标签化的转座复合体及其在高通量测序中的应用。提供了一种转座酶识别元件,为如下结构:X(m)Y(f)N(n);X(m)表示转座酶识别区,为双链核酸结构;Y(f)表示间隔区,为单链DNA结构;N(n)表示样本标签,为单链DNA结构。高分子量DNA采用标签化的转座复合体进行处理后,得到大量标签化的DNA片段。将各个高分子量DNA得到的标签化DNA片段混合,得到混合样本;采用具有分子标签的载体进行捕获;采用核酸外切酶进行处理,然后释放转座酶;采用stLFR技术进行建库,得到DNA文库。可用于高通量测序平台混合测序。
