本发明公开了一种灰黄霉素发酵生产培养基,由以下重量百分比的原料配制而成:籼米粉6%,玉米粉5%,地瓜粉3%,磷酸二氢钾0.6%,碳酸钙0.4%,硝酸钠0.1%,硫酸铵0.1%,硫酸亚铁0.1%,氯化钠0.8%,氯化钾0.7%,硫酸镁0.1%,泡敌GPE型0.05%,剩余量为水。本发明发酵液粘度降低,发酵单位稳定在28000‑30000μg/ml,发酵周期缩约45小时,降低了发酵耗能,降低成本,收率高。
Description
技术领域 本发明涉及原料药制备方法,特别涉及一种灰黄霉素发酵生产培养基。
背景技术 灰黄霉素为抗生素类原料药,英文名Griseofulvin,中文别名灰霉素、微晶灰黄霉素,英文别名Fulcin、Fulcine、Fulvicin、Fungivin、Grifulvin、Grisactin、Grisefulin、Griseofulvinum,【CAS】126-07-8,本品为白色或类白色的微细粉末;本产品主要对毛发癣菌、小孢子菌、表皮癣菌等浅部真菌有良好抗菌作用。
灰黄霉素适用于各种癣病治疗,包括头癣、须癣、体癣、股癣、足癣和甲癣。
目前,制备灰黄霉素的方法,a、灰黄霉素发酵培养一定周期加热灭活、板框固液分离、滤饼菌丝体制粒干燥。b、丙酮萃取得到灰黄霉素丙酮萃取液;c、灰黄霉素丙酮萃取液活性炭脱色,脱色液浓缩结晶得到灰黄霉素粗品;d、灰黄霉素粗品用丙酮溶解,灰黄霉素丙酮溶解液活性炭脱色,脱色液再浓缩结晶得到灰黄霉素重结晶体;e、灰黄霉素重结晶体丁醇洗涤,再用乙醇洗涤、干燥、超微粉碎得灰黄霉素原料药。
发明内容 本发明的目的是提供一种灰黄霉素发酵生产培养基,使生产过程中耗能低、降低成本、发酵周期短,提高发酵单位,提高生产效率。
本发明采用的技术方案如下:
一种灰黄霉素发酵生产培养基,由以下重量百分比的原料配制而成:籼米粉6%,玉米粉5%,地瓜粉3%,磷酸二氢钾0.6%,碳酸钙0.4%,硝酸钠0.1%,硫酸铵0.1%,硫酸亚铁0.1%,氯化钠0.8%,氯化钾0.7%,硫酸镁0.1%,泡敌GPE型0.05%,剩余量为水。
本发明的优点在于:1、调整发酵培养基中碳源成分及配比,经生产过程监测数据显示碳源配比为籼米粉6%,玉米粉5%,地瓜粉3%,可使发酵液粘度降低,发酵单位稳定在28000-30000μg/ml,发酵周期缩约45小时,降低了发酵耗能,降低成本;
2、与对比配方相比,调整磷酸二氢钾为0.6%,碳酸钙0.4%,发酵液pH由6.8降低到5.8,经取样在显微镜下观察,发酵液中菌株生长比对照试验配方中的菌株生长速度显著增快;
3、加入适量的GPE型泡敌可改善发酵罐内气液混合效果,提高发酵罐内氧气的速率,有利于菌株生长提高收率;
4、本试验处方已在工业化大生产应用,且比对照试验配方过滤,收率高约16%。
具体实施方式 下面对本发明作进一步说明。
一种灰黄霉素发酵生产培养基,由以下重量百分比的原料配制而成:籼米粉6%,玉米粉5%,地瓜粉3%,磷酸二氢钾0.6%,碳酸钙0.4%,硝酸钠0.1%,硫酸铵0.1%,硫酸亚铁0.1%,氯化钠0.8%,氯化钾0.7%,硫酸镁0.1%,泡敌GPE型0.05%,剩余量为水。
使用上述培养基生产灰黄霉素方法如下:
a、将上述培养基配置后装入发酵罐121~126℃,维持30分钟消毒,待罐温降至30度,接种、发酵,培养条件:初始培养基pH=5.8,培养温度:29℃±1℃,空气流量:2500~3600m3/h,罐压:0.01~0.05MPa,在发酵中期补料,培养至340小时,加热灭活,用板框压滤机将菌丝体与发酵液分离,菌丝体加氧化钙制粒、干燥,装入萃取柱加丙酮萃取;
b、丙酮萃取得到灰黄霉素丙酮萃取液;
c、灰黄霉素丙酮萃取液活性炭脱色,脱色液浓缩结晶得到灰黄霉素粗品;
d、灰黄霉素粗品用丙酮溶解,灰黄霉素丙酮溶解液活性炭脱色,脱色液再浓缩结晶得到灰黄霉素重结晶体;
e、灰黄霉素重结晶体丁醇洗涤,再用乙醇洗涤、干燥、超微粉碎得灰黄霉素原料药。
对比实验:
试验配方(本发明所用培养基)
籼米粉6%,玉米粉5%,地瓜粉3%,磷酸二氢钾0.6%,碳酸钙0.4%,硝酸钠0.1%,硫酸铵0.1%,硫酸亚铁0.1%,氯化钠0.8%,氯化钾0.7%,硫酸镁0.1%,泡敌GPE型0.05%,剩余量为水。
对比配方(本公司以前使用的培养基)
籼米粉15%,磷酸二氢钾0.4%,碳酸钙0.6%,硝酸钠0.1%,硫酸铵0.1%,硫酸亚铁0.1%,氯化钠0.8%,氯化钾0.7%,硫酸镁0.1%,剩余量为水。
按上述两配方配料、消毒后,将种子培养液压至发酵罐内培养、中期补料后培养、灭活,菌丝体与发酵液分离、制粒、萃取、脱色、结晶、洗涤、干燥、超微粉碎得灰黄霉素原料药。通过三次试验比较结果如表:

试验结论:通过试验配方与对比配方比较,试验配方中的发酵液滤速明显提高,过滤收率比对比配方高约16%,消后pH由6.8降低到5.8,发酵效价稳定在28000-30000μg/ml,发酵周期缩短约45小时,每年节约150万元,为企业提高效率、提高产量,创造了经济效益近1000万利润。
