Description

Qi and blood tonifying beverage composition and preparation method thereof
Technical Field
The invention belongs to the field of processing of functional foods, and particularly relates to a qi and blood tonifying beverage composition and a preparation method thereof.
Background
In the cloud of the Huangdi's Canon of Huang Di Nei Jing, the theory of blood as the mother of qi and qi as the commander of blood in TCM shows that qi and blood depend on each other and accompany each other, so as to jointly maintain and promote life activities. The theory of blood syndrome, theory of yin yang, water, fire, blood and qi: the "qi-guarding person means blood", and the book Ling Shu & Ying sanitary meeting says: the "nutrient qi moving in the blood vessels" refers to the qi in blood vessels that can move blood, and the qi is not attached to the blood because the blood can carry the gas, so the qi will not disperse and get rid of the blood. Qi is attached to blood, and blood carries qi and continuously provides food essence for the functional activities of qi to supplement nutrition, so that exuberance of qi causes exuberance of blood, deficiency of blood causes qi deficiency, and blood degassing is also lost, which is also known as qi deficiency in blood diseases. Therefore, patients with blood deficiency are often accompanied by qi deficiency such as shortness of breath, fatigue, laziness to speak, etc.
Qi deficiency refers to the condition of weakness of the zang-fu organs and tissues, manifested as shortness of breath, fatigue, mental fatigue, and pulse deficiency. It is usually caused by weakness due to chronic diseases, fatigue, and physical weakness due to old age. The specific clinical manifestations include poor disease resistance, aversion to cold, cold limbs, spontaneous perspiration, dizziness, tinnitus, listlessness, fatigue, palpitation, shortness of breath and hypoevolutism.
Blood deficiency refers to the weak condition of the face, palpebral margin, lips, pale tongue and thready pulse due to blood deficiency, malnutrition of the zang-fu organs, meridians and tissues. It is usually caused by excessive blood loss, insufficient blood production, or excessive anxiety, and dark consumption of yin blood. The clinical manifestations include lusterless sallow complexion, dry skin, dry hair, dry nails, blurred vision, numbness of hands and feet, insomnia, dreaminess, amnesia, palpitation, absentmindedness, etc.
Because qi and blood have a close relationship of interdependence, breeding and mutual interaction, qi and blood often affect each other when pathological changes occur. Qi-blood deficiency refers to the condition of both qi deficiency and blood deficiency.
With the change of life style and the explosion of social pressure, modern people have a large number of sub-healthy populations presenting symptoms of qi and blood deficiency among many young and middle-aged people. However, the qi and blood deficiency of the people is not caused by the passive qi and blood generation caused by insufficient nutrient substances, but caused by insufficient transportation and transformation of five cereals due to improper life style, and the normal transportation and transformation of qi and blood in the body are influenced by adverse factors such as long-term air-conditioning environment, sedentary and restlessness and the like. Therefore, the traditional Chinese medicine has the effects of tonifying qi and enriching blood, and is assisted with qi stagnation to promote the circulation of qi and blood and increase the nourishing and warming capacity of blood, so that the circulation and transformation of qi and blood of a human body are adjusted to be normal, five cereals can fully nourish four limbs, and the human body tends to be mild. Aiming at people with qi deficiency and blood stasis constitution, relatively mild and medicinal and edible materials are selected, and the food therapy effect is realized through a reasonable formula. Therefore, the king sun Simiao in the cloud of thousand gold essences: the whole life is not enough for those who do not know proper diet. If you are ill, you take a life-saving diet to treat it. If the diet therapy fails to cure, the medicine is activated. "

Experimental example 1 evaluation of blood-enriching efficacy of different formulations on cyclophosphamide-induced blood-deficient mice
Animal grouping and administration
After the mice are bred adaptively for 7 days, the mice are randomly divided into 9 groups according to the weight, namely a normal group, a model group and an administration group 1-7# group, wherein each group comprises 10 mice, and the number of the mice is half that of the mice. Wherein the administration group ig administers 4 ml/kg of the composition prepared in each example and comparative example per day -1 For 14 days. The normal group and the model group ig were given an equal volume of physiological saline for 14 days. The model group and the administration group were administered daily on day 15 of administrationip cyclophosphamide 40 mg/kg -1 80 mg/kg of ig cyclophosphamide on day 18 for 3 days -1 (ii) a Normal group ip equal amount of saline. 2h after the last administration, the anal temperature of the mice is measured by one time; blood is taken from eyeballs by a blood collecting tube, heparin is used for anticoagulation, and peripheral hemogram, thymus index, spleen index and cell membrane ATP enzyme activity are measured.
Peripheral hemogram measurement: the detection is performed using a fully automatic blood analyzer, which mainly includes white blood cell count (WBC), Red Blood Cells (RBC), hemoglobin content (HGB), red blood cell Hematocrit (HCT), and platelet count (PLT).
Thymus index and spleen index determination: the mice after blood collection were sacrificed by taking off the cervical vertebrae, and the thymus and spleen were taken and weighed. Spleen Index (SI) and Thymus Index (TI) were calculated. The calculation formula is as follows: spleen mass and thymus mass were divided by the corresponding mouse body weight, respectively.
Determination of ATPase Activity: the determination is carried out by applying the principle of an inorganic phosphate radical method according to the specification of an ATP enzyme kit.
The measurement results are shown in tables 2 to 4.
TABLE 2 Effect of qi and blood drinks with different formulas on peripheral hemogram of blood deficiency model mouse: (

Figure BDA0002984136490000121
Note: comparison with Normal group 1 P<0.01; comparison with model group 2 P<0.01, 3 P<0.05
From the above results, it was found that the qi and blood replenishing drink 1-5# composition of the present invention significantly increased the body temperature level (P <0.01) of blood-deficient mice, and increased the hemogram levels of the mice, particularly the effects of example 2 and example 1, higher than those of the other groups. The indication shows that the prescription can exert the maximum effect only within the range of proper compatibility proportion. For example, in example 3, the effect of enriching the blood is affected because the consumption of perilla leaves is low, which causes weak qi-moving force, while in example 4, the effect of enriching the blood is greatly reduced because perilla leaves are reused, which causes too strong qi-moving force. In a comparative example, a perilla leaf-lack group and a peanut red coat-lack group are designed, compared with example 3 with less perilla leaf usage, when perilla leaf is not used at all (comparative example 1), the blood enriching effect is further reduced, which prompts the importance of qi-moving drugs on the blood enriching effect while enriching blood, and embodies the formulation idea of 'tonifying without stagnation'; in example 5, the amount of peanut skin was significantly increased compared to example 1, but the blood-enriching effect was not improved due to the decrease in the amount of perilla, and in comparative example 2, the peanut skin, which is the essential blood-enriching drug of the present invention, was removed, and thus the group exhibited no blood-enriching effect at all.

Note: comparison with Normal group 1 P<0.01; comparison with model group 2 P<0.01, 3 P<0.05
From the results, the qi and blood replenishing drink 1-5# composition can remarkably increase Na of blood deficiency model mice + -K + ATPase and Ca 2+ -Mg 2+ The improvement effect of ATPase, especially 1-2# composition, is most obvious. The dosage of qi-moving herbs in the recipe-perilla leaf will be higher or lower to cause the decrease of curative effect.

The results show that the qi-blood drink can obviously increase the thymus index of a blood deficiency model mouse, has obvious effect of reducing the increased spleen index, and the strength of the drug effect is related to the prescription proportion.
Test example 2 evaluation of blood-enriching efficacy of different formulations on rats with blood deficiency caused by exsanguination
Animal grouping and administration
After the rats were bred for 7 days, the rats were randomly divided into 9 groups by weight, namely a normal group, a model group and a drug administration 1-7# group, wherein each group contains 8 females. Administration group ig the compositions prepared in examples and comparative examples were administered at a dose of 3 ml/kg/day -1 The normal group and the model group ig were given an equal volume of physiological saline. The administration is carried out 1 time per day for 30d continuously, the model is started at 18d, and the model group and the administration group are bled for 5ml/kg per day and continuously bled for 10 d. Blood is taken from fundus venous plexus 2h after the last administration, heparin is used for anticoagulation, and peripheral hemogram, thymus index, spleen index and cell membrane ATP enzyme activity are measured.
Peripheral hemogram measurement: the detection is carried out by a full-automatic blood analyzer, which mainly comprises Red Blood Cells (RBC), hemoglobin content (HGB), red blood cell Hematocrit (HCT) and platelet count (PLT).
Determination of ATPase Activity: the determination is carried out by applying the principle of an inorganic phosphate radical method according to the specification of an ATP enzyme kit.
The measurement results are shown in tables 5 to 6.

Note: comparison with Normal group 1 P<0.01; comparison with model group 2 P<0.01, 3 P<0.05
From the above results, it can be seen that the compositions of examples 1 to 5 of the qi and blood replenishing drink of the present invention can significantly increase the blood levels of the blood-deficient rats, and particularly the compositions of examples 1, 2 and 5 have the most significant effects. Since this model is blood deficiency due to blood loss, the formulation of example 5 with stronger blood-enriching efficacy showed better blood-enriching effect. The components of examples 3 and 4 are characterized in that the amount of the qi-moving drug perilla leaf is changed compared with the formulation of example 1, but the perilla leaf is not used at all (comparative example 1), so that stagnation is easily caused, the blood-enriching efficacy cannot be exerted to the best, and the fact that the amount of the perilla leaf needs to be controlled within a certain range is suggested, and the blood-enriching effect of the composition is reduced when the amount of the perilla leaf is higher or lower. In contrast, comparative example 2 shows that the blood-enriching efficacy is almost not achieved by removing peanut skin in the formula.

From the above results, it was found that the compositions of examples 1 to 4 were capable of significantly increasing Na content in blood-deficiency model mice + -K + ATPase and Ca 2+ -Mg 2+ ATP enzyme, the composition of example 5 has a low dosage of peanut skin, and the effect is not obvious, and in comparative example 1, on the basis of the formula of example 1, perilla leaf is deleted, the blood enriching effect is also influenced to a certain extent, and the curative effect is slightly reduced.
Test example 3
Animal grouping and administration
After the experiment, all rats were placed in a constant temperature water tank [ (25 + -1) DEG C, water depth 40cm]And 3d, swimming for 5min every day. 1 day before grouping, exhaustive swimming is carried out after a weight with the body mass of minus 5% at the tail of the rat (the rat finally sinks to the nose tip and is immersed in water after swimming, and the rat can not return to the water surface after 10 seconds, namely the exhaustive swimming), rats with large difference of negative-gravity exhaustive swimming time (from swimming to the time of the occurrence of the exhaustive swimming) and without swimming are removed, and qualified rats with the negative-gravity exhaustive swimming time of 10-50 min are reserved. The qualified rats were randomly divided into a normal group, a model group, an administration group of examples and a comparative example, 8 rats each, according to body mass. The rats in the normal group are normally raised without special treatment; the other groups of rats adopt a 'diet irregularity + fatigue' method to construct the qi deficiency animal model: 9 a.m. each day: 00 exhaustive swimming in a constant temperature water tank by a weight with the tail part accounting for minus 5 percent of the body mass, and feeding the livestock in a feed limiting amount after 1 day of fasting for 21 days continuously. The administration group rats were gavaged with 3 ml/kg while molding -1 The qi and blood replenishing drink compositions of examples 1 to 5 and comparative examples 1 to 2 of the present invention, rats of the normal group and the model group were each gavaged with water of equal volume once a day for 21 days continuously. The negative gravity swimming time of rats was measured after the 21d dosing. The results are shown in Table 7.

Note: comparison with Normal group 1 P<0.01; 2 P<0.05 comparison with model group 3 P<0.01, 4 P<0.05
The above results show that the qi and blood replenishing beverage composition can obviously prolong the negative gravity swimming time of the rats with qi deficiency models, and has good effects of replenishing qi and improving the tolerance of organisms, but the qi replenishing effect is reduced by removing the qi-moving perilla leaves or the blood replenishing peanut skin. Comparing examples 1 and 2, it can be seen that, under the condition of the same dosage of other medicines in the prescription, the peanut red skin and the perilla in example 2 are increased, and the qi-moving and blood-enriching effects are enhanced, so that better tolerance effect is shown, while in example 5, the peanut red skin dosage for enriching blood is increased, and the perilla dosage for promoting qi is decreased, so that the effect is slightly poor. The effect is also worse by removing the perilla in comparative example 1, but the blood-enriching effect cannot be better exerted by adding no peanut red skin in comparative example 2, so that the generation of qi and blood is passive, and thus, compared with a model group, almost no improvement is caused.
