Abstract
The invention discloses a selenium-rich beverage formula for enhancing immunity, which comprises the following raw materials in parts by weight based on 100 parts by weight: 0.003-0.007 part of selenium-rich shepherd's purse powder, 0.6-1.0 part of blueberry concentrated juice, 0.000001-0.00001 part of burdock extract, 0.03-0.07 part of potassium sorbate, 0.005-0.012 part of sucralose and the balance of water. The preparation method comprises (1) weighing the raw materials for use; (2) placing selenium-rich shepherd's purse powder, blueberry concentrated juice, burdock extract, potassium sorbate, sucralose and water in a shearing tank for shearing to obtain a mixed solution; (3) homogenizing the mixed solution, filtering, and sterilizing to obtain the selenium-rich beverage. The selenium-rich capsella bursa-pastoris powder is added to provide necessary trace elements for a human body, the blueberry concentrated juice and the burdock extract are added to further enhance the effect of human immunity, and anthocyanin in the blueberry can prevent myopia and improve eyesight; the burdock has the special kidney tonifying effect.
Description
Technical Field
The invention relates to the technical field of beverages, in particular to a selenium-rich beverage formula for enhancing immunity and a preparation method thereof.
Background
Selenium is honored as the 'fire species of life' by the medical and nutritional communities at home and abroad, and enjoys the reputations of 'longevity element', 'king of cancer', 'heart guard spirit', 'natural antidote' and the like. Selenium is one ten million in human tissues, but the selenium determines the existence of life, and the selenium has great effect on human health and cannot be replaced by other substances. The selenium deficiency can directly lead to the reduction of the immunity of human bodies, and more than forty diseases threatening the health and the life of human bodies are proved to be related to the selenium deficiency of human bodies by clinical medicine, such as cancers, cardiovascular diseases, liver diseases, cataract, pancreatic diseases, diabetes, reproductive system diseases and the like. Both animal experiments and clinical practice indicate that sufficient selenium should be continuously obtained from the diet, and if the selenium cannot be supplemented in time, the disease-removing capacity is reduced.
China is one of the countries with serious selenium deficiency, the areas with selenium deficiency in China occupy 72 percent of the total area, and 5 months and 17 days are set as the selenium supplement days of the whole people by the Ministry of health. The common research of Chinese scientists and foreign scientists finds that: selenium deficiency is the source of ten thousand diseases, more than forty diseases are directly related to selenium deficiency in human beings, more than one hundred diseases are related to selenium deficiency, and in addition, the anti-aging mechanism and the immunity regulation function of human beings are closely related to selenium. The Hubei Enshi, Wei Wuling mountain range, the Hubei Gao land has forest coverage rate of nearly 70 percent and is called the 'selenium city of the world' brand. On this unusually thick land, selenium-rich plants are growing. The plant rich in selenium in the region can be used for preparing beverage.
However, the selenium-rich beverage at present has low selenium content and bitter taste, and cannot meet the market demand.
Therefore, the problem to be solved by the technical personnel in the field is how to provide a selenium-rich plant beverage with high selenium content, enhanced immunity and good taste and a preparation method thereof.

The invention has the beneficial effects that: the selenium-rich capsella bursa-pastoris powder is added to provide necessary trace elements for a human body, the blueberry concentrated juice and the burdock extract are added to further enhance the effect of human immunity, and anthocyanin in the blueberry can prevent myopia and improve eyesight; the burdock has the special kidney tonifying effect.

The effects of the raw materials in the invention are as follows:
shepherd's purse: sweet in taste, warm in nature, non-toxic, harmonizing spleen, inducing diuresis, stopping bleeding, improving eyesight. It can be used for treating dysentery, edema, gonorrhea, chyluria, hematemesis, hematochezia, metrorrhagia, menorrhagia, conjunctival congestion, and pain.
Blueberry: the anthocyanidin in fructus Myrtilli can promote regeneration of rhodopsin in retinal cell, prevent severe myopia and retinal detachment, and improve vision. Meanwhile, the anthocyanin has the capability of accelerating the regeneration of the rhodopsin, which is an indispensable component for ensuring good vision.
Blueberry has the function of resisting free radicals, the free radicals can be generated in a plurality of environments and worsen in a human body to cause various diseases related to the free radicals, the occurrence of the diseases can be prevented by removing the free radicals, the damage of the free radicals to cell membranes, DNA and other cell components can be reduced, the functional disorder in the body can be prevented, and the immunity and disease resistance of the human body can be enhanced.
Anthocyanidin in blueberry fruit is a very strong antioxidant, and can help prevent the formation of intra-arterial plaque and various cancers (cervical cancer and the like) and reduce the possibility of cancer. The natural blueberry anthocyanin is the most effective antioxidant, and researches on USDA show that the blueberry has the strongest antioxidant capability in more than 40 kinds of fruits and vegetables which are frequently eaten by people. This means that eating blueberries can have high antioxidant capacity, thereby delaying human aging.
The blueberry has high content of antioxidant, and can improve mental power and reduce incidence rate of Alzheimer disease. The blueberry is rich in anthocyanin, and has the effects of preventing dysfunction, improving short-term memory, improving the balance and coordination of the old and the like.
Burdock: the burdock contains various vitamins and minerals required by human bodies, such as inulin, cellulose, protein, calcium, phosphorus, iron and the like, wherein the content of carotene is 150 times higher than that of carrot, and the content of protein and calcium is the first of rhizomes. The radix Arctii contains inulin and volatile oil, arctic acid, multiple polyphenols and aldehydes, and is rich in cellulose and amino acids.
The burdock also has (1) antibacterial effect: the burdock whole plant contains antibacterial components, wherein the leaves contain the most antibacterial components and mainly resist staphylococcus aureus. (2) The hypoglycemic effect: the burdock fruit water extract can obviously and durably reduce the blood sugar of a rat and can increase the carbohydrate tolerance. (3) Anti-aging and oxygen radical scavenging effects: during the normal active metabolism of human life, oxygen free radicals, which are substances harmful to health and promote cell aging, are generated. They can promote the formation and accumulation of age spots, which are pigments that produce lipofuscin. The appearance of senile plaques on the body surface indicates that cells in the body have entered the aging stage. The bovine lateral root contains peroxidase, which can enhance the activity of cellular immune mechanism, eliminate oxygen free radicals in vivo, prevent lipofuscin pigment from generating and accumulating in vivo, resist aging, and provide an inner protective environment for resisting and eliminating oxygen free radicals for the organism. (2) placing selenium-rich shepherd's purse powder, blueberry concentrated juice, burdock extract, potassium sorbate, sucralose and water in a shearing tank for shearing to obtain a mixed solution;
(3) homogenizing the mixed solution, filtering, and sterilizing to obtain the selenium-rich beverage.
The preparation method is simple, easy to operate and more suitable for large-scale industrial production.
Preferably, the preparation method of the burdock extract in the step (2) comprises the following steps:
step (21) weighing burdock and 60 wt% ethanol solution respectively according to the mass ratio of 1 (5-7) at the temperature of 60-65 ℃, carrying out reflux extraction on the burdock for 3 times with 60 wt% ethanol for 2 hours each time to obtain burdock slag and filtrate, and recovering ethanol in the filtrate to obtain burdock liquid for later use;
step (22) comprises the following steps of (1) mixing burdock slag with water by mass: (6-9), decocting the burdock dregs with water at 80-100 ℃ for 3 times, each time for 2 hours, filtering, combining the obtained filtrate with the burdock liquid obtained in the step (21), concentrating and drying to obtain the burdock extract.
Preferably, in the step (2), the shearing speed is 500-600r/min, and the time is 8-10 min.
Preferably, in the step (3), the homogenizing speed is 1000-.
Preferably, in the step (3), the sterilization temperature is 85-100 ℃ and the time is 25-35 min.
According to the technical scheme, compared with the prior art, the selenium-rich beverage formula for enhancing the immunity and the preparation method thereof are disclosed, and the selenium-rich beverage prepared by compounding the selenium-rich raw material, the blueberry concentrated juice and the burdock extract has high nutritional value, high selenium content and good taste, and can improve the immunity of the organism, so that the effect of maintaining the blood and delaying the aging is achieved.

step (22) comprises the following steps of (1) mixing burdock slag with water by mass: and 8, decocting the burdock dregs with water at 85 ℃ for 3 times, each time for 2 hours, filtering, combining the obtained filtrate with the burdock liquid obtained in the step (21), concentrating and drying to obtain the burdock extract.

The selenium-enriched beverage for enhancing immunity has the following formula: 0.003g of selenium-enriched shepherd's purse powder, 0.6g of blueberry concentrated juice, 0.000001g of burdock extract, 0.03g of potassium sorbate, 0.005g of sucralose and 99.362g of water.
The preparation method of the selenium-enriched beverage for enhancing immunity comprises the following steps:
(1) weighing the raw materials according to the formula for later use;
(2) placing selenium-rich shepherd's purse powder, blueberry concentrated juice, burdock extract, potassium sorbate, sucralose and water in a shearing tank, and shearing at 500r/min for 8min to obtain a mixed solution;

The selenium-enriched beverage for enhancing immunity has the following formula: 0.007g of selenium-enriched shepherd's purse powder, 1.0g of blueberry concentrated juice, 0.00001g of burdock extract, 0.07g of potassium sorbate, 0.012g of sucralose and 98.911g of water.
The preparation method of the selenium-enriched beverage for enhancing immunity comprises the following steps:
(1) weighing the raw materials according to the formula for later use;
(2) placing selenium-rich shepherd's purse powder, blueberry concentrated juice, burdock extract, potassium sorbate, sucralose and water in a shearing tank, and shearing at 600r/min for 10min to obtain a mixed solution;

Performance testing
1. Animal experiments: influence on phagocytic function of mouse abdominal cavity macrophages
Taking 70 mice, randomly dividing into 7 groups, and respectively drenching the products prepared in the examples 1-5 and the comparative examples 1-2 of the invention, wherein the product is taken 3 times a day, 10mL each time, and is continuously taken for 1 month; injecting 5% of chicken blood erythrocytes into the abdominal cavity of all mice after administration, wherein each mouse is 0.2mL, killing the mice by a cervical dislocation method after 2 hours, immediately injecting 0.5mL of physiological saline into the abdominal cavity, gently kneading the abdominal cavity for 2 minutes, shearing the abdominal skin, opening a small opening in a muscle layer, taking abdominal cavity liquid and a push sheet on a clean glass slide, incubating for 30 minutes at 37 ℃, performing microscopic examination after staining, and directly observing and counting under a microscope, wherein the average number of macrophages in each mouse is 2000. Calculating phagocytosis rate and phagocytosis index, phagocytosis rate: macrophage number/2000 × 100% for phagocytosing chicken red blood cells; specific data are shown in table 1:
as can be seen from table 1, the products prepared in embodiments 1 to 5 of the present invention can significantly improve the phagocytic rate of mouse peritoneal macrophages, which indicates that the phagocytic rate of chicken red blood cells by mouse peritoneal macrophages can be enhanced, and the cellular immunity of mice can be improved, so that the products of the present invention have the effect of improving the human immunity.
2. The sensory evaluation results of the product of the invention are shown in table 2:
from the table 2, the product prepared in the invention meets GB7101-2015, is also suitable for infants to eat, and can obviously enhance the immunity through the table 1-2.

step (22) comprises the following steps of (1) mixing burdock slag with water by mass: (6-9), decocting the burdock dregs with water at 80-100 ℃ for 3 times, each time for 2 hours, filtering, combining the obtained filtrate with the burdock liquid obtained in the step (21), concentrating and drying to obtain the burdock extract.
6. The method for preparing the selenium-enriched beverage formula for enhancing immunity as claimed in claim 4, wherein in the step (2), the shearing speed is 500-600r/min, and the time is 8-10 min.
