The invention extracts honeysuckle, chrysanthemum, dandelion, sterculia seed and medlar respectively, adds the extracts of all the components into auxiliary materials, mixes the extracts and prepares the granular tea. According to the characteristics of the components, the extraction and concentration temperatures (the concentration temperature of honeysuckle and chrysanthemum extracting solution is 55-60 ℃, the concentration temperature of dandelion extracting solution is 50-55 ℃, and the concentration temperature of boat-fruited sterculia seed and medlar extracting solution is 45-50 ℃) are respectively controlled, so that the active ingredients or the active parts are prevented from being heated unstably for a long time to cause chemical structure damage. Meanwhile, in the extraction process of the active ingredients of each substance, the material-liquid ratio is determined according to the viscosity of the extracting solution when each component is extracted, and when root medicinal materials and medicinal materials containing more polysaccharide are extracted by water, if the extracting solution is less, the extraction is incomplete, and in addition, the filtration and the concentration are difficult; if the amount of the extract is too large, the extraction rate is not greatly improved, and energy is wasted during concentration.
Through the technical scheme, compared with the prior art, the invention has the following advantages:
(1) directly acts on throat, is beneficial to the direct absorption of mucosa, immediately enters the submucosa, directly participates in blood circulation, quickly plays the effects of clearing heat, cooling blood, purging fire for removing toxin, nourishing yin and moistening throat and achieves the purposes of clearing away the lung-heat and stomach-dryness fire and relieving sore throat;
(2) the preparation method is simple, and has lower requirements on process equipment;
(3) compared with similar products, the product has the characteristics of convenient carrying, easy drinking and the like, and can be drunk in any occasions;
(4) the health-care tea has the advantages of small dosage, good curative effect, small toxic and side effects, convenience in use, low price and easiness in drinking, has the health-care effect in daily drinking, and is suitable for various crowds.
Detailed Description
The technical solutions in the embodiments of the present invention will be clearly and completely described below, and it is obvious that the described embodiments are only a part of the embodiments of the present invention, and not all embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example 1
(1) Mixing honeysuckle and drinking water according to a mass material-liquid ratio of 1:20, soaking for 2h, heating and leaching at 90 ℃, extracting twice, extracting for 2h for the first time and extracting for 1.5h for the second time, filtering while hot, combining extracting solutions of the two times, centrifuging (25 ℃, 4000r/min and 10min), and concentrating under reduced pressure (steam pressure of 0.2MPa and temperature of 58 ℃) to obtain a honeysuckle extract with a relative density (the reference ratio is the density of water) of 1.3(60 ℃).
(2) Mixing chrysanthemum and drinking water according to a mass material-liquid ratio of 1:25, soaking for 2h, heating and leaching at 90 ℃, extracting twice, extracting for 2h for the first time and extracting for 1.5h for the second time, filtering while hot, combining extracting solutions of the two times, centrifuging (25 ℃, 4000r/min and 10min), and concentrating under reduced pressure (steam pressure of 0.2MPa and temperature of 58 ℃) to obtain chrysanthemum extract with relative density (the reference ratio is the density of water) of 1.3(60 ℃).
(3) Mixing herba Taraxaci with drinking water at a mass ratio of 1:15, soaking for 2h, heating and leaching at 90 deg.C, extracting twice, extracting for 2h for the first time and 1.5h for the second time, filtering while hot, mixing the two extractive solutions, centrifuging (25 deg.C, 4000r/min, 10min), concentrating under reduced pressure (steam pressure 0.2MPa, temperature 53 deg.C) to obtain herba Taraxaci extract with relative density (water density) of 1.3(60 deg.C).
(4) Mixing semen Scaphii Lychnophori and drinking water at a mass ratio of 1:20, soaking for 2 hr, heating and leaching at 90 deg.C, extracting twice, the first extraction for 2 hr and the second extraction for 1.5 hr, filtering while hot, mixing the two extractive solutions, centrifuging (25 deg.C, 4000r/min, 10min), concentrating under reduced pressure (steam pressure of 0.2MPa, temperature of 50 deg.C) to obtain semen Scaphii Lychnophori extract with relative density (water density) of 1.3(60 deg.C).
(5) Mixing fructus Lycii and drinking water at a mass ratio of 1:20, soaking for 3 hr, heating and leaching at 80 deg.C, extracting twice, the first extraction for 2 hr and the second extraction for 1.5 hr, filtering while hot, mixing the two extractive solutions, centrifuging (20 deg.C, 4000r/min, 10min), concentrating under reduced pressure (steam pressure of 0.2MPa, temperature of 50 deg.C) to obtain fructus Lycii extract with relative density (water density of reference ratio) of 1.3(60 deg.C).
(6) Weighing 12g of maltodextrin, 60g of soluble starch, 0.01g of natural menthol, 1g of sodium carboxymethylcellulose, 0.1g of potassium sorbate and 0.1g of sodium citrate, and uniformly mixing to obtain auxiliary materials for later use.
(7) Weighing 4g of honeysuckle extract, 5g of chrysanthemum extract, 4g of dandelion extract, 5g of boat-fruited sterculia seed extract and 8g of medlar extract to obtain main medicine components for later use.
(8) Adding the main medicine components prepared in the step (7) into 74g of auxiliary materials, uniformly mixing the main medicine components, preparing into a held mass and soft materials which can be scattered by light pressing, extruding, sieving by a 16-mesh sieve to prepare into granular substances, drying the prepared granular substances in a drying box at 65 ℃ for 70min, collecting granules, sieving by a medicine detection sieve (the granules which pass through a No. 1 sieve but cannot pass through a No. 5 sieve), collecting and bagging.
The medicine taking method comprises the following steps: it is taken with warm water twice a day, 1.5g each time, and 15 days is a treatment course.
Example 2
In the step (6), the auxiliary materials comprise the following components: 12g of maltodextrin, 57g of soluble starch, 0.03g of natural menthol, 0.67g of sodium carboxymethyl cellulose, 0.1g of potassium sorbate and 0.2g of sodium citrate.
The main medicine components in the step (7) are as follows: 5g of honeysuckle extract, 5g of chrysanthemum extract, 5g of dandelion extract, 5g of boat-fruited sterculia seed extract and 10g of medlar extract.

The results in table 1 show that the granular tea prepared by the technical scheme of the invention is superior to the comparative examples in the aspects of forming rate, moisture absorption rate and dissolubility, and the formula and the process of the invention are reasonable.
Experiment 2 efficacy of in vitro simulated treatment of chronic pharyngitis
The bacterial detection rate of chronic pharyngitis in clinic is as follows: 54.4% of beta hemolytic streptococcus, 23.3% of staphylococcus aureus, 28.8% of klebsiella pneumoniae, 22.8% of staphylococcus epidermidis and the like. Therefore, the experiment examines the different degrees of inhibition effects of the honeysuckle extract, the chrysanthemum extract, the boat-fruited sterculia seed extract, the dandelion extract, the medlar extract and the compatible traditional Chinese medicine combined granulated tea in the embodiment 6 of the invention on the clinically common pathogenic microorganism bacterial suspension.
Subject: honeysuckle extract, chrysanthemum extract, sterculia scaphigera extract, dandelion extract, medlar extract and the granulated tea prepared in example 6, which are prepared in example 1;
detecting an object: minimum Inhibitory Concentrations (MIC) of Streptococcus hemolyticus B, Staphylococcus aureus, Klebsiella pneumoniae, and Staphylococcus epidermidis;

And (4) observing results: and (5) performing a plate streaking experiment, culturing for 24h, and observing the result to determine the minimum inhibitory concentration.

As can be seen from the results in Table 2, the positive control bacterial suspensions of each group grew well, and the negative control bacterial suspensions grew aseptically; the extract extracts have different degrees of inhibition effects on beta hemolytic streptococcus, staphylococcus aureus, klebsiella pneumoniae and staphylococcus epidermidis, and the minimum inhibition concentration of the traditional Chinese medicine composite granular tea in the example 6 is low, namely the dilution factor is maximum, compared with the extract extracts, so that the synergistic effect is achieved through reasonable proportion. The traditional Chinese medicine composite particle tea has broad-spectrum antibacterial effect.
Experiment 3 animal experiments
Subject: the SD rats weighing 180-200 g are 40 males and divided into 4 groups, and each group comprises 10 males;

The experimental method comprises the following steps: the rats are bred in cages in groups, water is freely drunk, administration is carried out for 15 days continuously, blood is taken from femoral artery to an Ep tube on the next day after the last administration, and then the rats in each group are killed by intraperitoneal injection of anesthetic.
The detection indexes and the method are as follows: measuring the content levels of Prostaglandin (PGE), interleukin-1 beta (1L-1 beta), tumor necrosis factor (TNF-alpha) by radioimmunoassay, and using the method according to the kit instructions;
the analysis method comprises the following steps: statistical analysis was performed using SPSS 22.0 statistical software, with the results expressed as mean ± standard deviation (x ± s), and with comparison using LSD-t test, significant differences were found when P <0.05 and very significant differences were found when P < 0.01.
The results of the experiment are shown in table 3.

As shown in Table 3, PGE, 1L-1. beta. and TNF-. alpha.were significantly increased in the model control group compared with the blank control group, indicating that the molding was successful (P < 0.05). Compared with a blank control group, the content of the serum of the model control group is obviously increased, and the statistical significance is achieved, and compared with the model control group, the content of PGE, 1L-1 beta and TNF-alpha (P is less than 0.01) in the serum can be obviously reduced by the traditional Chinese medicine combined granular tea group; compared with the sarcandra glabra buccal tablet, the traditional Chinese medicine granule tea group is superior to the sarcandra glabra buccal tablet group. Therefore, the traditional Chinese medicine composite granular tea prepared by the invention has good anti-inflammatory effect on chronic pharyngitis.
Experiment 4 clinical experiment
First, general data of the experimental subjects: among 180 patients with chronic pharyngitis, 83 men and 97 women are aged 18-67 years old.
Clinical manifestations of the patients: pharyngeal discomfort, foreign body sensation, itching, burning sensation, dryness or irritation, with cough and nausea; the throat part is checked to see that mucous membrane is diffusely congested, dilated, dark red, accompanied by a small amount of viscous secretion; leucocytes and a small amount of lymphocytes infiltrate the blood vessels under the mucosa and around the mucosal glands, even the lymph follicles swell, often accompanied by yellowish white punctate exudates.
The treatment group and the control group are randomly divided, the treatment group comprises 90 cases, the control group comprises 90 cases, and the age, clinical condition and the like of the two groups have no obvious difference and are comparable.
Second, therapeutic methods
1. Control group: the compound sarcandra glabra buccal tablet is taken orally.
2. Treatment groups: the Chinese medicinal granule tea of the embodiment 6 of the invention is taken after being mixed with water.
Thirdly, usage and dosage:
1. control group
The usage and dosage are as follows: it is administered orally 2 tablets (small tablets) at a time, every 2 hr, 6 times daily.
2. Treatment group
The application method and the dosage are as follows: the preparation is taken twice a day, one dose is taken each time, and each dose contains 1.5g of the medicine, and is taken with warm water.
Fourthly, a course of treatment:
the course of treatment is as follows: 15 days is a course of treatment, and three courses of treatment are taken.
Fifth, observe the index
The improvement in clinical symptoms was recorded before and after treatment.
Sixthly, treatment course evaluation standard:
the main indexes for examining the normal chronic pharyngitis are as follows: the pharyngeal portion is checked to see that mucous membrane has no phenomena of diffuse hyperemia, vasodilatation and dark red color, and the viscous secretion disappears; there were no leukocytes and no lymphocytes infiltrating around the submucosal vessels and mucosal glands, the lymphoid follicles recovered normal, and the yellowish-white punctate exudates disappeared.
