Abstract
本发明提供一种大鼠牙周炎模型建立方法,属于牙周病学技术领域,向多个待建模大鼠注射药物,建立牙周炎大鼠模型;将多个患有牙周炎大鼠划分为硬食喂养组和软食喂养组,向硬食喂养组的牙周炎大鼠喂养硬食,向软食喂养组的牙周炎大鼠喂养软食;分离牙周炎大鼠的双侧磨牙区上颌骨标本,扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立。通过对大鼠待建模组织行影像学分析,证实了通过改变食物硬度来检测牙周炎大鼠局部牙周组织病理变化,充分模拟了临床牙列缺损伴牙周炎患牙的病理学过程,为牙周疾病及伴有牙周疾病的牙列缺损患者义齿设计研究提供了创新的方法。

技术领域
本发明涉及牙周病学技术领域,具体而言,涉及一种大鼠牙周炎模型建立方法。
背景技术
当今社会“缺牙”-牙列缺损或缺失,已成为困扰我国中老年人的首要口腔问题,对患者的咀嚼功能、外貌面容及颞下颌功能等都会产生不良影响。根据当代口腔医学观点,应尽可能地维护患者留存牙齿以预防老年人过早进入牙列缺失阶段,而牙列缺损的中老年患者常伴有不同程度的慢性牙周炎,因此利用牙周炎患者余留牙作为基牙来设计义齿存在很大风险,这极大地增加了修复缺失牙的难度。如何在此类患者现有口腔条件下,充分利用剩余牙齿的稳定、支持作用来恢复牙列完整性,减轻缺牙对口腔健康的进一步损害,延长修复体使用寿命,以达到提高患者生存质量的目的,是口腔医生与患者共同关心的话题。
因此,通过建立牙周炎动物模型,并模拟不同咀嚼力对于牙列缺损伴牙周炎患者的治疗具有重要意义。通过文献检索发现,目前有学者尝试研究牙周炎伴咬合创伤动物模型的建立,但尚未发现有通过改变食物硬度来建立牙周炎大鼠力学载荷复合模型的相关研究,因此本发明具有一定创新性。
发明内容
本发明的目的在于提供一种大鼠牙周炎模型建立方法,能够建立牙周炎力学载荷复合模型,提供患有牙周炎后,牙齿咬合力的数据模型。
本发明的实施例可以采用以下技术方案实现:
本发明实施例提供一种大鼠牙周炎模型建立方法,其包括向多个待建模大鼠注射药物,建立牙周炎大鼠模型;将患有牙周炎大鼠划分为硬食喂养组和软食喂养组,向所述硬食喂养组的牙周炎大鼠喂养硬食,向所述软食喂养组的牙周炎大鼠喂养软食,其中,所述硬食喂养组和软食喂养组的牙周炎大鼠数量相等;分离所述牙周炎大鼠的双侧磨牙区上颌骨标本,扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立。
可选地,所述向多个待建模大鼠注射药物,建立牙周炎大鼠模型包括:麻醉多个所述待建模大鼠;分别在多个所述待建模大鼠的双侧上颌第一磨牙腭侧近中龈沟、根分叉对应黏膜及第一、第二磨牙间龈***三处位点注射脂多糖。

本发明实施例提供的大鼠牙周炎模型建立方法,采用与人类患有牙周炎病症相似的大鼠进行建模,能使得到的数据更接近人类数据,数据的可参考性更高。建模时,先对待建模大鼠注射药物,建立牙周炎大鼠模型;然后对患有牙周炎的大鼠按喂养硬食和喂养软食划分,使其分为硬食喂养组和软食喂养组,喂养一段时间后,分离待建模大鼠的双侧磨牙区上颌骨标本,通过micro-CT扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立,通过硬食喂养和软食喂养得到两组力学载荷复合模型,以进行数据分析和对比,得出患有牙周炎后,咀嚼硬食使牙齿承担过大咬合力对牙周炎的影响,填补现有模型建立存在的这一缺漏,本实施例通过改变食物硬度来建立牙周炎大鼠力学载荷复合模型,通过对大鼠待建模组织行影像学分析,证实了通过改变食物硬度来检测牙周炎大鼠局部牙周组织病理变化,充分模拟了临床牙列缺损伴牙周炎患牙的病理学过程,以进一步推进了牙周病研究的深度和广度,并为伴有牙周疾病的牙列缺损患者义齿设计提供了创新的方法。

牙周病是常见的牙齿疾病,目前是造成成年人牙缺失的主要病因,而在牙列缺损形成后牙周炎往往也并未能得到良好的控制,因此这两种口腔常见疾病在中老年人中常常同时存在,对于修复医生来说如何利用患有牙周炎的余留牙作为基牙进行修复设计是个难题。
牙周炎患者的牙周储备力与健康牙齿相比显著降低,若再施加过大的咬合力,则会出现咬合创伤加重牙槽骨吸收,这个过程中牙周组织具体发生了怎样的变化,变化的机制是什么,是口腔研究的热点问题。
常用的牙周炎建模方法为单纯结扎丝结扎,通过在实验牙颈部或龈沟内放置缝合线、不锈钢丝等来造成局部菌斑堆积诱发牙周炎。该方法需定期检查,若脱落则需再次手术安置,全麻手术具有一定的风险,局部安置也存在一定的刺激。
通过查阅文献得到以下几种加力方法:施加正畸的牵引力、抬高咬合、改变食物硬度或者在邻间隙施加楔力。其中抬高咬合制造早接触从而实现咬合创伤是学者们常用的方式,具体操作方法包括有制作金属冠、粘接异物等,但都与本实施例想要模拟的状态有所区别。
对于牙周炎患者来说,正常硬度的食物已属于一种过大的咬合负担,而添加过硬的食物更会使咀嚼力超过牙周潜力。对于牙列缺损患者而言,若不注意食物硬度,余留牙往往需要承担过大的咬合力,造成咬合创伤。因而改变食物硬度,通过大鼠自由进食行使咀嚼功能,可以更好地模拟复杂的咀嚼运动,间接完成过大力学载荷,符合本实施例建模目的。
大鼠属于单牙列啮齿类动物,其磨牙形态、牙周软/硬组织等组织学表现与人类近似,与犬、猪等动物相比价格低廉、便于饲养,与小鼠等动物相比体型较大、易于操作,且经前人大量实验,发现其在牙周炎表现上与人类十分近似,因此本实验考虑选取大鼠来建立动物模型。因此,利用大鼠模型对牙周病患者在不同咀嚼载荷下牙周疾病的发生发展机制进行进一步研究十分必要。
具体地,请参照图1,本实施例提供一种大鼠牙周炎模型建立方法,其包括:
S100:向多个待建模的大鼠注射药物,建立牙周炎大鼠模型。
向多个待建模的大鼠注射药物,使多个待建模大鼠患牙周炎,建立牙周炎大鼠模型。
具体地,S101:向多个待建模大鼠分别喂食10%葡萄糖水,10%葡萄糖水是作为辅助刺激因素诱导建模。
S102:麻醉多个待建模大鼠。
使多个待建模大鼠吸入3%异氟烷麻醉,使多个待建模大鼠处于麻醉状态。
S103:分别在多个待建模大鼠的双侧上颌第一磨牙腭侧近中龈沟、根分叉对应黏膜及第一、第二磨牙间龈***三处位点注射脂多糖。
具体地,分别向多个待建模大鼠缓慢注射3μL(浓度为10μg/μL)的脂多糖(LPS),注射时需缓慢注射,并停留数秒,目的是减少试剂流失。
隔天再分别向多个待建模大鼠缓慢注射浓度为10μg/μL的脂多糖;
连续注射18天~22天。
S110:将多个患有牙周炎大鼠划分为硬食喂养组和软食喂养组,向硬食喂养组的牙周炎大鼠喂养硬食,向软食喂养组的牙周炎大鼠喂养软食,其中,硬食喂养组和软食喂养组的牙周炎大鼠数量相等。
具体地,每天向硬食喂养组的牙周炎大鼠喂养硬食,向软食喂养组的牙周炎大鼠喂养软食。
每天投食足量常规的硬饲料或软饲料,并使牙周炎大鼠自由饮水。
一般硬食食物,牙周炎大鼠咀嚼硬食,以模拟患有牙周炎后牙齿承担过大咬合力的情况。
软食用于作为对比模型,能更充分说明对比的情况。
其中,硬饲料:在原饲料的基础成分中提高粗纤维的占比约3-5%,增大原饲料粉剂的颗粒大小,使用较大压强压制,成品饲料颗粒硬度≥20kg/cm2。
软饲料:将原饲料的基础成分的颗粒研磨至更细,并且加入适当水分混合成糊状,制作成条形后常温干燥24h使之定型,但保留一定水分。
连续喂养7天~10天,达到需要的建模时间。当然,喂养天数也可根据实际需要具体设置。
S120:分离牙周炎大鼠的双侧磨牙区上颌骨标本,通过micro-CT扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立。
在该步骤之前,使牙周炎大鼠吸入3%异氟烷,使牙周炎大鼠处于麻醉状态,然后断头处死牙周炎大鼠。然后再取每个牙周炎大鼠的双侧上颌骨组织,通过micro-CT组织影像学扫描局部组织以检测牙周炎大鼠的双侧磨牙区上颌骨组织。
扫描得到牙周炎大鼠力学载荷复合模型,每个牙周炎大鼠对应一个力学载荷复合模型,根据力学载荷复合模型进行数据分析和处理,以进行牙周病的研究。

以下通过一个具体示例进行大鼠牙周炎模型建立方法的说明:
1、牙周炎大鼠模型建立:
6只待建模大鼠,10%葡萄糖水喂养,3%异氟烷吸入麻醉下,在双侧上颌第一磨牙腭侧近中龈沟、根分叉对应黏膜及第一、第二磨牙间龈***3处位点分别使用微量进样器注射3μL(浓度为10μg/μL)LPS,缓慢注射,并停留数秒,减少试剂流失,隔天注射,共18次。
2、不同咀嚼力模型的建立:
将上述建立好的牙周炎大鼠分为硬食喂养组和软食喂养组,每组3只,每天投食足量常规的硬饲料或软饲料,自由饮水,连续喂养7天。
为提高对比度,还可设置3只建立好的牙周炎大鼠为对照组,对照组的3只牙周炎大鼠按照上述牙周炎大鼠模型建立的方法进行,对照组为正常硬度饲料喂养,自由饮水,连续喂养7天。
2、影像学分析:于3%异氟烷吸入麻醉下断头处死所有大鼠,分离双侧磨牙区上颌骨标本,送Micro-CT扫描。
结果显示:与对照组大鼠相比,软食喂养组大鼠与硬食喂养组大鼠均有明显弧形牙槽骨吸收,远中根处尤为明显,近中根处牙周膜间隙增宽;相比于软食喂养组大鼠,硬食喂养组大鼠则吸收更为明显,且牙根与牙槽骨表面显得较为粗糙。
在经micro-CT扫描所得二维图像中进行,对每只大鼠左右两侧样本的腭侧牙槽嵴最低点到咬合平面的平均距离测量2次,然后取均值进行对比分析(图2~图4)。结果显示软食喂养组大鼠与硬食喂养组大鼠腭侧牙槽嵴最低点到咬合平面的最大距离明显大于对照组,统计学差异显著(P<0.05),如图5所示,大鼠左右侧上颌第一磨牙腭侧最低点距咬合平面最大距离的均值统计(*,P<0.05);软食喂养组多个大鼠腭侧牙槽嵴最低点到咬合平面的最大距离的平均距离小于硬食喂养组多个大鼠腭侧牙槽嵴最低点到咬合平面的最大距离的平均距离,但未见统计学差异(P>0.05)。

Claims (8)
1.一种大鼠牙周炎模型建立方法,其特征在于,所述方法包括:
向多个待建模大鼠注射药物,建立牙周炎大鼠模型;
将多个患有牙周炎大鼠划分为硬食喂养组和软食喂养组,向所述硬食喂养组的牙周炎大鼠喂养硬食,向所述软食喂养组的牙周炎大鼠喂养软食,其中,所述硬食喂养组和软食喂养组的牙周炎大鼠数量相等;
分离所述牙周炎大鼠的双侧磨牙区上颌骨标本,扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立。
2.根据权利要求1所述的大鼠牙周炎模型建立方法,其特征在于,所述向多个待建模大鼠注射药物,建立牙周炎大鼠模型包括:
麻醉多个所述待建模大鼠;
分别在多个所述待建模大鼠的双侧上颌第一磨牙腭侧近中龈沟、根分叉对应黏膜及第一、第二磨牙间龈***三处位点注射脂多糖。
3.根据权利要求2所述的大鼠牙周炎模型建立方法,其特征在于,所述麻醉多个所述待建模大鼠包括:
使多个所述待建模大鼠吸入3%异氟烷麻醉。
4.根据权利要求2所述的大鼠牙周炎模型建立方法,其特征在于,所述分别在多个所述待建模大鼠的双侧上颌第一磨牙腭侧近中龈沟、根分叉对应黏膜及第一、第二磨牙间龈***三处位点注射脂多糖包括:
分别向多个所述待建模大鼠缓慢注射浓度为10μg/μL的脂多糖;
隔天再分别向多个所述待建模大鼠缓慢注射浓度为10μg/μL的脂多糖;
连续注射18天~22天。
5.根据权利要求2所述的大鼠牙周炎模型建立方法,其特征在于,在所述麻醉多个所述待建模大鼠之前,所述方法包括:
向多个所述待建模大鼠分别喂食10%葡萄糖水。
6.根据权利要求1所述的大鼠牙周炎模型建立方法,其特征在于,所述将多个患有牙周炎大鼠划分为硬食喂养组和软食喂养组,向所述硬食喂养组的牙周炎大鼠喂养硬食,向所述软食喂养组的牙周炎大鼠喂养软食,其中,所述硬食喂养组和软食喂养组的牙周炎大鼠数量相等包括:
每天向所述硬食喂养组的牙周炎大鼠喂养硬食,向所述软食喂养组的牙周炎大鼠喂养软食;
连续喂养7天~10天。
7.根据权利要求1所述的大鼠牙周炎模型建立方法,其特征在于,所述分离所述牙周炎大鼠的双侧磨牙区上颌骨标本,扫描局部组织确认牙周炎大鼠力学载荷复合模型的建立之前,所述方法包括:
使所述牙周炎大鼠吸入3%异氟烷。
8.根据权利要求1所述的大鼠牙周炎模型建立方法,其特征在于,通过micro-CT组织影像学扫描所述局部组织以检测所述牙周炎大鼠的双侧磨牙区上颌骨组织。
