Abstract
本发明提供了一种全营养配方食品及应用。所述全营养配方食品,按重量100g计包括蛋白质20~40g、碳水化合物30~40g、脂肪10~25g、膳食纤维10~15g、复合维生素2~8g和复合矿物质2~8g。其中,所述膳食纤维包括水溶性人参膳食纤维。本发明通过在全营养配方食品中引入人参水溶性膳食纤维(SDF),促进短链脂肪酸的生成,对肠道菌群有益,搭配其他的营养成分的摄入,保证机体消化吸收良好且营养充足,有效改善营养不足的情况。

技术领域
本发明涉及特殊医学用途食品领域,具体涉及一种全营养配方食品及应用。
背景技术
营养不良作为一种普遍的衰弱性疾病,会导致人体免疫力低下、代谢异常以及患病风险增加,常发生在住院患者人群中。特殊医学用途配方食品(简称特医食品)作为向住院患者提供营养支持的一种肠内营养方式,广泛应用于临床中。特医食品是为了满足进食受限、消化吸收障碍、代谢紊乱或者特定疾病状态人群对营养素或者膳食的特殊需要,专门加工配制而成的一种配方食品。特医食品作为病人的“饭”给患者提供必须的能量和营养支持,可分为三类:全营养配方食品、非全营养配方食品、特定全营养配方食品,其中全营养配方食品可作为单一营养来源满足目标人群营养需求。
但目前国内的全营养配方食品种类较少,大部分依赖进口,无法满足临床需求,因此根据相应的标准,亟需开发出一种全营养配方食品,有效改善营养不良人群的生理需要和营养需求,促进特医食品的产业发展。
发明内容
有鉴于此,本发明的目的在于提供一种全营养配方食品及应用。所述全营养配方食品可以提高患者的健康水平,易于患者消化吸收,维持期正常生理代谢活动。
为达到此目的,本发明采用以下技术方案:
第一方面,本发明提供一种全营养配方食品,按重量100g计包括蛋白质20~40g、碳水化合物30~40g、脂肪10~25g、膳食纤维10~15g、复合维生素2~8g和复合矿物质2~8g;
所述膳食纤维包括水溶性人参膳食纤维。
优选地,按全营养配方食品100g计,所述蛋白质包括乳清蛋白10~30g,大豆蛋白10~30g。
优选地,所述碳水化合物为麦芽糊精。
优选地,按全营养配方食品100g计,所述脂肪包括MTC粉15~20g,亚麻籽油粉10~15g。
优选地,按全营养配方食品100g计,所述复合维生素包括维生素A400~800mg RE、维生素D 5~10μg、维生素E10~15mg、维生素K1 30~80μg、维生素B1 1~1.5mg、维生素B21~1.5份mg、维生素B3 7~15mg、维生素B53~5mg、维生素B6 1~1.5mg、维生素B9 100~400μg、维生素B12 1~2.4μg、维生素C 50~100mg、生物素20~40μg。

与现有技术相比,本发明的有益效果为:
本发明通过在全营养配方食品中引入人参水溶性膳食纤维(SDF),其可以在结肠的菌群作用下发酵成短链脂肪酸,从而有效调节肠道菌群,搭配其他的营养成分(包括蛋白质、碳水化合物、脂肪、复合维生素和复合矿物质)的摄入,保证机体消化吸收良好且营养充足,有效改善营养不足的情况。

下面将结合本发明实施例,对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
针对现有技术中,全营养配方食品种类较少无法满足临床需求的问题,本发明提供了一种全营养配方食品,按总量100g计包括:膳食纤维10~15g、蛋白质20~40g、碳水化合物30~40g、脂肪10~25g、复合维生素2~8g和复合矿物质2~8g。
其中,所述膳食纤维包括水溶性人参膳食纤维,在本发明中,所述人参膳食纤维是以提取完活性成分之后的人参渣为原料,进行酶解,纯化所得,富含糖类、蛋白质及人体所必需的氨基酸。
肠道作为人体消化吸收的重要场所,已有多项研究表明,人体营养状况与肠道菌组成有着密切的关系,肠道微生物的组成是动态的,受饮食特性的功能状态影响,当机体发生营养不良时,肠道菌群会发生变化,有益菌减少,有害菌增多。而本发明通过在全营养配方食品中引入人参水溶性膳食纤维(SDF),其可以在结肠的菌群作用下发酵成短链脂肪酸,从而有效调节肠道菌群,搭配其他的营养成分(包括蛋白质、碳水化合物、脂肪、复合维生素和复合矿物质)的摄入,保证机体消化吸收良好且营养充足,有效改善营养不足的情况。
所述乳清蛋白和大豆蛋白作为优质蛋白,更易被机体吸收,可以快速提高血液中氨基酸的含量,俗称"快蛋白",能够快速释放氨基酸而使血液中的氨基酸含量增加,从而促进蛋白质合成反应,保证机体可以获得稳定、持续的蛋白质供应。因此,在本发明的一些实施方案中,所述全营养配方食品按总量100g计,蛋白质包括乳清蛋白10~30g,大豆蛋白10~30g。
碳水化合物作为机体代谢不可缺少的基础营养素之一,不仅是营养物质,而且有些还具有特殊的生理活性,对于机体的正常运转具有极其重要的意义。特医食品中最为常用的碳水化合物包括麦芽糊精、蔗糖或葡萄糖中的任意一种或多种。其中,麦芽糊精是以淀粉或者淀粉质为原料,经酶轻度水解、精制、喷雾干燥而成的不含游离淀粉的淀粉衍生物,包括多种水解度不同的衍生物,应用更加广泛。在本发明的一些实施方案中,所述碳水化合物优选麦芽糊精。
中链脂肪酸(MCT)通常指由6~12碳链脂肪酸所形成的甘油三酯,其具有高能和易吸收的特点,被广泛用于食品行业。亚麻籽是亚麻科亚麻属一年生或多年生草本植物亚麻的种子,由其压榨获取的油中含有的人体必需脂肪酸-α-亚麻酸含量极高。因此,本发明的一些实施方案中,所述全营养配方食品按总量100g计,脂肪包括MCT粉15~20g,亚麻籽油粉10~15g。
维生素可以提供机体所需的营养成分,保证营养均衡,因此,在本发明的一些实施方案中,所述全营养配方食品按总量100g计,复合维生素包括维生素A 400~800mg RE、维生素D 5~10μg、维生素E 10~15mg、维生素K130~80μg、维生素B1 1~1.5mg、维生素B2 1~1.5份mg、维生素B3 7~15mg、维生素B5 3~5mg、维生素B6 1~1.5mg、维生素B9 100~400μg、维生素B121~2.4μg、维生素C 50~100mg、生物素20~40μg。其中,维生素A的来源包括醋酸维生素A,维生素D的来源包括胆钙化醇,维生素E的来源包括dl-α-醋酸生育酚,维生素K1的来源包括植物甲萘醌,维生素B1的来源包括盐酸硫胺素,维生素B2的来源包括核黄素,维生素B3的来源包括烟酰胺,维生素B5的来源包括D-泛酸钙,维生素B6的来源包括盐酸吡哆醇,维生素B9的来源包括叶酸,维生素B12的来源包括氰钴素,维生素C的来源包括L-抗坏血酸,生物素的来源包括D-生物素-1%。
矿物质是机体极为重要的营养元素,必须从食物中获得足量的矿物质,才能维持良好的健康状态。因此,本发明的一些实施方案中,所述全营养配方食品按总量100g计,复合矿物质包括钠500~1400mg、钾600~2000mg、铜400~800μg、镁100~300mg、铁6~12mg、锌6~12mg、锰1.5~4.5mg、钙500~1000mg、磷300~700mg、碘40~120μg、氯500~2000mg、硒20-60μg。
按照本发明,在一些实施方案中,所述全营养配方食品按总量100g计,包括麦芽糊精30~40g,乳清蛋白10~30g,大豆蛋白10~30g,MTC粉15~20g,亚麻籽油粉10~15g,水溶性人参膳食纤维10~15g,维生素A400~800mg RE,维生素D5~10μg,维生素E10~15mg,维生素K1 30~80μg,维生素B1 1~1.5mg,维生素B2 1~1.5份mg,维生素B3 7~15mg,维生素B5 3~5mg,维生素B6 1~1.5mg,维生素B9 100~400μg,维生素B12 1~2.4μg,维生素C50~100mg,生物素20~40μg,钠500~1400mg,钾600~2000mg,铜400~800μg,镁100~300mg,铁6~12mg,锌6~12mg,锰1.5~4.5mg,钙500~1000mg,磷300~700mg,碘40~120μg,氯500~2000mg,硒20-60μg。
在本发明的另一些实施方案中,所述全营养配方食品可以根据实际需要适当添加其他的营养组分,具体可选自铬、钼、氟、胆碱、肌醇、牛磺酸、左旋肉碱或核苷酸中的任意一种或多种。其中,铬可维持机体生长发育和调节血糖,钼解毒,氟可参与钙和磷的代谢,胆碱促进脑部发育和提高记忆力,肌醇可降低胆固醇,促进毛发生长,牛磺酸促进脑发育、保护视网膜,左旋肉碱提高运动耐力,促进疲劳恢复,保护心脑血管,核苷酸调节代谢。
具体地,在本发明的一些实施方案中,按全营养配方食品100g计,包括铬5-150μg、钼18-150μg、氟0-0.7μg、胆碱70-557mg、肌醇14-469mg、牛磺酸0-67mg、左旋肉碱4.2mg—无上限说明、核苷酸7mg—无上限说明。
在本发明中,所述全营养配方食品的剂型可以为粉剂、注射剂、片剂、颗粒剂或胶囊中的任意一种。
上述全营养配方食品按比例将各组分混合均匀即可,在本发明的一些实施方案中,优选将蛋白质、脂肪、碳水化合物和膳食纤维混合1~1.5h后,再添加复合维生素和复合矿物质充分混匀。在本发明中,所述混合在混匀机中进行。
为了进一步说明本发明,下面通过以下实施例进行详细说明。本发明以下实施例中所用的实验原料均可从市场上购买或者按照本领域技术人员熟知的常规制备方法制备得到。其中,人参购买自吉林省抚松县。
制备例1
本制备例提供一种水溶性人参膳食纤维,制备方法如下:
将人参粉碎至10-20目的颗粒或1-3mm的薄片,先经过80℃纯水浸提3-5次,得到的人参渣烘干后粉碎,用pH=8.2的MES-Tris缓冲液配制成固液比为(1:30)的人参渣混悬液,加入2.5mL耐高温α淀粉酶在1.5L混悬液中,使用电磁搅拌器300rmp,100℃搅拌30min,冷却至60℃,再加入5.0mL蛋白酶继续搅拌混合物60min,然后将混合物的pH值调整到4.5,再加入10.0mL淀粉葡糖苷酶搅拌60min。酶解后反应物8000×g离心20min,收集上清液即为水溶性人参膳食纤维(SDF),再经浓缩、烘干、打粉后备用。
实施例1
本实施例提供一种含SDF的全营养食品配方和不含SDF的基础配方,具体配方如下表1所示(表1中的100g指的是全营养配方的总质量):

按照上述配方,采用等量递加法,将乳清蛋白、大豆蛋白、MTC粉、亚麻籽油粉、任选的人参水溶性膳食纤维和麦芽糊精混合1h后,然后添加复合维生素和复合矿物质继续充分混匀,分别得到基础配方和含SDF的全营养配方。
动物分组及模型建立
雄性Wistar大鼠SPF级,体质量180-200g,适应性喂养7d,遵循动物实验伦理建立营养不良的大鼠模型,大鼠42只,于恒温恒湿的SPF级环境内,适应性喂养7d,随机分为正常组(K),模型组(M),阳性药组(Y),基础配方低剂量组(F1D),基础配方高剂量组(F1G);SDF配方低剂量组(F2D),SDF高剂量组(F2G),共7组,每组6只。所有组都正常饮水(121℃,15min高温高压灭菌),正常组每日给予(蛋白质12.5%,脂肪4%,灰分3.89%,20g/72.02kcal/只),模型组每日给予(蛋白质10%,脂肪3%,灰分3.11%,10g/36.01kcal/只)。连续饲养5周,造模期间每日测量大鼠的体重,造模最后一天眼眶取血,检测血清白蛋白,总胆固醇,判断模型是否建立成功。
本试验通过体重,血清白蛋白和总胆固醇作为评价营养不良模型是否建立成功的标志,测试结果如表2所示(与正常组相比,**p<0.01为差异极显著):

由表2可知,模型组大鼠血清白蛋白含量和体质量与正常对照组相比显著降低(P<0.01),同时模型组大鼠总胆固醇的含量与正常对照组相比明显降低,以上结果表明,营养不良大模型构建成功。
给药方法
按照《中国居民膳食营养素参考摄入量》推荐人体每天需摄入2300kcal,动物与人体的每公斤剂量折算系数表计算,折算成大鼠剂量,设置基础配方低剂量组和SDF配方低剂量组每天按10g·kg-1的浓度灌胃相应配方,基础配方高剂量组和SDF配方高剂量组每天按20g·kg-1的浓度灌胃相应配方。阳性药采用佳膳优选,给药浓度1.165g/mL。正常组、模型组分别灌胃蒸馏水;配方食品组每天按10mL/(kg·bw)灌胃1次,连续30d。
上述各组别在灌胃期间大鼠体重的变化情况如图1所示,可以看出随着饲养天数的逐渐增加,各组配方食品均会导致对大鼠体重增加,与模型组相比,以F2G组和Y组体重增长最明显。
血液和组织样本采集
末次给药后,断粮不断水24h。麻醉,心脏取血处死,取血,肝,脾,胸腺,称重。将血液室温静置30min,然后在4℃条件下离心(2500rmp/min)30min,分离血清,保存于-80℃。
以胸腺指数、脾脏指数和脾脏指数作为判断全营养配方食品对机体细胞免疫和体液免疫功能强弱的初步观察指标。结果如下表3所示(每列相同上标值表示无显著性差异(P>0.05),不同上标值有显著差异(P<0.05)):

由表3数据可知,与模型组相比,F2G组的胸腺指数显著升高(P<0.05),其余各组的胸腺指数有升高但无显著性差异(P>0.05);各试验组的脾脏指数相比于模型组有升高但无显著性差异(P>0.05);F1G和F3G组显著升高了模型组大鼠的肝脏指数(P<0.05),各组全营养配方食品均可提高模型组大鼠的胸腺指数。
血细胞的测定
取适量血液于加有抗凝剂的离心管内,上机前血液颠倒混匀采用全自动血细胞分析仪检测。
测试结果如下表4所示(每列相同上标值表示无显著性差异(P>0.05),不同上标值有显著差异(P<0.05)):

由表4数据可知,与模型组相比,F2D组显著提高血液中的红细胞总数(P<0.01),F2G组能显著提高血红蛋白的含量(P<0.01)。白细胞总数,淋巴细胞总数,中性粒细胞总数是机体用来抵抗细菌、病毒的免疫细胞。与模型组相比,F2G的白细胞和中性粒细胞总数显著升高(P<0.05)。
生化指标的测定
测定血清中ALB,PA(上海酶联生物科技有限公司),TP,TC,TG(北京瑞达恒辉科技发展有限公司),IgA,IgG,IgM(上海酶联生物科技有限公司)指标检测按照试剂盒提供的说明书进行规范操作。
不同组别配方食品对大鼠血清总蛋白,白蛋白和前白蛋白的影响
血清白蛋白(ALB)作为血液中最丰富的蛋白质,具有为组织提供营养支撑、抗氧化、运输和排毒的作用,在发生炎症和营养不良状态下,血清白蛋白的水平都会下降,因此是营养不良评估最广泛的指标;血清总蛋白(TP)可以反映肝脏的合成功能,血清前白蛋白(PA)作为一种肝脏合成的糖蛋白,在营养状况评估方面是两项很灵敏的指标。
测试结果如图2(a)~(c)所示,可以看出与正常组比,模型组大鼠血清TP、ALB含量显著降低(P<0.05)。与模型组相比,F1D、F2G组极显著升高ALB的含量(P<0.001),Y、F2D组显著升高ALB的含量(P<0.01),F1G组显著升高了ALB的含量(P<0.05),F2D和F2G组显著升高了血清中TP的含量(P<0.05),同时F2D、F2G组显著升高PA的含量(P<0.05,P<0.01),综合评判,SDF对提高大鼠血清蛋白水平有显著作用。
不同组别配方食品对免疫球蛋白的影响
免疫球蛋白A、G和M是由抗原受到刺激之后所产生的动物蛋白,其中IgM是在初次免疫应答中首先产生的免疫球蛋白,占血清中免疫球蛋白总量的5%~10%;IgA具有中和内毒素,抗菌的作用,占血清免疫球蛋白的10%~15%;IgG是血清中免疫球蛋白的主要成分,约占总含量的75%,具有中和病毒,抗菌和免疫调节的功能。研究证实,营养状况与免疫功能有关,营养不良可导致某些免疫功能的丧失。
测试结果如图3(a)~(c)所示,可以看出与正常组比,模型组大鼠血清IgG和IgM含量显著降低(P<0.001,P<0.01),同时IgA的含量也低于正常组但无统计学差异。与模型组相比,各营养配方组均极显著升高IgG的含量(P<0.001),F1D、F1G组显著升高了IgM的含量(P<0.05);F2G组显著升高IgA和IgM的含量(P<0.05,P<0.01)。
不同组别配方食品对短链脂肪酸的影响
配制乙酸、丙酸、丁酸、异丁酸、戊酸、异戊酸、己酸的梯度浓度标准液,制作标准曲线。取一定质量的大鼠粪便,按1:3(W/V)的比例加入去离子水中,涡旋混合,4℃提取12h,再15000rpm离心10min(4℃)。吸取上清液200μL,加入40μL含有2g/L内标2-乙基丁酸溶液(TE007025ML,国药化学试剂有限公司,溶于25%的偏磷酸中),涡旋混合,并在冰浴中放置35min,4℃离心15000rpm,10min。上清液经0.22μm滤膜过滤,在Agilent7890A气相色谱***上进行测试。DB-FFAP色谱柱(30m×0.32mm×0.32μm),载气为氮气,流量2.2mL/min,FID检测器,进样温度250℃,初始温度为60℃,10℃/min升温到170℃,再以8℃/min升至212℃。
测试结果如图4所示,可以看出与模型组相比,F2D和F2G组可以促进短链脂肪酸的生成,对肠道菌群有益。表明本发明提供的配方中,通过水溶性膳食纤维(SDF)搭配其他的营养成分的摄入,有利于保证机体消化吸收良好且营养充足,有效改善营养不足的情况。

Claims (10)
1.一种全营养配方食品,其特征在于,按重量100g计包括蛋白质20~40g、碳水化合物30~40g、脂肪10~25g、膳食纤维10~15g、复合维生素2~8g和复合矿物质2~8g;
所述膳食纤维包括水溶性人参膳食纤维。
2.根据权利要求1所述的全营养配方食品,其特征在于,按全营养配方食品100g计,所述蛋白质包括乳清蛋白10~30g,大豆蛋白10~30g。
3.根据权利要求1所述的全营养配方食品,其特征在于,所述碳水化合物为麦芽糊精。
4.根据权利要求1所述的全营养配方食品,其特征在于,按全营养配方食品100g计,所述脂肪包括MTC粉15~20g,亚麻籽油粉10~15g。
5.根据权利要求1所述的全营养配方食品,其特征在于,按全营养配方食品100g计,所述复合维生素包括维生素A 400~800mg RE、维生素D 5~10μg、维生素E10~15mg、维生素K1 30~80μg、维生素B1 1~1.5mg、维生素B2 1~1.5份mg、维生素B3 7~15mg、维生素B5 3~5mg、维生素B6 1~1.5mg、维生素B9 100~400μg、维生素B12 1~2.4μg、维生素C 50~100mg、生物素20~40μg。
6.根据权利要求1所述的全营养配方食品,其特征在于,按全营养配方食品100g计,所述复合矿物质包括钠500~1400mg、钾600~2000mg、铜400~800μg、镁100~300mg、铁6~12mg、锌6~12mg、锰1.5~4.5mg、钙500~1000mg、磷300~700mg、碘40~120μg、氯500~2000mg、硒20-60μg。
7.根据权利要求1所述的全营养配方食品,其特征在于,按重量100g计包括麦芽糊精30~40g,乳清蛋白10~30g,大豆蛋白10~30g,MTC粉15~20g,亚麻籽油粉10~15g,水溶性人参膳食纤维10~15g,维生素A400~800mg RE,维生素D5~10μg,维生素E10~15mg,维生素K1 30~80μg,维生素B1 1~1.5mg,维生素B2 1~1.5份mg,维生素B3 7~15mg,维生素B5 3~5mg,维生素B6 1~1.5mg,维生素B9 100~400μg,维生素B12 1~2.4μg,维生素C50~100mg,生物素20~40μg,钠500~1400mg,钾600~2000mg,铜400~800μg,镁100~300mg,铁6~12mg,锌6~12mg,锰1.5~4.5mg,钙500~1000mg,磷300~700mg,碘40~120μg,氯500~2000mg,硒20-60μg。
8.根据权利要求1所述的全营养配方食品,其特征在于,还包括铬、钼、氟、胆碱、肌醇、牛磺酸、左旋肉碱或核苷酸中的任意一种或多种。
9.根据权利要求8所述的全营养配方食品,其特征在于,按全营养配方食品100g计,包括铬5-150μg、钼18-150μg、氟0-0.7μg、胆碱70-557mg、肌醇14-469mg、牛磺酸0-67mg、左旋肉碱4.2mg—无上限说明、核苷酸7mg-无上限说明。
10.根据权利要求1所述的全营养配方食品,其特征在于,剂型为粉剂、注射剂、片剂、颗粒剂或胶囊。
