背景技术
随着我国老年化进程的加速以及物质生活水平的不断丰富,人们越来越关注保健、健康以及如何应对衰老产生的问题。现代细胞生物学研究表明,衰老与氧化损伤有密切联系,也就是自由基对细胞和***的有害攻击;自由基氧化应激学说已越来越被人群接受,成为最有说服力的衰老学说之一,许多由老龄化引起的疾病,例如恶性肿瘤、自身免疫性疾病、白内障、视网膜病变等,其发病机制都与机体参与氧化应激有关,所以在维护健康和预防、治疗疾病方面,抗氧化剂的作用及从天然物质中寻找有效地、抗氧化化合物越来越受到人们的关注。
胶原蛋白具有三螺旋结构,生物相容性较好,可生物降解;牛骨通常含有12-48%的水分,13-26%的蛋白质、4-33%的脂肪以及21-48%的灰分;牛骨中含有丰富的蛋白质,通过先进加工手段以及深层次加工,可以生产出高附加值的功能性多肽、牛骨胶原蛋白等产品,牛骨胶原蛋白经进一步酶解、分离可以获得牛骨胶原蛋白肽粉,牛骨胶原蛋白肽粉具有分子量小,吸收快、脂肪含量低等优点,是保健食品、配方食品和化妆品生产加工的重要原料,经研究表明,牛骨胶原蛋白肽和一般胶原蛋白一样具有一定的免疫活性、能够预防和治疗骨质疏松、具备降血压、降血脂以及抗氧化等生物功能活性。

本发明主要目的是提供一种抗氧化效果好的含有复合胶原蛋白的固体冲剂,通过抗氧化活性成分的合理组合配伍,抗氧化保健功效突出,辅料的选择和配伍进一步丰富了营养,提高抗氧化功效,使得固体冲剂在口感、风味、营养保健功效、分散性和溶解性方面具有独特的优势,另外,加工步骤简单、生产成本低、便于工业化生产。
为了实现本发明的目的,本发明的技术方案如下:
一种含有复合胶原蛋白肽的抗氧化固体冲剂,其特征在于是由以下重量份的活性成分制备而成:牛骨胶原蛋白肽粉5-10份、海参胶原蛋白肽粉5-10份、大豆分离蛋白粉30-50份、大蒜粉10-15份、辅料50-70份。
作为优选,各原料的重量份配比为牛骨胶原蛋白肽粉5份、海参胶原蛋白肽粉10份,大豆分离蛋白粉40份、大蒜粉12份、辅料50份。
作为优选,各原料的重量份配比为牛骨胶原蛋白肽粉10份、海参胶原蛋白肽粉5份、大豆分离蛋白粉40份、大蒜粉12份、辅料70份。
作为优选,各原料的重量份配比为牛骨胶原蛋白肽粉8份、海参胶原蛋白肽粉8份、大豆分离蛋白粉40份、大蒜粉12份、辅料60份。
进一步地,辅料为麦芽糊精10-30份、木糖醇20-25份、苹果酸10-15份、抗结剂5-8份。
在其中一个实施例中,抗氧化胶原蛋白肽固体冲剂中,牛骨胶原蛋白肽粉的制备方法为:将市售牛骨胶原蛋白粉先按1∶10加入蒸馏水制成溶液,加入以1∶1复配的复合蛋白酶和风味蛋白酶液,加酶量为2000U/g条件下水解60-120min,至水解度为8-10%;高温灭酶10min,水解液经离心得上清液、经4KDA渗透膜过滤、浓缩、冻干得到牛骨胶原蛋白肽粉。
在其中一个实施例中,抗氧化胶原蛋白肽固体冲剂中,海参蛋白胶原蛋白肽粉的制备方法为:将市售刺参,清水浸泡脱除盐分,洗净后浸入0.8%HCl浸泡60-120min,捞出沥干水分、洗至中性,使用打浆机进行打浆处理,加入30-40倍体积的纯净水后采用胶体磨进行研磨细化处理,得浆状物;将上述浆状物加热至100℃后,保持10分钟,冷却至40℃,加入酸性蛋白酶2000u/g酶解2-6h,高温灭酶后收集上清液,加入复合蛋白酶2000U/g继续酶解2-4h,灭酶离心后得上清液、经4KDA渗透膜过滤、浓缩、冻干得到海参胶原蛋白肽粉。
本发明还提供了一种抗氧化蛋白肽组合物的制备方法,包括如下步骤:(1)按重量份称取各原料;(2)采用混合机将各原料混合均匀,使其变异系数小于5%;(3)将混合均匀的原料经过辐射杀菌、密封包装。
本发明含有复合胶原蛋白肽的抗氧化固体冲剂中组方原理和有益效果为:
(1)牛骨胶原蛋白肽粉、海参胶原蛋白肽均具有抗氧化作用,但是申请人发现通过将二者组合,在抗氧化功效方面具有协同增效的作用,组合以后的效果远远优于单一原料;试验证实,组合以后清除自由基能力明显提升,能显著降低血中脂质过氧化物(MDA)含量,提高血清超氧化物歧化酶(SOD)活力等。
(2)牛骨胶原蛋白肽粉、海参胶原蛋白肽均为动物源蛋白肽;而大豆分离蛋白作为植物源蛋白质,既可作为蛋白肽的载体,又补充了植物蛋白的营养,协同发挥蛋白肽粉的营养和抗氧化保健作用;
(3)大蒜粉中富含大蒜素,具有广谱抑菌功效,同时其还具有降脂、降血糖、防癌抗癌、提高免疫力等功效,同时大蒜粉具有较强的抗氧化功效和矫味功能,与复合胶原蛋白肽等组合,能在显著增强抗氧化功效的同时,改善胶原蛋白多肽带来的不良刺激;
(4)辅料的添加改善了冲剂的口感、分散性、稳定性、溶解性和抗结性能;具备固体冲剂优良的性能;
(5)生产加工方法简单易行、成本低、便于实现工业化生产。
具体实施方式
为了更好地理解本发明的本质,下面结合实施例对本发明优选实施方式进行详细说明。需要。需要理解的是以下实施例的给出仅是为了起到说明的目的,并不是用于对本发明的范围进行限制。本领域的技术人员在不背离本发明的宗旨和精神的情况下,可以对本发明进行各种修改和替换。
实施例1
一种含有复合胶原蛋白肽的抗氧化固体冲剂,由以下重量份的原料制备而成:牛骨胶原蛋白肽粉5-10份、海参胶原蛋白肽粉5-10份、大豆分离蛋白粉30-50份、大蒜粉10-15份、辅料50-70份;
辅料为麦芽糊精10-30份、木糖醇20-25份、苹果酸10-15份、抗结剂5-8份;
制备方法包括如下步骤:(1)按重量份称取各原料;(2)采用混合机将各原料混合均匀,使其变异系数小于5%;(3)将混合均匀的原料经过辐射杀菌、密封包装。
实施例2
一种含有复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉5份、海参胶原蛋白肽粉10份,大豆分离蛋白粉40份、大蒜粉12份、辅料50份;
辅料为麦芽糊精10份、木糖醇25份、苹果酸10份、抗结剂5份;
制备方法包括如下步骤:(1)按重量份称取各原料;(2)采用混合机将各原料混合均匀,使其变异系数小于5%;(3)将混合均匀的原料经过辐射杀菌、密封包装。
实施例3
一种含有复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉10份、海参胶原蛋白肽粉5份、大豆分离蛋白粉40份、大蒜粉12份、辅料70份。
辅料为麦芽糊精30份、木糖醇20份、苹果酸15份、抗结剂5份;
制备方法包括如下步骤:(1)按重量份称取各原料;(2)采用混合机将各原料混合均匀,使其变异系数小于5%;(3)将混合均匀的原料经过辐射杀菌、密封包装。
实施例4
一种含有复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉8份、海参胶原蛋白肽粉8份、大豆分离蛋白粉40份、大蒜粉12份、辅料60份;
辅料为麦芽糊精20份、木糖醇22份、苹果酸12份、抗结剂6份;
制备方法包括如下步骤:(1)按重量份称取各原料;(2)采用混合机将各原料混合均匀,使其变异系数小于5%;(3)将混合均匀的原料经过辐射杀菌、密封包装。
实施例5牛骨胶原蛋白肽粉的制备
牛骨胶原蛋白肽粉的制备方法为:将市售牛骨胶原蛋白粉先按1∶10加入蒸馏水制成溶液,加入以1∶1复配的复合蛋白酶和风味蛋白酶液,加酶量为2000U/g条件下水解60-120min,至水解度为8-10%;高温灭酶10min,水解液经离心得上清液、经4KDA渗透膜过滤、浓缩、冻干得到牛骨胶原蛋白肽粉。
实施例6海参胶原蛋白肽粉的制备
将市售刺参,清水浸泡脱除盐分,洗净后浸入0.8%HCl浸泡60-120min,捞出沥干水分、洗至中性,使用打浆机进行打浆处理,加入30-40倍体积的纯净水后采用胶体磨进行研磨细化处理,得浆状物;将上述浆状物加热至100℃后,保持10分钟,冷却至40℃,加入酸性蛋白酶2000u/g酶解2-6h,高温灭酶后收集上清液,加入复合蛋白酶2000U/g继续酶解2-4h,灭酶离心后得上清液、经4KDA渗透膜过滤、浓缩、冻干得到海参胶原蛋白肽粉。
实施例7
为了探讨本发明抗氧化胶原蛋白肽组合物在抗氧化方面的作用,按照常规实验方法进行了清除自由基以及小鼠过氧化脂质、对小鼠SOD活力的方面的实验。
(1)清除自由基
清除超氧阴离子自由基能力测定采用邻苯三酚自氧化法、清除羟基自由基能力的测定采用结晶紫法、DPPH自由基清除能力的测定同样采用常规方法。
组1:牛骨胶原蛋白肽溶液1.5%(m/v)
组2:海参胶原蛋白肽溶液1.5%(m/v)
组3:牛骨胶原蛋白肽溶液0.75%(m/v)+海参胶原蛋白肽溶液0.75%(m/v)
组4:牛骨胶原蛋白肽溶液0.5%(m/v)+海参胶原蛋白肽溶液1.0%(m/v)
组5:牛骨胶原蛋白肽溶液1.0%(m/v)+海参胶原蛋白肽溶液0.5%(m/v)

实验表明,组3-5中组合物在清除自由基能力方面能力与组1和2均存在显著性差异,说明将牛骨胶原蛋白肽粉与海参胶原蛋白肽粉组合以后在抗氧化方面具有协同功效。
(2)对脂质过氧化物与血清SOD的影响
昆明种小鼠60只,雌雄混半,体重为25-30克。实验前三天内小鼠不需要禁食,饲喂同一种饲料,同一饮水;实验前小鼠禁食12小时,自由饮水。随机将小鼠分为对照组、实验1组、实施2组、实验3组、实验4组、实验5组各10只。对照组每次灌水5毫升,实验1组每次灌组1的溶液5毫升,实验2组每次灌组2的溶液5毫升,实验3组每次灌组3的溶液5毫升,实验4组每次灌组4的溶液5毫升,实验5组每次灌组5的溶液5毫升,其他饮食均相同,连续30天。第31天,小鼠尾尖上采血测定MDA的含量,并摘眼球采血,离心,取血清,测定血清超氧化物歧化酶(SOD)活力和谷胱甘肽过氧化物酶。

实验发现,在服用本发明复合抗氧化胶原蛋白肽组合物1个月后,血清中SOD明显升高,同时脂质过氧化物的产物-丙二醛含量明显降低,这表明复合抗氧化胶原蛋白肽组合物比单一的组分抗氧化效果更好。

Claims (8)
1.一种含有复合胶原蛋白肽的抗氧化固体冲剂,其特征在于是由以下重量份的原料制备而成:牛骨胶原蛋白肽粉5-10份、海参胶原蛋白肽粉5-10份、大豆分离蛋白粉30-50份、大蒜粉10-15份、辅料50-70份。
2.根据权利要求1所述的复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉5份、海参胶原蛋白肽粉10份,大豆分离蛋白粉40份、大蒜粉12份、辅料50份。
3.根据权利要求1所述的复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉10份、海参胶原蛋白肽粉5份、大豆分离蛋白粉40份、大蒜粉12份、辅料70份。
4.根据权利要求1所述的复合胶原蛋白肽的抗氧化固体冲剂,所述各原料的重量份配比为牛骨胶原蛋白肽粉8份、海参胶原蛋白肽粉8份、大豆分离蛋白粉40份、大蒜粉12份、辅料60份。
5.根据权利要求1所述复合胶原蛋白肽抗氧化固体冲剂,所述辅料为麦芽糊精10-30份、木糖醇20-25份、苹果酸10-15份、抗结剂5-8份。
6.根据权利要求1所述复合胶原蛋白肽抗氧化固体冲剂,所述的牛骨胶原蛋白肽粉的制备方法为:将市售牛骨胶原蛋白粉先按1∶10加入蒸馏水制成溶液,加入以1∶1复配的复合蛋白酶和风味蛋白酶液,加酶量为2000U/g条件下水解60-120min,至水解度为8-10%;高温灭酶10min,水解液经离心得上清液、经4KDA渗透膜过滤、浓缩、冻干得到牛骨胶原蛋白肽粉。
