Abstract

本发明属于医用敷料的药物技术领域,具体涉及具有破损皮肤修复功能的纤维敷料及其制备方法。本发明的制备方法包括如下步骤,(1)称取聚乙烯醇溶于30%的乙酸中均匀搅拌为2wt%的溶液,再加入等量的壳聚糖粉末在磁力搅拌器下均匀混合,完成基础的电纺溶液制备;(2)之后加入熊果酸,均匀搅拌;(3)以锡箔纸为基材,进行静电纺丝,纺织完成后的电纺膜静置干燥,制得所述纤维敷料。本发明打破传统外敷材料的理念,将低成本、高治疗效果的中药加入外敷材料中,使其材料具有生物活性从而在外敷过程中发挥抗炎、抗氧化、促修复的作用。

Description

具有破损皮肤修复功能的纤维敷料及其制备方法
技术领域
本发明属于医用敷料的药物技术领域,具体涉及具有破损皮肤修复功能的纤维敷料及其制备方法。
背景技术
糖尿病患者伤口易感染不易愈合的问题是临床上待解决的棘手问题。目前,治疗糖尿病创面的常规方法是手术清创和包扎创面。传统的纱布或棉垫,在吸收了伤口产生的体液后,会出现纱布干燥的情况。并且,修复过程中,皮肤新生组织会从纱布空隙中生长进去,所以在更换新的纱布时容易造成二次伤害。
至今,随着技术研究的发展,已有许多种类的敷材在市面贩售,例如:凡士林纱布、半渗透膜敷料、透明薄膜式敷料、亲水性凝胶、亲水性敷料、藻酸盐敷料、亲水性纤维敷料、海绵敷料、抗菌敷料、胶原蛋白、接触式敷料(又称作非黏附性敷料)与复合性材料等。传统敷料虽然种类众多,但保湿、抗菌、吸收、促进愈合、生物兼容性、脱敏性仍较差,需要进一步改良。
和传统材料的拉伸、相分离等方法相比,静电纺丝已成为制取纳米纤维最重要、最有效的方法。然而大多数报道的生物材料敷料缺乏生物活性。目前材料中添加促增生的系列生长因子显现出良好的治疗效果,同时生长因子对组织的副作用、递送***的低效性及价格昂贵限制其临床推广。
发明内容
根据现有技术上存在的缺陷,结合目前的研究前沿,本发明提供了具有破损皮肤修复功能的纤维敷料及其制备方法。伤口愈合过程是一个复杂低炎症的慢性疾病过程,本发明加入具有抗菌抗炎的生物活性物质将大大提高材料的性能和促进伤口愈合。
本发明是采用以下的技术方案实现的:
本发明提供了一种具有破损皮肤修复功能的纤维敷料的制备方法,包括如下步骤,
(1)称取聚乙烯醇溶于30%的乙酸中均匀搅拌为2wt%的溶液,再加入等量的壳聚糖粉末在磁力搅拌器下均匀混合,完成基础的电纺溶液制备;
(2)之后加入熊果酸,均匀搅拌;
(3)以锡箔纸为基材,进行静电纺丝,纺织完成后的电纺膜静置干燥,制得所述纤维敷料。
具体的,所述步骤(2)中熊果酸的终浓度为1×10-3~5×10-3g/ml。
其中,所述步骤(3)中静电纺丝的参数设定为电压20KVp,针头与收集器距离为10cm,流速为0.5ml/min,环境温度控制在25-29℃,湿度50-58%R,收集时间为两小时。
本发明还提供一种上述方法制备的具有破损皮肤修复功能的纤维敷料。
与现有技术相比,本发明的有益效果:
本发明打破传统外敷材料的理念,将低成本、高治疗效果的中药加入外敷材料中,使其材料具有生物活性从而在外敷过程中发挥抗炎、抗氧化、促修复的作用,促进伤口加快恢复来缓解病人的痛苦。

具体实施方式
为了使本发明目的、技术方案更加清楚明白,下面结合附图,对本发明作进一步详细说明(如图1所示)。下述实施例中所述实验方法,如无特殊说明,均为常规方法;实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件或者按照产品说明书进行;所述试剂和材料,如无特殊说明,均可从商业途径获得。
1、下列实施例采用的材料如下:
生物医药专用静电纺丝机CPS-60K02VIT,购自韩国首尔春巴机电有限公司。
接触角测量仪OCA25,购自北京奥德利诺仪器有限公司。
2、Ⅱ型糖尿病(T2DM)小鼠
4周龄Ⅱ型糖尿病小鼠在12:12小时白昼晚上交替,室温状况下喂养,喂养高糖模型饲料,适应性喂养1周,自由饮食。
一、制备具有破损皮肤修复功能的纤维敷料
实施例1~实施例5
(1)称取0.2g聚乙烯醇溶于10ml 30%的乙酸中均匀搅拌为2wt%的溶液,再加入0.2g的壳聚糖粉末在磁力搅拌器下均匀混合1小时,完成基础的电纺溶液制备;
(2)之后实施例1~实施例5分别加入不同质量的熊果酸(0.01g、0.02g、0.03g、0.04g、0.05g),均匀搅拌;
(3)以锡箔纸为基材,静电纺丝的参数设定为电压20KVp,针头与收集器距离为10cm,流速为0.5ml/min,环境温度控制在25-29℃,湿度50-58%R,收集时间为两小时,纺织完成后的电纺膜静置干燥。
二、效果检测
1、通过扫描电子显微镜分别对实施例1~5制备的纤维敷料进行表面形貌检测,从图2可以看出,纺织后获得的纳米纤维膜的纤维直径小和纤维间孔径均匀。
2、表面张力检测
利用接触角测量仪,对实施例1~5制备的纤维敷料的表面张力检测,结果如图3所示。可以看出,所有纤维敷料的表面张力随着时间下降,并引起接触角也随着时间发生相应的变化(表面张力降低,接触角减小)。实施例1的纤维敷料的初始接触角角度最大,纤维敷料对水的吸湿性最差;实施例3、4的敷料对水的吸湿性接近,而实施例5的辅料的吸湿性最强。
3、Ⅱ型糖尿病(T2DM)小鼠伤口愈合效果检测
将样本组模型小鼠的伤口处敷贴实施例1~5制备的纤维敷料(每组4只),对照组小鼠不敷贴任何材料,在伤口愈合前期、中期、后期,分别观察样本组和对照组小鼠,每个样本组小鼠,伤口愈合速度均快于对照组。
当然,以上仅为本发明的优选实施例而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

Claims (4)

1.一种具有破损皮肤修复功能的纤维敷料的制备方法,其特征在于,包括如下步骤,
(1)称取聚乙烯醇溶于30%的乙酸中均匀搅拌为2wt%的溶液,再加入等量的壳聚糖粉末在磁力搅拌器下均匀混合,完成基础的电纺溶液制备;
(2)之后加入熊果酸,均匀搅拌;
(3)以锡箔纸为基材,进行静电纺丝,纺织完成后的电纺膜静置干燥,制得所述纤维敷料。
2.根据权利要求1所述的制备方法,其特征在于,所述步骤(2)中熊果酸的终浓度为1×10-3~5×10-3g/ml。
3.根据权利要求2所述的制备方法,其特征在于,所述步骤(3)中静电纺丝的参数设定为电压20KVp,针头与收集器距离为10cm,流速为0.5ml/min,环境温度控制在25-29℃,湿度50-58%R,收集时间为两小时。
