Description
Technical Field
The invention belongs to the technical field of cosmetics production, and particularly relates to preparation and application of a composition for removing acne and beautifying skin.
Background
Acne is a chronic inflammatory skin problem of the pilosebaceous unit, and the occurrence of acne is mainly closely related to factors such as hyperseborrhea, blockage of pilosebaceous ducts, bacterial infection and inflammatory reaction. After adolescence, the level of androgen, particularly testosterone, in a human body is rapidly increased, the development of sebaceous glands is promoted, and a large amount of sebum is produced. Meanwhile, abnormal keratinization of the pilosebaceous canal causes blockage of the canal, obstruction of sebum discharge and formation of a keratotic plug, namely, comedo. A number of microorganisms in the hair follicle, particularly propionibacterium acnes, multiply in number, and lipases produced by propionibacterium acnes break down sebum to produce free fatty acids, while chemotactic inflammatory cells and mediators, ultimately inducing and exacerbating the inflammatory response. It is manifested as acne (white or black), papule, pustule, and nodule, and severe cases form scar, which is chronic in course and easy to repeat.
Due to the improvement of living standard, such as the improvement of meat consumption, and the attention of people on beauty, the incidence rate of acne has obvious growth trend. Acne is a skin disease with a very high incidence rate, which is better at puberty, and according to a research in the United states, the incidence rate of acne in people of 12-24 years is 85 percent. Acne lesions contain a large number of propionibacterium acnes, which can produce enzymes that break down sebum, thereby causing alterations in the sebum composition. Such as triglycerides in sebum, can be broken down to produce glycerol and free fatty acids. Recent studies have shown that sebaceous gland cells themselves can also produce small amounts of free fatty acids. The generation of free fatty acid is increased in skin lesions of acne patients, so that excessive sebum overflow can be caused, the hair follicle mouth is abnormally keratinized, sebum is accumulated to form acne, the number of propionibacterium acnes secondary to hair follicles is increased sharply, sebaceous gland ducts are ruptured, inflammatory acne is caused, white comedo and black comedo appear, and great mental trouble is caused to patients.
At present, products for treating acne which are adopted on the market mainly comprise corticosteroids, chemical antibiotics, hormone antagonists, astringents, chemical bactericides, anti-inflammatory agents and the like, although inflammation can be eliminated in a short time, because pathogenic factors of acne cannot be fundamentally solved, secretion of sebaceous glands cannot be inhibited, hair follicle blockage cannot be fundamentally eliminated, symptoms and root causes are treated, acne usually recurs, and the phenomenon of repeated confusion occurs. In addition, pathogenic bacteria easily generate drug resistance to chemical bactericides, and the recurrent acne is more difficult to treat, forms intractable acne, and can be accompanied with uneven pox marks and pigmentation, thereby generating new and unaesthetic phenomena for the skin.

1. The diameter measurement result of each group of inhibition zone
The Propionibacterium acnes not only can decompose triglyceride in sebum, but also can generate a large amount of free fatty acid to stimulate hair follicles and peripheral tissues to generate inflammation, and can also induce monocytes and keratinocytes to secrete cell factors to cause or aggravate acne inflammation, so that the Propionibacterium acnes can repair skin lesions in a manner of inhibiting or killing the Propionibacterium acnes.
1) And preparing a culture medium: respectively weighing 33g of lecithin-Tween nutrient agar culture medium and 29.3g of liquid thioglycollate culture medium, adding 1L of ultrapure water, stirring, heating, boiling to dissolve completely, subpackaging, sterilizing at 120 deg.C under high pressure for 30min, cooling to 60 deg.C-70 deg.C, and keeping warm in a water bath for use. 2) Recovery of propionibacterium acnes and preparation of bacterial suspension: taking propionibacterium acnes freeze-dried powder, fusing an ampoule bottle under an alcohol lamp, adding 1mL of 0.85% physiological saline, uniformly stirring, inoculating onto 3 plates containing a culture medium, and culturing for 3-4 days under anaerobic conditions at 37 ℃. The slant colonies were scraped with an inoculating loop, and the suspension was diluted appropriately with physiological saline to a concentration of 106-. Taking the bacterial suspension under aseptic operation, and uniformly smearing the bacterial suspension on a culture medium by using a coating rod. Then, the Oxford cup is lightly placed on the surface of the solidified culture medium by using sterile forceps, and the Oxford cup (the outer diameter is 8mm) is lightly pressed to ensure good contact. Adding 200 mu L of test solution into an Oxford cup, culturing for 48h in an incubator at 37 ℃, taking out, and measuring the diameter of the effective bacteriostatic zone of the test substance in each culture dish. A matrix control group and a positive control group of 0.1% triclosan (DP300) are also provided. Each set of experiments was repeated twice, and samples were taken twice each time for testing. Antibacterial effect judgment standard: the high sensitivity is that the diameter of the bacteriostatic zone is more than 20 mm; the diameter of the inhibition zone is 10-20mm, and the drug is moderate-sensitivity; the diameter of the inhibition zone is less than 10mm, and the drug resistance is achieved.
The bacteriostatic effect of each group on propionibacterium acnes is shown in table 1
Group of | Zone of inhibitionDiameter/mm (X + -S, n ═ 4) |
Matrix control group | 8.2±0.1 |
EXAMPLE 1 group | 27.8±0.2 |
EXAMPLE 2 group | 33.6±0.1 |
0.1% DP300 Positive control | 32.5±1.0 |
As can be seen from Table 1, the diameter of the inhibition zone of the blank control matrix group is about 8mm, and the diameter of the inhibition zone of the positive control group added with 0.1% DP300 is 32.5mm, which indicates that the matrix control group has no interference to the inhibition test result, the groups of examples 1 and 2 have better inhibition effect on Propionibacterium acnes, and the inhibition effect of example 2 is stronger than that of example 1.
