the preparation procedure is as in example 1.
First, physicochemical test
The physical and chemical tests of the blue light-resistant repair stock solutions prepared in examples 1 to 3 and comparative examples 1 to 5 were carried out, respectively, and the results were as follows:
1. appearance: the stock solution of comparative example 1 was a colorless transparent viscous liquid, and the remaining stock solution was a yellowish transparent viscous liquid.
2. pH value: the stock solutions of examples 1-3 and comparative examples 1-5 all had a pH range between 5.5 and 7.5.
3. Heat resistance stability test (48 ℃ C.. times.72 h): all stock solutions were qualified for heat stability.
4. Cold resistance stability test (-15 ℃ C.. times.72 h): cold resistance stability all stock solutions were acceptable.
Second, safety and efficacy testing
(1) Testing the human body safety patch:
the experimental steps are as follows: the selected area is not more than 50mm2And a qualified patch device with the depth of about 1mm, wherein a patch test is carried out on the back of 30 volunteers, the stock solution samples prepared in examples 1-3 and comparative examples 1-5 and a blank control sample (purified water) are respectively placed on the patch device in sequence, 0.02mL of the samples of examples 1-3 and comparative examples 1-5 is selected, the purified water is placed in a control hole to serve as a blank control sample, then a low-sensitization adhesive tape is used for being attached to the back of each tester for testing, and the test is carried out by lightly pressing the palm of the hand to be uniformly attached to the skin for 24 hours.
The experimental results are as follows: after the test is carried out for 24 hours, 30 minutes (after the pressure mark disappears) after the spot pasting device on the back of the subject is removed, the back skin of the subject is observed to have no red or stabbing pain phenomenon or suspected erythema reaction, and the result shows that the stock solution containing the blue light resisting repair composition is safe and has no adverse reaction phenomenon.
(2) Evaluation test of blue light resistance:
test samples: stock samples prepared in examples 1-3 and comparative examples 1-5, incubators with blue light source, culture medium containing human keratinocytes, opsin-3 fluorescent marker, light microscope.
The experimental steps are as follows: 8 culture mediums containing human keratinocytes are selected and numbered respectively, then the same amount of stock solution samples prepared in examples 1-3 and comparative examples 1-5 are added in sequence, the same amount of opsin (opsin-3) fluorescent marker is added, finally the mixture is placed in an incubator with a blue light source for culturing for 24 hours, the culture medium is taken out after 24 hours, and the density of fluorescence in the culture medium is observed on a microscope.
The experimental results are as follows: the fluorescence intensity of the culture medium of the stock solution samples of examples 1-3 is reduced in sequence, and the fluorescence density of example 1 is the maximum and occupies 93 percent of the area of the culture medium; the fluorescence density of the medium of example 2 was 73%; the fluorescence density of the medium of example 3 was 65%; the fluorescence density range of comparative example 1 was the smallest, there was no fluorescence range at all, and the fluorescence density was 0%; the fluorescence density of the medium of comparative example 2 was 52%; the fluorescence densities of the media of comparative example 3 and comparative example 4 were very close, about 2%, and the fluorescence density was very weak, and the fluorescence density of the media of comparative example 5 was 59%. Since human keratinocytes secrete a protein called opsin (opsin-3) and decrease or even do not secrete the protein under the irradiation of blue light, if the added sample has no efficacy of resisting blue light, the opsin (opsin-3) is decreased or even not, and the protein can be completely seen by a fluorescence labeling method. From the above results, the blue light resisting efficacy of the stock solution samples in examples 1 to 3 is obviously better than that of the stock solution samples in comparative examples 1 to 5, and the blue light resisting efficacy of the samples in examples 1 to 3 shows a trend of being weaker and weaker with the decrease of the concentration of the sophora japonica seed extract, so that the blue light resisting repair composition provided by the invention has a synergistic blue light resisting repair effect by the combined application of the sophora japonica seed extract, the centella asiatica extract and the selaginella tamariscina extract.
(3) Lactic acid sting repair efficacy test:
test samples: stock samples prepared in examples 1-3 and comparative examples 1-5, 10% v/v lactic acid solution.
Subject: 8 healthy skin volunteers between 30 and 45 years of age were selected.
The experimental steps are as follows: selecting 8 volunteers, respectively coating 10% lactic acid solution on one side of each arm to stimulate, so as to cause the skin of the arm to have red and stabbing pain, and then respectively using the samples of examples 1-3 and comparative examples 1-5 to repair the stimulated skin, wherein the corresponding volunteer 1 uses the stock solution sample of example 1, the corresponding volunteer 2 uses the stock solution sample of example 2, the corresponding volunteer 3 uses the stock solution sample of example 3, the corresponding volunteer 4 uses the stock solution sample of comparative example 1, the corresponding volunteer 5 uses the stock solution sample of comparative example 2, the corresponding volunteer 6 uses the stock solution sample of comparative example 3, the corresponding volunteer 7 uses the stock solution sample of comparative example 4, and the corresponding volunteer 8 uses the stock solution sample of comparative example 5.
Recording time: the skin repair of arm skin was recorded for 1h, 3h, 7h, and 12h, respectively.
The experimental results are as follows:
TABLE 1 lactic acid sting repair results
Figure GDA0002545493060000181
As can be seen from table 1, the stock solutions of examples 1 to 3 had a reduced skin-repairing effect with a reduced amount of the anti-blue light repairing composition, so that the stock solutions of example 1 had the best skin-repairing effect. Comparative example 1 does not contain the blue light-resistant repair composition and has no effect on skin repair, comparative example 2 only contains the sophora japonica seed extract and has poor skin repair effect, comparative examples 3-4 respectively only contain the centella asiatica extract and the selaginella tamariscina extract and have reduced skin repair effect, and comparative example 5 contains the sophora japonica seed extract and the selaginella tamariscina extract and has better skin repair effect than comparative examples 3-4 but still weaker than example 3, so that the sophora japonica seed extract, the centella asiatica extract and the selaginella tamariscina extract in the blue light-resistant repair composition synergistically exert good skin repair and blue light-resistant effects.
In conclusion, the blue light resisting and repairing stock solution containing the blue light resisting repairing composition is safe and low in irritation, and can effectively resist the damage of blue light to skin cells, improve the phenomenon of rough skin, repair damaged skin cells and achieve the effects of delaying skin aging and reducing skin pigmentation after being frequently used.
