Description

Oral liquid for clearing away plague and toxic materials and preparation method thereof
Technical Field
The invention belongs to the technical field of veterinary medicines, and in particular relates to a pestilence-clearing and detoxifying oral liquid and a preparation method thereof.
Background
The oral liquid for clearing away the plague and detoxifying is a traditional Chinese veterinary medicine compound preparation which is most commonly used in prevention and control of poultry diseases in recent years, has the efficacy of clearing away the heat-evil and expelling superficial evils and mainly treats exogenous fever of the poultry. The prescription consists of radix isatidis, radix scutellariae, fructus forsythiae, fructus gardeniae, radix scrophulariae, radix rehmanniae and other medicinal materials. Traditional Chinese medicine suggests that fowl biological diseases are usually caused by invasion of heat toxin from shallow to deep, from exterior to interior and from heart lung to lower body, and the disease is caused by excessive heat evil in qi system, and the heat evil in blood system is easy to damage body fluid, so animals are ill: the general manifestations are: thirst, difficulty in acting, and stiffness of the extremities. The morbidity of animals is mainly acute, and the mortality rate is relatively high. Radix Isatidis in the oral liquid prescription for clearing away heat and toxic materials can detoxify and moisten throat; radix Scutellariae has effects of clearing lung-heat and purging heat; fructus forsythiae has effects of clearing heat and resisting bacteria, and eliminating upper middle-upper coke gas; fructus Gardeniae is used as adjuvant medicine for eliminating triple energizer heat syndrome, purging pathogenic fire and removing toxic substances; the brown participation of Sheng Di Huang can cool blood and reduce fire, thereby making up the kidney, and being an auxiliary drug. Clinical application in many countries proves that the preparation has obvious curative effect on prevention and control of poultry virus and germ, but the traditional production process has the defects of low extraction rate of medicinal material active ingredients, high loss in impurity removal and concentration links, high energy efficiency and the like. Meanwhile, due to the long-term use of various chemicals, antibiotics and the like, animals are extremely easy to generate drug resistance, although the dosage is continuously increased, the curative effect is not obviously improved or even continuously reduced.
For example, the traditional process generally has the defects of low dissolution rate of active ingredients extracted from decoction pieces, high loss rate of active ingredients except heterocyclic joints, unstable thermosensitive ingredients in a concentrating link, high loss rate of ingredients and the like, and also has the defects of high resource loss, high energy consumption, serious environmental pollution, high production cost and the like, so that the clinical and efficient exertion and the market competitiveness of the prescription are limited.
Therefore, how to innovate and improve the formulation of the oral liquid for clearing plague, and change the traditional preparation process, so as to provide a novel preparation process of the veterinary drug with simple and convenient process, high active ingredient yield, short extraction time, energy conservation and environmental protection, so as to enhance the exertion of clinical efficacy, which is a problem to be solved urgently.

2.2 test results
2.2.1 particle size of the crushed material
The yield of the active ingredients of the traditional Chinese medicinal materials is closely related to whether the traditional Chinese medicinal materials are crushed and extracted or not, the crushing meshes of the medicinal materials are different, and the yields of the active ingredients are also different, which is probably caused by the difference of the contact areas of the solvent and the medicinal materials. As the number of pulverized medicinal materials increases, the dissolution rate of active ingredients increases, because the solvent can be more fully contacted with the medicinal materials. The test results show that: when the crushing fineness is 100 meshes, the baicalin yield in the liquid medicine reaches the maximum value, but the granularity of the medicinal materials is too small, so that the problem of removing impurities in the later period is solved, and after comprehensive comparison, the crushing fineness of the medicinal materials is selected to be better than that of the medicinal materials passing through a 100-mesh sieve. The test results are shown in table 2:
TABLE 2 influence of crushing particle size on baicalin content
Crushing granularity (mesh) 50 80 100 120 200
Content (mg/mL) 6.79 6.95 6.98 6.82 6.55
2.2.2 extraction time
The extraction time in the extraction process of the traditional Chinese medicine is closely related to the yield of the active ingredients, and the longer the extraction time is, the longer the solvent is contacted with the medicine, the larger the dissolution rate of the active ingredients is, because the solvent can be contacted with the medicine more fully. Meanwhile, the cell walls can be broken gradually by adopting stage heating, the dissolution efficiency is improved, and the test result is obtained: when the extraction time is 60min and the temperature is raised in stages, the baicalin content of the obtained liquid medicine reaches the maximum value, and the test results are shown in the following table 3:
TABLE 3 influence of extraction time on baicalin content
Extraction time (min) 30 40 50 60 Example 1
Content (mg/mL) 5.23 5.65 6.04 6.65 6.98
2.2.3 extraction temperature
The yield of the active ingredients extracted from the traditional Chinese medicine is closely related to the extraction time and the extraction temperature, and the more the extraction time is sufficient, the more the active ingredients are dissolved out at a certain temperature. Meanwhile, as the extraction temperature is increased, the dissolution rate of the active ingredients is gradually increased, because the higher the temperature is, the more active ingredients in the medicinal materials move more severely and are more easily dissolved. Meanwhile, when the temperature is raised to 80-90 ℃, the content change is not obvious, but a certain dissolution rate can be improved by adopting a stage temperature raising method, so that the test results show that: when the temperature reaches above 90 ℃, the yield of the active ingredient tends to be balanced, so that the extraction temperature of 90 ℃ is selected to be better. As shown in table 4 below:
TABLE 4 influence of extraction temperature on baicalin content
Extraction temperature (. Degree. C.) 60 70 80 90 Example 1
Content (mg/mL) 5.69 5.88 6.44 6.65 6.98
2.2.4 Water addition
In the extraction process of the traditional Chinese medicinal materials, the water adding amount is extremely related to the yield of the active ingredients, and researches prove that the yield of the active ingredients is gradually increased along with the increase of the water adding amount, and the more the solvent amount is, the more the solvent amount is in full contact with the medicinal materials, so that the active ingredients are more soluble. As can be seen from the data shown in table 5 below, when the water addition amount reaches 10 times or more, the yield of the active ingredient tends to be maximum, and the factors such as energy consumption are comprehensively considered, and the test results show that: the water adding amount is 10 times of that of the medicinal materials, and is better.
TABLE 5 influence of the amount of added water on the baicalin content
Water addition multiple (mL) 600 1000 1600 2000 3000
Content (mg/mL) 4.98 5.24 5.68 6.15 6.23
The research shows that when plant cells are broken, the fiber structure is destroyed, the effective components in the cells can be fully exposed, and then the effective components are directly contacted with the organic reagent for extraction and separation, which shortens the extraction and separation time greatly compared with the extraction of Chinese medicinal decoction pieces, thereby improving the dissolution rate of the Chinese medicaments and reducing the resource loss.
In summary, the optimal technical parameters of the crushing and extracting process of the invention are as follows: pulverizing the medicinal materials, sieving with 100 mesh sieve, extracting for 60min with three-stage heating, extracting at 90deg.C with water amount 10 times of that of the medicinal materials. Under the condition of the technological parameters, the yield of the active ingredients in the obtained liquid medicine is highest.
