本发明属于动物传染性病防治技术领域,具体涉及一株突变的金黄色葡萄球菌噬菌体及其应用。本发明分离的金黄色葡萄球菌噬菌体4PHCISA25保藏在中国典型培养物保藏中心,保藏编号为CCTCC NO:M20211354。本发明提供了所述噬菌体在防治金黄色葡萄球菌及其生物被膜作为杀菌剂的作用,为临床治疗金黄色葡萄球菌感染疾病提供了具有应用潜力的杀菌剂制品。本发明的金黄色葡萄球菌噬菌体裂解范围广,裂解能力强,能够强有力地杀灭耐甲氧西林的金黄色葡萄球菌。

Description

一株突变的金黄色葡萄球菌噬菌体及其应用
技术领域
本发明属于动物传染病防治技术领域,具体涉及一株突变金黄色葡萄球菌噬菌体及其应用。
背景技术
金黄色葡萄球菌(Staphylococcus aureus,S.aureus)是G+菌,能够引起人和家畜多种感染性疾病,例如局部脓肿、败血症和脓毒血症等。金黄色葡萄球菌可以附着并持续存在于宿主组织(如骨骼和心脏瓣膜)上,引起骨髓炎和心内膜炎。该菌也是引起细菌性食物中毒和医院内感染常见的病原体之一,与食品行业及医药领域的卫生和安全休戚相关。近年来,由于抗生素的使用致使金黄色葡萄球菌耐受甲氧西林形成“超级细菌”(MRSA),每年导致大量感染“超级细菌”的患者发病并造成21.8%的感染患者死亡。MRSA菌株易于形成生物被膜(biofilm,BF),生物被膜基质保护宿主免受吞噬作用和其他先天性和适应性免疫和炎症***的影响。金黄色葡萄球菌通过其高度结构化的细胞外基质生物膜介导的抗性对大量抗菌剂的敏感性降低了10-1000倍。大量抗生素缺乏穿透细胞膜和细菌生物膜的能力。这些使得治疗金黄色葡萄球菌感染愈加艰难极具挑战。迄今为止,在临床试验中还没有专门针对靶向生物膜的获批药物。可见,防治金黄色葡萄球菌感染成为目前国际上医药、农业安全领域的急切任务之一。
随着抗生素在农业、医药和食品领域的过度使用和误用,导致了当前抗菌素的耐药性危机。在正在探索的众多选择中,作为细菌的天然病毒敌人—噬菌体疗法的复兴是一种有吸引力的替代或补充疗法。噬菌体对比抗生素有以下优势:作为专性细菌病毒对细菌宿主具有特异性,对非目标微生物没有毒性并且杀伤活性仅限于狭窄范围的致病菌株,避免了对人类和动物健康共生微生物群的附带损害。噬菌体提供动态剂量,当目标细菌宿主菌株存在时剂量增加,并随着目标菌的数量而减少。噬菌体通常能够独立于其多重耐药表型杀死细菌,表现出对抗生素耐药细菌的有效性。众所周知,噬菌体治疗安全性高,噬菌体主要由蛋白质和核酸组成具有免疫原性,对噬菌体静脉注射实验已经反复进行,并没有观察到其主要的副作用。迄今为止所报道的临床实验都表明它们没有表现出显着的毒性。基于以上优势使得噬菌体的应用具有了更广阔的前景。
除了最终导致细菌死亡的裂解循环的烈性噬菌体外,一些噬菌体也可以经历所谓的溶原循环即病毒基因组与细菌染色体整合并复制的情况,可以积极促成水平基因转移导致抗菌素耐药性和毒力基因的传播,这种类型的噬菌体被称为温带噬菌体。因此,温和的噬菌体不被认为是噬菌体治疗的良好候选者。截至2018年在国家生物技术信息中心(NCBI)数据库已报道了200多株金黄色葡萄球菌噬菌体。对其中127株长尾病毒科噬菌体的生物信息学分析,其中大多数(125株噬菌体,98.4%)的基因组中存在溶原决定基因簇是温和噬菌体。因此,对金黄色葡萄球菌温带噬菌体的溶原决定基因簇进行基因工程改造,获得烈性噬菌体是进一步应用控制金黄色葡萄球菌的有用策略。研究表明,焦磷酸盐可用于包括质粒和病毒在内的染色体外元件的DNA突变。通过将噬菌体颗粒暴露于多轮螯合剂(如乙二胺四乙酸,即EDTA)、柠檬酸钠或焦磷酸钠)中,可以轻松分离噬菌体缺失突变体。焦磷酸钠通过结合位于噬菌体颗粒表面的阳离子对噬菌体衣壳产生不稳定作用,这会导致构象缺陷并最终导致活力丧失从而产生突变噬菌体。因此,急需随机突变转化为烈性噬菌体是本领域亟需解决的技术问题。
发明内容
本发明的目的在于克服现有技术的缺陷,筛选得到一株突变的金黄色葡萄球菌噬菌体(4PHCISA25),该噬菌体突变后不具有整合作用,不会介导细菌耐药性和毒力基因水平基因的转移。可望作为一种新型金黄色葡萄球菌的防治菌剂,表明该噬菌体4PHCISA25为广谱型能够有效抑制29株耐甲氧西林的金黄色葡萄球菌。
本发明的第二个目的是在于提供了一株突变金黄色葡萄球菌噬菌体在降解金黄色葡萄球菌生物被膜的应用。
本发明的另一个目的是提供一株突变金黄色葡萄球菌噬菌体在皮肤脓肿抑菌剂中的应用。
为了实现上述技术目的,本发明采用以下技术措施:
本发明以金黄色葡萄球菌为宿主菌分离出一株裂解能力强、裂解范围广的温和噬菌体,通过焦磷酸钠对其进行突变成烈性噬菌体。研究其生物学特性,为该噬菌体在兽医临床应用提供理论基础。申请人将筛选的噬菌体命名为金黄色葡萄球菌噬菌体4PHCISA25,Staphylococcus aureus bacteriophage 4PHCISA25于2021年11月01日送交中国.武汉.武汉大学中国典型培养物保藏中心保藏,保藏编号为CCTCC NO:M20211354。
本发明提供的噬菌体4PHCISA25为有尾噬菌体目长尾噬菌体科,在4℃-60℃和pH为5-9的条件下效价保持稳定。在25℃保存6个月活性稳定。在4℃保存9个月后活性稳定。该菌株对紫外光有一定的抵抗作用,存活率保持相对稳定。对酒精不耐受,即在酒精存在条件下10min即可灭活。
本发明还提供一种所述金黄色葡萄球菌噬菌体4PHCISA25在防治金黄色葡萄球菌生物被膜中的应用。以及在制备金黄色葡萄球菌感染疾病抑菌剂中的应用,例如可用于抑制耐甲氧西林金黄色葡萄球菌造成的皮肤脓肿。
本发明的有益效果如下:
(1)现有技术表明经焦磷酸钠突变的噬菌体不具有整合作用,而本发明提供的噬菌体4PHCISA25应用安全性高。
(2)本发明的噬菌体热稳定性好。本发明提供的噬菌体4PHCISA25优于专利文献201810715693 0中的金黄色葡萄球菌噬菌体SH-St 15644热稳定性(4℃-40℃稳定),以及专利文献CN2015109752592的金黄色葡萄球菌噬菌体qdsa001的热稳定性(4℃-50℃稳定)。
(3)本发明的噬菌体在不同MOI的条件下,本发明的噬菌体抑菌能力强。在本发明中,噬菌体4PHCISA25在MOI=(0.01-100)的条件下对金黄色葡萄球菌均具有很好的抑菌效果。
(4)本发明噬菌体的宿主谱宽。本发明提供的噬菌体4PHCISA25对29株耐甲氧西林金黄色葡萄球菌均具有良好的裂解效果。
(5)本发明的噬菌体的降解生物被膜能力强。本发明中噬菌体4PHCISA25对金黄色葡萄球菌所形成中强黏附的生物被膜作用4h后,显著抑制了生物被膜。
(6)本发明的噬菌体的体内治疗效果好。本发明中噬菌体4PHCISA25显著增强了小鼠皮肤愈合速度,脓肿面积和治愈时间显著小于MRSA感染组,且差异具有统计学意义。
综上所述,利用本发明提供的金黄色葡萄球菌噬菌体4PHCISA25能有效抑制和裂解金黄色葡萄球菌,利用本发明的噬菌体制备发的生物制剂(生物菌剂)不仅能作为杀菌剂使用,还可靶向针对生物膜,可广泛应用于养殖、医药和食品安全领域。

SEQ ID NO:1是金黄色葡萄球菌噬菌体的核苷酸序列。
下面结合具体实施例对本发明作进一步的详细阐释。但本发明实施的范围并不仅限于实施例阐释的范围。
实施例1金黄色葡萄球菌噬菌体4PHSA25的分离和筛选
详细步骤如下:
(1)样品采集
猪子***拭子样品采集于广西壮族驻自治区某猪场。
(2)金黄色葡萄球菌噬菌体的分离
猪子***拭子样品用生理盐水浸泡过夜于4℃,8000r离心10min,0.22μm滤器过滤上清。此滤液为用于分离噬菌体的前处理样品,于4℃保存过夜。在10mL TSB液体培养基(胰蛋白胨17g,氯化钠5g,大豆胨3g,葡萄糖2.5g,磷酸氢二钾2.5g,pH7.3±0.2)中按体积比为1:100的比例加入100ul宿主菌(金黄色葡萄球菌SA25),37℃培养使其增殖到对数期。在前述对数期菌液中加入10mL前处理的粪水样品和40mLTSB液体培养基,37℃振荡培养3h~4h,使噬菌体进一步增殖。将增殖的噬菌体液在4℃,8000r离心10min,再用0.22μm滤器过滤上清。采用双层平板法观察有无噬菌斑。具体步骤为:取300μL宿主菌液、1mL过滤后的噬菌体样品和6mL半固体培养基加入10mL灭菌的离心管内,颠倒混匀倒入提前预备的TSA固体平板,上层琼脂凝固后于37℃倒置过夜培养。
(3)噬菌体的纯化
用灭菌白枪头在形成噬菌斑的上层琼脂中挑取大且透亮的单一噬菌斑,将带有噬菌体的枪头在1mL SM缓冲液中吹打混匀,并于4℃冰箱中静置。静置数小时后将噬菌体液振荡混匀,10倍比稀释,取300μL SA25与1mL噬菌体稀释液和6mL半固体培养基加入10mL灭菌的离心管内,颠倒混匀倒入提前预备的TSA固体平板,待上层琼脂凝固后于37℃倒置培养12h。如此重复5次以上步骤直至得到的噬菌斑大小形态均一。
实施例2噬菌体4PHSA25的电镜观察
采用磷钨酸负染色法对噬菌体染色观察(Clokie and Kropinski 2009)。首先对噬菌体进行浓缩,将噬菌体4PHSA25进行大量增殖,取1mL噬菌体原液加入100mL对数生长期的宿主菌菌液(金黄色葡萄球菌SA25)中震荡培养3-4h直至菌液变澄清。4℃下8000r离心10min,取上清用0.22μm滤器过滤除菌。过滤后的噬菌体用长针头加入5mL30%的蔗糖溶液进一步配平,4℃、30000r离心2h浓缩噬菌体,弃上清,沉淀加入200μL TEN溶液重悬。将浓缩后重悬的噬菌体送至透射电镜平台,用常规磷钨酸(PTA)染色制样干燥后用透射电镜观察噬菌体形态。用图像处理软件ImageJ进行长度测量。
结果如图2所示,本发明所分离的噬菌体4PHSA25属于有尾噬菌体目长尾噬菌体科,头部呈类椭圆形,头部直径长80nm±5,宽40nm±5,有条长尾,尾长260nm±5。
实施例3:噬菌体基因组的提取与全基因组测序
噬菌体基因组利用蛋白酶K/SDS的方法提取。取1mL超离后浓缩的噬菌体原液加入RNase20ul、DNaseI 2ul、Buffer10ul,混匀后于37℃裂解1h。按噬菌体浓缩液体积的1/5加入10%的SDS到0.5%,50ul蛋白酶K颠倒混匀后56度水浴1h。加入1mL体积比为25:24:1的酚:氯仿:异戊醇(现用现配,避光贮存)的蛋白质抽提液后颠倒混匀,在13000r,离心10min,先用1mL断枪头再用200uL断枪头吸取上清到新的2mLEP管中,重复3-4次。最后一次将收集的上清转移至10mL EP管内,按照体积比为1:2的比例加提前预冷的无水乙醇中,-20度静置过夜。次日用断枪头将絮状沉淀分装入1.5mL离心管内,13000r/min离心5min。1mL75%无水乙醇洗涤DNA沉淀,13000r/min离心5min,重复3-4次;在超净台晾干成透明状后用50ul去离子水重悬上述DNA白色沉淀,4℃溶解DNA。-20℃保存备用,将所得DNA样品交于商业测序公司测序。
噬菌体4PHSA25的全基因测序结果来看,噬菌体基因组为双链DNA,序列全长为45314bp(见序列表SEQ ID NO:1)。对测序结果注释分析:噬菌体4PHSA25含有整合酶、阻遏蛋白及溶原模块(Lysogeny module),即可判定4PHSA25为一株溶原性噬菌体。
实施例4噬菌体4PHSA25的突变试验
将野生型噬菌体4PHSA25(108PFU/mL)与300mM焦磷酸钠(pH7.4)混合,在42℃下孵育40min后与金黄色葡萄球菌宿主菌株混合培养。将1mL SM缓冲液(Na Cl 5.8g,MgSO4·7H2O 2.0g,明胶0.1g,1.0mol/L Tris-HCl(pH7.5)50.0mL)添加至噬菌体的平板中,并在室温下轻微摇动(60rpm)孵育1h。收集液体4℃10,000rpm离心10分钟,使上清液中含有尚存的噬菌体。SM缓冲液中的噬菌体再次用相同浓度的焦磷酸钠处理,并在相同的条件下孵育,重复整个突变过程30次。仔细观察假定的噬菌体缺失突变体形成的裂解噬菌斑,并与野生型噬菌体形成的裂解噬菌斑进行比较。挑取每个假定的“透明裂解噬菌斑”并将其悬浮在200μL SM缓冲液中。PCR测序噬菌体溶原模块(Lysogeny module)是否产生突变位点,测序成功突变的噬菌体进行整合菌株的筛选,具体操作步骤如下:取对数前期的宿主菌(例如金黄色葡萄球菌SA25)300uL,加入1mL突变噬菌体(4PHCISA25),铺双层平板,37℃孵育,待长出单克隆后,挑取单克隆进行PCR检测,用3轮进行纯化。
噬菌体4PHCISA25阻遏蛋白编码基因中的Leu 1个碱基替换,TTA密码子的1个碱基替换到终止密码子TAA。以整合酶基因(1200bp)为目的基因扩增PCR(正向引物F;gttcgtgtacgtttgagaatgg,反向引物R;ctcctgtaagaagaagcgc反应体系:2×Rapid TaqMaster Mix 10μL,上下游引物各1μL,模板2μL,ddH2O 6μL共20μL体系。反应条件95℃5min,95℃15s,55℃15s,72℃30s,30个循环,72℃10min.),经3轮纯化后突变噬菌体未筛选到整合菌株,说明筛选得到的突变噬菌体不再具有整合作用,因此本发明成功筛选得到一个烈性噬菌体。
实施例5噬菌体4PHCISA25宿主谱的测定
试验选择30种金黄色葡萄球菌菌株(为常规应用菌株:例如SA1、SA2、SA3、SA4、SA5、SA6、SA7、SA8、SA9、SA10、SAA、SA12、SAB、SAC、SA15、SA16、SA17、SA18、SA19、SA20、SA21、SA22、SA23、SA24、SA25、SA26、SA27、SA28、SA29、SA30,实验室保存,实施本发明并不限于上述菌株)和8种其他菌属(为常规应用的菌株:例如鼠伤寒沙门氏菌1344、甲型副伤寒沙门氏菌201007、大肠杆菌E02、粪肠球菌EF3964、巴氏杆菌A型HB03、巴氏杆菌D型HB05、波士杆菌D8D9、肺炎克雷伯菌kp9,实验室保存,实施本发明并不限于上述菌株)来对噬菌体4PHCISA25进行宿主谱分析(结果见表1),具体步骤如下:
分别培养上述菌株(入上所述的30种金黄色葡萄球菌菌株和8种其他种属菌株)至对数生长期。取300μL培养至对数期的菌液和6mL半固体培养基加入10mL灭菌的离心管内,颠倒混匀倒入提前预备的TSA固体平板,室温放置干燥。10倍比稀释噬菌体4PHCISA25,取10uL噬菌体稀释液依次滴加至上层琼脂表面,待液体表面干燥后于37℃下倒置培养过夜观察对菌株的裂解能力,试验重复三次,取平均值。以宿主菌的噬菌体滴度为1,其他菌株的噬菌体滴度与宿主菌的噬菌体滴度的比值为EOP(Efficiency of plating)。
结果如表1所示,本发明的噬菌体4PHCISA25可裂解30株金黄色葡萄球菌中的29株,表现出广谱性。此外,噬菌体4PHCISA25不能感染其他革兰氏阳性和革兰氏阴性细菌,这表明该噬菌体对金黄色葡萄球菌具有较高的宿主特异性。

实施例6噬菌体4PHCISA25裂菌能力的评价
(1)挑取金黄色葡萄球菌菌株SA25,以单菌落接种于5mL TSB培养基(常规培养基)中,220r/min、37℃摇床中振荡培养15h。次日在5mL TSB液体培养基中按体积比为1:100的比例转接金黄色葡萄球菌,然后在220r/min,37℃摇床中培养振荡3h。培养液10倍比稀释取100ul涂平板计数,备用。
细菌的数量(CFU/mL)计算公式=单菌落个数×稀释梯度×10
(2)取噬菌体4PHCISA25原液(噬菌体4PHCISA25原液的制备方:以金黄色葡萄球菌SA25为宿主菌,取100ul金黄色葡萄球菌SA25、100ul 106PFU/mL 4PHCISA25与6mL半固体培养基混合,混匀后倒入提前预备的TSA固体平板(常规),待上层琼脂凝固后于37℃下倒置培养12h使噬菌体增殖。将上层琼脂加入5mL SM缓冲液(常规),取5mL增殖液于离心管中,4℃下12000r离心10min,0.22μm滤器过滤上清即为噬菌体原液。噬菌体10倍比稀释取10ul点斑法测效价,备用。
噬菌体的效价(PFU/mL)=噬菌斑个数×稀释倍数×100
(3)取对数期金黄色葡萄球菌菌液,在100孔板中将100uL对数期的宿主菌与100uL不同MOI(0.01、0.1、1.0、10、100)噬菌体混合,37℃低速摇动。同时设对照组,加入等体积TSB培养基或磷酸盐缓冲液(PBS,PH=7.5)缓冲液作为阴性对照,加入等体积金黄色葡萄球菌菌液作为阳性对照。通过全自动生长曲线仪器测定OD 600值,每隔1h测定一次直至12h,该试验重复三次。
结果如图3所示:与不加噬菌体的阳性对照相比,噬菌体4PHCISA25在MOI=(0.01-100)感染金黄色葡萄球菌时均表现出有效的杀菌特性,且显著延迟了细菌的再次增殖。在8h后试验组吸光值增大,说明金黄色葡萄球菌菌量有所上升,对噬菌体4PHCISA25产生抗性菌株,不能完全杀死细菌,但是试验组明显低于阳性对照组,且当MOI值足够高时,对宿主菌的抑制能力也更强。
实施例7噬菌体4PHCISA25一步生长曲线的测定
挑取金黄色葡萄球菌SA25单菌落接种于5mL TSB培养基中,220r/min、37℃摇床中振荡培养15h。次日在20mL TSB液体培养基中按体积比为1:100的比例转接金黄色葡萄球菌,37℃下培养3-4h使其达到对数期,以最佳MOI=0.01加入实例6中制备的噬菌体4PHCISA25混匀后于37℃温孵10min后,在8000r/min离心10min,洗涤沉淀3次,以彻底洗去未吸附的噬菌体。用等体积的TSB培养基重悬沉淀,置于摇床37℃振荡培养,自0min起每隔10min取样200ul,于4℃,12000r/min下离心10min,吸取上清液至新的1.5mLEP管中10倍比稀释点斑法测定噬菌体的滴度。
结果如图4所示:噬菌体4PHCISA25的潜伏期约为10min;10~100min处于裂解期;爆发期约为90min;裂解量约为470PFU/cell。
实施例8:噬菌体4PHCISA25的pH稳定性的测定
取1.5mL离心管分别加入900uL不同pH值(3~12)的PBS缓冲溶液,将100uL(1010PFU/mL)噬菌体加入其中37℃水浴1h,10倍比稀释采用点斑法测定不同PH处理组噬菌体滴度。每个试验重复三遍。
结果如图5所示:噬菌体4PHCISA25在pH值为5.0-9.0范围内有较高的活性,而pH低于5.0或者大于11.0时,则生物学活性显著下降。当pH值为3和12时,则噬菌体完全失活失去感染宿主的能力。
实施例9噬菌体4PHCISA25的热稳定性的测定
取1.5mL离心管加入1mL(1010PFU/mL)噬菌体,分别调节恒温水浴锅(4℃、20℃、40℃、50℃、60℃、70℃和80℃),待水浴锅温度稳定后分别作用20min、40min和60min,以10倍比稀释采用点斑法测定不同时间噬菌体滴度。每个试验重复三遍。
结果如图6所示:噬菌体4PHCISA25的效价在4-50℃基本保持原有活性,50℃~60℃效价开始降低,70℃温孵40min,80℃温孵20min噬菌体基本被杀灭。噬菌体4PHCISA25对高温(70℃以上)耐受性较差,适合在60℃以下环境中生存。
实施例10噬菌体4PHCISA25的紫外、75%酒精稳定性的测定
紫外稳定性测定:在6孔板中每孔加入1mL(1010PFU/mL)噬菌体液,置于(20w,90cm)紫外灯下分别作用5min、10min、15min、30min,作用后将噬菌体液放置暗处稳定30min,每组设定3个重复,以1 0倍比稀释采用点斑法测定不同照射组噬菌体滴度。
酒精稳定性测定:取1.5mL无菌EP管加入990ul 75%酒精,将10ul(1010PFU/mL)噬菌体液加入处理,同时设置对照将10ul噬菌体原液加入990ul ddH2O。每隔10min取样测定一次直至30min,每组设定3个重复,以10倍比稀释采用点斑法测定不同时间噬菌体滴度。
结果如图7所示:噬菌体4PHCISA25在不同时间紫外(20w,90cm)照射下存活率保持相对稳定,在紫外下照射30min噬菌体效价高达109PFU/mL,表明该噬菌体对紫外光有一定的抵抗作用(图7a)。噬菌体4PHCISA25在75%酒精作用下10min基本检测不到噬菌体的存在,表明该噬菌体对酒精不耐受(图7b)。
实施例11噬菌体4PHCISA25对金黄色葡萄球菌生物膜的抑制
试验方法如下所述:
(1)将金黄色葡萄球菌按体积比1:1000振荡培养15h,次日用NaCl-Free培养基(常规)将金黄色葡萄球菌按体积比为1:100进行稀释。
(2)在96孔板中每孔加入200uL稀释菌液,同时设置培养基对照组,37℃静置孵育24h。
(3)孵育结束后,轻轻吸出培养液,每孔用200ul PBS洗涤1遍。
(4)在试验组中向96孔板中每孔分别加入100μL(109PFU/mL)的噬菌体稀释液;对照组对形成生物膜的孔中加入100μL NaCl-Free培养基。恒温培养箱37℃培养4h。
(5)孵育结束后,用PBS洗涤96孔板1遍,晾干。用200uL甲醇固定30min。弃甲醇,PBS洗涤1遍,自然晾干。
(6)用200μL的1%结晶紫染色15min。弃结晶紫,PBS洗涤3遍直至溶液呈无色,晾干。
(7)加入150uL的33%冰乙酸,放水平摇床摇15min,用酶标仪OD590nm读取数值。
结果如图8所示:当噬菌体4PHCISA25的浓度为108PFU/mL时,加入噬菌体后(编号为SA1、SA9、SAA、SAC、SA21)OD590nm显著下降,其中SA1下降64.66%、SA9下降45.65%、SAA下降47.77%、SAC下降69.47%、SA21下降43.28%。由此可知,噬菌体4PHCISA25对金黄色葡萄球菌形成的生物被膜表现出良好的抑制效果。
实施例12噬菌体4PHCISA25治疗金黄色葡萄球菌感染小鼠的效果评估
(1)试验动物
6周SPF级雌性balb/c鼠,购自湖北宜昌三峡大学。
(2)金黄色葡萄球菌皮肤脓肿模型的构建
用棉签蘸取硫化钠去除小鼠背部毛,用酒精棉球擦拭表面剩余硫化钠并消毒背部皮肤。去毛面积约为2cm×2cm占体表面积15%左右。按实例6方案制备金黄色葡萄球菌菌液,调节菌量至1×108CFU/mL。用吸入式***对小鼠进行麻醉处理,用1mL注射器皮下注射100μL(1×108CFU/mL)金黄色葡萄球菌悬液。24h后局部感染形成皮下脓肿。
(3)应用4PHCISA25治疗皮肤脓肿裸鼠
注射菌液1h后在相同位置皮下注射100μL噬菌体4PHCISA25治疗(MOI=10),并设置阳性对照组(注射100μl无菌SM缓冲液)。每天拍照记录小鼠治疗效果,使用量尺测量小鼠皮肤脓肿的长(L)和宽(W),计算脓肿面积area(A)=π(L×W)/2,绘制小鼠背部皮肤感染的脓肿面积曲线。
单独使用噬菌体皮下注射并没有对小鼠造成皮肤损伤,可以安全应用。噬菌体4PHCISA25治疗组小鼠的皮肤脓肿面积和治愈时间小于MRSA感染组(图9a),显著增强了小鼠皮肤愈合速度。噬菌体4PHCISA25治疗组24h即可显著抑制小鼠皮肤脓肿(图9b)。噬菌体治疗组小鼠皮肤脓肿平均细菌裁量为7.81×107CFU/g;MRSA感染组小鼠鼠皮肤脓肿平均细菌载量为8.87×108CFU/g;与感染组相比,噬菌体治疗组在24小时后导致细菌数量减少1.1log,差异具有统计学意义(见图9c)。表明本发明的噬菌体4PHCISA25是治疗MRSA引起的皮肤感染的强有力的生物杀菌剂。
