Abstract

The invention relates to a black tea extract for inhibiting the proliferation of human colorectal cancer cells SW480 and application thereof, belonging to the technical field of tea processing.

Description

Black tea extract for inhibiting SW480 proliferation of human colorectal cancer cells and application thereof
Technical Field
The invention belongs to the technical field of tea processing, and particularly relates to a black tea extract for inhibiting the proliferation of human colorectal cancer cells SW480 and application thereof.
Background
Colorectal cancer is one of the common malignant tumors in the world. The incidence rate of colorectal cancer in China has increased by 3 to 4 percent in every 30 years. The morbidity and the mortality of colorectal cancer in China respectively account for 18.6 percent and 20.1 percent of the total morbidity and mortality in the world, and are in the 1 st level. According to the national tumor registration data report of the national cancer center, the colorectal cancer incidence rates of cities and rural areas of China are respectively 3 rd and 5 th of all malignant tumors, and the disease death rates of the colorectal cancer are respectively 4 th and 5 th. Although some new methods, such as cetuximab, bevacizumab and other molecular targeting drugs, emerge from the aspect of colon cancer treatment at present, the colon cancer treatment effect is still poor overall, and no satisfactory non-invasive treatment scheme exists.
Black tea is the major tea species produced and consumed in the world, and is consumed in higher amounts than green tea and oolong tea. Researches show that the black tea has various biological activities, such as antioxidation, atherosclerosis prevention, weight reduction, inflammation resistance, tumor resistance, metabolic syndrome regulation and the like. The invention takes black tea as a raw material to prepare a black tea extract capable of inhibiting the proliferation of human colorectal cancer cells SW 480. Through the literature search, the same public reports as the invention are not found.
Disclosure of Invention
The invention aims to provide a black tea extract capable of inhibiting the proliferation of human colorectal cancer cells SW480 and application thereof,
in order to realize the purpose, the invention is realized by the following technical scheme:
the black tea extract is prepared by a water extraction method, and specifically comprises the following steps of carrying out superfine grinding on black tea, extracting the ground tea for 1-60 minutes by using a proportion of boiling water to 1g to 1-200m L, filtering to obtain filtrate, repeating the extraction step for 2 times, combining the filtrates, concentrating and drying the filtrate to obtain the black tea extract.
Further preferably, the drying is vacuum freeze drying.
Further preferably, the drying is carried out by drying at 40-80 ℃.
The black tea extract is applied to products for inhibiting the proliferation of human colorectal cancer cells SW480, and the black tea extract is freeze-dried powder or dried powder.
Application of black tea extract in products for inhibiting SW480 proliferation of human colorectal cancer cells, wherein the black tea extract is obtained by the preparation method.
The black tea extract is an application in food and health care products, and the black tea extract is freeze-dried powder or dried powder.
Use of a black tea extract in food and health care products, the black tea extract being obtained by the above preparation method.
The invention has the beneficial effects that: the black tea extract is rich in tea polyphenols, tea pigment, protein and polysaccharide, can inhibit proliferation of human colorectal cancer cell SW480, and can be applied in food and health product.
Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, preferred embodiments of the present invention will be described in detail below to facilitate understanding of the skilled person.
[ example 1 ]
A black tea extract is prepared by water extraction method, which comprises micronizing black tea, extracting with pulverized tea at boiling water ratio of 1g to 10m L for 40 min, filtering to obtain filtrate, repeating the above extraction steps for 2 times, mixing filtrates, concentrating the filtrate under reduced pressure, and vacuum freeze drying to obtain black tea extract (lyophilized).
[ example 2 ]
A black tea extract is prepared by water extraction method, which comprises micronizing black tea, extracting with pulverized tea at boiling water ratio of 1g to 10m L for 40 min, filtering to obtain filtrate, repeating the above extraction steps for 2 times, mixing filtrates, concentrating the filtrate under reduced pressure, and oven drying the concentrated solution at 65 deg.C to obtain black tea extract (oven drying).
Carrying out the analysis
The black tea extracts obtained in examples 1 and 2 were subjected to cell test to examine the effect of the black tea extracts on the proliferation of human colorectal cancer cells SW480 by the MTT method, and the results were as follows: as can be seen from table 1, the relative survival rates of human colorectal cancer cells SW480 cultured with different concentrations of black tea extract (lyophilized or oven dried) were all decreased with the increase of the culture time, indicating that the black tea extract can inhibit the proliferation of human colorectal cancer cells SW 480. SW480 cells are cultured by black tea extracts (freeze-dried) with different concentrations, and the effect of inhibiting cancer cell proliferation is displayed only after the culture time is longer than 24 hours; SW480 cells were cultured with black tea extract (oven dried) at different concentrations for 24h to show the effect of inhibiting cancer cell proliferation. The dose effect relationship of the black tea extracts (freeze-dried or oven-dried) with different concentrations on the inhibition effect on the proliferation of human colorectal cancer cells SW480 is not obvious.
TABLE 1 Effect of Black tea extract on the relative survival of human colorectal cancer cells SW480

Note: the difference is not significant when capital letters are marked with the same shoulder in the same row (P >0.05), and the difference is significant when capital letters are marked with different shoulders in the same row (P < 0.05); the difference is not significant when the lower case letters in the same row are marked with the same shoulder mark (P >0.05), and the difference is significant when the lower case letters in the same row are marked with different shoulders (P < 0.05).
The research result shows that: the black tea extract can inhibit proliferation of human colorectal cancer cells SW480, can be directly processed into powder, capsule, buccal tablet, beverage, etc., and is convenient for use and carrying; can also be used as additive in food.
Finally, it is noted that the above-mentioned preferred embodiments illustrate rather than limit the invention, and that, although the invention has been described in detail with reference to the above-mentioned preferred embodiments, it will be understood by those skilled in the art that various changes in form and detail may be made therein without departing from the scope of the invention as defined by the appended claims.
