Abstract
The invention discloses an anti-aging composition for promoting collagen effect and application thereof. The composition comprises collagen, chitosan and zinc, wherein the chitosan is low-molecular-weight chitosan, the chitosan and the zinc can promote the absorption and synthesis of the collagen, and the chitosan and the zinc are added into the composition consisting of the collagen, so that the composition not only can keep respective efficacy and functions, but also can play a role in synergy, and can promote the absorption and synthesis efficiency of the collagen to improve the skin moisturizing function, increase the skin elasticity of a human body and reduce the skin wrinkle depth, and has the function of resisting aging. And by adjusting the dosage proportion of the collagen, the chitosan and the zinc, the synergistic interaction can be realized.
Description
Technical Field
The invention relates to the technical field of food, cosmetics and medicines, in particular to an anti-aging composition for promoting collagen function and application thereof.
Background
Skin is the largest organ of the human body and suffers from aging as well as other organs of the human body. Skin aging is classified into exogenous and endogenous causes. Environmental pollution, bad living habits such as alcoholism and smoking and ultraviolet irradiation in sunlight are exogenous factors, and endogenous factors of skin aging are mainly caused by that the normal physiological circulation balance of the skin is broken due to the fact that the cell activity of a human body is reduced, the cell metabolism is slowed down, the repairing capability is poor and the immunity is reduced along with the increase of the age of the human body, collagen in the skin is continuously reduced, moisturizing factors are continuously lost, skin pigments are increased, and the capacities of resisting oxidation and removing free radicals of the skin are reduced. The aging factors of the skin can lead the skin to lose softness, elasticity and luster, and lead the skin to have a series of aging phenomena such as wrinkles and the like.
Collagen is a biological macromolecule, is a main component in the connective tissue of animals, is also a functional protein with the largest content and the widest distribution in mammals, accounts for 25 to 30 percent of the total amount of protein, and has good biocompatibility, biodegradability and bioactivity, so the collagen is widely applied to the fields of food, medicine, tissue engineering, cosmetics and the like. For example, collagen is extracted from animal skin, the skin contains proteoglycan such as hyaluronic acid and chondroitin sulfate besides collagen, and the proteoglycan contains a large amount of polar groups, is a moisturizing factor and has the effect of preventing tyrosine in the skin from being converted into melanin, so the collagen has the effects of purely natural moisturizing, whitening, crease resistance, freckle removal and the like, and can be widely applied to beauty products. The chemical composition and structure of collagen make it the basis of beauty. The collagen is similar to the structure of human skin collagen, is water insoluble fibrous glycoprotein, contains abundant amino acids and hydrophilic groups in molecules, has certain surface activity and good compatibility, and has good moisturizing effect because the molecules contain abundant hydroxyl groups. At a relative humidity of 70%, it can still retain 45% of its own weight of moisture. Chitosan is a primary derivative of chitin. The chemical structure of the fiber is a macromolecular alkaline polysaccharide polymer with cations, and the polysaccharide polymer is the only edible fiber with positive charges and cations in the nature; has the physical and chemical properties of stable physical properties, good solubility, hydrophilic hygroscopicity, biodegradability, biocompatibility, film-forming property, cohesiveness, air permeability and the like. Particularly, because of the pure natural activity and no toxic or side effect of the chitosan, the chitosan has good affinity to human bodies, can promote the organisms to synthesize and secrete a large amount of immunoglobulin antibodies, and can regulate and improve the immunity of the human bodies; adsorbate endotoxins, harmful pigments, and radiatins; chelating harmful heavy metal ions in vivo, such as lead, mercury and cadmium, and discharging out of the body along with metabolites; regulating pH value of human body, enhancing quality and quantity of immunocompetent cells, and activating normal somatic cells; promoting secretion of male and female hormone; inhibiting the formation of lipid peroxide and delaying aging; inhibiting cancer cell metastasis; strengthening liver function, regulating blood fat, blood sugar and blood pressure, so that it can be extensively used in health-care food and medicine additive. Zinc is one of essential trace elements for human body, plays an extremely important role in the growth and development process of human body, is usually praised as 'flowers of life' and 'sources of intelligence', is used for supplementing zinc, namely supplementing the essential trace elements for human body, and plays an extremely important role in growth and development, immunity, vitamins and the like of human body. Zinc is present in a number of enzyme systems, such as carbonic anhydrase, respiratory enzyme, lactate dehydrogenase, superoxide dismutase, alkaline phosphatase, DNA and RNA polymerase, and the like, and is an essential substance for the synthesis of nucleic acids, proteins, carbohydrates and vitamin A utilization. The three substances are commonly applied in a few examples, and most of the applications only use the respective properties of the three substances, and do not show the synergistic effect of the collagen, the chitosan and the zinc to enhance the absorption and synthesis efficiency of the collagen to improve the skin moisturizing function, increase the skin elasticity of a human body and relieve the anti-aging effect of the skin wrinkle depth in the fields of food, cosmetics or medicines.
Disclosure of Invention
In order to solve the problems, the invention provides an anti-aging composition for promoting collagen effects, which takes chitosan and zinc as promoters, enhances the absorption and synthesis efficiency of collagen to improve the skin moisturizing function, increases the skin elasticity of a human body and relieves the skin wrinkle depth, and can also play a synergistic effect and an interaction role by adjusting the dosage proportion of the collagen, the chitosan and the zinc.
The specific technical scheme of the invention is as follows:

3. Composition according to claim 1, wherein the collagen has a molecular weight of 200 to 10000Da, preferably 300 to 5000Da, more preferably 800 to 3000Da.
4. The composition according to claim 1, wherein the chitosan is a low molecular weight chitosan having a molecular weight of 200-20000 Da, preferably 300-5000 Da, more preferably 800-3200 Da.
5. The composition of claim 1, wherein the zinc is selected from the group consisting of: zinc sulfate, zinc gluconate, zinc glycinate, zinc oxide, zinc lactate, zinc citrate, zinc chloride, zinc acetate and zinc carbonate, preferably zinc citrate.

The composition according to any one of claims 1 to 5, wherein the collagen is present in an amount of 1000 to 100000 parts, preferably 10000 to 100000 parts, and more preferably 50000 to 100000 parts; 10-10000 parts of chitosan, preferably 500-10000 parts of chitosan, and further preferably 2500-10000 parts of chitosan; the amount of the zinc is 0.1 to 100 parts, preferably 1 to 100 parts, and more preferably 5 to 100 parts.
7. An article of manufacture which is a food, cosmetic or pharmaceutical product, preferably a food product which is a health food.
8. The composition according to any one of claims 1 to 7, wherein the composition is contained in an amount of 30% by mass or more in the product.
9. The product of claim 7, further comprising an adjuvant, wherein the adjuvant is a food, cosmetic or pharmaceutical acceptable adjuvant.
10. The composition according to any one of claims 1 to 9 has application in the field of food, cosmetics or pharmaceuticals, preferably the food is a health food.
ADVANTAGEOUS EFFECTS OF INVENTION
The composition provided by the invention comprises collagen, chitosan and zinc, wherein the collagen, the chitosan and the zinc can interact, the chitosan and the zinc can promote the absorption and synthesis of the collagen, and the chitosan and the zinc are added into the composition consisting of the collagen, so that the composition not only can keep respective efficacy and function, but also can play a role in synergy, and can promote the absorption and synthesis efficiency of the collagen to improve the skin moisturizing function, increase the skin elasticity and relieve the skin wrinkle depth, and is applied to anti-aging in the fields of food, cosmetics or medicine.

The present invention provides a composition comprising collagen, chitosan and zinc.
In a preferred embodiment of the present invention, wherein the collagen is selected from the group consisting of: collagen type I, collagen type II, collagen type III, collagen type V, collagen type XI, oligopeptide, recombinant humanized collagen, preferably collagen type I.
The type I collagen consists of three peptide chains, two alpha (I) chains and one alpha (II) chain. Has the strongest effect on the body functions.
The type II collagen is a naturally occurring protein, and is present in hyaline cartilage and articular cartilage of the human body.
The type III collagen is the main collagen in human skin, fascia and tendon, is homotrimer, or is protein composed of three identical peptide chains (monomers).
The peptide chain of the type V collagen consists of 2 α 1 chains and 1 α 2 chains or 3 α 1 chains, in 3 assembly forms. The type V collagen is distributed around cells and type I collagen, and may act as a bridge between the basement membrane and connective tissue.
The type XI collagen is heterotrimer, and its composition is α 1 (XI) α 2 (XI) α 3 (XI), and type VI collagen is a minor component of cartilage, plays an important role in the formation of cartilage collagen fiber and the composition of cartilage matrix, and often coexists with type II collagen.
The oligopeptide refers to collagen obtained by hydrolysis or enzymolysis, has a molecular weight generally below 1000 daltons, and is also called small peptide, oligopeptide or small molecule active peptide.

In a preferred embodiment of the present invention, wherein the collagen has a molecular weight of 200 to 10000Da, preferably 300 to 5000Da, and more preferably 800 to 3000Da. For example, the collagen molecular weight may be 200Da, 300Da, 400Da, 500Da, 600Da, 700Da, 800Da, 1000Da, 1500Da, 2000Da, 2500Da, 3000Da, 3500Da, 4000Da, 4500Da, 5000Da, 6000Da, 7000Da, 8000Da, 9000Da, 10000Da, and the like.
In a preferred embodiment of the present invention, the molecular weight of the chitosan is 200-20000 Da, preferably 300-5000 Da, more preferably 800-3200 Da. The molecular weight of the collagen and chitosan refers to a weight average molecular weight.
The invention provides a product, which comprises the composition, wherein the composition contains more than 30% of the composition in percentage by mass in the product.
In a preferred embodiment of the present invention, the product is a food, a cosmetic or a pharmaceutical product.
In a preferred embodiment of the present invention, the product further comprises an auxiliary material, and the auxiliary material is a food, cosmetic or pharmaceutical acceptable auxiliary material. The embodiment 1 is an anti-aging composition for promoting collagen function, which comprises the following components in parts by weight: 10000 parts of collagen, 500 parts of chitosan and 1 part of zinc.
The composition for promoting collagen effect can be made into tablet, capsule, granule, powder, pill, beverage, oral liquid, etc. by conventional method in the art.
Example 2
The embodiment 2 is an anti-aging composition for promoting collagen function, which comprises the following components in parts by weight: 50000 parts of collagen, 3000 parts of chitosan and 8 parts of zinc.

Example 3
This example 3 is a commercial collagen powder of the same type.
The preparation of the compositions described in examples 1-2 specifically comprises: weighing the raw materials according to the proportion, mixing the raw materials, stirring the mixture for 30 to 60 minutes at the speed of 5 to 7r/min, sieving the mixture by a sieve of 30 to 100 meshes, filling the mixture, and entering a qualified product area of a storehouse after the finished product is inspected to be qualified.

Selecting 9 volunteer women, wherein the average age is 50 years old and the skin is similar; every 3 people take the same collagen powder 5 g each time, and take the collagen powder in the morning and at night every day for three months continuously.

as can be seen from the above table, after three months of administration, the anti-aging composition for promoting collagen function described in examples 1-2 has an obvious moisturizing function, and the effect is superior to that of similar products on the market.
Test example 2
When the anti-aging composition for promoting collagen effect prepared in examples 1-2 is taken together with the similar product in the market, i.e., collagen powder, the skin elasticity increasing rate effect is tested by a skin elasticity tester.
Selecting 9 volunteer women, wherein the average age is 50 years old and the skin is similar; every 3 persons take the same collagen powder 5 g each time, once every morning and evening, and continuously for three months.
The skin elasticity increase rate was measured as shown in the following table:
as can be seen from the above table, after three months of administration, the collagen-promoting anti-aging composition described in examples 1-2 has an obvious effect of increasing skin elasticity, and the effect is superior to that of the similar products on the market.
Test example 3
When the anti-aging composition for promoting collagen effect prepared in examples 1-2 is taken with the similar product in the market, namely collagen powder in example 3, the effect of the wrinkle depth rate is tested by a skin wrinkle tester and compared.
Selecting 9 volunteer women, wherein the average age is 50 years old and the skin is similar; every 3 persons take the same collagen powder 5 g each time, once every morning and evening, and continuously for three months.
Skin wrinkle depth rate measurements are shown in the following table:
as can be seen from the above table, after three months of administration, the collagen-promoting anti-aging composition described in examples 1-2 has an obvious skin wrinkle depth reduction effect, and the effect is superior to that of the similar products on the market.
In conclusion, the composition disclosed by the invention takes chitosan and zinc as the promoters, can keep respective effects and play a role in synergy, can promote the absorption and synthesis efficiency of collagen to improve the skin moisturizing function, increase the skin elasticity of a human body and relieve the skin wrinkle depth, and has the anti-aging effect.
Although the invention has been described in detail hereinabove by way of general illustration and by way of specific embodiments and tests, it will be apparent to those skilled in the art that modifications and improvements can be made thereto. Therefore, it is intended that all such modifications and improvements be made without departing from the spirit of the invention.
Claims (10)
1. A composition comprising collagen, chitosan, and zinc.
2. The composition of claim 1, wherein the collagen is selected from the group consisting of: type I collagen, type II collagen, type III collagen, type V collagen, type XI collagen, oligopeptide, recombinant humanized collagen, preferably type I collagen.
3. Composition according to claim 1, wherein the collagen has a molecular weight of 200 to 10000Da, preferably 300 to 5000Da, more preferably 800 to 3000Da.
4. The composition according to claim 1, wherein the chitosan is a low molecular weight chitosan having a molecular weight of 200-20000 Da, preferably 300-5000 Da, more preferably 800-3200 Da.
5. The composition of claim 1, wherein the zinc is selected from the group consisting of: zinc sulfate, zinc gluconate, zinc glycinate, zinc oxide, zinc lactate, zinc citrate, zinc chloride, zinc acetate and zinc carbonate, preferably zinc citrate.
6. The composition according to any one of claims 1 to 5, wherein the collagen is present in an amount of 1000 to 100000 parts, preferably 10000 to 100000 parts, and more preferably 50000 to 100000 parts; the chitosan accounts for 10 to 10000 parts, preferably 500 to 10000 parts, and further preferably 2500 to 10000 parts; the amount of zinc is 0.1 to 100 parts, preferably 1 to 100 parts, and more preferably 5 to 100 parts.
7. An article of manufacture which is a food, cosmetic or pharmaceutical product, preferably a food product which is a health food.
8. The composition according to any one of claims 1 to 7, wherein the composition is contained in an amount of 30% by mass or more in the product.
9. The product of claim 7, further comprising an adjuvant, wherein the adjuvant is a food, cosmetic, or pharmaceutical acceptable adjuvant.
10. The composition according to any one of claims 1 to 9 has application in the field of food, cosmetics or pharmaceuticals, preferably the food is a health food.
