Description

Composition for resisting propionibacterium acnes and regulating skin microecology and application
Technical Field
The invention belongs to the technical field of medical biology, and particularly relates to a composition for resisting propionibacterium acnes and regulating skin microecology and application thereof.
Background
The skin is the largest organ of the human body, part of microorganisms inhabited on the skin provide ecological protection for the skin, and skin diseases such as the overgrowth of propionibacterium acnes can be caused when the skin flora is unbalanced, so that the skin is acnes. Propionibacterium acnes is a multi-drug resistant bacterium, the growth of which is easy to generate a bacterial film, is one of skin invasive factors, and is easier to grow anaerobically and resist the action of antibiotics. In general, drugs such as antibiotics and hormones are mostly adopted for treating acne, but the effect is not good.
The skin is an ecosystem consisting of niches (micro-ecosystems) in different locations. About one billion bacteria are symbiotic on the surface of the skin per square centimeter, forming a microecological "shield". Most skin infectious diseases can use a microecological method to adjust normal flora of the skin, promote microecological balance, improve microenvironment (ecological niche), adjust skin barriers, consolidate the planting force of the normal flora, balance the flora number and conform to the microecological law, thereby achieving the purposes of treating and preventing diseases. In recent years the skin micro-ecology has become a hot spot in the skin field, and a new consensus has also been formed: the skin barrier, the skin microecology and the skin immunity jointly form a first defense system of an organism, resist the stimulation of various external factors and maintain the normal functions of the skin and the organism. The prior art also lacks drugs that effectively regulate the skin's micro-ecology.
Therefore, it is desirable to provide a composition for regulating skin micro-ecology against propionibacterium acnes with good effects.

Application example 1: example 2 Regulation of skin Microecology by the compositions prepared
2 patients with facial acne were selected, 1 in each of men and women, with a course of 1-3 years, and after cleansing the face every morning and evening, an aqueous solution containing 10% of the composition prepared in example 2 was used for 4 weeks continuously. In addition, 1 case of each of the healthy control group 2, male and female was set.
Applying a cotton swab in a sterilized phosphate buffer solution (PBS, the same below) to an acne skin lesion area (about 4cm multiplied by 4cm) on a cleaning face for more than 4 hours from inside to outside with slight force, immediately shearing off a cotton swab head by using a sterile scissors after collection, placing the cotton swab head into a test tube containing 5mL of sterile conveying liquid, screwing down the cotton swab head by using a screw cap, taking the cotton swab head back to a laboratory, and collecting the treated acne patient in the skin lesion improvement area by the method; healthy adult specimens were collected at the center of the forehead as above. The staphylococcus epidermidis and propionibacterium acnes which are more than the total number of normal skin colonies on the skin of an acne patient are taken as representative flora, the flora is counted, and the facial pH value of a tested person is tested (the pH value of the normal skin is 5.0-7.0), and the experimental results are shown in the following table:
table 4: counting result and pH value of viable bacteria of facial skin flora
Staphylococcus epidermidis Propionibacterium acnes pH value
Before treatment 4.36 4.96 4.84
After treatment 3.22 3.52 5.55
Healthy controls 3.38 3.69 5.60
Note: the number in the table is the viable bacteria technique per square centimeter of skin, namely CFU/ML/cm2And is indicated by log 10.
Table 4 shows the viable count results and pH values of facial flora, and after 2 weeks using the aqueous solution containing 10% of the composition prepared in example 2, the population numbers of staphylococcus epidermidis and propionibacterium acnes on the face of acne patients were significantly decreased and restored to normal levels, and the facial pH values tended to normal levels, which indicates that the composition prepared in example 2 of the present invention has the effect of regulating skin micro-ecology.
The subject has no skin irritation and no local and systemic adverse reaction.
