Description

Emulsion type moisturizing and relieving spray water and preparation method thereof
Technical Field
The invention belongs to the technical field of skin care products, and particularly relates to emulsion type moisturizing and soothing spray water and a preparation method thereof.
Background
With age, the morphological structure and physiological function of the skin change. Skin aging is the result of multiple factors, and skin damage, infection, malnutrition, metabolic disorders, and drug abuse can affect skin condition. Healthy skin is smooth, moist and elastic, free radicals destroy collagen and active substances in normal cell membrane tissues to form fine lines and wrinkles, and the skin is evaporated, lacks water and loses elasticity to form the wrinkles. The root cause of skin aging is that the water in the body is lost too fast, cells cannot obtain sufficient water, the metabolism of the cells is influenced, the division and the propagation of the cells are influenced, and a large number of aged tissue cells cannot be normally updated, so that the skin is rough and has wrinkles. Meanwhile, excessive oxygen radicals in the body are an important reason for accelerating skin aging, and the generation of the oxygen radicals can be originated from the metabolism of the human body and can also be caused by a plurality of factors such as ultraviolet irradiation, environmental pollution, bad living habits and the like. The free radicals are originally intermediates of human metabolism and are effective defense systems of organisms; but when the free radicals are excessive and cannot be cleared in time, the aging process of the body is accelerated.
The conventional spray water takes the atomization effect into consideration, the formula mainly takes water as the main component, a small amount of polyhydric alcohol humectant is added to play a basic skin care effect, and a slightly better formula can also be added with a small amount of water-soluble active substance so as to have certain functionality. But even the most basic skin care, even if only moisturizing and moisturizing are performed, the mere use of water as an input is not always effective, just as in autumn and winter when the lips are cracked, the use of water to moisturize the lips is just frosting on the snow, and then people know why is the use of the lip balm? Because oil is needed to moisturize the dry skin, the moisture is locked in the skin and is not excessively lost. After all, skin care is based on water-oil balance, and the oil needed by skin needs to be supplemented while water is fully supplemented, so that water and oil are supplemented at the same time. How do to supplement with water and oil? Many spray products which claim to supplement both water and oil are mainly prepared from water and oil, but emulsion type spray is rarely seen, because the emulsion type is known to mean an emulsifier, and the conventional emulsifier generally tends to be suitable for cream, emulsion and other products with higher viscosity.

Test example
Samples prepared according to the technical schemes of example 1, example 2 and example 3, and samples prepared according to the technical schemes of comparative examples 1 to 5 were selected and tested as follows:
and (4) moisture retention performance test:
30 subjects aged 25-40 years were tested using a skin moisture content test probe Corneometer CM 825 from CK, germany. The inner forearm of the subject should be kept free of any cosmetic product 2-3 days before using the test sample. The test area mark should be made on the inner side of the forearm of the test subject, the area of the test area is 3cm multiplied by 3cm, a plurality of areas can be marked on the same arm at the same time, and the interval between each test area is at least 1cm. The test areas of the products of the examples and comparative examples should be randomly distributed over the left and right arm mark areas during application to ensure that the test area locations of all test samples are statistically balanced. The test sample is sprayed once in an amount of 1mg, and the test sample is uniformly coated in the test area. The test subjects used the test samples in the test area daily, morning and evening during the test period, and after use the test samples were stored in a home freezer. Before each instrument test, the testee needs to clean the inner sides of the forearms of both hands uniformly, the cleaning method is to wipe the forearms clean by using dry facial tissues, then sit still for 20min in a room meeting the standard, cannot drink water and beverages, expose the forearms, place the forearms in a test state and keep relaxed. Test time was before use, after 4 weeks of use.
TABLE 1 skin moisture content (MMV)
Figure BDA0003744813470000181
As can be seen from table 1, the raw materials such as avocado oil, olive oil unsaponifiable matter, cetearyl alcohol, sodium hyaluronate, etc. have a significant effect on the improvement of moisturizing performance. Specifically, in combination with comparative examples 1 and 5, the conventional sodium hyaluronate does not bring about a good improvement in moisturizing performance, because the ultra-low molecular hyaluronic acid can be absorbed instantaneously and combined with water molecules in the skin, but water is slowly lost without a protective barrier formed by cationic hyaluronic acid in the skin. Similarly, the absorption channel opened by ultralow molecules entering the skin instantly is lacked, the speed of cationic hyaluronic acid entering the skin is very low, and the moisturizing effect of the skin is influenced, in combination with comparative examples 2 and 3, avocado oil, olive oil unsaponifiable matter and cetearyl alcohol which are used as traditional emulsifiers or emollients show a good moisturizing effect, which is probably related to the fact that the skin absorption channel is opened and the protective effect of skin care products is improved, and meanwhile, the spherical liquid formed by the avocado oil and the olive oil unsaponifiable matter is wrapped by the high molecular weight cetearyl alcohol and is matched with an emulsion type spraying mode for use, so that the moisturizing effect of the skin is improved.
Anti-inflammatory performance test:
IL-8 is a cytokine secreted by Th1 cells, mainly mediates the generation of immune response auxiliary antibodies related to cytotoxicity and local inflammation, participates in the generation of cellular immunity and delayed hypersensitivity type inflammation, has the main biological effects of chemotaxis and activation of neutrophils, promotes the lysosome enzymatic activity and phagocytosis of the neutrophils, has certain chemotaxis on basophils and T cells, generates morphological change after the neutrophils are contacted with IL-8, directionally swims to a reaction part and releases a series of active products, and the effects can cause local inflammatory reaction of an organism and achieve the purposes of sterilization and cell injury.
Experimental materials: ku812 cells are provided by the cell center of the chinese academy of sciences; RPMI1640, IMDM medium and penicillin and streptomycin were purchased from Gibco; fetal bovine serum was purchased from HyClone; DMSO, PMA and A12387 are available from Sigma; IL-8 kits were purchased from R & D systems; mu.Q μ ant enzyme-labeled Detector was purchased from BIO-TEK.
Experimental methods sample preparation: the spray water obtained in examples 1 to 3 and comparative examples 1 to 5 was prepared in DMSO or double distilled water at a concentration of 25mg/mL by dilution with a medium containing 10% fetal bovine serum.
Ku812 cell density was adjusted to 2.2X 10 using IMDM medium containing 10% fetal bovine serum 6 Cell suspension of one/mL, seeded in 96-well plates at 200. Mu.L per well. Setting cell blank control group, example group and comparative example group, each group was set with 3 parallel wells at 37 ℃ and 5% CO 2 The culture box is cultured and kept still for 10min, the samples are added into the groups of the examples and the comparative examples at the final concentration of 50 mu g/mL, the cells are placed in the culture box for pre-incubation, after 15min, the stimulants PMA and A12387 are added into the cells of each group to make the final concentration of 5nm and 1 mu m respectively, and the DMSO culture medium with the same volume is added into the blank groups of the cells. After 2h, the cells were centrifuged, the cell supernatant was collected, and the IL-8 content in the supernatant was determined. In addition, a model group was set and treated in the same manner as in the example group or the comparative example group, but the sample was not added for treatment.
TABLE 2IL-8 assay results

Figure BDA0003744813470000201
In combination with table 2, the products prepared in examples 1 to 3 of the present application have significant effects of inhibiting skin inflammation, compared to the model group and the comparative example group. Specifically, avocado oil, olive oil unsaponifiables, cetearyl alcohol, sodium hyaluronate and other raw materials can play a role in inhibiting type IV hypersensitivity (delayed hypersensitivity) by inhibiting IL-8, and specifically, for example, avocado oil and olive oil unsaponifiables can be competitively combined with surface specific receptors of nucleated leucocytes (PMNs), thereby avoiding cell shape change, reducing PMN degranulation and further inhibiting PMNs, and shutting down and reducing inflammatory reactions; cetearyl alcohol and sodium hyaluronate can bind with IL-8 receptor, weaken neutrophil and reduce local inflammatory reaction probability. In addition, as shown in fig. 1, fig. 2 and fig. 3, based on the new combination with anti-inflammatory effect formed by polyglycerol ester and isomers of vegetable fatty acid, avocado oil and italian olive oil unsaponifiable matter, 10% of grease is stabilized in the foreign body without adding any thickening stabilizing agent, and meanwhile, a stable liquid crystal structure is formed, and the percentage of interlayer water in the liquid crystal structure can reach 70%, and after the liquid crystal structure is sprayed on the skin, the liquid crystal lattice is broken, and the water can be immediately soaked into the skin, so that a 'refreshing' feeling is given. The formula is simple, and the water-soluble active substance is free of PEG, soap, phosphate and alcohol, has no encumbrance, has moisturizing, nourishing, relieving and anti-inflammatory effects, and can be verified to have anti-inflammatory activity by measuring the expression of an inflammation marker interleukin-8 (IL-8). The emulsion type moisturizing and relieving spray water provided by the invention can ensure that the emulsion is layered on the premise of not destroying the emulsified state without intentionally adjusting the density difference between the emulsion and the external phase, and presents the appearance of a double-layer spray water of a microemulsion formed by an upper emulsion, a lower external phase aqueous solution and a small amount of emulsion.
