Abstract
本发明涉及一种创面修复功能性敷料及其制备方法和用途,属于医用制品技术领域。包括生物活性玻璃,海藻酸钙纤维,壳聚糖,蒸馏水、PH调节剂、芦荟提取物、二氯苯氧氯酚、皮肤粘合剂、聚乙烯醇、胶原蛋白、六亚甲基双胍、褐藻寡糖、苦参、黄柏、卡波姆、三乙醇胺。本发明提供一种创面修复功能性敷料及其制备方法和用途,其具有较好的吸收渗液的作用,一方面形成了对创面愈合的湿润微环境,另一方面,其底部设置成多孔隙结构有利于联合发挥第一功能敷料层和第二功能敷料层的护创功效,达到对伤口最佳的愈合效果,避免频繁揭除敷料换药,降低了伤口二次损伤的风险。

技术领域
本发明涉及一种创面修复功能性敷料及其制备方法和用途,属于医用制品技术领域。
背景技术
伤口是由机械、电、热、化学等因素造成的或由一种医学或生理***形成的皮肤缺陷。根据宏观分类,伤口可以被分成开放性伤口(open wound)和闭合性伤口(closewound)。其中,闭合性伤口主要包括挫伤、血肿和挤压伤。该类伤口的治疗方法往往不同于开放性伤口,因此在文中不做赘述。本文所介绍的敷料,主要针对开放性伤口(简称伤口)的治疗。伤口也可以根据病理学、受伤部位、致伤原因或伤口颜色等条目进行分类。一般来说,根据愈合时间长短,可以分为急性伤口和慢性伤口;根据伤口表面色泽,可以分为红、黄、黑三类;根据受伤原因,可以分为机械性或创伤性伤口、热损伤或化学损伤伤口、溃疡性伤口以及放射性伤口。
伤口愈合是指机体组织遭受创伤后进行再生修复的复杂过程。以皮肤伤口的愈合过程为例,包括伤口早期的炎症反应、伤口收缩、肉芽组织增生和表皮再生等阶段。伤口在不同的愈合阶段,对护理伤口的敷料要求也不相同。随着伤口愈合理论的发展和成熟,各类新型敷料层出不穷地出现。但无论是单一材料的敷料,还是复合型材料的敷料,均无法将复杂的伤口完全护理至愈合,而需要采取多种敷料联合护理。这种经常更换敷料护理伤口,常常会导致伤口的二次损伤或感染,有时甚至会加重伤口的感染。因此,现有敷料在护理一些复杂伤口时常常表面出很大的不足
发明内容
有鉴于此,本发明提供一种创面修复功能性敷料及其制备方法和用途,其具有较好的吸收渗液的作用,一方面形成了对创面愈合的湿润微环境,另一方面,其底部设置成多孔隙结构有利于联合发挥第一功能敷料层和第二功能敷料层的护创功效,达到对伤口最佳的愈合效果,避免频繁揭除敷料换药,降低了伤口二次损伤的风险。

作为本发明的进一步改进生物活性玻璃是采用百合科植物叶子作为生物模板而制得,所述生物活性玻璃具有纳米微孔结构。

所述PH调节剂包括柠檬酸、磷酸、磷酸钠和柠檬酸钠中的至少一种,所述皮肤粘合剂包括羧甲基纤维素钠和α-氰基丙烯酸酯中的至少一种。
所述壳聚糖的制备方法包括如下步骤:
1)除钙:将昆虫甲壳粉碎后,置于质量浓度为10-20%的稀酸中于30-50℃浸煮4-6小时,过滤,取滤饼水洗1-3次后干燥得脱钙产物A待用;
2)脱蛋白:将脱钙产物A、复合酶和水置于反应容器中,控制反应液PH为7.5-8.5,于
40-55℃搅拌水解2-3小时后,过滤,取滤饼水洗干燥得脱蛋白产物B待用;所述复合酶为胰蛋白酶、木瓜蛋白酶和碱性脂肪酶的混合物,三者的质量比为1:1:2;
3)纯化:将脱蛋白产物B置于混合溶剂中,并加入中性盐,于60-80℃搅拌溶解1-3小时后,过滤,滤液经减压干燥去除溶剂,再经水洗去除中性盐,干燥得所需甲壳素;所述混合溶剂为己酸、乙二酸和吡啶的混合物,三者的质量比为1:0.8:0.5;
4)脱乙酰:将步骤(3)所得甲壳素和15-20%NaOH水溶液加入石英玻璃反应容器中,置于紫外光下照射2-3小时,同时予以超声处理,反应结束后降温至室温,离心,沉淀物水洗至中性,于50-60℃干燥24小时后得壳聚糖。
作为本发明的进一步改进一种创面修复功能性敷料制备方法包括
1)将壳聚糖和聚乙烯醇进行混合,溶解于体积比为20-40%乙酸水溶液中,得到浓度为10%的纺丝液,90℃水浴加热并搅拌4-6小时至溶解,静置冷却2h,再将质量为壳聚糖5-30%的海藻酸钙纤维加入到纺丝液中,混合后的溶液放在70℃恒温振荡器中磁力搅拌均匀,静置脱泡后得壳层溶液;
2)将生物活性玻璃、蒸馏水、芦荟提取物、二氯苯氧氯酚、胶原蛋白、六亚甲基双胍、褐藻寡糖、苦参、黄柏、卡波姆、三乙醇胺进行混合在在80-110的温度下,搅拌、真空消泡,倒入模具中,再采用100Gy/S的电子束辐照8-10分钟后,采用去离子水浸泡,每隔12小时换一次水,重复浸泡3次后加入PH调节剂、皮肤粘合剂,在与壳层溶液进行混合即得所需创面修复功能性敷料。
作为本发明的进一步改进创面修复功能性敷料适用于烧伤、切伤、手术创面、***、褥疮、窦道、脂肪液化性伤口、糖尿病足或各类皮肤表面溃疡。

下面将对本发明的优选实施例进行详细的描述。
实施例一
本发明提供一种创面修复功能性敷料及其制备方法和用途,其包括生物活性玻璃10份,海藻酸钙纤维25份,壳聚糖15份,蒸馏水50份、PH调节剂、芦荟提取物3份、二氯苯氧氯酚1份、皮肤粘合剂、聚乙烯醇2份、胶原蛋白1份、六亚甲基双胍2份、褐藻寡糖15份、苦参1份、黄柏3份、卡波姆5份、三乙醇胺3份。

2)将生物活性玻璃、蒸馏水、芦荟提取物、二氯苯氧氯酚、胶原蛋白、六亚甲基双胍、褐藻寡糖、苦参、黄柏、卡波姆、三乙醇胺进行混合在在80-110的温度下,搅拌、真空消泡,倒入模具中,再采用100Gy/S的电子束辐照8-10分钟后,采用去离子水浸泡,每隔12小时换一次水,重复浸泡3次后加入PH调节剂、皮肤粘合剂,在与壳层溶液进行混合即得所需创面修复功能性敷料。

实施例二

1.一种创面修复功能性敷料,其特征在于:包括生物活性玻璃10-15份,海藻酸钙纤维25-40份,壳聚糖15-25份,蒸馏水50-90份、PH调节剂、芦荟提取物3-15份、二氯苯氧氯酚1-10份、皮肤粘合剂、聚乙烯醇2-8份、胶原蛋白1-8份、六亚甲基双胍2-10份、褐藻寡糖15-25份、苦参1-15份、黄柏3-12份、卡波姆5-15份、三乙醇胺3-10份;
所述生物活性玻璃是采用百合科植物叶子作为生物模板而制得,所述生物活性玻璃具有纳米微孔结构;
所述壳聚糖的制备方法包括除钙、脱蛋白、纯化、脱乙酰。

5.根据权利要求4所述的一种创面修复功能性敷料,其特征在于:所述PH调节剂包括柠檬酸、磷酸、磷酸钠和柠檬酸钠中的至少一种,所述皮肤粘合剂包括羧甲基纤维素钠和α-氰基丙烯酸酯中的至少一种。
6.根据权利要求4所述的一种创面修复功能性敷料,其特征在于:所述壳聚糖的制备方法包括如下步骤:
1)除钙:将昆虫甲壳粉碎后,置于质量浓度为10-20%的稀酸中于30-50℃浸煮4-6小时,过滤,取滤饼水洗1-3次后干燥得脱钙产物A待用;
2)脱蛋白:将脱钙产物A、复合酶和水置于反应容器中,控制反应液PH为7.5-8.5,于40-55℃搅拌水解2-3小时后,过滤,取滤饼水洗干燥得脱蛋白产物B待用;所述复合酶为胰蛋白酶、木瓜蛋白酶和碱性脂肪酶的混合物,三者的质量比为1:1:2;
3)纯化:将脱蛋白产物B置于混合溶剂中,并加入中性盐,于60-80℃搅拌溶解1-3小时后,过滤,滤液经减压干燥去除溶剂,再经水洗去除中性盐,干燥得所需甲壳素;所述混合溶剂为己酸、乙二酸和吡啶的混合物,三者的质量比为1:0.8:0.5;
4)脱乙酰:将步骤(3)所得甲壳素和15-20%NaOH水溶液加入石英玻璃反应容器中,置于紫外光下照射2-3小时,同时予以超声处理,反应结束后降温至室温,离心,沉淀物水洗至中性,于50-60℃干燥24小时后得壳聚糖。
7.根根据权利要求1、2、3、5、6中的任意一项所述的一种创面修复功能性敷料制备方法,其特征在于:包括步骤如下:
1)将壳聚糖和聚乙烯醇进行混合,溶解于体积比为20-40%乙酸水溶液中,得到浓度为10%的纺丝液,90℃水浴加热并搅拌4-6小时至溶解,静置冷却2h,再将质量为壳聚糖5-30%的海藻酸钙纤维加入到纺丝液中,混合后的溶液放在70℃恒温振荡器中磁力搅拌均匀,静置脱泡后得壳层溶液;
2)将生物活性玻璃、蒸馏水、芦荟提取物、二氯苯氧氯酚、胶原蛋白、六亚甲基双胍、褐藻寡糖、苦参、黄柏、卡波姆、三乙醇胺进行混合在在80-110的温度下,搅拌、真空消泡,倒入模具中,再采用100Gy/S的电子束辐照8-10分钟后,采用去离子水浸泡,每隔12小时换一次水,重复浸泡3次后加入PH调节剂、皮肤粘合剂,在与壳层溶液进行混合即得所需创面修复功能性敷料。
8.根据权利要求1-3任一项所述的一种创面修复功能性敷料用途,其特征在于:所述创面修复功能性敷料适用于烧伤、切伤、手术创面、***、褥疮、窦道、脂肪液化性伤口、糖尿病足或各类皮肤表面溃疡。
