Description
Technical Field
The invention belongs to the technical field of ancient botany experimental tools in ancient biology specialties, and particularly relates to a replaceable phytolithic epidermis stripping needle.
Background
Ancient botany is a branch of ancient biology and is the science of studying the plant kingdom during the period of local history. The basis of ancient botany is the identification of phytolith, and the species of the phytolith can be determined for further research. Plant fossil identification is mainly determined by combining plant macroscopic characteristics (such as size, leaf vein number, leaf margin characteristics, leaf division condition and the like) and microscopic characteristics (including cell shape and size, cell wall characteristics, pore number, pore composition characteristics, upper epidermis and lower epidermis differences and the like) and comparing the characteristics with known species. Macroscopic feature observation is mainly performed through a magnifying glass or a solid microscope.
The microscopic characteristics need to be completed through ancient plant epidermis experiments, and the current general experimental method comprises the steps of taking down the plant fossil epidermis from the surrounding rock, removing the surrounding rock by HCI and HF, oxidizing the epidermis by concentrated nitric acid and saturated potassium chlorate, soaking the epidermis in ammonia water for a short time, separating the upper epidermis and the lower epidermis of the plant fossil by using a tool in the soaking process, removing residual organic matters in the upper epidermis and the lower epidermis, cleaning, dividing the epidermis according to the required size, and observing the epidermis under an optical microscope or a scanning electron microscope to achieve the research purpose.
The epidermis for the experiments is usually very small, a few millimeters or less, so the exfoliation process must be performed under a solid microscope. Because the phytolithic fossils are preserved in rocks for millions of years, the upper epidermis and the lower epidermis of plant epidermal tissues are very tight under the action of long-time compaction, and the epidermis stripping of the phytolithic fossils is difficult work.
And at present, no tool specially used for peeling the plant fossil epidermis is provided, and the peeling is mostly finished by means of a dissecting needle and a bamboo stick. The stripping step is usually to hold the bamboo stick with the left hand and press one side of the epidermis, and the right hand strips the upper and lower epidermis with a dissecting needle, and then remove the residual organic matter therein. The bamboo sticks are not acid and alkali resistant, can cause certain pollution to samples and are seriously consumed. The existing dissecting needle is usually long, is not beneficial to being used under a microscope, is difficult to operate when being used for removing residues on the upper epidermis and the lower epidermis, and can only be realized by repeated washing. The experiment effect is not good, the operation is difficult, and the experiment time is wasted.
In view of the above, it is very necessary to invent a replaceable plant fossil epidermolysis needle.
