When the formulation of the present invention is a powder or a spray, lactose, talc, silica, aluminum hydroxide, calcium silicate, polyamide powder, or a mixture thereof can be used as a carrier component, and particularly, when the formulation is a spray, a propellant such as chlorofluorocarbon, propane/butane, or dimethyl ether can be contained.
In the case where the dosage form of the present invention is a solution or emulsion, as a carrier ingredient, a solvent, a solubilizer or an emulsifier may be used, and for example, water, ethanol, isopropanol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1, 3-butyl glycol oil, and in particular, cottonseed oil, peanut oil, corn seed oil, olive oil, castor oil, and sesame oil, aliphatic esters of glycerin, polyethylene glycol or fatty acid esters of sorbitan may be used.
When the formulation of the present invention is a suspension, as a carrier component, a liquid diluent such as water, ethanol or propylene glycol, a suspending agent such as ethoxylated isostearyl alcohol, polyoxyethylene sorbitol ester or polyoxyethylene sorbitan ester, microcrystalline cellulose, aluminum metahydroxide, bentonite, agar or tragacanth, and the like can be used.
On the other hand, when the formulation of the present invention is a capsule, the formulation may be in the form of an alginate (alginate) capsule, an agar (agar) capsule, a gelatin (gelatin) capsule, a wax (wax) capsule, or a double capsule, but is not particularly limited thereto.
The cosmetic composition of the present invention may further contain commonly used adjuvants such as a hydrophilic or lipophilic gelling agent, a hydrophilic or lipophilic active agent, a preservative, an antioxidant, a solvent, an aromatic agent, a filler, a blocking agent, a pigment, a deodorant, a dye, and the like, in addition to the giant knotweed root extract and 2,3-butanediol or the giant knotweed root extract and the deamidated collagen.
In order to achieve the above object, another aspect of the present invention provides use of a composition comprising a Polygonum Cuspidatum root extract (Polygonum Cuspidatum root extracts) as an active ingredient in the manufacture of a cosmetic composition for enhancing skin elasticity or improving wrinkles.
In order to achieve the above objects, another aspect of the present invention provides a pharmaceutical external preparation composition for skin elasticity enhancement or wrinkle improvement, comprising an extract of polygonum cuspidatum root as an active ingredient.
The medical external product composition is a composition prepared by mixing the giant knotweed rhizome extract and 2,3-butanediol or deamidated collagen.
The terms "polygonum cuspidatum root extract", "2, 3-butylene glycol", "deamidated collagen", "skin elasticity enhancement" and "wrinkle improvement" used in the present invention are as described above.
In addition to the above components, the pharmaceutical external product composition of the present invention may further comprise pharmaceutically acceptable carriers, excipients or diluents as necessary. The pharmaceutically acceptable carrier, excipient or diluent is not limited as long as the effect of the present invention is not impaired, and examples thereof include fillers, extenders, binders, wetting agents, disintegrants, surfactants, lubricants, sweeteners, flavoring agents, preservatives, and the like.
The pharmaceutical external preparation composition of the present invention may be exemplified by disinfectant cleaners, bath lotions, ointment liquids, wet tissues, coating agents, and the like, but is not limited thereto, and the method of preparation, volume, method of use, constituent components, and the like of the pharmaceutical external preparation can be appropriately selected from techniques known in the art.
In order to achieve the above objects, in another aspect, the present invention provides a use of a composition comprising an extract of polygonum cuspidatum root as an active ingredient for manufacturing a pharmaceutical external product for skin elasticity enhancement or wrinkle improvement.
In order to achieve the above objects, according to still another aspect of the present invention, there is provided a method for enhancing skin elasticity or improving wrinkles, comprising the step of administering a composition comprising an extract of polygonum cuspidatum root to a subject.
The composition is prepared by mixing rhizoma Polygoni Cuspidati extract with 2,3-butanediol or deamidated collagen.
The terms "polygonum cuspidatum root extract", "2, 3-butanediol", "deamidated collagen", "skin elasticity enhancement" and "wrinkle improvement" used in the present invention are as described above.
The term "administering" as used herein means introducing the composition of the present invention into a patient by any suitable method, and the route of administration of the composition may be any general route that can reach the target tissue. The route of administration of the composition can be performed by applying the composition to the skin based on the property of the composition of the present invention that it is effective in enhancing skin elasticity or improving wrinkles.
The composition of the present invention may be administered daily or intermittently, and the number of administrations per day may be divided into 1 or 2 to 3. In addition, the composition of the present invention can be used alone or in combination with other drugs for the purpose of skin elasticity enhancement or wrinkle improvement. In view of all the above, it is preferable to add an amount that achieves the maximum effect with the minimum amount in the case where there is no side effect, and the amount to be added can be easily determined by those skilled in the art.
The term "individual" as used herein means any animal including mice, chevrons, and domestic animals, including humans, in which wrinkles or skin wrinkles occur or may occur. Specifically, mammals including humans can be used.
Effects of the invention
The composition for enhancing skin elasticity or improving wrinkles, which comprises the polygonum cuspidatum root extract and 2,3-butanediol or deamidated collagen as active ingredients, is safe to the skin and has a synergistic effect in promoting collagen synthesis, and therefore, can be used in cosmetics, external pharmaceutical products, and the like for enhancing skin elasticity or improving wrinkles.

Detailed Description
The present invention will be described in more detail below with reference to examples. These examples are intended to illustrate the present invention more specifically, and the scope of the present invention is not limited to these examples.
Example 1: culture of human fibroblasts
Fibroblasts from human foreskin (forekin) were seeded into the bottom of the culture dish, then put in DMEM medium (Dulbecco's modified eagles medium) (DMEM, gibcoBRL, u.s.a.) containing penicillin (100 IU/ml), streptomycin (100 ug/ml), and 10% fetal bovine serum (total bovine serum; FBS, gibcoBRL, u.s.a.) and cultured in an incubator (New Brunswick Scientific, UK) containing 5% carbon dioxide at 37 ℃. When the surface of the culture dish was covered with fibroblasts to an extent of about 70 to 80%, the cells were collected by treating with 0.25% trypsin (Gibco BRL, u.s.a.) and cultured, and a part of the cells were stored frozen in liquid nitrogen for subculture.
Example 2: evaluation of enhancement of collagen Synthesis
The collagen synthesis enhancing effect was evaluated at the cellular level by adding the polygonum cuspidatum root extract, 2,3-butanediol, deamidated collagen, separately or in combination to the culture medium of human-derived fibroblasts. The type i procollagen hydroxy-terminal-extended peptide (PICP), which is typically most synthesized in fibroblasts, was quantitatively determined at the time of evaluation.
Specifically, human fibroblasts were treated at 1x10, respectively 5 The/well was aliquoted into cell culture plates (24 well cell culture plates) and cultured in DMEM containing 10% fetal bovine serum for 24 hours. After the cells were replaced with DMEM containing no serum, each sample was diluted with a culture medium and treated for 48 hours. 20ul of the culture supernatant was removed and the degree of collagen increase at each concentration was measured at 450nm using a PICPEIA KIT (Procollagen type I C-Peptide Enzyme linked immunosorbent assay KIT). Quantitative measurements were performed by plotting PICP standard concentrations, and experiments were performed in 3 cases for each concentration, and the results were averaged and shown. As a positive control group, TGF-. Beta.s (10 ng/ml) were used.
As a result, as shown in fig. 1, when the polygonum cuspidatum root extract, 2,3-butanediol, and deamidated Collagen were treated alone, the expression of Collagen type (Collagen type) 1 was increased, which was a result of satisfying the known effects of the respective raw materials.
However, it was confirmed in fig. 2 that the collagen expression level was significantly increased in the case of the treatment in which the polygonum cuspidatum root extract and 2,3-butanediol or the polygonum cuspidatum root extract and deamidated collagen were mixed, as compared with the case of the single treatment in fig. 1.
Thus, the cosmetic composition of the present invention exhibits an effect of enhancing skin elasticity by increasing the expression level of collagen, and as to the effect, when the polygonum cuspidatum root extract and 2,3-butanediol or deamidated collagen are mixed, the effect is more enhanced than when each is treated separately.
Example 3: compatibility comparison of Polygonum cuspidatum root extract and polyol
In order to evaluate the degree of precipitation or stabilization of the polygonum cuspidatum root extract, the compatibility with 7 types of polyols used in cosmetic materials was evaluated.
Specifically, examples 1 and 2 were prepared by adding 0.5% or 5.0% of the polygonum cuspidatum root extract to 2,3-butanediol and stirring at 900rpm for 1 hour at room temperature.
In addition, comparative examples 1 to 12 were produced in the same manner as in example 1, except that the polyol was different. The difference in stability of the polygonum cuspidatum root extract tends to be more pronounced at low temperatures than at high temperatures or temperature cycling conditions, and therefore the compatibility of examples 1 and 2 and comparative examples 1 to 12 was confirmed after storage for 4 weeks under normal temperature (25 ℃) conditions after stirring. In the case of freezing (-20 ℃ C.), thawing and freezing were repeated 5 times during the period. The stability under each condition was visually observed with emphasis on precipitation and discoloration and shown in tables 1 and 2 below.

As a result, as shown in tables 1 and 2,3-butanediol was most excellent in compatibility with the extract of alpine rush root as described in examples 1 and 2.
The excellent degree of ethoxydiglycol and dimethyl isosorbide are similar, but in the case of ethoxydiglycol, there is a problem of skin irritation when used in a small amount, and when used at a high content (comparative example 10), the color changes to pink, and the color change further deepens as the shelf life becomes longer. Dimethyl isosorbide shows a light pink color when 0.5% of the polygonum cuspidatum root extract is used (comparative example 12), and the degree of discoloration increases with increasing content of the polygonum cuspidatum root extract.
From this, it was confirmed that when the polygonum cuspidatum root extract and 2,3-butanediol were used together, the results were more excellent not only in terms of efficacy but also in terms of the compatibility and stability of the raw materials.
Example 4: evaluation of collagen Synthesis Capacity of Polygonum cuspidatum root extract and various polyols
In addition to 2,3-butanediol, 1,3-butanediol, ethoxydiglycerol, and dimethyl isosorbide, which were evaluated to have excellent compatibility with the polygonum cuspidatum root extract in table 2, were additionally evaluated and the collagen synthesis ability when mixed with the polygonum cuspidatum root extract was confirmed.
As a result, as shown in fig. 3, it was confirmed that when the polygonum cuspidatum root extract and 2,3-butanediol were mixed and used, the collagen synthesis ability was significantly enhanced, but the collagen synthesis ability was rather inhibited in the other mixed sample with polyhydric alcohol.
Production example 1: preparation of essence composition
In order to evaluate the formulation stability of the cosmetic comprising the polygonum cuspidatum root extract and 2, 3-butylene glycol or deamidated collagen, cosmetic compositions were prepared with the components and contents described in table 3 below.
Specifically, in example 3, the raw material 3 was dissolved in the raw material 1 at the composition ratio shown in table 3, and then the raw materials 4 to 10 were added and dissolved. After the dissolution of the raw material 3 was confirmed, the raw materials 4 to 9 were dissolved in this order, and then the raw material 10 was slowly added and stirred uniformly. The essences were prepared by measuring the raw materials 11 to 13 in a separate beaker, dissolving by heating to 40 to 45 ℃, and then gradually adding to the aqueous phase while stirring uniformly for 15 minutes. Comparative examples 13 and 14 were produced in the same manner as in example 3.
Essence type cosmetics (unit: wt%) of the experimental group containing 2, 3-butylene glycol and 1, 3-butylene glycol and the control group containing no butylene glycol were prepared using the components and contents shown in table 3 below.

Example 5: evaluation of formulation stability
The cosmetic compositions of example 3, comparative example 13 and comparative example 14 were evaluated for compatibility. As for the storage conditions, temperature conditions with significant differences among experiments were selected and observed again at normal and low temperatures, and for comparison, additional observation was also performed at high temperature conditions, as in the previous polyol compatibility evaluation.
Specifically, the cosmetic composition was stored in a glass container for 4 weeks at high temperature (45 ℃ C.), normal temperature (25 ℃ C.), and refrigerated storage (0 ℃ C.). In the case of freezing (-20 ℃ C.), thawing and freezing were repeated 5 times. The presence or absence of separation and precipitation of the phase (phase) was visually observed under the respective conditions, and is shown in table 4 below.

As a result, as shown in Table 4, in example 3 using 2,3-butanediol, no precipitation occurred under all storage conditions, and a uniform and stable formulation was maintained. However, in the case of comparative example 13 in which 1, 3-butanediol was used, slight deposition occurred during repetition of the freezing conditions, and the amount of deposition increased with the number of repetitions. In addition, in the case of comparative example 14 in which no polyol was used, precipitation was observed under most of the storage conditions.
The above experimental results confirmed that the polyhydric alcohol contributes to the stability of the formulation of the polygonum cuspidatum root extract, and the stability of the cosmetic can be significantly improved when used together with 2, 3-butylene glycol in the polyhydric alcohol.
Example 6: evaluation of skin stability
In order to confirm the skin stability of the above-mentioned substances as cosmetics, tests were carried out with respect to skin irritation.
Specifically, the subject was 20 healthy adult volunteers having no skin disease at the experimental site, and the I Q Ultra was used TM The patch test units (Chemotechnique MB Diagnostics AB, vellinge, sweden) performed stimulation tests. When the primary stimulation index was calculated using the following calculation formula, a sample exceeding 0.20 (in the case where a positive stimulation reaction corresponding to 1 point was observed at a frequency exceeding 20% of all the subjects or in the case where the reaction corresponds to this case) was judged as a sample that could induce stimulation, and 1% Sodiu m Lauryl Sulfate (CAS Number: 151-21-3) aqueous solution was used as a standard substance. In summary, in the case of the mixed composition comprising the polygonum cuspidatum root extract of the present invention and 2, 3-butylene glycol or deamidated collagen, the skin elasticity-enhancing or wrinkle-improving effect by promoting collagen synthesis is more excellent and the formulation is more stable than the case of including the same as the individual components.
As is apparent from the above description, those skilled in the art can implement the present invention in other specific forms without changing the technical idea or essential features thereof. In this regard, the above-described embodiments are merely exemplary in all aspects, and the present invention is not limited thereto. In the present invention, the scope of the present invention is defined by the meaning of the claims to be described later, and all modifications and variations derived from the equivalent concept are included in the scope of the present invention.

Claims (6)

1. Use of a composition comprising an extract of the root of Polygonum cuspidatum as an active ingredient in the manufacture of a cosmetic for enhancing skin elasticity or improving wrinkles.
2. The use according to claim 1, wherein,
the composition further comprises 2, 3-butylene glycol or deamidated collagen.
3. The use according to claim 2, wherein,
the above composition promotes the synthesis of procollagen type I C-terminal propeptide (PICP) type 1.
4. The use according to claim 1, wherein,
the composition further comprises one or more selected from the group consisting of water, surfactant, humectant, alcohol, thickener, chelating agent, pigment, antiseptic and perfume.
5. Use of a composition comprising an extract of the root of Polygonum cuspidatum as an active ingredient for the manufacture of a pharmaceutical external product for enhancing skin elasticity or improving wrinkles.
6. The use according to claim 5, wherein,
the composition further comprises 2, 3-butylene glycol or deamidated collagen.
