A solid beverage of a probiotic compound composition containing traditional Chinese medicine components is prepared from the following raw materials in percentage by mass: the probiotic composition is 15% (wherein, bifidobacterium BLa 803%, bifidobacterium longum BL 213%, lactobacillus bulgaricus 3%, lactobacillus plantarum LP 902%, bifidobacterium bifidum 2%, lactobacillus rhamnosus 2%), inulin 33%, fructo-oligosaccharide 14%, yam powder 5%, coix seed powder 2%, blueberry fruit powder 3%, skim milk powder 19%, erythritol 9%.
The preparation method comprises the following steps: step 1, checking: all the raw materials are inspected according to corresponding quality standards;
step 2, pretreatment of the traditional Chinese medicine raw materials: selecting and cleaning raw materials including Chinese yam and coix seed, drying (at 35 ℃), crushing, and sieving by a 24-mesh sieve to obtain fine powder for later use;
step 3, pretreatment of auxiliary materials: drying inulin, fructo-oligosaccharide, blueberry fruit powder, skim milk powder and erythritol (35 deg.C), and sieving with 24 mesh sieve;

1. Pilot test:
the results of pilot scale production were shown in tables 1 and 2, based on the formulation ratios listed in examples 1 to 8.

As can be seen from tables 1 and 2, the solid beverage of the probiotic compound composition containing the traditional Chinese medicine components meets the requirements of enterprise quality standards, the yield meets the regulations, and the production process of the product is reasonable.
2. And (3) sensory index inspection:
1) a small amount of the solid beverage of the probiotic compound composition containing the traditional Chinese medicine components prepared in the embodiments 1-8 is respectively taken and subjected to sensory index inspection according to GB/T29602-2013 (solid beverage) standards, and the operation steps are as follows: a50 mL sample was poured into a colorless transparent container and placed in a bright place, and the state, color, smell and taste were observed, and the results are shown in Table 3.

As can be seen from tables 3 and 4, the state, color, smell and taste of the solid beverage of the probiotic compound composition containing traditional Chinese medicine components all meet corresponding indexes.
3. Physical and chemical index and microorganism index test
Physical and chemical index inspection: a small amount of each of the probiotic composition-containing solid beverages prepared in examples 1 to 8 was subjected to physical and chemical index tests, and the results are shown in tables 5 and 6. The method comprises the steps of detecting the moisture content according to a GB 5009.3 detection method, detecting the lead content according to a GB 5009.12 detection method, detecting other pollutant limit amount according to a GB 2762 detection method, and detecting mycotoxin limit amount according to a GB 2761 detection method.
And (3) microorganism index detection: a small amount of the solid beverage of the probiotic compound composition containing the traditional Chinese medicine components prepared in the embodiments 1-8 is taken, and the results of the microbial indicator tests are shown in tables 6 and 5. The method for testing the coliform group bacteria comprises the following steps: GB 4789.3MPN counting method, and the mould testing method is GB 4789.15.

As can be seen from tables 5 and 6, the solid beverage containing the probiotic compound composition containing the traditional Chinese medicine components has the water content, the total number of lactic acid bacteria, total arsenic, coliform group bacteria and staphylococcus aureus which meet the corresponding standards, and lead, mould and salmonella are not detected.
And (4) experimental conclusion: through the pilot-scale verification results of experimental examples, all quality indexes of the solid beverage of the probiotic compound composition containing the traditional Chinese medicine components meet the requirements of enterprise quality standards, the yield meets the regulations, and the production process of the product is proved to be reasonable. Through sensory index detection experiments, physicochemical indexes and microbial index detection experiments, all indexes in the obtained probiotic product accord with national standards, and the probiotic product not only has extremely high nutrient content, but also can be eaten at ease.
The following describes the application effect of the product:
constipation clinical effect observation of solid beverage containing probiotic compound composition containing traditional Chinese medicine components
1. Clinical data
300 constipation patients were selected as subjects. The selected patients were included in the treatment group 240 and the control group 60 in the order of treatment using a random number table. The general data of sex, age, course of disease, etc. of two groups of patients have no obvious difference, P is more than 0.05, and the statistical significance is avoided, and the groups have comparability.
2. Diagnostic criteria
The traditional Chinese medicine diagnosis and syndrome differentiation standards and bases are as follows: according to the guideline of clinical research on constipation caused by new traditional Chinese medicine treatment, the standard of syndrome differentiation of heat secret syndrome: dry stool, difficult defecation, prolonged time interval, anal fissure, hemafecia, bitter taste in the mouth, scanty and dark urine, red tongue with yellow coating, and slippery and rapid pulse.
Western diagnosis standard and basis: according to the guideline for clinical research on novel Chinese medicine for treating constipation, the constipation diagnosis standard is as follows: (1) the defecation time is prolonged, and the interval of each defecation is more than 72 hours; (2) the stool is dry and hard, even like the sheep feces or the lump, the defecation is strenuous or the stool is not dry and hard to be discharged. Functional constipation can be diagnosed by eliminating organic lesions through necessary examination. Functional constipation is more than one month and can be brought into test cases according with the heat syndrome of traditional Chinese medicine.
3. Case exclusion criteria: (1) the examination proves that the intestinal canal is caused by rectal and colonic organic lesions (such as tumor, Crohn's disease, colon polyp, intestinal tuberculosis, etc.). (2) Those under 18 years of age, pregnant or lactating women, are allergic to the medicine. (3) Combined with serious primary lesions of cardiovascular, liver, kidney and hematopoietic systems, patients with psychosis. (4) If the composition does not meet the inclusion criteria and is not administered according to the prescription, the curative effect or the data are not complete and the curative effect and safety are not judged.
4. The test method comprises the following steps: (1) dosage and treatment course: example 1 was used for treatment group 1, example 2 was used for treatment group 2, example 3 was used for treatment group 3, example 4 was used for treatment group 4, example 5 was used for treatment group 5, example 6 was used for treatment group 6, example 7 was used for treatment group 7, and example 8 was used for treatment group 8, 2 times/d, 2 g/time, and consecutive 5 days. The treatment effect of each group is compared after 1 treatment course.
5. Observation index

(2) And (3) observation of curative effect: the defecation interval time, the defecation quality, the defecation labor-consuming condition and the defecation regularity.
6. Method for evaluating therapeutic effect

(2) Evaluation standard of constipation curative effect: the clinical recovery: normal stool, all other symptoms disappeared. 7. Statistical analysis
Analyzing and processing each data by adopting statistical software SPSS 22.0, and expressing the measurement data by adopting the mean value plus or minus standard deviation of a sample; the comparison among groups adopts t test; the comparison of the counting data and the inter-group rate adopts non-parameter test, and the difference is shown as P less than 0.05, which has statistical significance.
8. Results
The results are shown in tables 7 to 8.

9. Conclusion
The solid beverage containing the probiotic compound composition containing the traditional Chinese medicine components has a remarkable curative effect on constipation treatment, and no relapse of patients in a treatment group is observed in the follow-up process; in addition, no adverse reaction and toxic or side effect are found in the process of verifying the curative effect, and the preparation is worthy of clinical popularization and application.
The product is added with a probiotic composition (wherein, bifidobacterium BLa80, bifidobacterium longum BL, lactobacillus bulgaricus, lactobacillus plantarum LP90, bifidobacterium bifidum and lactobacillus rhamnosus), inulin, fructo-oligosaccharide, mulberry leaf powder or yam powder, coix seed powder, blueberry fruit powder, skim milk powder and erythritol, and the probiotic has the advantages of convenient taking, quick effect, pure taste and easy storage through the mixture ratio of the components; the preparation method has simple process, the whole process flow of the probiotics is completely sterilized without a terminal, the sanitation control in the whole production process is carried out in a production environment with 10 ten thousand grade cleanliness, the product sanitation index can effectively meet the quality requirement, and the preparation method has the advantages of simple preparation process, strong production operability and low cost, and is suitable for industrial mass production.

The solid beverage of the probiotic compound composition containing traditional Chinese medicine components according to claim 1, wherein the solid beverage comprises the following components in percentage by mass: 14-26% of probiotic composition, 31-36% of inulin, 14-26% of fructo-oligosaccharide, 4-16% of mulberry leaf powder or yam powder, 1-10% of coix seed powder, and the sum of the mass percentages of the components is 100%.
3. The solid beverage containing the probiotic compound composition containing traditional Chinese medicine components as claimed in claim 1, wherein the solid beverage comprises any one or more of blueberry fruit powder, skim milk powder and erythritol.

The solid beverage of the probiotic compound composition containing traditional Chinese medicine components according to claim 3, wherein the solid beverage comprises the following components in percentage by mass: 14-16% of a probiotic composition, 31-33% of inulin, 14-16% of fructo-oligosaccharide, 4-6% of mulberry leaf powder or yam powder, 1-3% of coix seed powder, 1-3% of blueberry fruit powder, 19-21% of skim milk powder and 9-11% of erythritol, wherein the sum of the mass percentages of the components is 100%.

The solid beverage of the probiotic compound composition containing traditional Chinese medicine components according to claims 1 to 4, characterized in that the probiotic composition comprises: one or more of Bifidobacterium BLa80, Bifidobacterium longum BL, Lactobacillus bulgaricus, Lactobacillus plantarum LP90, Bifidobacterium bifidum, and Lactobacillus rhamnosus, and the effective viable count of each strain is not less than 3 × 10 10 CFU/g。
6. The solid beverage of the probiotic compound composition containing traditional Chinese medicine components according to any one of claims 1 to 4, characterized in that the raw material of the mulberry leaf powder is young mulberry buds of the first crop before Qingming.

step 2, auxiliary material pretreatment: drying inulin, fructo-oligosaccharide, blueberry fruit powder, skim milk powder and erythritol, and sieving with a 24-50-mesh sieve for later use;
step 3, crushing the traditional Chinese medicine components: drying the fine powder obtained in the step 1 at a low temperature of-20 to-10 ℃, and then carrying out ultra-low temperature ultra-micro crushing at a temperature of-35 to-30 ℃ to obtain ultra-micro powder for later use;
step 4, sieving the superfine powder obtained in the step 3, wherein the particle size of the superfine powder is not more than 75 microns;
step 5, weighing: weighing the inulin, the fructo-oligosaccharide, the blueberry fruit powder, the skim milk powder and the erythritol which are processed in the step 2 according to the mass parts, weighing the mulberry leaf powder or the yam powder and the coix seed powder which are processed in the step 4 according to the mass parts, and weighing the probiotic composition according to the mass parts for later use;
step 6, mixing: and (5) uniformly stirring the components weighed in the step (5) to obtain the traditional Chinese medicine probiotic compound composition solid beverage.
