Abstract
本发明涉及一种抑制人结直肠癌细胞SW480增殖的红茶提取物及其应用,属于茶加工技术领域。该红茶提取物经如下步骤得到:红茶超微粉碎,用粉碎后的茶叶:沸水=1g:1‑200mL的比例浸提40分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经传统的浓缩、干燥后即得。细胞试验表明,此红茶提取物能显著抑制人结直肠癌细胞SW480的增殖,能作为爱吃红肉人士的健康伴侣,应用于食品和保健品中。

技术领域
本发明属于茶加工技术领域,具体的说,涉及一种抑制人结直肠癌细胞SW480增殖的红茶提取物及其应用。
背景技术
结直肠癌是世界常见的恶性肿瘤之一。30年来,我国结直肠癌发病率年均上升3%~4%。我国结直肠癌发病率和死亡例数分别占全世界发病和死亡总例数的18.6%和20.1%,均居第1位。根据国家癌症中心全国肿瘤登记数据报告,我国城市和农村地区结直肠癌发病率分别列所有恶性肿瘤的第3位及第5位,病死率分别居第4位和第5位。尽管当前结肠癌治疗方面涌出一些新的方法,如西妥昔单抗、贝伐单抗等分子靶向药物,但总体上来看结肠癌治疗效果仍然较差,尚无满意的非侵入性治疗方案。
红茶是世界生产和消费的主要茶类,其消费量高于绿茶和乌龙茶。研究表明红茶具有多种生物活性,如抗氧化、防止动脉粥样硬化、减肥、抗炎症、抗肿瘤及调节代谢综合征等。本发明以红茶为原料,制备一种具有抑制人结直肠癌细胞SW480增殖的红茶提取物。经文献检索,未见与本发明相同的公开报道。
发明内容
本发明是目的在于提供一种能抑制人结直肠癌细胞SW480增殖的红茶提取物及其应用,
为实现上述目的,本发明是通过以下技术方案实现的:
所述的红茶提取物,采用水浸提法制备而得,具体包括以下步骤:红茶超微粉碎,用粉碎后的茶叶:沸水=1g:1-200mL的比例浸提1-60分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经浓缩、干燥后即得红茶提取物。
进一步优选,所述干燥采用真空冷冻干燥。
进一步优选,所述干燥采用40-80℃条件下烘干处理。
红茶提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,所述红茶提取物为冻干粉或烘干粉。
红茶提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,所述红茶提取物是利用上述制备方法获得。
红茶提取物在食品和保健品中的应用,所述红茶提取物为冻干粉或烘干粉。

本发明的有益效果:该红茶提取物富含茶多酚、茶色素、蛋白质及多糖,能抑制人结直肠癌细胞SW480的增殖,可应用于食品和保健品中。
具体实施方式
为了使本发明的目的、技术方案和有益效果更加清楚,下面将对本发明的优选实施例进行详细的说明,以方便技术人员理解。
【实施例1】
红茶提取物,采用水浸提法制备而得,具体步骤如下:红茶超微粉碎,用粉碎后的茶叶:沸水=1g:10mL的比例浸提40分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液减压浓缩后,经真空冷冻干燥,得到红茶提取物(冻干)。
【实施例2】
红茶提取物,采用水浸提法制备而得,具体步骤如下:红茶超微粉碎,用粉碎后的茶叶:沸水=1g:10mL的比例浸提40分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液减压浓缩后,浓缩液在65℃下烘干,得到红茶提取物(烘干)。
实施分析
将实施例1、2得到的红茶提取物进行细胞试验用MTT法检测红茶提取物对人结直肠癌细胞SW480增殖的影响,其结果如下:从表1可看出,随着培养时间的延长,用不同浓度的红茶提取物(冻干或烘干)培养的人结直肠癌细胞SW480相对存活率均降低,表明红茶提取物能抑制人结直肠癌细胞SW480增殖。采用不同浓度的红茶提取物(冻干)培养SW480细胞,培养时间大于24h后才显示抑制癌细胞增殖的作用;而采用不同浓度的红茶提取物(烘干)培养SW480细胞,培养时间为24h时即显示抑制癌细胞增殖的作用。不同浓度的红茶提取物(冻干或烘干)对人结直肠癌细胞SW480增殖的抑制作用剂量效应关系不明显。

研究结果表明:该红茶提取物能抑制人结直肠癌细胞SW480的增殖,能直接加工成粉剂、胶囊、含片及饮料等,使用方便,便于携带;也能够作为添加剂用于食品中。

1.一种红茶提取物,其特征在于:其制备方法包括以下步骤:红茶超微粉碎,用粉碎后的茶叶:沸水=1g:1-200mL的比例浸提1-60分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经浓缩、干燥后,即得红茶提取物。
2.根据权利要求1所述的一种红茶提取物,其特征在于:所述干燥采用真空冷冻干燥。
3.根据权利要求1所述的一种红茶提取物,其特征在于:所述干燥采用40-80℃条件下烘干处理。
4.红茶提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,其特征在于:所述红茶提取物为冻干粉或烘干粉。
5.红茶提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,其特征在于:所述红茶提取物是利用权利要求1-3任一项所述的制备方法获得。
