Sleep is one of important contents in daily life of people, and the sleep time is about more than 3 times in a life of a person. A study by the world health organization has shown that about 27% of people worldwide suffer from sleep disorders, about 35.2% of people in western countries have varying degrees of insomnia, and the incidence of insomnia in chinese adults is 38.2%. Insomnia affects the life and work of patients to different degrees, and causes the decline of the attention, judgment, memory and daily working ability of the patients, and causes depression, anxiety or fear of the psychology of the patients. In addition, insomnia is also an independent risk factor for coronary heart disease, diabetes, and the like.
There are many ways to improve insomnia, such as taking hypnotics. However, the current drugs for clinically treating insomnia mainly comprise melatonin receptor agonists, benzodiazepine receptor agonists, antidepressant drugs with hypnotic effect and the like, but the drugs usually have strong side effects such as dizziness, reaction retardation or somnolence after being taken for a long time.
The traditional Chinese medicine aromatherapy has four characteristics of accuracy, uniqueness, quick effect and naturalness, and has three main effects of treating diseases, synergistic effect of treating diseases and rehabilitation and conditioning core. The theory of traditional Chinese medicine indicates that aromatic drugs permeate into human body through skin, channels and collaterals and other ways, and play a role in opening and relaxing striae, promoting blood circulation to remove blood stasis, and smoothing and balancing yin and yang. The fragrant medicines in the traditional Chinese medicine aromatherapy mainly utilize the volatile substances to achieve the purpose of preventing and treating diseases. At present, rose, lavender, bergamot and chamomile are taken as main aromatic essential oils for treating insomnia in western countries, and although some aromatic traditional Chinese medicines or essential oils are used for treating insomnia by an inhalation method at present, reasonable compatibility under the guidance of the traditional Chinese medicine theory is lacked, so the invention provides the composition for regulating sleep, the compound essential oil and the application thereof, wherein the reasonable compatibility is under the guidance of the traditional Chinese medicine theory.

Animal behavioral testing experiment:
experimental animals: 72 SPF-class S-day rats weighing 180-220 g are normally bred in a normal-temperature quiet animal environment.
Grouping experiments: after 7 days of adaptive feeding, the rats were randomly divided into 9 groups of 8 rats each with half of male and female according to body weight, including a blank group, a model group, a positive group, 3 administration groups and 3 control groups.
Establishing a model: the blank group had no any external stimulation, and the other groups had their forelimbs restrained by medical adhesive tape to restrict their movement, and kept holding the rat tail every day, which was located at the position away from the proximal end 1/3 for 45 minutes every day, and kept in a fighting state for 20 consecutive days, with no bleeding on the skin.
Administration: the rats in the model group and each administration group were injected with 450mg/kg of p-chlorophenylalanine (PCPA) dissolved in physiological saline for 2 consecutive days from the model establishment day 16. From the 7 th day of tail clamping stimulation, the positive group is perfused with 1mg/kg diazepam solution dissolved by normal saline; perfusing normal saline with the same volume as the diazepam solution into the stomach in the blank group and the model group; aromatherapy was given once daily for 14 consecutive days to 3 dosing groups and 3 control groups. Specifically, the compound essential oils of examples 1 to 3 were administered to three groups of administration groups for aromatherapy inhalation. The aromatherapy sniffing method is used by an aromatherapy sniffing device, and the aromatherapy sniffing device main body consists of a large transparent organic glass box with the specification of 80 x 65cm and a cover and four small transparent organic glass boxes with the specification of 30 x 30 cm. The small boxes are evenly placed at the bottom of the large box, and the aromatherapy atomizer used for containing essential oil is placed in the center of the large box. Rats were placed in the aromatherapy sniffing device, and the compound essential oil provided in examples 1-3 was added to the spray pot of the aromatherapy atomizer, respectively, with sniffing time of 1h per group. And testing after the sniffing is finished.
Index detection: the rat open field test was performed within 1 hour after the end of the dosing. The light of the laboratory is kept constant and dim, the lowest brightness at a distance of 1.5cm for distinguishing the fine activities of the rats is taken as the standard, the room temperature is 20 +/-1 ℃, the humidity is 60-70%, and the laboratory is kept quiet. The bottom surface of the open field is black, and a camera is arranged right above the central area of the open field. The rat is placed in a laboratory to adapt for 10 minutes, the rat is rapidly placed in the center of an open field after the beginning of the experiment, the head of the rat is fixed towards one side, a video analysis system is started immediately after the rat is released, and the video is automatically collected. The adaptability of each rat in the open field box is measured for 3 minutes, the measurement time is 5 minutes, fecal particles and urine left by the rat are removed after the measurement is finished, the environment in the box is cleaned by low-concentration alcohol, and the next 1 rat is carried out after the alcohol is volatilized and smells less. Taking less than 2cm/s as rest, taking more than 20cm/s as fast movement, and measuring by adopting a single-target tracking mode, and recording the maximum speed, the average speed, the movement speed, the rest time, the low-speed movement time and the high-speed movement time of each rat.
After the treatment of each group of rats is finished, the rat is killed by cutting off the head, the hypothalamus and hippocampus tissues of the rat are quickly taken out, the rat is weighed after being washed by phosphate buffer solution, the phosphate buffer solution with 9 times of volume is added, and an electric homogenizer is used for homogenate. Centrifuging at 4 deg.C for 10 min at 3500 r/min, collecting supernatant, and storing at-20 deg.C. The enzyme-linked immunosorbent assay is adopted to strictly operate according to the instruction of a corresponding kit, and the content of 5-hydroxytryptamine (also known as serotonin, 5-HT for short), glutamic acid (Glutamic acid for short), gamma-aminobutyric acid (GABA for short) and the ratio of glutamic acid to gamma-aminobutyric acid (Glu/GABA) in hypothalamus and hippocampus tissues of each group of rats is determined.
Wherein, 5-HT, Glu and GABA are all neurotransmitters. Specifically, 5-HT has functions of regulating sleep and maintaining mental stability. GABA is an inhibitory transmitter, maintains the balance of excitation inhibition in brain, and hypofunction can cause insufficient inhibition function in brain, and causes headache, anxiety, nervousness, irritability and other conditions. GLu is involved in higher functions of the brain, glutamate plays an important role in learning, memory, neuronal plasticity and brain development, and GLu together with GABA regulates the function of other neurotransmitters. That is, the levels of 5-HT, Glu and GABA in the hypothalamus and hippocampus tissues can reflect the mental and neurological status of the organism.

As can be seen from Table 1, compared with the blank group, the model group and the positive group, the examples 1 to 3 in the administration group significantly reduce the moving distance, the maximum speed and the average speed of the rat in the open field experiment and simultaneously increase the rest time of the rat in the open field experiment, and the examples and the corresponding comparative examples show that the effect of the examples is better than that of the comparative examples, namely the compound essential oil provided by the invention can reduce the moving distance, the maximum speed and the average speed of the rat in the open field experiment and increase the rest time of the rat in the open field experiment, and has more obvious effect compared with the control group only containing the agilawood essential oil and the tangerine peel essential oil.
Table 2 shows the statistics of the 5-HT, Glu, GABA and Glu/GABA content in the hypothalamus and hippocampus tissues of each group of rats: (
