Abstract
The invention discloses application of endophytic fungus Peniophora sp.A871 in preparation of antibacterial drugs. The invention discovers that the endophytic fungus Peniophora sp.A871 extract has obvious inhibiting effect on staphylococcus aureus and bacillus subtilis, and has good application and development prospect. Therefore, the invention provides a candidate drug for researching and developing a new antibacterial drug and provides a scientific basis for developing and utilizing natural active substances derived from endophytic fungi of plants.
Description
Technical Field
The invention belongs to the technical field of biological medicines, and particularly relates to an application of an aquilaria malabarica endophytic fungus Peniophora sp.A871 extract in preparation of antibacterial drugs.
Background
The plant endophytic fungi has rich species diversity and genetic diversity, so that a plurality of new or rare fungal species are discovered from the plant endophytic fungi so far, and the plant endophytic fungi become research hotspots at home and abroad. Researches show that the active compounds produced by the endophytic fungi of plants far exceed the range of plant metabolites, can produce various active substances with unique structure types and potential application prospects, and become important resources for finding new natural active substances.
At present, the infection caused by the increasingly strong pathogenic microorganisms, especially drug-resistant strains, is found to be more serious in clinical treatment, and many traditional antibiotics basically lose the treatment efficacy, so that the search for novel antibacterial drugs is a key field of new drug development in the world. (1) Inoculating endophytic fungus Peniophora sp.A871 to a slant culture medium, and culturing at 28 deg.C for 5d to obtain activated strain. The slant culture medium is obtained by the following method: decocting potato 200g in distilled water for 20min, filtering to obtain juice, and adding glucose 20g and KH2PO4 3g、MgSO40.75g, vitamin B1 10mg,20g of agar, supplementing water to 1L, naturally adjusting the pH value, and sterilizing for later use.
(2) Inoculating the strain A871 subjected to activation culture in the step (1) into a seed culture medium, and culturing for 7d under the conditions of 28 ℃ and 120r/min oscillation to obtain a seed solution, wherein the seed culture medium is obtained by the following method: decocting potato 200g in distilled water for 20min, filtering to obtain juice, and adding glucose 20g and KH2PO4 3g、MgSO40.75g, vitamin B110mg, adding water to 1L, naturally adjusting pH, and sterilizing.
(3) Inoculating the seed liquid obtained in the step (2) into a fermentation culture medium in an inoculation amount of 10% (v/v), and culturing at 28 ℃ and 120r/min for 7d to obtain a fermentation culture. The fermentation medium is the same as the seed culture medium.
(4) Preparing an extract: and (3) filtering the fermentation culture obtained in the step (3) to remove mycelium, extracting the filtrate for 3 times by using ethyl acetate, and concentrating the extract under reduced pressure to obtain the endophytic fungus Peniophora sp.A871 extract.
Example 1: determination of activity of endophytic fungus Peniophora sp.A871 extract against staphylococcus aureus and bacillus subtilis
The endophytic fungus Peniophora sp.A871 extract was dissolved in DMSO and diluted to a concentration of 2mg/mL to obtain a sample dilution.
Blending Staphylococcus aureus or Bacillus subtilis in logarithmic growth phase with normal saline to give a solution with a concentration of 106Absorbing 1mL of prepared bacterial liquid into a culture dish, pouring LB culture medium cooled to a proper temperature, uniformly mixing, clamping a filter paper sheet with the thickness of 1.5mm and the diameter of 6mm by using a sterile forceps, placing the filter paper sheet on a bacterial-containing flat plate, respectively dropwise adding 5 mu L of the sample diluent on the filter paper sheet to serve as a sample group, replacing the sample diluent with DMSO to serve as a blank control group, using ampicillin as a positive control group, placing the flat plate in a 37 ℃ incubator for culturing for 24h, and measuring the diameter of an antibacterial ring by using a cross measurement method.
