The invention is suitable for the field of health products, and provides a traditional Chinese medicine enzyme and a preparation method thereof, wherein poria cocos, dendrobium, radix ophiopogonis and medlar are adopted for compatibility according to a specific proportion, traditional Chinese medicine extracts are prepared by processes of alcohol extraction, water decoction, concentration and the like, then the traditional Chinese medicine extracts are mixed with egg powder, and the traditional Chinese medicine enzyme is prepared by three times of fermentation; the prepared traditional Chinese medicine enzyme is easy to absorb by a human body, has good effects of resisting fatigue and enhancing immunity of the organism, can well condition the sub-health state of the organism, has good palatability, can be taken for a long time, can be industrialized, and has high application value.

putting the traditional Chinese medicine extract and the egg powder into a fermentation container according to the weight ratio of (9-15) to (1-2), mixing, sequentially adding a primary fermentation microbial inoculum, a secondary fermentation microbial inoculum and a tertiary fermentation microbial inoculum, and performing tertiary fermentation to obtain the feed additive; the traditional Chinese medicine extract is prepared by the following method: mixing and crushing poria cocos, dendrobium, radix ophiopogonis and medlar according to the weight ratio of (1-2) to (2-3) to (0.5-1) to (1-1.5), adding ethanol to soak for 2-3 days, then performing reflux extraction for 1-3 hours, and separating to obtain an alcohol extract and filter residues; adding water into the filter residue, and decocting for 1-3 hours to obtain a decoction; and (3) combining the alcohol extract and the decoction, and concentrating until the density is 1.10-1.20.
The embodiment of the invention also provides the traditional Chinese medicine enzyme prepared by the preparation method of the traditional Chinese medicine enzyme.
The embodiment of the invention provides a preparation method of a traditional Chinese medicine enzyme, which comprises the steps of carrying out compatibility use on the traditional Chinese medicine raw materials of poria cocos, dendrobe, radix ophiopogonis and medlar according to a specific proportion, carrying out processes of alcohol extraction, water decoction, concentration and the like to obtain a traditional Chinese medicine extract, mixing the traditional Chinese medicine extract with egg powder, and carrying out three-time fermentation to obtain the traditional Chinese medicine enzyme; the prepared traditional Chinese medicine enzyme is easy to absorb by a human body, has good effects of resisting fatigue and enhancing the immunity of the organism, is beneficial to improving and conditioning the sub-health state of the organism, has good taste and wide market prospect.

And adding a third fermentation microbial inoculum into the secondary fermentation product, and fermenting for 1-2 days at the temperature of 25-35 ℃ to obtain the microbial inoculum.
Wherein the primary fermentation microbial inoculum is saccharomycetes, the secondary fermentation microbial inoculum is probiotics, and the tertiary fermentation microbial inoculum is acetic acid bacteria. In the primary fermentation process, yeast is added, and the fermentation temperature is controlled to be 32-40 ℃ for 12-24 hours, so that the yeast is propagated in large quantity and generates more ethanol, alcohol-soluble components in the traditional Chinese medicinal materials can be dissolved out more fully, and the leaching rate of effective components of the traditional Chinese medicinal materials is improved.
In the secondary fermentation process, probiotics are added, the fermentation temperature is controlled to be 30-45 ℃, and fermentation is carried out for 10-30 days, so that the probiotics can be propagated in a large amount, the traditional Chinese medicine components in the primary fermentation product are fully decomposed, secondary metabolites of the probiotics are generated, and the carbon source in the fermentation product can be digested to be changed into small molecular substances which are easily absorbed by the intestinal tract of a human body. In the embodiment of the invention, the adopted probiotics can be active microorganisms such as bifidobacterium, lactobacillus acidophilus and the like which have positive benefits on human bodies and animal bodies.
In the third fermentation process, acetic acid bacteria are added, and the fermentation temperature is controlled to be 25-35 ℃ for fermentation for 1-2 days, so that the ethanol conversion speed in the secondary fermentation product can be accelerated, the pH value of the fermentation system is quickly reduced, an acidic environment which is favorable for growth and propagation of probiotics is formed, and the growth of mixed bacteria is effectively inhibited; the fermentation conditions are favorable for the growth and the propagation of the acetic acid bacteria.
In the embodiment of the invention, the egg powder is prepared by the following method:
extracting egg white solution from eggs, and freezing the egg white solution at the temperature of-10 ℃ below zero to-15 ℃ below zero for 5 to 10 hours;
and taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 20-30 min, drying for 20-45 min at the temperature of 55-65 ℃, taking out and crushing to obtain the egg white powder.
In the embodiment of the invention, the mixing mass ratio of the frozen egg white solution, the edible alkali and the water is (1-5): (0.1-0.2): (15-20). Wherein the edible alkali is a mixture of sodium bicarbonate and sodium carbonate. Preferably, the mixing mass ratio of the sodium bicarbonate to the sodium carbonate is 3:1.
The egg white is frozen, added with alkali, stirred, mixed, dried at the temperature of 55-65 ℃ and crushed, so that the egg white is denatured to become loose in structure, can not form crystals, is easy to hydrolyze by protease and is beneficial to digestion and absorption by human bodies. In addition, according to the invention, the egg powder of the poultry egg and the traditional Chinese medicine extract are fermented for three times, so that not only can the effective components in the traditional Chinese medicine materials be extracted, but also the effective components and the protein can be decomposed into small molecular substances which are easier to be absorbed by a human body, and the curative effects of the product on enhancing the immunity of the human body and resisting fatigue are improved.
In the preferred embodiment of the invention, when the mixing mass ratio of the traditional Chinese medicine extract to the egg powder is 10:1.5, the prepared traditional Chinese medicine ferment has the best curative effect, can well enhance the immunity of the organism and condition the sub-health state of the human body, and has good taste.
In a preferred embodiment of the present invention, in order to make the prepared traditional Chinese medicine ferment have better taste, a proper amount of flavoring agents such as honey, syrup, etc. can be added to mask the bitter taste of the traditional Chinese medicine therein.
In the embodiment of the present invention, the above traditional Chinese medicine enzyme can be used as an active ingredient, and pharmaceutically acceptable excipients can be added to prepare a dosage form such as a granule, an oral liquid, a tablet, a capsule, and the like, which is not limited in the present invention.

Example 1
The preparation method of the traditional Chinese medicine enzyme provided by the embodiment of the invention comprises the following steps:
mixing and crushing 100g of poria cocos, 200g of dendrobium, 50g of radix ophiopogonis and 100g of medlar, adding ethanol with the mass fraction of 70% into the mixture, soaking the mixture for 2 days at room temperature, performing reflux extraction for 1 hour, and separating the mixture to obtain an ethanol extract and filter residues; adding water with the mass 6 times of that of the filter residue into the filter residue, and decocting for 1 hour to obtain a decoction; mixing the above alcoholic extractive solution and decoction, and concentrating to density of 1.10 to obtain Chinese medicinal extract.
Extracting egg white solution from egg, and freezing at-10 deg.C for 5 hr; taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 20min (wherein the mixing mass ratio of the egg white solution, the edible alkali and the water is 2:0.1:15), drying for 20min at the temperature of 60 ℃, taking out, and crushing to obtain the egg powder.
Mixing the prepared traditional Chinese medicine extract and egg powder in a weight ratio of 9:1 in a fermentation container, adding yeast accounting for 0.05% of the mass of the mixture of the traditional Chinese medicine extract and the egg powder, and fermenting at 32 ℃ for 12 hours to obtain primary fermentation product; adding probiotics accounting for 0.01 percent of the mass of the primary fermentation product into the primary fermentation product, and fermenting for 15 days at the temperature of 30 ℃ to obtain a secondary fermentation product; adding acetic acid bacteria accounting for 0.02% of the mass of the secondary fermentation product into the secondary fermentation product, and fermenting at 25 ℃ for 1 day to obtain the traditional Chinese medicine ferment.
Example 2
The preparation method of the traditional Chinese medicine enzyme provided by the embodiment of the invention comprises the following steps:
150g of poria cocos, 300g of dendrobium, 100g of radix ophiopogonis and 150g of Chinese wolfberry are mixed and crushed, the mixture is added into ethanol with the mass fraction of 70% to be soaked for 3 days at room temperature, reflux extraction is carried out for 1.5 hours, and alcohol extract and filter residue are obtained through separation; adding water with the mass 6 times of that of the filter residue into the filter residue, and decocting for 1.5 hours to obtain a decoction; mixing the above alcoholic extractive solution and decoction, and concentrating to density of 1.12 to obtain Chinese medicinal extract.
Extracting egg white solution from egg, and freezing at-15 deg.C for 5 hr; taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 30min (wherein the mixing mass ratio of the egg white solution, the edible alkali and the water is 1:0.1:15), drying for 45min at the temperature of 55 ℃, taking out, and crushing to obtain the egg powder.
Mixing the prepared traditional Chinese medicine extract and egg powder in a fermentation container according to a weight ratio of 10:2, adding yeast accounting for 0.1% of the mass of the mixture of the traditional Chinese medicine extract and the egg powder, and fermenting at 35 ℃ for 18 hours to obtain primary fermentation product; adding probiotics accounting for 0.02% of the mass of the primary fermentation product into the primary fermentation product, and fermenting for 10 days at the temperature of 40 ℃ to obtain a secondary fermentation product; adding acetic acid bacteria accounting for 0.05% of the mass of the secondary fermentation product into the secondary fermentation product, and fermenting at 25 ℃ for 2 days to obtain the traditional Chinese medicine ferment.

mixing and crushing 200g of poria cocos, 250g of dendrobium, 80g of radix ophiopogonis and 120g of medlar, adding 70% ethanol by mass fraction, soaking for 1 day at room temperature, performing reflux extraction for 3 hours, and separating to obtain an ethanol extract and filter residues; adding water with the mass 6 times of that of the filter residue into the filter residue, and decocting for 2 hours to obtain a decoction; mixing the above alcoholic extractive solution and decoction, and concentrating to density of 1.20 to obtain Chinese medicinal extract.
Extracting egg white solution from egg, and freezing at-10 deg.C for 10 hr; taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 30min (wherein the mixing mass ratio of the egg white solution to the edible alkali to the water is 5:0.2:20), drying for 30min at the temperature of 65 ℃, taking out, and crushing to obtain egg powder.
Mixing the prepared traditional Chinese medicine extract and egg powder in a fermentation container according to a weight ratio of 15:1.5, adding yeast accounting for 0.1% of the mass of the mixture of the traditional Chinese medicine extract and the egg powder, and fermenting at 40 ℃ for 12 hours to obtain primary fermentation product; adding probiotics accounting for 0.05 percent of the mass of the primary fermentation product into the primary fermentation product, and fermenting for 20 days at the temperature of 45 ℃ to obtain a secondary fermentation product; adding acetic acid bacteria accounting for 0.13% of the mass of the secondary fermentation product into the secondary fermentation product, and fermenting at 25 ℃ for 1 day to obtain the traditional Chinese medicine ferment.
Example 4
The preparation method of the traditional Chinese medicine enzyme provided by the embodiment of the invention comprises the following steps:
mixing and crushing 170g of poria cocos, 230g of dendrobium, 70g of radix ophiopogonis and 110g of Chinese wolfberry, adding 70% by mass of ethanol, soaking for 2 days at room temperature, performing reflux extraction for 2 hours, and separating to obtain an ethanol extract and filter residues; adding water with the mass 6 times of that of the filter residue into the filter residue, and decocting for 3 hours to obtain a decoction; mixing the above alcoholic extractive solution and decoction, and concentrating to density of 1.15 to obtain Chinese medicinal extract.
Extracting egg white solution from egg, and freezing at-15 deg.C for 6 hr; taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 25min (wherein the mixing mass ratio of the egg white solution, the edible alkali and the water is 2:0.1:15), drying for 45min at the temperature of 60 ℃, taking out, and crushing to obtain the egg powder.

The traditional Chinese medicine ferment of the comparative example 6 is prepared without a fermentation process, specifically, the prepared traditional Chinese medicine extract and the egg powder are mixed according to the weight ratio of 10:1.5, and the rest raw materials and process conditions are the same as those of the example 6.
Product performance testing
First, acute oral toxicity test
1. Preparation of test solutions: weighing 25g of the traditional Chinese medicine enzyme samples provided by embodiments 1-6 respectively into the volumetric flasks numbered 0-6, adding ultrapure water to the volumetric flasks at a constant volume of 100mL, and uniformly mixing to obtain test solutions.
2. Pre-testing: the test liquid with the dose of 5000mg/kg body weight is adopted to carry out oral gavage test on the test animal, and the test animal is not dead, so the formal test design dose is 5000mg/kg body weight.
3. And (3) formal test: selecting 70 healthy SPF-grade Kunming mice with the weight of 18-20 g, randomly and averagely dividing the mice into 7 groups, wherein each group comprises 10 mice and half of the mice, and the number of the mice is 0-6, wherein the group 0 is a blank control group, and the mice are subjected to oral gavage for 1 time by using a gavage needle according to the weight of 0.2mL/10g to give ultrapure water; groups 0 to 6 were administered with No. 0 to 6 test solutions through oral gavage 1 time by gavage needles in an amount of 0.2mL/10g body weight, i.e., group 0 was administered with No. 0 test solution, group 1 was administered with No. 1 test solution, group 2 was administered with No. 2 test solution, group 3 was administered with No. 3 test solution … …, and so on, and group 6 was administered with No. 6 test solution. Before the gavage, animals in each group are fasted for 16 hours, water is freely drunk, and after the gavage, normal diet and conventional management are given; the mice are kept at the room temperature of 22 +/-1 ℃ and the humidity of 60 +/-5% for the same days, the poisoning symptoms and death conditions of the mice are observed and recorded every day, and after 14 days of test completion, data statistics are carried out on the test results.
Wherein, in the test process, the poisoning means that the mouse has symptoms of tremor, convulsion, salivation, diarrhea, lethargy or coma and the like in the test process.
During this test, the diet activity of each group of animals was normal and no obvious toxicant symptoms and death were observed. Therefore, the mouse with the traditional Chinese medicine enzyme provided by the embodiment of the invention has acute toxicity by mouth50>It was found to be safe at 5000mg/kg body weight.
Second, evaluation of therapeutic efficacy

And 1 hour after the administration on the 10 th day from the start of the experiment, each group of animals was placed in a 10 ℃ swimming pool, and the time for each group of mice to sink to the water bottom was observed as the duration of swimming, and the test results are shown in table 1 below.

As can be seen from table 1, the traditional Chinese medicine ferment prepared in the embodiment of the present invention can significantly prolong the continuous swimming time of the mouse, effectively relieve the fatigue of the mouse, and enhance the immunity of the mouse, compared to the traditional Chinese medicine ferment prepared in the comparative examples 1 to 6. Moreover, the addition of the egg powder, the extraction of the traditional Chinese medicine extract by combining the alcohol extraction and water decoction processes and the preparation of the traditional Chinese medicine ferment by three-time fermentation are beneficial to improving the effects of resisting fatigue and enhancing the immunity of the organism.
2. Clinical trial
240 sub-healthy volunteers aged 18 to 65 years who exhibited fatigue, muscle and joint soreness, dizziness, headache, palpitations, chest distress, sleep disturbance, inappetence, abdominal discomfort, loose stools, sexual dysfunction, aversion to cold and heat, susceptibility to colds, dry eyes were selected, and volunteers were randomly divided into 12 groups of 20 groups, i.e., example 1 group, example 2 group, example 3 group, example 4 group, example 5 group, example 6 group, comparative example 1 group, comparative example 2 group, comparative example 3 group, comparative example 4 group, comparative example 5 group, and comparative example 6 group.
In the test, the traditional Chinese medicine enzymes provided by the embodiments 1-6 of the invention are respectively taken by the embodiments 1-6, 2 times a day, 10mL each time, half an hour after breakfast and supper, and 7 days are a course of treatment. Comparative examples 1 to 6 the Chinese herbal ferment provided in comparative examples 1 to 6 was administered, respectively, in the same manner as in the above example group.
The judgment standard of the curative effect is as follows: and (3) curing: the symptoms disappear or substantially disappear. The method has the following advantages: the symptoms are obviously relieved. And (4) invalidation: the symptoms are not obviously improved.

And it can be seen that the addition of the egg powder, the extraction of the traditional Chinese medicine extract by combining the alcohol extraction and water decoction processes and the preparation of the traditional Chinese medicine ferment by three-time fermentation are all beneficial to improving the effects of resisting fatigue and enhancing the immunity of the organism.
Typical case

putting the traditional Chinese medicine extract and the egg powder of the poultry eggs in a fermentation container according to the weight ratio of (9-15) to (1-2), mixing, adding a primary fermentation microbial inoculum, and fermenting at the temperature of 32-40 ℃ for 12-24 hours to obtain a primary fermentation product;
adding a secondary fermentation microbial inoculum into the primary fermentation product, and fermenting for 10-30 days at the temperature of 30-45 ℃ to obtain a secondary fermentation product;
adding a third fermentation microbial inoculum into the secondary fermentation product, and fermenting for 1-2 days at the temperature of 25-35 ℃ to obtain the fermented product;
wherein the traditional Chinese medicine extract is prepared by the following method:
mixing and crushing poria cocos, dendrobe, radix ophiopogonis and medlar according to the weight ratio of (1-2) to (2-3) to (0.5-1) to (1-1.5), adding ethanol to soak for 2-3 days, then performing reflux extraction for 1-3 hours, and separating to obtain an alcohol extract and filter residues;
adding water into the filter residue, and decocting for 1-3 hours to obtain a decoction;
combining the alcohol extract and the decoction, and concentrating until the density is 1.10-1.20 to obtain the tea beverage;
the egg powder is prepared by the following method:
extracting an egg white solution from the poultry egg, and freezing the egg white solution at the temperature of minus 10 ℃ to minus 15 ℃ for 5 to 10 hours;
and taking out the frozen egg white solution, adding edible alkali and water, stirring and mixing for 20-30 min, drying for 20-45 min at the temperature of 55-65 ℃, taking out and crushing to obtain the egg white powder.
2. The method for preparing traditional Chinese medicine ferment of claim 1, wherein the primary fermentation agent is yeast, the secondary fermentation agent is probiotic bacteria, and the tertiary fermentation agent is acetic acid bacteria.
3. The preparation method of the traditional Chinese medicine enzyme according to claim 1, wherein the addition amount of the primary fermentation microbial inoculum accounts for 0.05-0.15% of the mass of the mixture of the traditional Chinese medicine extract and the egg powder;
the addition amount of the secondary fermentation inoculant accounts for 0.01-0.05% of the mass of the primary fermentation product;
the addition amount of the tertiary fermentation inoculant accounts for 0.01-0.13% of the mass of the secondary fermentation product.
4. The preparation method of the traditional Chinese medicine enzyme according to claim 1, wherein the mixing mass ratio of the frozen egg white solution, the edible alkali and the water is (1-5): (0.1-0.2): (15-20).
5. The method for preparing traditional Chinese medicine ferment of claim 1, wherein the dietary alkali is a mixture of sodium bicarbonate and sodium carbonate.
6. The preparation method of the traditional Chinese medicine enzyme according to claim 1, wherein the mixing mass ratio of the poria cocos, the dendrobe, the radix ophiopogonis and the medlar is 1:2:0.5: 1.2.
