Abstract
The invention discloses a method for improving whitening effect of pseudo-ginseng cream, belonging to the technical field of cosmetics, and the method comprises the steps of adding chitosan oligosaccharide, algal polysaccharide and pseudo-ginseng extract into the pseudo-ginseng cream, wherein the addition amount of the chitosan oligosaccharide in the pseudo-ginseng cream is 3-8 wt%, the addition amount of the algal polysaccharide in the pseudo-ginseng cream is 0.05-0.2 wt%, and the addition amount of the pseudo-ginseng extract in the pseudo-ginseng cream is 0.5-2 wt%; the pseudo-ginseng facial cream disclosed by the invention can effectively promote skin microcirculation, is beneficial to absorption of pseudo-ginseng active ingredients, deeply whitens and removes freckles, and can effectively fade and remove various stubborn spots and inhibit generation of new pigments; meanwhile, the skin care product is rich in various nutrient substances such as polysaccharide, protein and the like, effectively nourishes the skin and makes the skin color uniform and bright.

Technical Field
The invention relates to the technical field of cosmetics, in particular to a method for improving whitening effect of pseudo-ginseng cream.
Background
Melanin is a major cause of human skin pigmentation, and the molecular mechanism of action of most whitening raw materials is to improve skin color by reducing the production of melanin, wherein inhibition of tyrosinase activity is one of the major strategies for reducing melanin production. However, some traditional effective whitening raw materials such as kojic acid and hydroquinone can cause side effects such as skin sensitivity and canceration after long-term use.
Notoginseng, also known as Tianqi, is an Araliaceae plant of Umbelliferae and is mainly distributed in Yunnan, Guangxi, Jiangxi, Sichuan and other places. The pseudo-ginseng of the same name takes the root of the pseudo-ginseng as a medicinal part and has the effects of removing blood stasis, stopping bleeding, reducing swelling and relieving pain. Can be used for treating hemoptysis, hematemesis, epistaxis, hematochezia, metrorrhagia, traumatic hemorrhage, thoracico-abdominal pain, and traumatic swelling and pain. Has been recognized as having remarkable efficacies of promoting blood circulation by removing blood stasis, relieving swelling and relieving pain since ancient times, and has the reputations of 'Jinbuhuan' and 'Nanguo Shencao'.
Pseudo-ginseng is a famous and precious medicinal material from China and China, and the main components of the pseudo-ginseng comprise notoginsenoside, sanchinin, polysaccharide, flavone, protein and the like, and the pseudo-ginseng is used as a medicine by main roots and rhizomes (cut pseudo-ginseng), and the pseudo-ginseng can stop bleeding, remove blood stasis, relieve swelling and pain, promote blood circulation, dissipate blood stasis, resist aging, reduce fat and the like when being used unprocessed. Generally, pseudo-ginseng is used as a raw material in a cosmetic formula, and the effects of promoting blood circulation, removing blood stasis and resisting aging of the pseudo-ginseng are mainly utilized. However, the whitening effect of pseudo-ginseng cosmetics, especially pseudo-ginseng cream, is not good.
Disclosure of Invention
The invention aims to provide a method for improving the whitening effect of pseudo-ginseng cream, and makes up the defect of the pseudo-ginseng cream in the aspect of whitening effect.

Furthermore, the addition amount of the chitosan oligosaccharide in the pseudo-ginseng cream is 3-8 wt%, the addition amount of the algal polysaccharide in the pseudo-ginseng cream is 0.05-0.2 wt%, and the addition amount of the pseudo-ginseng extract in the pseudo-ginseng cream is 0.5-2 wt%.
The chitosan oligosaccharide can penetrate into skin follicular orifice to eliminate melanin and mottle caused by microorganism accumulation, and can inhibit activity of melanin forming enzyme (tyrosinase) and also eliminate melanin caused by metabolism disorder. The addition of chitosan oligosaccharide can also increase immunity of human body to bacteria and fungi, inhibit growth of original bacteria, and eliminate dermatitis and acne caused by microorganism invasion. Free amino in the chitosan oligosaccharide molecule can also generate chelation with metal ions to form complex or precipitate, so as to block the metal ions and improve the product quality. After the chitosan oligosaccharide with the molecular weight of 3000 is added into the pseudo-ginseng cream, the chitosan oligosaccharide has positive charges, so that the skin surface of a human body has negative charges, the absorption of the human body to the pseudo-ginseng cream is promoted, and the whitening effect is improved. The mechanism of action of algal polysaccharide in inhibiting the activity of the neuraminidase is as follows: (1) competitive inhibition: the algal polysaccharide can compete with the substrate for the active site of tyrosinase, such as metal ions such as copper ions on the active site can be chelated; (2) non-competitive inhibition: the algal polysaccharide can be combined with tyrosine residues outside the active center of the enzyme, so that the product after the combination of the substrate and the enzyme cannot be further converted; (3) mixed inhibition: can be combined with active sites of enzyme and amino acid residues outside active centers. The inhibition mechanism of the algal polysaccharide is mixed inhibition, and the algal polysaccharide has good inhibition effect when the concentration of the polysaccharide is 52mmol/L, and does not generate toxic action on human cells. The notoginseng extract contains polyphenol, flavonoid, polysaccharide and other components, and meanwhile, the arbutin content is high, and the notoginseng extract interacts with chitosan oligosaccharide and algal polysaccharide, so that the whitening effect of the notoginseng cream is improved.
Further, the molecular weight of the chitosan oligosaccharide is 2000-5000, preferably 2000-4000, and more preferably 3000.
Furthermore, the molecular weight of the algal polysaccharide is 8-35 kDa.
Further, the preparation method of the pseudo-ginseng extract comprises the following steps:
(1) pretreating Notoginseng radix to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with ethanol, adjusting the pH value to 4-7, heating to 50-55 ℃, adding enzyme, and performing enzymolysis for 50-150 min to obtain an enzymolysis pseudo-ginseng extract solution;
(3) and adding ethyl acetate into the panax notoginseng extract solution after enzymolysis, performing ultrasonic treatment, filtering, centrifuging the filtrate, concentrating and drying to obtain the panax notoginseng extract.
Further, the pretreatment process in the step (1) comprises the steps of placing the pseudo-ginseng at a temperature of-20 to-30 ℃, standing for 20 to 30 hours, and crushing and sieving with a sieve of 80 to 100 meshes.
Further, the enzyme added in the step (2) accounts for 0.1-0.3 wt% of the pseudo-ginseng powder.
Further, the enzymes added in the step (2) are cellulase and pectinase, and the mass ratio of the cellulase to the pectinase is (2-3): 1.

The pseudo-ginseng extract disclosed by the invention contains polyphenol, flavonoid, polysaccharide and the like, can inhibit tyrosinase activity or inhibit proliferation of melanocytes to reduce melanin whitening components, and meanwhile, due to the improvement of an extraction method, the arbutin content in the pseudo-ginseng extract is increased, so that the whitening effect is better. The cell wall of pseudo-ginseng cells can be damaged by using cellulase hydrolysis, the massive exudation of active ingredients contained in the pseudo-ginseng cells is promoted, and the concentration of compounds in a solution is increased by combining the action of ultrasound, so that the extraction rate is effectively improved; meanwhile, pectic substances wrapped around the cells can be effectively decomposed by using the pectinase, so that the diffusion resistance is reduced, and the extraction rate is further improved. Meanwhile, the ethyl acetate is added in the ultrasonic extraction process, so that the interfacial tension between the extraction solution and cell membranes can be remarkably reduced, the extraction solution can enter cell walls through capillary penetration to dissolve out more effective components, and more importantly, arbutin can be specifically enriched, the content of arbutin in the pseudo-ginseng extract is increased, and the whitening effect of the pseudo-ginseng cream is improved.
The invention discloses the following technical effects:
the whitening mechanism of the cosmetic is to accelerate the decomposition of the existing melanin, inhibit the generation of new melanin and inhibit the activity of tyrosinase. Tyrosinase is a key enzyme in the melanin synthesis process, inhibits the activity of tyrosinase, can directly inhibit the synthesis of melanin, and achieves the whitening effect. Inhibiting tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2) in melanin formation process, and blocking or inhibiting melanin synthesis. A large number of research results show that phenols, flavonoids, polysaccharides, volatile oil, organic acids, triterpenoid saponins and the like in a plurality of plant extracts are better sources of tyrosinase inhibitors and melanin synthesis inhibitors. The pseudo-ginseng contains a large amount of phenols, flavonoids and polysaccharides, and also contains a small amount of arbutin, oleanolic acid, ursolic acid, myricetin-3-galactoside and other components, wherein the arbutin and oleanolic acid contained in the pseudo-ginseng have good activity of inhibiting the activity of the tyrosinase, but the pseudo-ginseng-containing cosmetics have poor whitening effect due to low content. Therefore, the invention increases the content of arbutin in the extract and improves the whitening effect of the pseudo-ginseng cream by adjusting the preparation method of the pseudo-ginseng extract, active ingredients contained in the pseudo-ginseng extract can rapidly permeate into skin, effectively inhibit the activity of tyrosinase in the skin and block the formation of melanin while not influencing the cell proliferation concentration, and accelerate the decomposition and excretion of the melanin by directly combining the active ingredients with the tyrosinase, thereby reducing the pigmentation of the skin, removing color spots and freckles, generating no toxic, irritant, allergic and other side effects on the melanocyte, and having the functions of sterilization and inflammation diminishing.
The pseudo-ginseng facial cream disclosed by the invention can effectively promote skin microcirculation, is beneficial to absorption of pseudo-ginseng active ingredients, deeply whitens and removes freckles, and can effectively fade and remove various stubborn spots and inhibit generation of new pigments; meanwhile, the skin care product is rich in various nutrient substances such as polysaccharide, protein and the like, effectively nourishes the skin and makes the skin color uniform and bright.

In the embodiment of the invention, the raw materials of the pseudo-ginseng cream are conventional additives in the field, and the preparation method of the pseudo-ginseng cream is a conventional technical means in the field, is not essential to the invention, and is not described herein again.
The chitosan oligosaccharide in the embodiment of the invention is purchased from Aoxing biological limited company in Zhejiang, and the algal polysaccharide is purchased from Hagal biological technology limited company in Qingdao.
Example 1
Dissolving chitosan oligosaccharide with molecular weight of 2000 in water-soluble Vc solution with concentration of 0.05mg/mL to prepare chitosan oligosaccharide solution with concentration of 10mg/mL, preparing algal polysaccharide with molecular weight of 20kDa into solution with concentration of 52mmol/L, adding chitosan oligosaccharide solution with concentration of 3wt permillage (calculated by chitosan, the same as in the following examples), algal polysaccharide solution with concentration of 0.15 wt% (calculated by algal polysaccharide, the same as in the following examples) and notoginseng extract with concentration of 1 wt% in the preparation process of the notoginseng cream, and then preparing the notoginseng cream.
The preparation method of the notoginseng extract in the embodiment comprises the following steps:
(1) placing Notoginseng radix in refrigerator at-20 deg.C, standing for 20 hr, pulverizing with pulverizer, and sieving with 80 mesh sieve to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with 70% of ethanol according to a mass ratio of 1: 8, mixing, adjusting the pH value to 4, heating to 55 ℃, adding 0.1 wt% of cellulase and pectinase (the mass ratio of the cellulase to the pectinase is 2: 1), performing enzymolysis for 120min, inactivating the enzyme at 70 ℃ for 15min, and naturally cooling to room temperature to obtain an enzymolysis panax notoginseng extract solution;
(3) adding 0.03 wt% ethyl acetate into the enzymolysis Notoginseng radix extract solution, performing ultrasonic treatment at 600W for 20min, filtering with 100 mesh filter cloth, centrifuging the filtrate at 2000rpm for 5min, vacuum concentrating under 0.085Mpa, and freeze drying to obtain Notoginseng radix extract. In this example, the content of arbutin in the notoginseng extract is 6.35 mg/g.
Example 2
Dissolving chitosan oligosaccharide with the molecular weight of 3000 in water-soluble Vc solution with the concentration of 0.05mg/mL to prepare chitosan oligosaccharide solution with the concentration of 10mg/mL, preparing algal polysaccharide with the molecular weight of 35kDa into solution with the concentration of 50mmol/L, adding 3wt per thousand chitosan oligosaccharide solution, 0.15 wt% algal polysaccharide solution and 1 wt% pseudo-ginseng extract in the preparation process of the pseudo-ginseng cream, and then preparing the pseudo-ginseng cream.
The preparation method of the notoginseng extract in this example is the same as that of example 1. In this example, the content of arbutin in the notoginseng extract is 7.10 mg/g.
Example 3
Dissolving chitosan oligosaccharide with the molecular weight of 3000 in water-soluble Vc solution with the concentration of 0.05mg/mL to prepare chitosan oligosaccharide solution with the concentration of 10mg/mL, preparing algal polysaccharide with the molecular weight of 35kDa into solution with the concentration of 50mmol/L, adding 6wt per thousand chitosan oligosaccharide solution, 0.05 wt% algal polysaccharide solution and 0.5 wt% pseudo-ginseng extract in the preparation process of the pseudo-ginseng cream, and then preparing the pseudo-ginseng cream.
The preparation method of the notoginseng extract in this example is the same as that of example 1. In this example, the content of arbutin in the notoginseng extract is 7.21 mg/g.
Example 4
Dissolving chitosan oligosaccharide with the molecular weight of 5000 in a water-soluble Vc solution with the concentration of 0.05mg/mL to prepare a chitosan oligosaccharide solution with the concentration of 10mg/mL, preparing algal polysaccharide with the molecular weight of 20kDa into a solution with the concentration of 50mmol/L, adding 4wt per thousand chitosan oligosaccharide solution, 0.2 wt% algal polysaccharide solution and 2 wt% pseudo-ginseng extract in the preparation process of the pseudo-ginseng cream, and then preparing the pseudo-ginseng cream.
The preparation method of the notoginseng extract in the embodiment comprises the following steps:
(1) placing Notoginseng radix in refrigerator at-30 deg.C, standing for 30 hr, pulverizing with pulverizer, and sieving with 100 mesh sieve to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with 70% of ethanol according to a mass ratio of 1: 6, mixing, adjusting the pH value to 6, heating to 50 ℃, adding 0.2 wt% of cellulase and pectinase (the mass ratio of the cellulase to the pectinase is 3: 1), performing enzymolysis for 50min, inactivating the enzyme at 70 ℃ for 15min, and naturally cooling to room temperature to obtain an enzymolyzed panax notoginseng extract solution;
(3) adding 0.05 wt% ethyl acetate into the enzymolysis Notoginseng radix extract solution, performing 500W ultrasound for 30min, filtering with 100 mesh filter cloth, centrifuging the filtrate at 3000rpm for 3min, vacuum concentrating under 0.090Mpa, and freeze drying to obtain Notoginseng radix extract. In this example, the content of arbutin in the notoginseng extract is 6.83 mg/g.
Example 5
Dissolving chitosan oligosaccharide with the molecular weight of 3000 in water-soluble Vc solution with the concentration of 0.05mg/mL to prepare chitosan oligosaccharide solution with the concentration of 10mg/mL, preparing algal polysaccharide with the molecular weight of 10kDa into solution with the concentration of 52mmol/L, adding 5wt per thousand chitosan oligosaccharide solution, 0.18 wt% algal polysaccharide solution and 0.9 wt% pseudo-ginseng extract in the preparation process of the pseudo-ginseng cream, and then preparing the pseudo-ginseng cream.
The preparation method of the notoginseng extract in the embodiment comprises the following steps:
(1) placing Notoginseng radix in refrigerator at-30 deg.C, standing for 30 hr, pulverizing with pulverizer, and sieving with 100 mesh sieve to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with 70% of ethanol according to a mass ratio of 1: 7, mixing, adjusting the pH value to 7, heating to 50 ℃, adding 0.2 wt% of cellulase and pectinase (the mass ratio of the cellulase to the pectinase is 2: 1), performing enzymolysis for 50min, inactivating the enzyme at 70 ℃ for 15min, and naturally cooling to room temperature to obtain an enzymolyzed panax notoginseng extract solution;
(3) adding 0.05 wt% ethyl acetate into the enzymolysis Notoginseng radix extract solution, performing 500W ultrasound for 30min, filtering with 100 mesh filter cloth, centrifuging the filtrate at 3000rpm for 3min, vacuum concentrating under 0.098Mpa, and freeze drying to obtain Notoginseng radix extract. In this example, the content of arbutin in the notoginseng extract is 7.41 mg/g.
Example 6
Dissolving chitosan oligosaccharide with molecular weight of 4000 in water-soluble Vc solution with concentration of 0.05mg/mL to prepare chitosan oligosaccharide solution with concentration of 10mg/mL, preparing algal polysaccharide with molecular weight of 8kDa into solution with concentration of 50mmol/L, adding chitosan oligosaccharide solution with concentration of 7wt per thousand, algal polysaccharide solution with concentration of 0.05 wt% and pseudo-ginseng extract with concentration of 0.8 wt% in the preparation process of the pseudo-ginseng cream, and then preparing the pseudo-ginseng cream.
The preparation method of the notoginseng extract in the embodiment comprises the following steps:
(1) placing Notoginseng radix in refrigerator at-30 deg.C, standing for 30 hr, pulverizing with pulverizer, and sieving with 100 mesh sieve to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with 70% of ethanol according to a mass ratio of 1: 6, mixing, adjusting the pH value to 6, heating to 55 ℃, adding 0.3 wt% of cellulase and pectinase (the mass ratio of the cellulase to the pectinase is 3: 1), performing enzymolysis for 150min, inactivating the enzyme at 70 ℃ for 15min, and naturally cooling to room temperature to obtain an enzymolyzed panax notoginseng extract solution;
(3) adding 0.01 wt% ethyl acetate into the enzymolysis Notoginseng radix extract solution, performing 700W ultrasonic treatment for 25min, filtering with 100 mesh filter cloth, centrifuging the filtrate at 2800rpm for 4min, vacuum concentrating under 0.090Mpa, and freeze drying to obtain Notoginseng radix extract. In this example, the content of arbutin in the notoginseng extract is 6.94 mg/g.
Example 7
Dissolving chitosan oligosaccharide with molecular weight of 7000 in water-soluble Vc solution of 0.05mg/mL to prepare chitosan oligosaccharide solution with concentration of 10mg/mL, preparing algal polysaccharide with molecular weight of 20kDa into solution with concentration of 50mmol/L, adding chitosan oligosaccharide solution of 4wt per thousand, algal polysaccharide solution of 0.2 wt% and notoginseng extract of 2 wt% in the preparation process of the notoginseng cream, and then preparing the notoginseng cream.
The preparation method of the notoginseng extract in this example is the same as that in example 4.
Example 8
The difference from example 4 is that the preparation method of the notoginseng extract in this example is:
(1) placing Notoginseng radix in refrigerator at-30 deg.C, standing for 30 hr, pulverizing with pulverizer, and sieving with 100 mesh sieve to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with 70% of ethanol according to a mass ratio of 1: 6, mixing, adjusting the pH value to 6, heating to 50 ℃, adding 0.2 wt% of cellulase and pectinase (the mass ratio of the cellulase to the pectinase is 3: 1), performing enzymolysis for 50min, inactivating the enzyme at 70 ℃ for 15min, and naturally cooling to room temperature to obtain an enzymolyzed panax notoginseng extract solution;

Verification example
1. Odor test
100 volunteers were arbitrarily selected, and the odor of the cream prepared in example 1 was scored by the volunteers, 10 was thick in the odor of the traditional Chinese medicine, 0 was odorless, and the evaluation results are shown in table 1. The results of examples 2 to 7 were similar in that example 8 had 12 unacceptable persons, example 9 had a large odor of the cream, and the number of unacceptable persons was 35.

As can be seen from Table 1, the pseudo-ginseng facial cream provided by the invention basically has no traditional Chinese medicine flavor, belongs to a light type and is accepted by people.
2. Test of use effect
Selecting 55 testers, dividing the testers into three age groups, wherein the age groups are 10-20 years old, and observing the whitening effect time and whether allergy occurs in 5 people; the whitening and freckle-removing mask is mainly used for 30 people in 20-50 years old and used for observing whitening, freckle-removing and allergy phenomena; more than 50 people and 20 people mainly observe the whitening and freckle removing effect time and whether allergy occurs, wherein the effect means that the area of the color spot is faded to be more than 90 percent, the color basically disappears, the effect means that the fading of the color spot is not more than 30 percent, and the color change is not obvious. The results are shown in Table 2.

As can be seen from table 2, after chitosan oligosaccharide, algal polysaccharide and notoginseng extract are added in the preparation process of the notoginseng cream, the whitening time of the notoginseng cream can be shortened, and the whitening effect can be improved.
Stability tests are carried out on the pseudo-ginseng cream in examples 1-6 by referring to the cosmetic industry standard QB/T1857-2013 skin moisturizing cream, and observation items comprise appearance changes (discoloration, fading, oil-water separation, caking, particle precipitation, oil-water separation and the like) and odor changes, so that the appearance and the odor of the pseudo-ginseng cream are not obviously changed, and the pseudo-ginseng cream meets the requirements listed in the standard QB/T1857-2013.

1. A method for improving whitening effect of pseudo-ginseng cream is characterized in that chitosan oligosaccharide, algal polysaccharide and pseudo-ginseng extract are added into the pseudo-ginseng cream.

The method for improving whitening effect of pseudo-ginseng cream according to claim 1, wherein the addition amount of chitosan oligosaccharide in the pseudo-ginseng cream is 3-8 wt%, the addition amount of algal polysaccharide in the pseudo-ginseng cream is 0.05-0.2 wt%, and the addition amount of pseudo-ginseng extract in the pseudo-ginseng cream is 0.5-2 wt%.
3. The method for improving whitening effect of pseudo-ginseng cream according to claim 2, wherein the molecular weight of the chitosan oligosaccharide is 2000-5000.
4. The method for improving the whitening effect of the pseudo-ginseng cream according to claim 2, wherein the preparation method of the pseudo-ginseng extract comprises the following steps:
(1) pretreating Notoginseng radix to obtain Notoginseng radix powder;
(2) mixing the pseudo-ginseng powder with ethanol, adjusting the pH value to 4-7, heating to 50-55 ℃, adding enzyme, and performing enzymolysis for 50-150 min to obtain an enzymolysis pseudo-ginseng extract solution;
(3) and adding ethyl acetate into the panax notoginseng extract solution after enzymolysis, performing ultrasonic treatment, filtering, centrifuging the filtrate, concentrating and drying to obtain the panax notoginseng extract.

The method for improving the whitening effect of the pseudo-ginseng cream according to claim 4, wherein the pretreatment process in the step (1) comprises the steps of placing pseudo-ginseng at a temperature of-20 ℃ to-30 ℃, standing for 20-30 hours, crushing and sieving with a sieve of 80-100 meshes.
6. The method for improving whitening effect of pseudo-ginseng cream according to claim 4, wherein the enzyme added in the step (2) accounts for 0.1-0.3 wt% of the pseudo-ginseng powder.
7. The method for improving whitening effect of pseudo-ginseng cream according to claim 4, wherein the amount of ethyl acetate added in the step (3) is 0.01 to 0.05 wt% of the solution of pseudo-ginseng extract after enzymatic hydrolysis.
8. The method for improving the whitening effect of the pseudo-ginseng cream according to claim 4, wherein the ultrasonic power in the step (3) is 500-800 w, and the ultrasonic time is 20-30 min.
9. The method for improving whitening effect of pseudo-ginseng cream according to claim 4, wherein in the step (3), the centrifugation speed is 2000-3000 rpm, and the centrifugation time is 3-5 min.
