Abstract
本发明公开了一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法,属于高通量测序技术领域。该检测方法包括以下步骤:提取检测对象的基因组DNA,对提取的基因组DNA进行定量并构建文库,对文库进行定量操作,上机测序后进行数据分析得到致病位点相关信息。本发明针对遗传性帕金森病致病基因突变检测,基于多因素算法对目标区域基因组进行设计,合成特异性探针与基因组DNA文库进行液相杂交,将目标区域序列进行捕获并富集后进行高通量测序,能够同时对一个样本中多个遗传性帕金森病致病基因突变进行分析,具有灵敏度高、针对性强、覆盖全面、通量大、准确性高等优点。

技术领域
本发明属于高通量测序技术领域,具体涉及一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法。
背景技术
帕金森病(Parkinson’s Disease,PD)是临床上最常见的神经***退行性疾病之一。其主要病理改变为中脑黑质致密部多巴胺(DA)能神经元进行性变性丢失,纹状体区多巴胺含量减少,以及残存的多巴胺能神经元内形成以α–突触共核蛋白(α–Syn)为主要成分的嗜酸性包涵体——路易小体(Lewy body)。帕金森病有两种类型,即家族性帕金森病与散发性帕金森病,大部分帕金森病患者为散发病例,仅有10%–15%的患者有家族史。
家族性帕金森病发病机制与多种致病基因突变有关,α–synuclein基因(PARK1/PARK4)、LRRK2基因(PARK8)、UCH–L1基因(PARK5)、GIGYF2基因(PARK11)和Htra2/Omi基因(PARK13)为常染色体显性遗传性帕金森病致病基因;Parkin基因(PARK2)、PINK1基因(PARK6)、DJ–1基因(PARK7)和ATP13A2基因(PARK9)为常染色隐性遗传性帕金森病致病基因。其中LRRK2基因是目前为止帕金森病患者中突变频率最高的帕金森病致病基因。散发性帕金森病易感基因有SNCA基因、GBA基因、LRRK2基因、DJ–1基因等。帕金森病并非单一因素引起,而是由多种因素包括遗传因素、环境因素、老年化、免疫/炎性因素等通过多种机制联合作用的结果。通过全面的基因检测方案,可以协助阳性携带者进入一些前沿的临床试验,希望随着医学的不断发展,这些携带突变的无症状者能够延缓甚至阻止症状性PD的出现。
发明内容
针对现有技术中遗传性帕金森病致病基因突变检测技术的限制,本发明的目的是,提供一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法,该方法具有灵敏度高、针对性强、覆盖全面、通量大、准确性高等优点。
为实现上述目的,本发明采用的技术方案是:
一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法,提取检测对象的基因组DNA后进行包括以下步骤的处理:
S1、将基因组DNA进行片段化;
S2、将片段化的基因组DNA进行末端修复和3'末端添加碱基A;
S3、将3'末端添加碱基A的产物连接扩增接头并纯化,以进行有效连接产物的富集;
S4、将连接产物进行Pre–PCR扩增反应,富集有效产物;
S5、用探针与上述富集的有效产物模板进行液相杂交后,并与磁珠结合,洗脱未结合探针与非特异性结合不稳定文库,捕获上述模板中目标区域DNA文库;
S6、将捕获的目标区域DNA文库分离后连接完整接头,进行Post–PCR扩增反应,捕获目标区域文库;
S7、对目标区域文库进行定量和质检操作;
S8、上机测序;
S9、数据分析得到致病位点相关信息;

PSMC3IP:基因编码在减数***重组中起作用的蛋白质,它是PSMC3IP/MND1复合体的一个亚基,可与PSMC3/TBP1相互作用,在减数***过程中刺激DMC1和RAD51介导的链交换,该基因编码的蛋白质还可以共激活由***、雄激素、糖皮质激素、孕激素和甲状腺核受体介导的配体驱动的转录,该基因的突变导致XX女性性腺发育不全。该基因的可变剪接导致多个转录物变体。PSMC3IP(PSMC3相互作用蛋白)是一种蛋白质编码基因,与PSMC3IP相关的疾病包括卵巢发育不全3和46,Xx性逆转1,其相关途径包括减数***和雄激素受体信号传导途径,与该基因有关的基因本体论(GO)注释包括DNA结合转录因子活性和核受体转录共激活因子活性。在减数***重组中起重要作用,刺激减数***过程中配对同源染色体所需的DMC1介导的链交换,复杂的PSMC3IP/MND1结合DNA,刺激DMC1的重组酶活性以及从双链DNA形成DMC1 D环,该复合物可稳定在单链DNA上形成的突触前RAD51和DMC1细丝,以捕获双链DNA,这种复合物刺激RAD51和DMC1促进的同源配对中的突触和突触前关键步骤,可能通过与PSMC3结合抑制HIV-1病毒蛋白TAT活性并调节蛋白酶体的活性,充当由核受体介导的激素依赖性转录的组织特异性共激活剂。
ATP6V0A1:基因编码液泡ATPase(V-ATPase)的成分,液泡ATPase是介导真核细胞内细胞器酸化的多亚基酶,V-ATPase依赖的细胞器酸化对于诸如蛋白分选、酶原激活、受体介导的内吞作用和突触囊泡质子梯度生成等细胞内过程是必需的。V-ATP酶由胞质V1结构域和跨膜V0结构域组成,V1域由三个A和三个B亚基,两个G亚基以及C,D,E,F和H亚基组成,V1域包含ATP催化位点,V0结构域由五个不同的亚基组成:a,c,c',c“和d。许多V1和V0亚基蛋白的其他同工型由多个基因或选择性剪接的转录变体编码。该基因编码三个A亚基蛋白质之一,并且编码的蛋白质与网格蛋白包被的囊泡相关,已经为该基因发现了编码不同同工型的三个转录物变体。ATP6V0A1(ATP酶H+转运V0亚基A1)是一种蛋白质编码基因,与ATP6V0A1相关的疾病包括精神发育迟滞、X连锁、综合症、常春藤型和下心肌梗塞,其相关途径包括溶酶体和突触囊泡循环,与该基因有关的基因本体论(GO)注释包括ATPase结合和质子转运ATPase活性,旋转机制,该基因的重要旁系同源物是ATP6V0A4,液泡ATPase的组装和活性所必需,在针对特定细胞器的酶的不同靶向和调节中的潜在作用(通过相似性)。H+-ATPase(也称为液泡ATPase,V-ATPase)是一种酶转运蛋白,其功能是酸化真核细胞中的细胞内区室它普遍存在,并存在于膜内细胞器中,如液泡、溶酶体和内体。
一些实施例中,基因组DNA来源于人类。
一些实施例中,提取基因组DNA的方法包括纯化柱纯化、磁珠纯化或酚氯仿提取。
一些实施例中,S7中定量的方法包括基于荧光定量原理的定量装置定量、Q–PCR定量或电泳。
一些实施例中,S1中基因组DNA片段化的方法包括超声破碎、转座酶酶切或限制性内切酶酶切。
一些实施例中,S5和S6中捕获方法包括液相探针捕获、固相芯片液相杂交捕获或PCR富集。
一些实施例中,S8中上机测序通过二代测序平台进行。
一些实施例中,S9中数据分析的具体操作如下:通过先对所得测序数据与人类基因组的参考序列进行对比,再对所测基因的每个突变位点情况进行分析,得到遗传性帕金森病致病突变相关信息。
一些实施例中,可以采用试剂盒的形式检测用于获取遗传性帕金森病致病基因突变相关信息。
图1示例性地描述了一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法(如采用试剂盒的形式),主要技术流程包括提取基因组DNA、文库制备、文库质检定量、上机测序及数据分析,具体步骤如下:
Step1:基因组DNA打断
1.将准备的基因组DNA按照反应体系转移至0.6ml MaxyClear SnaplockMicrocentrifuge Tube内,充分混匀,短暂离心后置于冰上待用;
2.提前打开超声打断仪Bioruptor Pico,待冷循环仪温度降至4℃后,设置参数ON30s,OFF 30s为1个循环,每10cycles为一轮,共进行3轮,每轮结束后将样品置于振荡器上充分混匀,短暂离心后进行下一轮打断(混匀越充分,打断样品片段大小越集中);
3.取1μl样品使用Agilent2100(DNA 1000Assay)进行片段检测,正常打断后样品检测主峰约在150bp–200bp。
Step2:末端修复、3’端加“A”
1.将打断后的样品完全转移至PCR管中,按如表1所示配置反应体系(此操作在冰盒上进行):

2.使用移液器吹打混匀(避免剧烈震荡混匀),将PCR管放回冰盒上备用;
3.运行PCR程序,设置PCR仪参数如下:
Heat lid 85℃、4℃for 1min、20℃for 30min、65℃for 30min、4℃for∞
4.程序运行完成后立即进行下步连接反应。
Step3:接头连接
1.参照表2将adapter提前稀释成合适的浓度:

2.在上述step2反应的PCR管中,按如表3配置反应体系(此操作在冰盒上进行):

3.轻轻吸打混匀6次,避免产生气泡,然后短暂离心;
4.运行PCR仪程序(要求无热盖),设置PCR仪参数如下:
Heat lid off、20℃for 15min、4℃for∞;
5.程序结束后,立即进行下步实验。
Step4:连接后纯化
1.将Agencourt AMPure XP磁珠拿至室温,震荡混匀备用;
2.在step3中程序运行完的PCR管中,按照0.8X体积的纯化磁珠进行实验,按表4配置反应体系:

3.轻轻吸打混匀6次;
4.室温静置孵育5–15min,将PCR管置于磁力架上3min使溶液澄清;
5.移除上清,PCR管继续放置在磁力架上,向PCR管内加入200μl 80%乙醇溶液,静置30s;
6.移除上清,再向PCR管内加入200μl 80%乙醇溶液,静置30s后彻底移除上清(可以使用10μl移液器移除底部残留乙醇溶液);
7.室温静置3–5min,使残留乙醇彻底挥发;
8.加入22μl的Nuclease–free water,将PCR管从磁力架取下,轻轻吸打重悬磁珠,避免产生气泡,室温静置2min;
9.将PCR管置于磁力架上2min使溶液澄清;
10.用移液器吸取20μl上清液,转移到新的PCR管中(置于冰盒上),在反应管上标记样本编号,准备下一步反应。
Step5:Pre–PCR反应
预先从﹣20℃保存的试剂盒中取出PCR Master Mix、TPE1.0和TPE2.0试剂,置于冰盒上溶解,混匀后置于冰上待用;注意TPE1.0和TPE2.0具体以收货单为准,按照下面所对应的体系进行配制。
1.在step4步骤10的管子中,按表5配置反应体系(此操作在冰盒上进行):

注:1.TPE2.0 index–8(10μM)中index号为1#–96#,记录好所使用的index号;
2.TPE2.0 index–8(10μM)为96孔板,使用前先离心。

注:1.TPE1.0 Index(10μM)中index号为1#–8#,记录好所使用的index号;
2.TPE2.0 Index–8(10μM)中index号为1#–12#,记录好所使用的index号。

注:TPE2.0 Index–6(10μM)中index号为1#–16#,记录好所使用的index号。
2.使用移液器轻轻吹打混匀,然后短暂离心;
3.将样品置于PCR仪上,启动PCR程序,如下:
4.PCR循环数根据投入DNA的量不同进行调整,参考数据如表8所示:

5.程序运行完成后立即进行磁珠的纯化。
Step6:PCR扩增后纯化
1.向step5反应后的PCR管中加入50μl Agencourt AMPure XP磁珠,用移液器吹打混匀,避免产生气泡;
2.室温孵育5–15min,把PCR管置于磁力架上3min使溶液澄清;
3.移除上清,PCR管继续放置在磁力架上,向PCR管内加入200μl 80%乙醇溶液,静置30s;
4.移除上清,再向PCR管内加入200μl 80%乙醇溶液,静置30s后彻底移除上清(可以使用10μl移液器移除底部残留乙醇溶液);
5.室温静置5min,使残留乙醇彻底挥发;
6.加入30μl的Nuclease–free water,将离心管从磁力架取下,使用移液器,轻轻吸打重悬磁珠;
7.室温静置2min,将200μl PCR管置于磁力架上2min使溶液澄清;
8.用移液器将上清液转移到新的200μl PCR管中(置于冰盒上),在反应管上标记好样本号,准备下一步反应;
10.取1μl样品使用Agilent 2100 Bioanalyzer system(Agilent DNA 1000Kit)进行文库片段长度测定,文库长度约在270bp–320bp间。
Step7:样品、探针液相杂交
文库准备
1.预先从﹣20℃保存的试剂盒中取出表9、10和11所示block试剂,放置于冰盒上溶解,溶解混匀后置于冰上或4℃待用;
2.按照以下体系将样品文库与各Hyb Block混匀,标记为B管;注意Block具体以收货单为准,按照下面所对应的体系进行配制。

3.将Hyb Buffer置于室温融化,融解之后会有沉淀出现,混匀后置于65℃恒温混匀仪内预热,完全溶解后(无沉淀及浑浊物)取20μl Hyb Buffer置于新的200μlPCR管内,盖好管盖,置于65℃恒温混匀仪孵育待用,标记为A管;
4.取5μl RNase Block与2μl Probe置于200μl PCR管内,轻轻吸打混匀,短暂离心后置于冰上或4℃待用,标记为C管。
操作步骤(一)
1.将B管放入真空浓缩离心机,打开PCR管盖,启动离心机,打开真空泵开关,开始浓缩(浓缩之前可以先用相同体积的水进行浓缩时间测试,计算样品单位体积减少的时间,确认浓缩速率,避免因浓缩时间过长导致过度干燥,造成样品损耗);
2.将B管浓缩至体积小于10μl,不足用Nuclease–free water补足,轻轻吸打混匀,短暂离心后置于冰上待用;
3.设置PCR仪参数如下:
Heat lid 105℃、95℃for 5min、65℃hold;
4.将PCR管B置于PCR仪上,运行以上程序;
5.PCR仪温度降至65℃时,将A管置于PCR仪上孵育,盖上PCR仪热盖;
6.5min后,将C管置于PCR上孵育,盖上PCR仪热盖(注意:保持A管和B管在PCR仪上不动);
7.2min后,把移液器调至13μl,从A管中吸取13μl Hyb Buffer移至C管中,吸取全部B管中样品移至C管中,轻轻吸打10次,充分混匀,避免产生大量气泡,密封管盖,盖上PCR仪热盖,65℃孵育过夜(16–24h)。
或者采用操作步骤(二)替代Step7的操作步骤(一),两种操作方式构建文库下机数据质量一致。
操作步骤(二)
1.将A管13μl与C管7μl混合在一起,标记为AC管;
2.将B管放入真空浓缩离心机,打开PCR管盖,启动离心机,打开真空泵开关,开始浓缩(浓缩之前可以先用相同体积的水进行浓缩时间测试,
计算样品单位体积减少的时间,确认浓缩速率,避免因浓缩时间过长导致过度干燥,造成样品损耗);
3.将B管浓缩至体积小于10μl,不足用Nuclease–free water补足,轻轻吸打混匀,短暂离心后置于冰上待用;
4.设置PCR仪参数如下:
Heat lid 105℃、95℃for 5min、65℃hold;
5.将PCR管B置于PCR仪上,运行以上程序;
6.PCR仪温度降至65℃时,将AC管置于PCR仪上孵育,盖上PCR仪热盖;
7. 2min后,把移液器调至20μl,,吸取全部AC管中样品移至B管中,轻轻吸打10次,充分混匀,避免产生大量气泡,密封管盖,盖上PCR仪热盖,65℃孵育过夜(16–24h)。
Step8:捕获磁珠准备
1.将捕获磁珠(Cap beads)从4℃取出,涡旋震荡重悬;
2.取50μl磁珠置于新的PCR管内,置于磁力架上1min使溶液澄清,移除上清;
3.从磁力架上取下PCR管,加入200μl Binding Buffer轻轻吸打数次混匀,重悬磁珠;
4.置磁力架上1min,移除上清;
5.重复步骤3–4两次,共清洗磁珠3次;
6.从磁力架上取下PCR管,加入200μl Binding Buffer轻轻吸打6次重悬磁珠待用。
注:捕获时所用的磁珠可为杭州惠煜医疗科技有限公司的Cap beads或DynabeadsMyOne Streptavidin T1 magnetic beads,不可使用其他型号的链霉亲和素磁珠如C1、M270、M280等替代,不同的链霉亲和素磁珠的反应条件不同不可随意更换,不可使用纯化磁珠Agencourt AMPure XP替代捕获使用的Cap beads或Dynabeads MyOne StreptavidinT1magnetic beads。
Step9:捕获目标区域DNA文库
实验前准备
提前分装Wash Buffer 2(每个捕获需要200μl×3=600μl),并置于恒温振荡混匀仪上65℃预热。
操作步骤
1.保持液相杂交产物在PCR仪上,将步骤Step8中第6步重悬后的200μl Cap beads加入到液相杂交产物中,用移液器吸打6次混匀,置于旋转混匀仪上室温结合30min;
2.将PCR管置于磁力架上2min使溶液澄清,移除上清液;
3.向液相杂交产物内加入200μl的Wash Buffer 1,轻轻吸打6次混匀,置于旋转混匀仪上清洗15min,然后短暂离心,将PCR管放于磁力架上2min,使溶液澄清,移除上清;
4.加入200μl的65℃预热的Wash Buffer 2,轻轻吸打6次混匀,置于恒温振荡混匀仪上65℃孵育10min,转速800转/min进行清洗;
5.短暂离心,将PCR管放于磁力架上2min,移除上清。使用Wash Buffer 2清洗2次,共计3次。最后一次彻底移除Wash Buffer 2(可用10μl移液器移除残留);
6.PCR管继续置于磁力架上,向PCR管内加入200μl 80%乙醇,静置30s后彻底移除乙醇溶液(可用10μl移液器移除残留),室温晾干;
7.向PCR管加入30μl Nuclease–free water,从磁力架上取下PCR管,轻轻吸打6次重悬磁珠待用。
Step10:Post–PCR反应
实验前准备
预先从–20℃保存的试剂盒中取出Post PCR Buffer、Post PCR Primer(25μM,forILM)试剂,放置冰上溶解,溶解混匀后置于冰上待用。取出Post PCR Polymerase简短离心后立即使用。
1.捕获后需要对DNA文库进行富集,根据表12配制反应体系:

2.将移液器调至40μl,轻轻吸打混匀6次,然后置于PCR仪上。
操作步骤
1.运行PCR仪程序:
2.PCR结束后向样品加入55μl Agencourt AMPure XP磁珠,用移液器轻轻吸打6次混匀;
3.室温孵育5min,把PCR管置于磁力架上3min使溶液澄清;
4.移除上清,PCR管继续置于磁力架上,加入200μl 80%无水乙醇,静置30s;
5.移除上清,再向PCR管内加入200μl 80%无水乙醇,静置30s后彻底移除上清(可用10μl移液器移除底部残留酒精);
6.室温放置5min,使得残留乙醇彻底挥发;
7.加入25μl Nuclease–free water,将PCR管从磁力架拿下,轻轻吹打混匀重悬磁珠,室温放置2min;
8.将PCR管置于磁力架上2min;
9.用移液器吸23μl上清液转移到1.5ml离心管,标记样品信息;
10.取1μl文库使用Qubit dsDNA HS Assay Kit进行定量,记录文库浓度,文库浓度约在1–10ng/μl;
11.取1μl样品使用Agilent 2100Bioanalyzer system(Agilent DNA 1000Kit)进行文库片段长度测定,文库长度约在270bp–320bp之间;
12.使用高通量测序平台进行测序。
在本文中所使用的术语“DNA”为脱氧核糖核酸(英文:Deoxyribonucleic acid,缩写为DNA)是一种由脱氧核糖核苷酸组成的双链分子。可组成遗传指令,引导生物发育与生命机能运行,其碱基排列顺序构成了遗传信息,所以在遗传病的诊断中具有重要的作用。
在本文中所使用的术语:“Q–PCR”为实时荧光定量核酸扩增(英文:Real-timeQuantitative PCR)。一种利用荧光检测达到实时检测PCR状况的PCR技术。
在本文中所使用的术语:“reads”为测得的DNA序列。
在本文中所使用的术语“高通量测序技术”指的是第二代高通量测序技术及之后发展的更高通量的测序方法。第二代高通量测序平台包括但不限于Illumina–Solexa(Miseq、Hiseq–2000、Hiseq–2500、Hiseq X ten等)、ABI–Solid和Roche–454测序平台等。随着测序技术的不断发展,本领域技术人员能够理解的是还可以采用其他方法的测序方法和装置进行本发明的检测。根据本发明的具体示例,可以将根据本发明实施例的核酸标签用于Illumina–Solexa、ABI–Solid和Roche–454测序平台等的至少一种进行测序。高通量测序技术如Miseq测序技术具有以下优势:(1)高灵敏度:高通量测序如Miseq的测序通量大,目前一个实验流程下来可以产生最多15G碱基数据,高的数据通量可以再测序序列数确定的情况下,使得每条序列获得高的测序深度,可以检测到含量更低的突变,同时因其测序深度高,其测序结果也更为可靠;(2)高通量、低成本:利用根据本发明的标签序列,通过一次测序可以检测上万份样本,从而大大降低成本。

Claims (9)
1.一种用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,提取检测对象的基因组DNA后进行包括以下步骤的处理:
S1、将基因组DNA进行片段化;
S2、将片段化的基因组DNA进行末端修复和3'末端添加碱基A;
S3、将3'末端添加碱基A的产物连接扩增接头并纯化,以进行有效连接产物的富集;
S4、将连接产物进行Pre–PCR扩增反应,富集有效产物;
S5、用探针与上述富集的有效产物模板进行液相杂交后,并与磁珠结合,洗脱未结合探针与非特异性结合不稳定文库,捕获上述模板中目标区域DNA文库;
S6、将捕获的目标区域DNA文库分离后连接完整接头,进行Post–PCR扩增反应,捕获目标区域文库;
S7、对目标区域文库进行定量和质检操作;
S8、上机测序;
S9、数据分析得到致病位点相关信息;
其中,所述探针通过遗传性帕金森病相关基因和/或相关突变点设计得到,包括SNCA、SNCB、SORL1、SQSTM1、TF、TMEM106B、TREM2、TRPM7、TTR、UBB、UBQLN2、VCP、VEGF、ZCWPW1、UCHL1、PINK1、LRRK2、ATP13A2、GIGYF2、HTRA2、FBX07、ATXN2、ATXN3、GBA、SYT11、STK39、MCCC1、DGKQ、BST1、ITGA8、LRRK2、CCDC62、MAPT、PARK2、PARK7ITPKB、IL1R2、SCN3A、SATB1、NCKIPSD、ALAS1、TLR9、DNAH1、BAP1、PHF7、NISCH、STAB1、ITIH3、ITIH4、ANK2、CAMK2D、ELOVL7、ZNF184、CTSB、PDLIM3、C8ORF58、BIN3、SH3GL2、FAM171A1、GALC、COQ7、TOX3、ATP6V0A1、PSMC3IP、TUBG2中的至少一种。
2.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,所述基因组DNA来源于人类。
3.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,提取所述基因组DNA的方法包括纯化柱纯化、磁珠纯化或酚氯仿提取。
4.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,S7中所述定量的方法包括基于荧光定量原理的定量装置定量、Q–PCR定量或电泳。
5.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,S1中所述基因组DNA片段化的方法包括超声破碎、转座酶酶切或限制性内切酶酶切。
6.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,S5和S6中所述捕获方法包括液相探针捕获、固相芯片液相杂交捕获或PCR富集。
7.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,S8中所述上机测序通过二代测序平台进行。
8.根据权利要求1所述的用于获取遗传性帕金森病致病基因突变相关信息的检测方法,其特征在于,S9中所述数据分析的具体操作如下:通过先对所得测序数据与人类基因组的参考序列进行对比,再对所测基因的每个突变位点情况进行分析,得到遗传性帕金森病致病突变相关信息。
