Abstract
The invention discloses a plant composition and application thereof in cosmetics, and relates to the technical field of cosmetics. The composition comprises ginseng, ganoderma lucidum, saussurea involucrata and truffle. Through the synergistic effect of ginseng, ganoderma lucidum, saussurea involucrata and truffle, free radicals can be effectively inhibited, glycosylation reaction and secretion of matrix metalloproteinase-1 (MMP-1) can be inhibited, generation of human type I collagen (COL-1) can be promoted, and the cosmetic is safe to use, has no toxic or side effect and can be well applied to anti-aging cosmetics.
Description
Technical Field
The invention relates to the technical field of cosmetics, in particular to a plant composition and application thereof in cosmetics.
Background
Aging is an activity that accompanies life occurrence and development, and is a process from the loss and degeneration of body from constituent substances, tissue structures to physiological functions. The aging process continues from fertilized eggs to death, and the aging characteristic is only apparent until a certain stage is reached. Physiological changes in the aging process of the human body are mainly reflected in the loss of tissue cells and constituent substances of the body, the slowing of the metabolic rate of the body and the hypofunction of the body and organs.
At present, the research on anti-aging related products is more and more aiming at the aging problem. The theory of free radicals suggests that the aging process of organisms is the result of the accumulation of free radicals produced by the tissue cells of the organism, which can cause DNA damage and thus mutations, inducing tumor formation. Free radicals are intermediate products of normal metabolism, have strong reaction capacity, can oxidize various substances in cells and damage biological membranes. And can crosslink macromolecules such as proteins, nucleic acids and the like to influence the normal functions of the macromolecules. Meanwhile, glycation theory indicates that glucose is an important cellular energy, but it can react randomly with proteins and DNA to destroy them. This process is called saccharification and it can lead to some undesirable consequences. The enzyme subjected to saccharification often fails to work properly, and the glycated protein is deformed and becomes unstable. As these processes are studied and well known, saccharification is considered one of the causes of aging. Chemical linkages are formed between proteins and other macromolecular substances due to crosslinking caused by saccharification. The materials that crosslink generally become stiffer, lack elasticity, and tend to tear or break. In the aging body, crosslinking can lead to arteriosclerosis, wrinkles and joint stiffness.
In addition, it has been shown that MMP-1 (matrix metalloproteinase-1) is closely related to skin aging of human body, and one of the main causes of skin aging is that the structure of dermis is changed, and the cause of skin dermis structure change is various, wherein, due to the action of some external factors, activation of Matrix Metalloproteinases (MMPs) in vivo can cause excessive degradation of collagen and elastin supporting skin structure in dermis of skin, so that skin has aging symptoms such as shrinkage, inelastic. Based on the important role of MMP-1 in the skin aging process, people gradually take inhibition of MMP-1 activity, inhibition of abnormal collagen degradation, reduction of skin inflammatory reaction and prevention of wrinkles generated by skin photoaging as a new way of delaying aging. Lim et al found that flavonoid compounds such as quercetin, kaempferol, naringin, apigenin, wogonin, etc. have MPP-1 activity and expression inhibiting effect, and can be used as main ingredient of medicine or cosmetic for preventing and treating skin inflammation and skin photoaging. In addition, the active ingredient of the water extract from the seeds of Cucurbita plant can inhibit degradation and degradation of ECM protein (especially collagen), inhibit ECM protease, especially MMPs, and can be used for reducing and preventing skin aging, reducing wrinkles and fine lines, increasing skin elasticity, making skin smooth and fine, and having antiinflammatory effect.
With the increasing demand of people for anti-aging products, the importance of safer and more natural anti-aging products is increasingly highlighted, and the anti-aging products of the existing plant compositions are usually more in components and not definite in curative effect, so that it is necessary to provide a plant composition with good anti-aging effect, and the requirements of definite anti-aging effect, safety and no toxic or side effect are met.
Disclosure of Invention
The invention aims to provide a plant composition and application thereof in cosmetics, which can effectively inhibit free radicals, inhibit the secretion of matrix metalloproteinase-1 (MMP-1) and promote the generation of human type I collagen (COL-1) through the synergistic effect of ginseng, ganoderma lucidum, saussurea involucrata and truffle, is safe to use, has no toxic or side effect, and can be well applied to anti-aging cosmetics.

In one aspect, the present invention provides a plant composition comprising ginseng, ganoderma lucidum, saussurea involucrata, and truffle.
Preferably, the mass ratio of ginseng, ganoderma lucidum, saussurea involucrata and truffle is 4:1-4:0.1-1:0.1-1.
Further preferably, the mass ratio of ginseng, ganoderma lucidum, saussurea involucrata and truffle is 4:2-4:0.5-1:0.5-1.
Most preferably, the mass ratio of ginseng, ganoderma lucidum, saussurea involucrata and truffle is 4:2:0.5:0.5.
In yet another aspect, the present invention provides a method for extracting the above plant composition, comprising the steps of:
mixing Ginseng radix, ganoderma, herba Saussureae Involueratae, and truffle with extraction solvent, extracting, and separating to obtain filtrate.
Preferably, the extraction solvent is water and/or alcohol solvent, more preferably at least one of water, methanol, ethanol, butanediol and isopropanol, still more preferably a mixed solvent of water and butanediol, still more preferably the mass ratio of water to butanediol is 6-8:2-4, most preferably water and butanediol in a mass ratio of 7: 3.
Preferably, the ratio of the extracted feed to the extracted feed is 1:10 to 80 (m/m), more preferably 1:20 (m/m).
Preferably, the extraction time is 1-3 hours, more preferably 2 hours.
In a further aspect, the present invention provides a plant extract obtained according to the extraction method described above.
In yet another aspect, the present invention provides the use of the above-described plant composition and/or plant extract in the preparation of an anti-aging cosmetic or in the preparation of an anti-aging medicament.
Preferably, the anti-aging includes scavenging DPPH radicals, inhibiting glycosylation reactions, inhibiting MMP-1 and collagen regeneration.
Finally, the invention provides an anti-aging cosmetic comprising the above plant composition and/or plant extract.
The beneficial effects of the invention are as follows:
The cosmetic raw material obtained by the preparation method is safe and environment-friendly, natural in components, free of toxic and side effects and definite in anti-aging effect, and has excellent effects of resisting oxidation, inhibiting glycosylation reaction, inhibiting MMP-1 and stimulating collagen COL-1 secretion through experimental verification.

The saussurea involucrata producing area is Xinjiang, the variety is Saus-surea involucrata (Kar. Et. Kir) Sch. -bip.
The variety of Yunnan in truffle production area is Tuber melanosporum.
Example 1
Ginseng, ganoderma lucidum, saussurea involucrata and truffle in weight ratio: 4:2:0.5:1.

Extraction process
Weighing the medicinal materials according to the proportions of examples 1-4 and comparative examples 1 and 2, respectively, and uniformly mixing and then weighing 5g. Adding 70g of pure water and 30g of butanediol according to a feed liquid ratio of 1:20 (m/m), stirring uniformly, heating in a water bath at 90 ℃ for 2h for extraction, cooling the feed liquid to 25 ℃ and carrying out suction filtration to obtain a plant formula extraction filtrate.
Result detection
1. DPPH free radical scavenging experiments
The experimental method comprises the following steps: and (3) extracting the filtrate from the plant formula to prepare 5.0%, 10%, 15% and 20% concentration solution to be tested. And (3) using absolute ethyl alcohol as a solvent to prepare DPPH solution for later use. A pipe A: 1mL of the solution to be tested is taken and evenly mixed with 1mL of DPPH solution with the mol/L of 2 multiplied by 10 -4; and B, pipe B: 1mL of solvent is taken and evenly mixed with 1mL of DPPH solution with the mol/L of 2 multiplied by 10 -4; c pipe: uniformly mixing 1mL of solvent with 1mL of liquid to be detected; after each tube was allowed to react for 30min in the dark, the absorbance value of A, B, C tubes was measured at 517 nm. The above experiments were carried out using the extracted filtrates obtained by the formulations of examples 1 to 4 and comparative examples 1 and 2,
DPPH free radical inhibition rate calculation formula: DPPH inhibition (%) = (b+c-se:Sub>A)/B.
The results were as follows:
The DPPH free radical inhibition experimental result shows that the plant composition formula provided by the embodiment of the invention has good free radical inhibition effect, and the DPPH free radical inhibition rate reaches more than 50% at the concentration of 10%, 15% and 20%, so that the effect is remarkable. Of these, the radical inhibition effect of example 1 is best, and can reach 83.79% at 20%.
2. Anti-glycation experiment
The experimental method comprises the following steps: adding deionized water into the plant formula extraction filtrate to prepare 5% of sample solution, and sequentially and gradually diluting into 2%, 1% and 0.5% of sample solution by using PBS (phosphate buffered saline). The prepared sample groups (test solution+bovine serum albumin-fructose reaction solution), negative control groups (PBS solution+bovine serum albumin-fructose reaction solution) and blank control groups (test solution+PBS solution) with different concentrations are placed in a 37 ℃ incubator for light-shielding incubation, and the fluorescence intensity of each group is detected on the 5 th day of culture. And spotting the mixed solution into a black 96-well plate, regulating the excitation wavelength to 370 nm and the emission wavelength to 440 nm by using an enzyme-labeled instrument, and finally calculating the inhibition rate of the sample to be tested according to a formula.

The detection result of the concentration of 5% shows that the in vitro anti-saccharification inhibition rate of other samples is basically not quite different and is between 30% and 36% except for the comparative example 1. Wherein example 1 still has an inhibition of 19.2% at a concentration of 0.5%.
3. Safe concentration test for screening cytotoxicity-CCK-8
The experimental method comprises the following steps: after incubation of 96-well plates in a 5% CO 2, 37℃incubator for 24h, the medium was aspirated, 100. Mu.L of the plant formulation extraction filtrate (formulated as 3%, 5% two concentrations, 3 duplicate wells each) was added, and 100. Mu.L of serum-free medium solution (Bioagrio) was added to the blank. The control group was dimethyl sulfoxide. Placing into a 5% CO 2, and incubating for 24 hours at 37 ℃.

4. Anti-aging effect experiment
4.1 MMP-1 vitality detection experiment
The experimental method comprises the following steps: the plant formula extraction filtrate was formulated to be 3% and 5% at two concentrations, and tested according to the instructions of MMP-1ELISA kit (BOSTER).
Experimental results for the formulation of example 1:
compared with the blank, the UVB radiation significantly up-regulates the secretion amount of MMP-1, and the model establishment is successful. The extract obtained by the formula of the embodiment is added to enable the secretion amount of MMP-1 of fibroblasts after UVB radiation to be down-regulated, which proves that the formula of the invention has the effect of inhibiting the secretion of MMP-1. 4.2 COL-1 Activity detection experiment
The experimental method comprises the following steps: the plant formula extraction filtrate is prepared into two concentrations of 3% and 5%, and detection is carried out according to the operation instruction of COL-1 kit (Shanghai enzyme-linked).
Experimental results for the formulation of example 1:
the results show that the formulation of the embodiment of the invention can up-regulate the COL-1 secretion of the fibroblast after UVB radiation, and has the effect of promoting the COL-1 secretion. Wherein at 3% concentration there is a more pronounced effect in promoting COL-1 secretion.
