Abstract
本发明提供一种调节免疫炎症微环境水凝胶支架及其制备方法和应用。所述调节免疫炎症微环境水凝胶支架的制备原料包括:阳离子高分子聚合物、交联水凝胶和抗炎细胞因子。本发明通过双功能水凝胶清除引发炎症反应的DAMP和持续释放抗炎细胞因子促进抗炎和组织重塑作用,协同增强对脊髓损伤后免疫炎症微环境的调控,以及更好地促进神经再生和功能恢复。

技术领域
本发明属于医用新材料技术领域,具体涉及一种调节免疫炎症微环境水凝胶支架及其制备方法和应用,特别涉及一种调控脊髓损伤免疫炎症微环境双功能调节免疫炎症反应水凝胶支架促进神经再生和功能恢复。
背景技术
脊髓损伤引起轴突破坏和神经元死亡,最终导致功能障碍。脊髓损伤分为原发性损伤和继发性损伤两个阶段。最初的创伤会导致原发性损伤,包括数分钟至数小时内的神经组织破坏以及神经元和神经胶质细胞坏死。继发性损伤过程是由原发性损伤引发的,并持续数周。继发性损伤主要包括氧化应激,继发的细胞凋亡,神经元脱髓鞘,免疫炎症反应和疤痕形成,进一步加剧的组织损伤和功能丧失;其中神经免疫炎症反应会在损伤后持续数天甚至数月,阻碍了神经***的修复。
脊髓损伤过程中释放损伤相关分子模式(DAMP)激活免疫细胞和非免疫细胞引起炎症和免疫反应。DAMPs激活单核细胞/巨噬细胞释放促炎性细胞因子(例如肿瘤坏死因子-(TNF-)α,白介素-(IL-)1β)和趋化因子(如诱导型一氧化氮合酶(iNOS)和基质金属蛋白酶)并诱导炎症细胞募集和组织进一步破坏。除巨噬细胞外,脊髓固有免疫细胞小胶质细胞也会被脊髓损伤后产生的DAMPs激活,并释放促炎和神经毒性因子,加剧了神经元丢失和继发性组织损伤。
简而言之,脊髓损伤后DAMP介导的炎症和免疫微环境阻碍了神经组织的再生和修复。因此,调控脊髓损伤免疫炎症治疗SCI(脊髓损伤)继发性损伤对于SCI修复很重要。已有研究人员发现,仅抑制一类DAMPs,如高迁移率族1(HMGB1)就可以显著减弱小胶质细胞介导的神经炎症,显着改善运动功能并减轻脊髓水肿。
尽管抑制HMGB1表达可能是SCI修复的潜在策略,但是单独调控HMBG1可能不足以为SCI治疗建立最佳的微环境。这主要是由于引发脊髓损伤免疫炎症微环境的DAMPs不仅包括HMGB1,还包括其他蛋白质,脂质过氧化产物,核酸和核苷酸衍生物。已有研究表明,阳离子聚合物,例如聚酰胺酰胺树状大分子,聚乙烯亚胺等,可以有效清除各种DAMP(例如细胞外DNA和HMBG1),从而减轻其引起的炎症。此外,将阳离子聚合物固定在支架上以限制全身暴露,从而提高生物安全性和DAMP中和能力。
此外,受损脊髓中巨噬细胞的发生不平衡极化:促炎性M1表型巨噬细胞的激活水平持续升高,而潜在的修复性M2表型巨噬细胞的激活水平降低,导致炎症持续时间延长和组织重塑失败。因此,减弱M1巨噬细胞/小胶质细胞的促炎反应和促进M2巨噬细胞/小胶质细胞的抗炎性质都是有效调节免疫微环境治疗SCI的有效方法。IL-10是一种关键的抗炎细胞因子,抑制单核细胞/巨噬细胞的炎症反应,调节小胶质细胞和巨噬细胞极化至M2表型,并促进神经元细胞存活,促进SCI小鼠和大鼠功能恢复中的作用。
发明内容
针对现有技术的不足,本发明的目的在于提供一种调节免疫炎症微环境水凝胶支架及其制备方法和应用,特别提供一种双功能调节免疫炎症反应/微环境水凝胶支架及其制备方法和应用。所述调节免疫炎症微环境水凝胶支架能调控脊髓损伤免疫炎症反应,同时促进神经再生和功能恢复。
为达此目的,本发明采用以下技术方案:
第一方面,本发明提供一种调节免疫炎症微环境水凝胶支架,所述调节免疫炎症微环境水凝胶支架的制备原料包括:阳离子高分子聚合物、交联水凝胶和抗炎细胞因子。
现有调控脊髓损伤后微环境大多采用单一策略,再生修复效果有限。此外,在病理状态对组织器官再生修复过程中,免疫炎症微环境在再生修复过程发挥的重要作用。因此,本发明研发了促进组织器官再生修复的新的干预策略。为了有效地重塑SCI免疫微环境促进神经再生修复,开发了一种新型的多重免疫调节功能调节免疫炎症微环境水凝胶支架,由阳离子高分子聚合物、交联水凝胶和抗炎细胞因子组成,其可通过清除DAMP减弱急性期炎症反应,并通过持续释放抗炎细胞因子促进抗炎和组织重塑。通过多策略联合调控脊髓损伤后免疫炎症微环境,抑制瘢痕形成,支持内源干细胞迁移和生长,促进神经再生和功能恢复。此外,通过功能水凝胶原位缓释递送生物活性分子并提高其稳定性,延长在损伤区的有效作用时间。相比于全身给药,降低治疗过程中生物活性分子的用量。
优选地,所述阳离子高分子聚合物包括PAMAM和/或PEI,优选为PAMAM。
优选地,所述PAMAM的代数为3-5,例如可以是3、4、5。
优选地,所述PAMAM的分子量为6909-28826,例如可以是6909、10109、12927、14214、14277、20112、20646、26252、28826等。
优选地,所述PAMAM的分子式包括C302H608N122O60、C622H1248N250O124或C1262H2528N506O252中的任意一种或至少两种的组合。
优选地,所述PAMAM的末端基团包括NH2。
优选地,所述PAMAM的末端基团数为32-128,例如可以是32、40、50、60、70、80、90、100、110、120、128等。
优选地,所述交联水凝胶为明胶复合胶原和/或透明质酸水凝胶。
优选地,所述明胶复合胶原为甲基丙烯酸酯化明胶水凝胶。
优选地,所述甲基丙烯酸酯化明胶水凝胶由以下制备方法制备得到:将甲基丙烯酸酐与明胶反应,纯化,得到甲基丙烯酸酯化明胶。
优选地,所述甲基丙烯酸酯化明胶的接枝率为60-95%,例如可以是60%、65%、70%、75%、80%、85%、90%、95%等。

在本发明中,所述水凝胶支架为阳离子高分子聚合物和交联水凝胶共交联形成的具有三维多孔结构形貌的支架,抗炎细胞因子负载在支架中,使得其可通过清除DAMP减弱急性期炎症反应,并通过持续释放抗炎细胞因子促进抗炎和组织重塑。
第二方面,本发明提供一种如第一方面所述的调节免疫炎症微环境水凝胶支架的制备方法,所述调节免疫炎症微环境水凝胶支架的制备方法为:将阳离子高分子聚合物、交联水凝胶和抗炎细胞因子混合,进行光固化反应,制备得到所述调节免疫炎症微环境水凝胶支架。
优选地,所述调节免疫炎症微环境水凝胶支架的制备方法包括以下步骤:
(1)将交联水凝胶、光引发剂和和溶剂混合,得到A溶液;将阳离子高分子聚合物和缓冲液,得到B溶液;将抗炎细胞因子和缓冲液混合,得到C溶液;
(2)将步骤(1)得到的A溶液、B溶液和C溶液混合后,进行光固化,得到所述调节免疫炎症微环境水凝胶支架。
优选地,所述A溶液中,所述交联水凝胶的浓度为0.08-0.16g/mL,例如可以是0.08g/mL、0.09g/mL、0.10g/mL、0.11g/mL、0.12g/mL、0.13g/mL、0.14g/mL、0.15g/mL、0.16g/mL等。
优选地,所述A溶液中,所述引发剂的浓度为0.002-0.01g/mL,例如可以是0.002g/mL、0.003g/mL、0.004g/mL、0.005g/mL、0.006g/mL、0.007g/mL、0.008g/mL、0.009g/mL、0.01g/mL等。

第三方面,本发明提供一种如第一方面所述的调节免疫炎症微环境水凝胶支架在制备调控脊髓损伤后免疫炎症微环境的材料和/或促进神经再生和功能恢复的材料中的应用。
相对于现有技术,本发明具有以下有益效果:
(1)本发明开发了一种新型的多重免疫调节功能调节免疫炎症微环境水凝胶支架,可通过清除DAMP减弱急性期炎症反应,并通过持续释放IL-10促进抗炎和组织重塑;
(2)本发明通过多策略联合调控脊髓损伤后免疫炎症微环境,抑制瘢痕形成,支持内源干细胞迁移和生长,促进神经再生和功能恢复;
(3)本发明通过功能水凝胶原位缓释递送生物活性分子并提高其稳定性,延长在损伤区的有效作用时间。相比于全身给药,降低治疗过程中生物活性分子的用量。

制备例
本制备例提供一种甲基丙烯酸酯化明胶,所述甲基丙烯酸酯化明胶由以下制备方法制备得到:将15g明胶和15mL甲基丙烯酸酐置于500mL的碳酸钠/碳酸氢钠缓冲溶液(0.1mol/L的碳酸钠溶液250mL和0.1mol/L的碳酸氢钠溶液250mL的混合液)中,在40℃条件下反应24h,烧杯纯水美国MFPI透析袋(截留分子量5000)透析去除副产物和未反应的反应物,制备得到接枝率90%甲基丙烯酸酯化明胶。
实施例1
本实施例提供一种清除和递送双功能水凝胶(Scav/Delv-GL),所述调节免疫炎症微环境水凝胶支架由以下制备方法制备得到:
(1)将1g的甲基丙烯酸酯化明胶、0.03g的光引发剂(苯基-2,4,6-三甲基苯甲酰基亚磷酸锂)和10mL的磷酸盐缓冲液混合,得到A溶液;将10mg的PAMAM-G3和1mL的磷酸缓冲液,得到B溶液;将50μg的IL-10和1mL的磷酸缓冲液,得到C溶液;
(2)将步骤(1)得到的A溶液,B溶液和C溶液以体积比为2:1:1进行混合后,在5W的可见光源进行100s的光固化,得到所述的调节免疫炎症微环境水凝胶支架。
其中,图1为本实施例提供的Scav/Delv-GL的扫描电镜图,如图1所示,Scav/Delv-GL支架的形貌为三维多孔结构,其孔径范围在20-200μm内。
对比例1
本对比例提供一种水凝胶支架(GL),所述水凝胶支架(GL)由以下制备方法制备得到:
将1g的甲基丙烯酸酯化明胶、0.03g的光引发剂(苯基-2,4,6-三甲基苯甲酰基亚磷酸锂)和10mL的磷酸盐缓冲液混合,得到A溶液。将A溶液与磷酸盐缓冲液以体积比1:1进行混合,在5W的可见光源进行100s的光固化,得到所述的对比例1水凝胶支架(GL)。
其中,图2为GL支架的扫描电镜图,如图2所示,GL支架的形貌为三维多孔状,其孔径为20-200μm。
对比例2
本对比例提供一种单清除功能水凝胶支架(Scav-GL),所述单清除功能水凝胶支架由以下制备方法制备得到:
将1g的甲基丙烯酸酯化明胶、0.03g的光引发剂(苯基-2,4,6-三甲基苯甲酰基亚磷酸锂)和10mL的磷酸盐缓冲液混合,得到A溶液。将10mg的PAMAM-G3和1mL的磷酸缓冲液,得到B溶液;将A溶液,B溶液与磷酸盐缓冲液以体积比2:1:1进行混合在5W的可见光源进行100s的光固化,得到所述的对比例2单清除功能水凝胶支架(Scav-GL)。

对比例3
本对比例提供一种单递送功能水凝胶(Delv-GL),所述单递送功能水凝胶由以下制备方法制备得到:
将1g的甲基丙烯酸酯化明胶、0.03g的光引发剂(苯基-2,4,6-三甲基苯甲酰基亚磷酸锂)和10mL的磷酸盐缓冲液混合,得到A溶液。将50μg的IL-10和1mL的磷酸缓冲液,得到C溶液;将A溶液,C溶液与磷酸盐缓冲液以体积比2:1:1进行混合在5W的可见光源进行100s的光固化,得到所述的对比例3的单递送功能水凝胶(Delv-GL)。
其中,图4为Delv-GL支架的扫描电镜图,如图4所示,Delv-GL支架的形貌为三维多孔状,其孔径为20-200μm。
试验例1
调节免疫炎症微环境水凝胶支架的基本性能测试
1、测试样品:实施例实施例1、对比例1-3制备得到的调节免疫炎症微环境水凝胶支架;
2、测试项目和测试方法:
(1)压缩模量:材料万能试验机(美国,Instron 5943);
(2)降解性能测试:体外在37℃条件下,在缓冲液中分别孵育0、7、14、21和28天后,去除缓冲液,用纯水洗涤3次后,真空-20℃冷冻干燥后称重;降解率为(初始质量—处理后剩余质量)/初始质量;
(3)DAMPs中蛋白质清除效率:与含有DAMPs的缓冲液(蛋白质含量C0)在37℃条件下孵育4h后,用BCA蛋白定量试剂盒测定缓冲液中未被清除的DAMPs的蛋白质的含量(C);
清除效率计算公式为:清除效率(%)=[(C0-C)/C0]×100%;
(4)HMGB1清除:与含有DAMPs的缓冲液在37℃条件下孵育4h后,用ELISA试剂盒测定缓冲液中未被清除的HMGB1含量;
(5)DAMPs中DNA清除:与含有DAMPs的缓冲液混合后,在37℃条件下孵育4h后,用Quant-iTTM PicoGreenTM dsDNA Assay试剂盒测定缓冲液中未被清除的双链DNA含量;
测试结果如下表1所示(或如图5-9所示),其中“-”代表未进行该项测试:

由表1测试数据可知,本发明制备得到的Scav/Delv-GL支架压缩模量约为3kPa(图5)。体外降解实验表明(图6),水凝胶降解以时间依赖性方式增加,孵育28天后,所有组的初始支架重量的约25%下降。在用Scav-GL支架处理后,DAMP溶液中的细胞外DNA和HMGB1蛋白以及总蛋白的量被有效捕获并去除,表明支架具有有效清除DAMPs的能力(图7-9)。
试验例2
调节免疫炎症微环境水凝胶支架的缓控释放性能测试
对上述实施例实施例1、对比例3制备得到的功能化调节免疫炎症微环境水凝胶支架进行缓控释放性能测试,通过ELISA进行检测;
其中,图10为不同支架在体外的缓控释放IL-10的对比图;在第1天,分别从Delv-GL和Scav/Delv-GL水凝胶中分别释放出约24.4%和23.7%的IL-10。在Hanks的平衡盐溶液(HBSS)中于37℃孵育21天后,最初封装的IL-10中约52.4%保留在Scav/Delv-GL支架中。
试验例3
调节免疫炎症微环境水凝胶支架调节脊髓损伤后炎症反应
(1)对上述实施例1和对比例1-3制备得到的功能化调节免疫炎症微环境水凝胶支架进行缓控释放性能测试,具体测试方法如下所示:通过免疫荧光染色进行检测:
其中,图11为植入不同支架的样本中IL-1β+和TNF-α+荧光染色区域对比图;脊髓损伤后,巨噬细胞和小胶质细免疫被DAMPs迅速激活,释放炎性因子并诱导持续数月的免疫反应,进一步加剧损伤。通过在损伤后7天对IL-1β和TNF-α免疫荧光染色发现植入Scav/Delv-GL和Scav-GL支架的样本中IL-1β+和TNF-α+染色区域相比于未植入支架(Control)或GL和Delv-GL明显减少。
其中,图12为IL-1β+的半定量免疫染色分析对比图;图13为TNF-α+的半定量免疫染色分析对比图,半定量免疫染色分析进一步证实Scav/Delv-GL和Scav-GL组的IL-1β和TNF-α荧光强度水平远低于其他组)。
上述结果表明通过在脊髓的损伤部位植入Scav/Delv-GL和Scav-GL支架来清除DAMPs可减少急性期促炎性细胞因子的分泌。
(2)对上述实施例1和对比例1-3制备得到的功能化调节免疫炎症微环境水凝胶支架进行免疫染色考察,具体测试方法如下所示:通过对ED1(反应性小胶质细胞/巨噬细胞标记物),F4/80(巨噬细胞标记物)和Iba-1(小胶质细胞的标记物)进行免疫染色考察脊髓损伤急性期病变区域巨噬细胞和小胶质细胞的分布和免疫表型;
其中,图14为通过对ED1进行免疫染色考察荧光染色图;图15为通过对F4/80进行免疫染色考察荧光染色图;图16为通过对Iba-1进行免疫染色考察荧光染色图;如图14-15所示,与其他四组相比,Scav/Delv-GL组中阳性染色的巨噬细胞和小胶质细胞的数量明显减少。与对照组和GL植入的脊髓损伤小鼠相比,植入Scav-GL支架显著减少了ED1,F4/80-和Iba-1阳性细胞的数量。此外,在Delv-GL组中观察到的Iba-1阳性小胶质细胞少于GL组。
(3)对上述实施例1和对比例1-3制备得到的功能化调节免疫炎症微环境水凝胶支架进行炎症细胞极化亚型测试,具体测试方法如下所示:通过免疫荧光染色进行检测:
其中,图17为iNOS免疫荧光染色图,图18为ARG-1免疫荧光染色图;如图17-18所示,促炎M1表型标志物iNOS和抗炎M2表型标志物ARG-1染色发现,与对照组和GL组相比,Scav/Delv-GL、Delv-GL和Scav-GL组的病变部位和周围组织区域的iNOS阳性细胞数量明显减少;ARG-1阳性细胞的数量增加。Scav/Delv-GL调控损伤急性期巨噬细胞和小胶质细胞活化表型。
试验例4
调控脊髓损伤免疫炎症微环境调节免疫炎症微环境水凝胶支架促进神经再生和运动功能恢复
通过Tuj-1免疫荧光染色以观察损伤部位的轴突生长和神经再生,具体测试方法如下所示:通过免疫荧光染色和实时定量PCR进行检测:
其中,图19为Tuj-1/GFAP免疫荧光共染图;由图19所示,手术后7天,Scav/Delv-GL和Delv-GL组的Tuj-1阳性新产生的神经元数量高于Scav-GL,GL和对照组;图20为治疗8周后,损伤区域中心的Tuj-1免疫荧光染色图;也表明了双功能支架(Scav/Delv-GL)的长期促进轴突再生作用。
其中,图21为水凝胶移植8周后Tuj-1基因表达情况;由图21所示,在所有组中,用双重免疫调节功能水凝胶处理的小鼠在病变部位的Tuj-1标记神经元数量最多。与植入GL支架的组相比,在植入Delv-GL和Scav-GL支架的小鼠的病变部位阳性染色的Tuj-1神经元显着增加。在脊髓损伤小鼠(Control)的损伤区域中心几乎看不到Tuj-1阳性细胞。Tuj-1表达趋势与上述免疫染色结果一致。Scav/Delv-GL组的样品的Tuj-1表达水平高于Delv-GL,Scav-GL,GL和SCI组的样品。
通过测量运动诱发电位(MEP)评估双功能水凝胶治疗SCI小鼠中神经回路的形成,其中,图22为Tuj-1运动诱发电位波形图;图23为运动诱发电位平均潜伏期图;图24为平均振幅图;如图22-24所示,在术后8周,植入Scav/Delv-GL支架的小鼠表现出更大的运动诱发反应改善,潜伏期缩短,平均振幅增加。其中,图25为小鼠运动功能评分图;如图25所示,BMS小鼠运动功能评分发现双功能调节免疫炎症微环境水凝胶支架发挥出协同促进运动功能恢复的作用。
申请人声明,本发明通过上述实施例来说明所述调节免疫炎症微环境水凝胶支架及其制备方法和应用,但本发明并不局限于上述实施例,即不意味着本发明必须依赖上述实施例才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明产品各原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
Claims (28)
1.一种调节免疫炎症微环境水凝胶支架,其特征在于,所述调节免疫炎症微环境水凝胶支架的制备原料包括:阳离子高分子聚合物、交联水凝胶和抗炎细胞因子;
所述阳离子高分子聚合物包括PAMAM和/或PEI;所述交联水凝胶为明胶复合胶原和/或透明质酸水凝胶;所述抗炎细胞因子包括IL-10、IL-4或IL-33中的任意一种或至少两种的组合。
2.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述阳离子高分子聚合物为PAMAM。
3.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述PAMAM的代数为3-5。
4.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述PAMAM的分子量为6909-28826。
5.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述PAMAM的分子式包括C302H608N122O60、C622H1248N250O124或C1262H2528N506O252中的任意一种或至少两种的组合。
6.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述PAMAM的末端基团包括NH2。
7.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述PAMAM的末端基团数为32-128。
8.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述明胶复合胶原为甲基丙烯酸酯化明胶水凝胶。
9.根据权利要求8所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述甲基丙烯酸酯化明胶水凝胶由以下制备方法制备得到:将甲基丙烯酸酐与明胶反应,纯化,得到甲基丙烯酸酯化明胶。
10.根据权利要求8所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述甲基丙烯酸酯化明胶的接枝率为60-95%。
11.根据权利要求9所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述甲基丙烯酸酐的体积和明胶的质量比为1mL:(0.8-2)g。
12.根据权利要求9所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述反应温度为37-50℃,所述反应时间为4-24h。
13.根据权利要求9所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述反应在溶剂存在下进行,所述溶剂包括水、NaCl溶液、磷酸盐缓冲液或Hank's平衡盐溶液中的任意一种或至少两种的组合。
14.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述调节免疫炎症微环境水凝胶支架的制备原料还包括:光引发剂、溶剂和缓冲液。
15.根据权利要求14所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述光引发剂包括2-羟基-4'-(2-羟乙氧基)-2-甲基苯丙酮、苯基-2,4,6-三甲基苯甲酰基亚磷酸锂或Irgacure中的任意一种或至少两种的组合。
16.根据权利要求14所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述溶剂为水。
17.根据权利要求14所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述缓冲液包括NaCl溶液、磷酸盐缓冲液或Hank's平衡盐溶液中的任意一种或至少两种的组合。
18.根据权利要求1所述的调节免疫炎症微环境水凝胶支架,其特征在于,所述水凝胶支架的凝胶形态具有三维多孔结构,所述多孔结构的孔直径为20-200μm。
19.根据权利要求1-18中任一项所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述调节免疫炎症微环境水凝胶支架的制备方法为:将阳离子高分子聚合物、交联水凝胶和抗炎细胞因子混合,进行光固化反应,制备得到所述调节免疫炎症微环境水凝胶支架。
20.根据权利要求19所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述调节免疫炎症微环境水凝胶支架的制备方法包括以下步骤:
(1)将交联水凝胶、光引发剂和溶剂混合,得到A溶液;将阳离子高分子聚合物和缓冲液混合,得到B溶液;将抗炎细胞因子和缓冲液混合,得到C溶液;
(2)将步骤(1)得到的A溶液、B溶液和C溶液混合后,进行光固化,得到所述调节免疫炎症微环境水凝胶支架。
21.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述A溶液中,所述交联水凝胶的浓度为0.08-0.16g/mL。
22.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述A溶液中,所述引发剂的浓度为0.002-0.01g/mL。
23.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述B溶液中,所述阳离子高分子聚合物的浓度为2-200mg/mL。
24.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,所述C溶液中,所述抗炎细胞因子的浓度为20-500μg/mL。
25.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,步骤(2)中,所述A溶液、B溶液和C溶液的体积比为(1-4):(0.5-2):(0.5-2)。
26.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,步骤(2)中,所述光固化的光源为可见光源或紫外光源。
27.根据权利要求20所述的调节免疫炎症微环境水凝胶支架的制备方法,其特征在于,步骤(2)中,所述光固化的能量为2-10W,所述光固化的时间为10-300s。
28.根据权利要求1-18中任一项所述的调节免疫炎症微环境水凝胶支架在制备调控脊髓损伤后免疫炎症微环境的材料和/或促进神经再生和功能恢复的材料中的应用。
