofludimus在制备戊型肝炎药物中的应用,为治疗慢性戊型肝炎患者提供一种新的药物和治疗思路,也可以为临床利巴韦林耐药的HEV患者治疗提供新方案。

含有HEV全长基因组的野生型HEV质粒(Kernow-C1 p6 gt3,GenBank:JQ679013.1)和HEV-p6Gluc复制子质粒(Wenshi Wang et al.,Gastroenterology,VOLUME 151,ISSUE6,P1251-1253;Wenshi Wang et al.,Science Signaling,2017,25;10(476);Wenshi Wanget al.,Hepatology.2018,67(6):2096-2112)以及携带RdRp突变(Y1320H、G1634R)的HEV基因组质粒(本实验室构建保存),分别采用限制性内切酶Mlu I进行酶切(按照表1加入酶切反应体系,将样品置于37℃水浴锅内,酶切1h),纯化回收线性化质粒(在酶切产物中加入5倍体积的BufferPB混合,将混合溶液加到吸附柱中,静置吸附2min后,12000r/min离心1min,向吸附柱中加入500μl BufferPW洗涤,12000r/min离心1min,将吸附柱空转1min后,向吸附柱中间加入30μlBuffer EB,静置2min后,12000r/min离心1min,回收质粒),利用T7体外转录试剂盒分别转录出病毒RNA(按照表2加入体外转录反应体系,37℃水浴锅中孵育2h,然后加入0.5μL TURBO DNase,去除残余DNA,37℃水浴锅中孵育15min后,加入30μLLiCl,在-20℃放2h沉淀RNA,4℃,12000r/min离心20min,轻轻弃去上清,1mL 70%乙醇洗涤一遍沉淀RNA,4℃,12000r/min离心10min,弃去乙醇,加入30μL DEPC重悬RNA),通过电穿孔技术在270V、975μF条件下分别将RNA电转进HEK 293T和Huh7细胞中(本实验室保存)(电转前将3×106个HEK 293T、Huh7分别在60mm培养皿中铺板,用含10%FBS的DMEM培养基在37℃,5%CO2培养箱中培养24h,弃去培养基,1mLPBS洗两遍。用1mL胰酶消化细胞1min,待细胞收缩变圆轻轻弃去胰酶,加入3mL含血清培养基,吹下细胞到15mL离心管中,1000r/min室温离心5min,弃去培养基,再用3mL PBS重悬细胞洗一遍,1000r/min离心5min,弃去PBS,用400μL Opti-MEM重悬细胞,然后将细胞转移到1.5mL无核酶EP中,加入3μg RNA混合后加入到4mm电穿孔杯(BTX)中进行电穿孔实验),分别构建HEK 293T-HEV-p6Gluc和Huh7-HEV-p6Gluc复制子模型、Huh7-HEV野生型和突变型复制模型。

下述实施例中使用的类器官HEV复制模型的构建方法如下:
将类器官在冷的Advance DMEM/F12培养基中收集。然后,在Opti-MEM中重悬类器官,500g,8℃离心5min,将200μL的Opti-MEM类器官悬浮液与6μg HEV RNA充分混合,加入到4mm的电穿孔杯中,按照以下程序进行电穿孔:电压700V、脉冲长度4ms,用Opti-MEM洗涤3遍以去除残留RNA,将类器官包埋于基质胶中,在37℃、5%CO2的EM中培养。
下述实施例中使用的WST-1细胞增殖及细胞毒性检测试剂盒为KeyGen公司生产的货号为C0036的产品。
下述实施例中使用的高斯荧光素酶检测试剂盒为Beyotime公司生产的货号为RG062M的产品。
下述实施例中使用的First-Strand cDNA Synthesis Kit,SYBR GreenqPCRMasterMix(2X)为APExBIO公司生产的货号分别为K1072、K1070的产品。
下述实施例中使用的FITC标记山羊抗兔IgG的二级抗体为Jackson公司生产的货号为111-585-003、商品名称为Alexa Flour 594-comjugated Affinipure GoatAnti-RabbitIgG(H+L)的二抗。
实施例1Vidofludimus抑制HEV复制子复制的体外实验
(1)HEV复制子模型细胞的培养
将含有HEV复制子的HEK 293T和Huh7细胞分别在60mm培养皿中进行培养,DMEM高糖培养基(Bio-Channel)+10%FBS(Bio-Channel),在37℃,5%二氧化碳培养箱中培养,待细胞长满后(约3×106个)铺板加药处理。
(2)实验分组和处理
将两种HEV复制子模型细胞分别接种到96孔板中,每孔2×104个细胞,加入10%FBS(Bio-Channel)+DMEM高糖培养基培养液,细胞贴壁24h后,分组加药处理,每组3个复孔,分组如下:
a、实验组:加Vidofludimus处理(培养基中的终浓度为0.2、1、5、10、50μM)72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(3)实验方法
将上述2个分组分别处理72h后,用高斯荧光素酶检测试剂盒检测高斯荧光素酶活性,分析Vidofludimus抑制HEV复制子复制情况。
(4)结果
实验结果如图1所示。由图1可知,Vidofludimus处理后,相比于空白对照组(1%DMSO),在有效浓度范围内高斯荧光素酶活性降低了,说明Vidofludimus对HEV复制子复制有抑制作用。Mean±SEM;****P<0.0001。
实施例2Vidofludimus对细胞安全性的体外实验
(1)Huh7-HEV复制子模型细胞的培养
复苏已电转HEV复制子RNA的Huh7细胞进行培养,首先从-80℃冰箱中取出冻存的细胞,快速将其放入37℃水浴锅中解冻,待细胞解冻后,将细胞转移到3mL完全培养基中(10%FBS+DMEM),1000r/min离心5min后弃去培养基,加入10%FBS(Bio-Channel)的DMEM高糖培养基(Bio-Channel),置于37℃,5%CO2培养箱中培养,待细胞长满后进行传代。细胞传代,首先弃去培养皿内的培养基,用无菌PBS清洗两遍,加入1mL胰酶(Bio-Channel)进行消化,待细胞回缩变圆后弃去胰酶,按照1:4比例加入完全培养基传代。
(2)实验分组和处理
将Huh7-HEV复制子细胞以每孔2×104个细胞接种到96孔板中,细胞贴壁24h后,分组加药处理,每组3个复孔,分组如下:
a、实验组:加Vidofludimus处理(培养基中的终浓度为0.2、1、5、10、50、200μM)72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(3)实验方法
将上述2个分组分别处理72h后,使用WST-1细胞增殖及细胞毒性检测试剂盒(KeyGen)检测细胞的增殖活性,通过GraphpadPrism 9计算出其半数毒性浓度(Half-maximal Cytotoxicity Concentration,CC50),分析Vidofludimus对Huh7细胞的安全性。
(4)结果
实验结果如图2所示。由图2可知,Vidofludimus对Huh7细胞的CC50为160μM。
实施例3Vidofludimus抑制野生型HEV RNA复制的体外实验
(1)Huh7-HEV复制模型细胞的培养
复苏已电转野生型HEV RNA的Huh7细胞进行培养,首先从-80℃冰箱中取出冻存的细胞,快速将其放入37℃水浴锅中解冻,待细胞解冻后,将细胞转移到3mL完全培养基中(10%FBS+DMEM),1000r/min离心5min后弃去培养基,加入10%FBS(Bio-Channel)的DMEM高糖培养基(Bio-Channel),置于37℃,5%CO2培养箱中培养,待细胞长满后进行传代。细胞传代,首先弃去培养皿内的培养基,用无菌PBS清洗两遍,加入1mL胰酶(Bio-Channel)进行消化,待细胞回缩变圆后弃去胰酶,按照1:4比例加入完全培养基传代。
(2)实验分组和处理
将已电转野生型HEV RNA的Huh7细胞以每孔5×104个细胞接种到24孔板中,细胞贴壁24h后,分组加药处理,每组2个复孔,分组如下:
a、实验组:加Vidofludimus处理(培养基中的终浓度为1、5μM)72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(3)实验方法
将上述2个分组分别处理72h后,利用TRIzol法提取含有野生型HEV的Huh7细胞总RNA,反转录为cDNA后作为模版(按照表3加入RNA变性体系,65℃加热5min,再按照表4加入反转录体系,轻轻混匀后,按照25℃,2min;45℃,50min;70℃,15min进行反转录),然后通过qPCR方法分别检测HEV RNA和GAPDH基因的转录水平(按照表5加入qPCR反应体系,反应程序见表6),引物序列见表7,分析Vidofludimus对HEV RNA复制水平的影响。

(4)结果
实验结果如图3所示。由图3可知,Vidofludimus处理后,相比于空白对照组(1%DMSO),Vidofludimus在5μM可以将HEV RNA水平降低到50%左右。Mean±SEM;****P<0.0001。
实施例4Vidofludimus抑制HEV ORF2蛋白表达的体外实验
(1)Huh7-HEV野生型和利巴韦林耐药相关的突变型HEV细胞复制模型的培养
将构建的野生型和突变型HEV复制模型细胞分别在60mm培养皿中,DMEM高糖培养基(Bio-Channel)+10%FBS(Bio-Channel),在37℃,5%二氧化碳培养箱中培养,待细胞长满后铺板加药处理。
(2)实验分组和处理
将野生型和三种突变型(Y1320H、G1634R单突变和双突变)HEV复制模型分别以每孔3×104个细胞接种到48孔板中,细胞贴壁24h后,分组加药处理,每组2个复孔,分组如下:
a、实验组:加Vidofludimus处理(培养基中的终浓度为0.04、0.2、1、5、20μM)72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(3)实验方法
将上述2个分组分别处理72h后,弃去细胞培养上清,利用4%多聚甲醛室温固定30min,PBS洗一次,0.3%TritonX-100对细胞打孔15min。5%脱脂牛奶室温封闭1h后,以兔源抗HEV ORF2蛋白(稀释比例1:3000,本实验室保存,Rabbit)为一抗,4℃孵育过夜,FITC标记山羊抗兔IgG(1:500,Jackson,111-585-003)为二抗,室温孵育1h,PBS洗三次。用Hoechst(1:500,invitrogen,33342)荧光染料染细胞核,然后用倒置荧光显微镜(Olympus)进行拍照,分析红色HEV ORF2蛋白表达水平的影响。并通过Image J对HEV ORF2蛋白阳性率进行分析,计算出其IC50。
(4)结果
实验结果如图4所示。由图4可知,Vidofludimus处理后,相比于空白对照组(1%DMSO),HEV ORF2蛋白表达降低,其IC50见表8。

实施例5Vidofludimus抑制类器官中HEV RNA复制的体外实验
(1)将包埋于基质胶中的类器官HEV复制模型以每孔3×104个细胞接种到24孔板中,高级DMEM/F12培养基(Invitrogen)培养24h后,分组加药处理,分组如下:
a、实验组:加Vidofludimus处理(培养基中的终浓度为25μM)72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(2)实验方法
将上述2个分组分别处理72h后,利用TRIzol法提取类器官HEV RNA定量,分析Vidofludimus对类器官中HEV RNA复制水平的影响。
(3)结果
实验结果如图5所示。由图5可知,Vidofludimus处理后,相比于空白对照组(1%DMSO),HEV RNA复制降低。
实施例6Vidofludimus抑制不同捐赠者肝脏类器官HEV-p6Gluc复制模型的作用
(1)实验分组和处理
将类器官HEV复制模型分组加药处理,分组如下:
a、实验组:加不同浓度的Vidofludimus进行处理72小时。
b、空白对照组:加与a组实验组1%DMSO处理。
(2)实验方法
将上述2个分组分别处理72h后,收取上清,用高斯荧光素酶检测试剂盒检测高斯荧光素酶活性,分析Vidofludimus在不同捐赠者肝脏类器官中抑制HEV复制子复制情况。
(4)结果
实验结果如图6所示。由图6可知,Vidofludimus处理后,相比于空白对照组(1%DMSO),HEV复制子***复制显著降低。
由以上实施例可知,本发明提供了一种Vidofludimus在制备戊型肝炎药物中的应用。本发明通过实验证明Vidofludimus可以抑制HEV RNA复制和HEV ORF2蛋白的表达,并且对临床利巴韦林治疗失败相关的HEV突变毒株同样具有抗病毒效果。因此Vidofludimus可以为治疗慢性戊型肝炎患者提供一种新的药物和治疗思路,也可以为临床利巴韦林耐药的HEV患者治疗提供新方案。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (9)
1.Vidofludimus在制备戊型肝炎药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述戊型肝炎由HEV引起。
3.根据权利要求2所述的应用,其特征在于,所述HEV为正戊肝病毒属病毒。
4.根据权利要求3所述的应用,其特征在于,所述正戊肝病毒属病毒包括正戊肝病毒属A种、B种、C种和D种。
5.一种用于治疗戊型肝炎的药物,其特征在于,所述药物包含Vidofludimus。
6.根据权利要求5所述的药物,其特征在于,所述药物中Vidofludimus的浓度为0.04~20μmol/L。
7.根据权利要求6所述的药物,其特征在于,所述药物中还含有药剂学能接受的辅料和载体。
8.根据权利要求7所述的药物,其特征在于,所述辅料和载体为稀释剂、赋形剂、填充剂、粘合剂、湿润剂、崩解剂、吸收促进剂、表面活性剂、吸附载体、润滑剂中的一种或多种。
9.根据权利要求8所述的药物,其特征在于,所述药物的剂型包括注射液、片剂、粉剂、颗粒剂、胶囊、口服液、膏剂、霜剂。
